Abstract

The purpose of this study is to determine the effect of the red dragon fruit peels (Hylocereus polyrhizus) extract on the growth of Salmonella pullorum. This research was conducted at the Laboratory of Microbiology, Veterinary Faculty, University of Syiah Kuala, Banda Aceh. Red dragon fruit peels that used in this study is obtained from  ripe fruit and Salmonella pullorum cultures is  obtained from  Microbiology Laboratory of Veterinary Faculty Unsyiah. Salmonella pullorum culture added evenly on the surface of Mueller Hinton Agar, the number of the bacteria have been standarized using Mc Farland'1 method. Discs soaked in the red dragon fruit peels extracts at different concentrations (20 mg/ml, 40 mg/mL, and 60 mg/ml). Ampicillin is used as positive controls and DMSO (dimethyl sulfoxide) was placed on the surface of Mueller Hinton Agar. The parameters in this study is the inhibition zone diameter. Data were analyzed descriptively. The results showed that the red dragon fruit peel extract is able to inhibit the growth of Salmonella  pullorum and the average of  inhibition diameter zone at a concentration 60 mg / ml is 9.6 mm, 40 mg / ml is 9.4 mm and 20 mg / ml is 9.3 mm

Abstract

Betta fish (Betta splendens) are one of the most popular ornamental fish due to their gorgeous colors and various shapes. This study aimed to find pathogenic bacteria in betta fish (Betta splendens) in fish farm in Bogor. Body swabs were collected from four decorative betta fish, and from tank water. Two of the fish were dead and two of which were exhibiting symptoms of disease. Bacterial isolation was performed on multiple agar media, and bacteria were identified based on the growth characteristics of the colonies, Gram staining, and biochemical testing. The results of the analysis revealed the presence of 16 bacterial isolates, including Edwardsiella sp., Escherichia coli, and Vibrio sp. from dead fish, whereas Citrobacter sp, Providencia sp, and Vibrio sp were detected in diseased fish. In aquarium water, Escherichia coli, Citrobacter sp Proteus sp and Providencia sp. were detected. Betta fish are susceptible to suffer from diseases if the water quality is low since maintaining and enhancing the health of betta fish is highly dependent on water quality.

Abstract

The aim of this study was to count the total sum of cellulolitic bacteria colony in  aceh cattle rumen. The sample used was aceh cattle rumen liquid which taken from salughtered house in Banda Aceh. Aceh cattle rumen liquid were diluted from 10-2 to 10-5, then inoculated to agar BHM-CMC media with pour plate method. Petri dish was entered to candle jar to be incubated in an incubator with temperature 37oC about 96 hours. Colony of cellulolitic bacteria which were grown then were observed by using colony bacteria criteria (colour, position, edge, and diameter of colony), counting total of bacteria which was grown, and Gram staining. The results showed that there is a white translucent colony morphology, growth inside agar, smooth periphery, Gram-positive bacil. The average number bacteria of total bacteria in the cattle rumen liquid was 4,81 x 105 cfu/ml

Abstract

Penelitian ini bertujuan mengetahui konsentrasi AFB1 pada jenis makanan olahan asal jagung yang dipasarkan di Banda Aceh. Pengambilan sampel dilakukan di pasar dan swalayan di Banda Aceh. Deteksi AFB1 pada sampel menggunakan metode enzyme-linked immunosorbent assay (ELISA). Berdasarkan penelitian, dapat disimpulkan bahwa dari 6 sampel makanan berbahan baku jagung, 5 sampel (83,33% ) terkontaminasi AFB1. Walaupun terkontaminasi AFB1 namun semua sampel masih aman dikonsumsi sesuai dengan persyaratan yang ditetapkan pemerintah yaitu maksimal 20 ppb.

Abstract

This research aim was to determine the level of bacterial contamination of Salmonella sp on quail egg. The samples used in this research was 24 shell of quail eggs obtained from eight stores in traditional markets in Ulee Kareung Banda Aceh. The shell was crushed with a mortar and then cultured on SCB media (selenite cysteine broth). Isolation and identification was conducted by using SSA media culture, Gram staining, and biochemical tests. The results of this research showed that the shell of quail eggs contained Salmonella sp. Therefore it can be concluded that the quail eggs sold in Ulee Kareung traditional market were contaminated by Salmonella sp.

Abstract

Shrimp is an easily polluted material, so it requires good handling. Contamination can occur during harvest, handling, or transportation. This study aims to detect Escherichia coli contamination in vaname shrimp sold at TPI Lampulo Banda Aceh. A total of 10 samples of vaname shrimp were obtained from 10 merchants at TPI Lampulo. The research method used is observational, and the data analysis is done descriptively. The detection of E. coli contamination was carried out by the total plate count (TPC) test method. Based on the study results, two of the 10 samples of vaname shrimp merchant detected E. coli bacterial contamination, namely the results of sample 01 merchant indicated the number of TPC 2x102 CFU/g and in 09 merchants indicated the number of TPC 11x101 CFU/g. The TPC results were indicated by the growth of E. coli colonies on eosin methylene blue agar (EMBA) media after incubation for 24 hours at 37 ºC. The number of the two detected positive samples showed less than 1x103 CFU/g by the provisions of SNI 2332.3:2015. Thus, it can be concluded that E. coli contamination was detected in vaname shrimp sold by the merchant at TPI Lampulo but did not exceed the SNI threshold, so it was safe for consumption.

Abstract

This study aimed to determine the total of A. niger found in the gastrointestinal tract of broilers after the addition of soybean dregs and fermented palm kernel meal by A.niger as probiotics (AKBIS Prob) in the fodder. This study used a complete randomized factorial design, which consists of two factors, namely factor AKBIS concentration and duration of AKBIS Prob. The treatment was divided into the group as a control P0, P1, P2 and P3 which consisted of five broilers, respectively given feed supplemented with AKBIS Prob 2, 4 and 6%. The provision of AKBIS Prob has done for 21 days and 35 days. On the 22nd and 36th day the contents of the caecum were taken. The total of A. niger was obtained using Total Plate Count (TPC) method. The data analysis used a factorial analysis of variance. The results showed that the concentration of 2, 4 and 6% did not affect (P 0.05) the total of A. niger in the caecum of broilers, while the provision of AKBIS prob in fodder for 21 and 35 days had a significant effect (P 0.05) to the total A. niger. It can be concluded that the provision of the concentration of AKBIS Prob up to 6% in the fodder decreases the total of A. Niger. The provision of AKBIS Prob for 35 days can increase the total of A. niger in the digestive tract of broilers.

Abstract

Leptospirosis is a zoonotic disease caused by bacteria Leptospira sp. which causes infection in animals and humans. Dogs infected with leptospirosis showed symptoms such as anorexia, fever, vomiting, weakness, diarrhea and often experience yellowing of the eye area and mucosa around the mouth (icteric) with fatal systemic complications and multi-organ dysfunction, especially in the kidneys and liver. Leptospirosis is an endemic disease in Jakarta. This study aims to identify risk factors that can contribute to canine mortality based on early clinical symptoms that are found when the dog is in an animal health service facility such as a veterinary clinic, veterinary hospital, or independent practice veterinarian. Method was used in this study is clinical manifestations and laboratory examinations and medical records of dogs with suspected leptospirosis. Criteria inclusions were based on aspects of the clinical symptoms of dogs in and around Jakarta. Analysis data used the chi-square with confidence of interval (CI) 95%. Dogs used during the study had ages for puppies (less than 1 year) totaling 13 or 32.50%, for adult dogs over 1 year amounted to 27 or 67.50%, 80% male dogs and 20% female. with 80% maintenance system not housed by the owner. And the conclusion is risk factors for clinical symptoms such as myalgia, symptomatic vomiting of the pulmonary area or shortness of breath and abdominal pain, conjunctival suffusion, anorexia and diarrhea contributed to the high mortality rate leptospirosis during study in dogs 2020.

Abstract

Avian Pathogenic Escherichia coli (APEC) is a cause of extraintestinal infections, particularly respiratory infections, pericarditis, and septicemia in poultry (colibacillosis). Research aimed at uncovering the virulence factors responsible for colibacillosis in poultry, especially laying hens in Indonesia, is still quite rare. The purpose of this study is to identify the virulence factors responsible for colibacillosis in E. coli strains resistant to ciprofloxacin. The virulence factors targeted in this research are the genes iss, iutA, iroN, ompT, and hlyF. The study utilized 327 E. coli isolates from the archives of the National Veterinary Drug Assay Laboratory, which were collected in 2021 in seven provinces.All isolates were tested for pathogenicity using Congo Red. Subsequently, E. coli strains determined to be pathogenic were tested for their sensitivity to ciprofloxacin using the agar dilution method. Isolates that were both resistant and pathogenic were then examined for the presence of the virulence genes using polymerase chain reaction (PCR). Based on the results of the Congo Red test, it was found that 59 isolates (18%) were pathogenic E. coli. Among the 54 pathogenic E. coli isolates subjected to sensitivity testing, 30 isolates were found to be resistant to ciprofloxacin. Subsequently, PCR analysis of these 30 ciprofloxacin-resistant pathogenic E. coli isolates revealed the presence of the iss gene in 29 isolates (96%), iutA in 23 isolates (76.6%), ompT in 19 isolates (63.3%), hlyF in 14 isolates (46.6%), and iroN in 11 isolates (36.6%). Eight isolates showed the presence of all five virulence genes, while the rest had one to three virulence genes. This study demonstrates that virulence genes are commonly found in ciprofloxacin-resistant pathogenic E. coli. This suggests a potential threat to the effectiveness of ciprofloxacin in managing colibacillosis in laying hens.

Abstract

Resistansi antibiotik merupakan masalah kesehatan penting di seluruh dunia yang berdampak pada kesehatan manusia dan hewan. Kehadiran gen resistan pada Isolat Staphylococcus aureus dapat berkontribusi pada pengembangan resistansi antibiotik. Penelitian ini bertujuan untuk mengidentifikasi keberadaan gen blaZ yang berasal dari isolat S. aureus yang resistan terhadap antibiotik penisilin G dari wilayah peternakan sapi perah di Provinsi Sulawesi Selatan. Sampel penelitian diperoleh dari 20 lokasi peternakan sapi perah yang tersebar di Kabupaten Enrekang. Identifikasi S. aureus dilakukan dengan metode fenotipik dan genotipik. Isolat S. aureus yang resistan terhadap antibiotik penisilin G berasal dari manusia (n=6), ternak sapi perah (n=10), dan produk dangke (n=2) diperoleh dari hasil uji sensitivitas terhadap antibiotik penisilin G (10 U) dengan metode difusi cakram Kirby-Bauer. Deteksi gen blaZ menggunakan metode polymerase chain reaction (PCR) konvensional. Hasil penelitian menunjukkan bahwa isolat yang berasal dari manusia, ternak sapi perah, dan produk dangke terdeteksi keberadaan gen blaZ masing-masing sebesar 100%. Gen blaZ yang terdapat pada isolat manusia, hewan, dan produk dangke berpotensi menyebar antar isolat dan berpotensi menimbulkan gangguan kesehatan manusia.
