Abstract

Penelitian ini dilakukan untuk membuktikan pengaruh ekstrak sambiloto terhadap apoptosis dan volume tumor bila diberikan secara oral dan diidentifikasi dengan metode TUNEL. Penelitian dilakukan dengan desain Randomized post test control group. Sebanyak 24 ekor mencit C3H yang telah tumbuh kanker mamma dibagi secara acak menjadi 4 kelompok masing-masing 6 ekor. Diberikan ekstrak sambiloto dengan dosis 5, 10 dan 15 mg per ekor per hari, dan kelompok kontrol. Pemberian ekstrak sambiloto dilakukan secara oral selama 14 hari. Pengukuran volume tumor dilakukan pada hari ke 1, 4, 8, 12 dan 15. Pada hari ke 15 mencit dimatikan dengan menggunakan eter. Jaringan kanker dibuat blok parafin untuk pemeriksaan apoptosis dengan metode TUNEL. Hasil Penelitian menunjukkan adanya peningkatan signifikan sel apoptosis (p=0,000), dengan dosis terbaik 15 mg/hari. Ada beda yang bermakna pada rerata kelipatan penambahan volume tumor (p=0,049). Hasil uji post hoc menunjukkan bahwa perbedaan terletak antara kelompok kontrol dengan kelompok dosis 10 mg/hr. Dapat disimpulkan bahwa pemberian ekstrak sambiloto secara oral meningkatkan sel yang mengalami apoptosis dengan pemeriksaan TUNEL namun belum dapat menurunkan volume tumor.

Abstract

Apoptosis adalah kematian sel terprogram (programed cell death) yang bertujuan untuk mempertahankan kestabilan populai sel. Kegagalan pengaturan apoptosis dapat menyebabkan sel membelah tanpa terkendali, yang disebut sebagai kanker. Peningkatan apoptosis merupakan suatu upaya yang dikembangkan sebagai terapi kanker. Pemeriksaan apoptosis dengan metode TUNEL(Terminal deoxyribonucleotidyl transferase dUTP nick end labeling) dapat memberikan gambaran proses apoptosis pada tingkat sel tunggal sehingga lebih spesifik dan memiliki akurasi tinggi. Sambiloto merupakan salah satu tanaman obat tradisional yang digunakan sebagai anti kanker. Beberapa penelitian yang dilakukan secara in vitro menunjukkan pengaruh andrografolid yang diisolasi dari sambiloto terhadap apoptosis sel kanker pada kultur. Hasil tersebut masih memerlukan penelitian lebih lanjut, terutama pengaruhnya bila diberikan secara oral. Abstract

Glutation merupakan antioksidan yang berperan dalam fungsi imun, dan diekspresikan secara genetik oleh urutan gen yang membentuk protein enzim Glutation Peroxidase (GPx1). Bila ekspresi gen berubah maka terjadi perubahan fungsi glutation dan kerentanan terhadap stress oksidatif. Metode yang digunakan adalah Kasus-kontrol. Sampel yang digunakan adalah sampel darah. Kelompok kasus adalah sampel darah pasien tuberkulosis paru sedangkan kelompok kontrol adalah sampel darah orang sehat. Pemeriksaan gen Glutation peroxidase (GPx1) menggunakan metode Polymerase Chain Reaction (PCR) untuk melihat pita DNA pada pasien tuberkulosis par serta elektroforesis produk PCR-RFLP gen GPx1 kelompok sampel tuberkulosis. Hasil penelitian menunjukkan bahwa tidak terdapat hubungan yang bermakna antara polimorfisme gen GPx1 (p=0,365) pasein tuberkulois dengan individu sehat, sehingga tidak dapat digunakan sebagai alat deteksi kerentanan terhadap stress oksidatif pada pasien tuberkulosis. Perlu penelitian lanjutan yang menggunakan sampel lebih besar dan populasi etnik yang berbeda.

Abstract

 Stunting is a form of malnutrition is characterized by short stature in children. The prevalence of toddler stunting in 2017 in South Sulawesi is 34%, while in  Sinjai Regency in 2016 is 34.6%, 2017 is 43.7%, and 2018 is 35.8%. The purpose of this study was to analyze the disparity of stunting risk factors in toddlers on the coast and mountain areas. This study used a case-control design. The sampling techniques are Fixed Disease Sampling and Stratified Proportional Random Sampling. The population consists of the entire toddler in Sinjai, whereas consists of 60 toddler stunting age 9-59 months and 60 normal toddler on the coast and mountain areas. The instrument used the Food Frequency Questionnaire (FFQ) sheet. The data were analyzed by univariate bivariate, Chi Square and multivariate with logistic regression. The Chi square test results showed that the risk factors stunting on the coast is energy intake (p = 0.03; OR = 2.99) and Fe (p = 0.03; OR = 2.99), while in the mountain is the protein intake (p = 0.01; OR = 6.5), Fe (p = 0.01; OR = 4) and Zn (p = 0.00; OR = 5.4). The logistic regression results indicate that stunting dominant risk factors in the coast area is the intake of proteins and in the mountain area is the intake of Fe. There is a disparity between the risk factors of stunting toddler in the coast  and mountain area. Poor nutritional intake increases the risk of stunting so it is necesary to increase the consumption of food sources of nutrients for toddlers.Stunting merupakan bentuk dari kurang gizi. Tahun 2017, stunting di Provinsi Sulawesi Selatan masih tinggi (34%) dan Kabupaten Sinjai tahun 2016 (34,6%), 2017 (43,7%), dan 2018 (35,8%). Tujuan penelitian untuk menganalisis disparitas faktor risiko stunting balita di wilayah pesisir pantai dan pegunungan. Penelitian ini merupakan penelitian kuantitatif menggunakan desain case control. Teknik sampling yaitu Fixed Desease Sampling dan Stratified  Proportional Random Sampling. Populasi adalah seluruh balita di Sinjai dan sampel yaitu balita stunting usia 9-59 bulan dan minimal sampel sebanyak 60 balita stunting di wilayah  pesisir pantai dan pegunungan serta 60 balita normal di wilayah pesisir pantai dan pegunungan. Instrumen menggunakan lembar FFQ Semikuantitatif. Data dianalisis univariat, bivariat dengan uji Chi Square dan  multivariat dengan regresi logistik. Hasil uji Chi Square menunjukkan faktor risiko stunting di wilayah pesisir pantai yaitu asupan energi dengan nilai p=0.03 OR=2.99 dan Fe dengan nilai p=0.03 OR=2.99 sedangkan di pegunungan yaitu asupan protein dengan nilai p=0.01 OR 6.5, Fe dengan nilai p=0.01 OR=4, dan Zn dengan nilai p=0.00 OR=5.4. Hasil regresi logistik menunjukkan faktor risiko stunting yang dominan di wilayah pesisir pantai yaitu asupan protein dan di wilayah pegunungan yaitu asupan Fe. Terdapat disparitas faktor risiko stunting pada balita antara wilayah pesisir pantai dan pegunungan.

Abstract

Dyslipidemia is a disorder of lipid metabolism that causes narrowing of blood vessels. Risk factors for coronary heart disease are characterized by an increase in cholesterol. Thus, prevention needs to be done by changing a balanced diet that is eating one fruit and vegetable, one with beet root that has a high antioxidant content. The purpose of this study was to examine the activity of beet root extract on rat cholesterol levels. This research method is experimental with post test design only randomized control group design. Treatment at a dose of 100 mg / kg, 200 mg / kg combined with 0.18 mg / kg simvastatin. The sample in this study was Wistar strain male rats aged 2-3 months with a weight of 100-200 grams, the technique of random sampling using the formula Federer as many as 5 animals / group and divided into 7 treatment groups. Beet root extract and atherogenic feed were given for 35 days. Cholesterol levels were examined by the CHOD-PAP (Cholesterol Oxidase-Peroxidase Aminoantipyrine Phenol) method using a clinicon 4010 photometer. Data were analyzed using the Kruskall Wallis and Mann Whitney tests. The effect of beet root extract activity on reducing cholesterol levels at a single dose of 100 mg/kg BW of 17.40% (p-value = 0.673), single dose 200 mg / kg BW of 21.59% (p-value = 0.009), combination dose of 100 mg / kgbb of 7.10% (p value = 0.009), a combined dose of 200 mg / kgbb of 18.65% (p value = 0.347). Beet root extract is effective in reducing rat cholesterol at a dose of 200 mg/kg BW.

Abstract

ENGLISHThe benefits of exclusive breastfeeding are in accordance with one of the goals of the Millennium Development Goals (MDGs), namely reducing child mortality and improving maternal health. The research objective is to analyze the implementation of input aspects of promotional efforts for exclusive breastfeeding including: 1) human resources (HR), 2) funding resources, 3) infrastructure, and 4) availability of Standard Operating Procedures (SOPs). This research uses qualitative approach which is focused on efforts to promote exclusive breastfeeding. The informants of the study were determined by purposive sampling and snowball sampling techniques. Data collection technique uses in-depth interview. The results of the study are: 1) midwives become official workers to promote exclusive breastfeeding, 2) the funding for exclusive breastfeeding promotion comes from the government, 3) the infrastructures to support exclusive breastfeeding are available sufficiently, and 4) standard operating procedures to support exclusive breastfeeding are not available yet. Finally, The Government is recommended to strengthen exclusive breastfeeding promotions by allocating special promotion workers in sufficient number, providing SOPs to promote exclusive breastfeeding. INDONESIAManfaat pemberian air susu ibu (ASI) eksklusif sesuai dengan salah satu tujuan dari Millenium Development Goals (MDGs) yaitu mengurangi tingkat kematian anak dan meningkatkan kesehatan ibu. Tujuan penelitian untuk menganalisis implementasi aspek input upaya promosi pemberian ASI eksklusif yang meliputi: Sumber Daya Manusia (SDM), sumber dana, sarana prasarana, dan ketersediaan SOP. Penelitian ini menggunakan pendekatan kualitatif yang difokuskan pada upaya-upaya promosi pemberian ASI eksklusif. Informan penelitian ditentukan dengan teknik purposive sampling dan snowball sampling. Teknik pengambilan data dengan wawancara mendalam. Hasil penelitian menunjukkan: 1) sumber daya manusia yang melakukan promosi ASI eksklusif adalah bidan, 2) sumber dana promosi ASI eksklusif dari pemerintah berupa dana Bantuan Operasional Kesehatan (BOK), 3) ketersediaan sarana prasarana promosi ASI eksklusif sudah memadai, dan 4) belum tersedianya SOP promosi pemberian ASI eksklusif. Oleh karena itu Dinas Kesehatan perlu membuat SOP yang jelas untuk promosi pemberian ASI eksklusif bagi petugas promosi kesehatan. Puskesmas perlu melakukan upaya perekrutan tenaga khusus untuk promosi kesehatan.

Abstract

Biology subjects in Pharmacy Vocational High Schools are not yet integrated with pharmacy subjects even though biology subjects have a close relationship with pharmacy fields related to organs and medicines in the human digestive system. This research aims to analyze the science process skills and learning outcomes in the inquiry learning model with a mini research strategy as a supporting of pharmacy science in Vocational High Schools. This research uses quasy experiment research with pretest posttest control group design. Analysis of learning outcomes using the independent sample t-test, N-Gain test, and N-Gain t-test to see differences in the value of learning outcomes between the control class and the experimental class. Analysis of science process skills uses the average score of observations of science process skills. To see whether the research can supporting Vocational Pharmacy Science is analyzed using descriptive analysis of interviews with vocational productive teachers. The result of the value of learning outcomes get data that the value of learning outcomes of the experimental class is better than the control class, while the science process skills of students in the experimental class are better than the control class. Descriptive analysis of interview productive teacher shows that this study can be used as a supported of pharmacy science in Vocational High Schools. Analysis of the value of student learning outcomes, science process skills and Interview productive teacher shows that inquiry learning models with mini research strategies can support subjects related to pharmacy.

Abstract

Abstract Wintergreen (Gaultheria fragantissima) is the source of the essential oil of wintergreen which is comprised predominantly of methyl salicylic. During this time wintergreen is not yet economically developed because of the lack of the cultivation technology. One of effort that is needed to be studied beside the cultivation tecnology and  the wintergren oil quality development is the product diversification. wintergreen comprised by far the highest concentration of both free salicylic and total salicylic acid which is expected of a plant known for the formation of the wintergreen oil, an essential oil consisting primarily of methyl salicylic. The concentration of total salicylate found in wintergreen is over 20 fold greater than the total salicylic concentration (salicylic and any derivates combined) found in Filipendula, and 100 fold greater than that found in Lemon Thyme. The active form of salicylic acid in wintergreen is Gaultherin. Gaultherin has many properties related to human health. Gaultherin consists of methyl salicylic conjugated to the disaccharide, primeverose. When plant tissues are disrupted the endogenous gaultherin is rapidly lost, presumably by enzymatic hydrolysis with the release of methyl salicylic. The problem is until this time there is not yet available an effective method for the gaultherin extraction. The difficulties faced on the gaultherin production are the fact that along the gaultherin extraction. and along with the disruption of the wintergreen tissue, the gaultherin is hydrolyzed into its individual components which are methyl salicylic and disaccharide. The alternative process for the gaultherin production from wintergreen is the gaultherin production process by enzyme inactivation and  extraction process in an inactivation extractor using a n organic solvent (alcoholic solvent extraction). This paper is intended to study the enzyme inactivation rate which is play an important role in the dimension determination of the process equipments. Enzyme inactivation can be describes as first orde reaction. The enzyme inactivation constant  kp is the temperature, pressure, and water concentration function. The dependency, model of  the temperature based on Arhenius type while the pressure dependency  is based on Eyring equation.   Keywords : gaultherin, inactivation. kinetics

Abstract

Hypertension is the degenerative disease that if not treated immediately, it causes other diseases such as diabetes mellitus, stroke, and so on indeed death as if it does not take any medical treatments.  The prevalence of hypertension on elderly in Central Java in 2018 was 38,4 %. 55-64 years old group was 58,4%, 65-74 years old was 61,6% and >74 years old was 65.6%. The aim of this research is to analyze saturated fat intake, sodium, protein and body mass index related to hypertension on elderly in Semarang Regency. The research used case control design. The sample of the research was 104 respondents obtained by purposive sampling technique. Technique for collecting the data were interview, observation, and questionnaire. Chi square and logistics regression test were used in analyzing the data. The result of the research showed that there were significant influences in the variables of Saturated fat intake (p= 0.019), sodium (p=0.006), protein (0.000), body mass index (0.000) concerning to hypertension on elderly in Semarang Regency. The most influential variable was sodium intake with p=0.006; Exp(B) /OR = 0.295. Thus,  the increase of information about the importance of hypertension prevention is needed.

Abstract

Diabetes Mellitus Type 2 is a pathophysiological disorder of insulin resistance. Coverage of type 2 diabetes mellitus in Yogyakarta Special Region 2017 (8,321 cases). In Kulon Progo Regency 2016 (1,355 cases), 2017 (1,235 cases), 2018 (1,532 cases). The purpose of the study was to analyze food consumption patterns with the incidence of type 2 diabetes mellitus in the Kulon Progo Regency, a special area of Yogyakarta. The study used a case-control study design. The research sample of 114 respondents was obtained by purposive sampling technique. Data collection techniques are interviews, observation, questionnaire. Data analysis in the study used the Chi-Square test and test logistic regression. The results of the study found that there was an influence on the variable of dietary consumption (p = 0,000) on the incidence of type 2 diabetes mellitus in the Kulon Progo district of Yogyakarta. The most influential variable is the pattern of eating consumption with p = 0,000; EXP (B) / OR = 0.004.
