Abstract

The pyrazole scaffold modification in various chemical structures on several studies has shown various biopharmacological activities. This study aims to predict the potential inhibition of pyrazole analogs derived benzene-sulfonylurea (4A, 4B, 4E, 5A, 5C, 5D) against the α-glucosidase (3A4A) and aldose reductase (3RX2) enzymes based on a molecular docking approach using Molecular Operating Environment (MOE) 2020.0102 software and evaluate pharmacokinetic profile (ADMET). In this study, the six test compounds were obtained from previous studies that have been proven antihyperglycemic. The results showed that all the 3,5-disubstituted benzene-sulfonylurea derivative pyrazole analogs are predicted to have low inhibitory activity against the α-glucosidase enzyme. Further, all compounds showed good aldose reductase inhibitor activity and had lower binding free energy values than tolrestat as the positive control (-6.82 kcal/mol). Compound 5C has the best potential inhibitory activity against the aldose reductase enzyme compared to the other test compounds, because it has the lowest binding free energy value (-8.76 kcal/mol) and interacts with important residues on the receptor forming four hydrogen bonds, namely the carbonyl group of SO2 with residues Trp111 and His110, and the carbonyl group of the amide with residues His110 and Tyr48, as well as 3 hydrophobic bonds, namely a pyrazole ring with residues Leu300, Trp219 and a furan ring with Phe122. ADMET properties of the compounds are also predicted. This information provides an opportunity for a 5C compound as an aldose reductase inhibitor agent to develop drug candidates with better and safer activities.

Abstract

Turbofan engines are crucial components in the aviation and manufacturing industries, where estimating the Remaining Useful Life (RUL) has a significant impact on operational efficiency and safety. This study aims to predict the RUL of turbofan engines using the Support Vector Regression (SVR) method, a machine learning approach that has proven effective in modeling nonlinear relationships between variables. Operational data related to turbofan engines include operational parameters, sensors, and maintenance records. The initial stage of this research involves data analysis based on unit number, time, operational control, and sensor parameters. This process begins with preprocessing to initialize the initial data values, normalize, and select sensors that have stagnant values, as these sensors do not affect the machine learning system. Subsequently, regression calculations are performed to compare predicted values and actual values using the Support Vector Regression method optimized with Grid Search Optimization. In this study, testing was conducted with Parameters C [1, 10, 50, 100] and ε [1, 5, 10, 50], resulting in the best model with an RMSE error of 19.56 and MAE of 14.73.

Abstract

This study developed interactive learning media using Macromedia Flash to enhance Occupational Health, Safety, and Environmental Awareness (K3LH) instruction within the Basic Electronics curriculum in vocational high schools. The research employed a Research and Development approach guided by the 4D model (Define, Design, Develop, and Disseminate) to produce a validated multimedia learning product aligned with the Merdeka Curriculum. During the Define stage, needs assessment and learner analysis revealed low student engagement and conceptual difficulties with electronics safety topics, underscoring the need for technology-based instructional innovation. The designed media integrated text, visuals, animations, audio, and interactive assessments to support autonomous and student-centred learning. Expert validation involving material and media specialists resulted in high validity scores of 94.7% and 94.8%, indicating strong curriculum alignment, conceptual clarity, interface organisation, and language quality. Field testing with Grade 10 Electronics Engineering students yielded a practicality score of 85.1%, confirming that the media was easy to operate, pedagogically relevant, visually engaging, and effective in supporting understanding and participation. These outcomes demonstrate that Macromedia Flash-based learning media remain highly functional for vocational classroom environments, particularly where technological infrastructure is limited. The study concludes that the developed media can serve as a robust instructional tool to strengthen student motivation, improve comprehension of K3LH concepts, and enrich technology-enhanced learning in vocational electronics education. Future research is recommended to expand implementation across different school contexts and migrate the product to modern digital platforms to improve scalability and accessibility.

Abstract

ABSTRACT. Pyrazoline is a well-known group of five-ring heterocyclic compound containing two nitrogen atoms that has a wide range of promising biological activities. The aim of this research is to synthesize 1,3,5-triaryl-2-pyrazoline derivatives with chloro substituent and pyrrolidine moiety and to explore their potency as inhibitors for cervical cancer. The synthesis was performed through three-steps reactions. Firstly, synthesis of chlorinated naphthalenyl chalcones through Claisen-Schmidt condensation. Secondly, synthesis of chlorinated naphthalenyl pyrazolines through nucleophilic addition of phenylhydrazine to the chalcone and followed by intramolecular cyclization. Thirdly, synthesis of pyrazoline derivatives by adding the pyrrolidine moiety through O-alkylation reaction. Then, the in silico studies through molecular docking, ADME profiling, and toxicity prediction were performed to explore the potency of the synthesized compounds as inhibitors of cervical cancer by inhibiting the activity of Sirtuin-1 (SIRT-1) (PDB ID: 4I5I). The structure of all pyrazoline derivatives (4a-4c) were confirmed by spectroscopic analysis including UV, FT-IR, HRMS, and NMR. Based on the docking study, compound 4a with binding free energy of -12.59 kcal/mol exhibited better potency as SIRT-1 inhibitor than previously reported inhibitor (EX527). Then, based on the ADME profiling and toxicity prediction, it can be concluded that compound 4a exhibited reasonable drug properties with low toxicity potency (predicted LD50 = 1000 mg/kg). Abstract

Lithocarpus bancanus is a medicinal plant used by communities in Riau Province, Indonesia. This plant is a tree locally known in the province as mempening. In this study, the cytotoxic activity profiles of the methanolic extract, n-hexane fraction, dichloromethane fraction, and ethyl acetate fraction from L. bancanus leaves were evaluated against human lung cancer cells (A549 cell line) using the MTT assay. IC₅₀ values were calculated from dose–response curves analyzed using the 4-parameter logistic (4PL) model. The results showed IC₅₀ values of 455.1, 361.9, 132.9, and 304.7 µg/mL for the methanolic extract, n-hexane fraction, dichloromethane fraction, and ethyl acetate fraction, respectively, with the dichloromethane fraction exhibiting relatively higher cytotoxic activity (IC₅₀ = 132.9 µg/mL). Cisplatin was used as the positive control and showed an IC₅₀ of 103.9 µg/mL. Meanwhile, the methanolic extract as well as the n-hexane and ethyl acetate fractions showed low cytotoxic activity, with IC₅₀ values greater than 300 µg/mL. Therefore, the dichloromethane fraction can serve as a basis for further isolation to identify bioactive anticancer compounds.

Abstract

Pyridazinones are a class of heterocyclic compounds with broad biological activities, one of which is as an anticancer. This study synthesized N-acetohydrazide substituted pyridazinone derivatives and evaluated their potential as breast cancer therapy through molecular docking studies. The target compound, 2-(3-(3-methoxyphenyl)-6-oxopyridazine-1(6H)-yl)acetohydrazide(3), was synthesized through three reaction steps: condensation to form the pyridqzinone core, functionalization of ethyl chloroacetate at the nitrogen position, and substitution of the ethoxy group with hydrazine hydrate. The yield obtained was 48.14%. The purity of the synthesized compound was confirmed through melting point determination and high-performance liquid chromatohraphy (HPLC) analysis, which showed a single dominant peak. Structural elucidation using Fourier-transform infrared (FTIR), mass spectrometry (MS), proton nuclear magnetic resonance (1H-NMR), and carbon-13 nuclear magnetic resonance (13C-NMR) verified the expected structure.  Molecular tethering studies against tyrosine kinase (PDB ID: 3ERT) showed that compound (3) has a binding free energy of -7.93 kcal/mol, with two hydrogen bonds formed with residues Glu353 and Leu387. These results indicate that compound (3) has not shown better inhibitory activity than tamoxifen. Nonetheless, this compound fulfils good physicochemical characteristics based on Lipinski's rule, so it remains promising for further development.
