Abstract

The objective of this study was to characterize the morphology of ovary from slaughtered Aceh cow and measuring the concentration of estradiol in the follicular fluid to examine the potential utilizing for in-vitro fertilization (IVF) development in Aceh cattle. In total, 30 pairs of ovaries were collected from 30 Aceh cattle slaughtered at Banda Aceh abattoir. Morphological characteristics of ovaries observed were the size of ovaries, weight of ovaries, number of follicles, the diameter of follicles, corpus luteal (CL), and volume of follicular fluid. The follicular fluid was aspirated using a disposable syringe and analyzed the estradiol concentrations using enzyme-linked immunosorbent assay (ELISA). The follicles were grouped into 3 categories based on the diameter (Ø) of follicles: small follicles (Ø 1 - 5 mm), medium follicles (Ø 5 - 8 mm), and large follicles (Ø 8 mm). The morphological characteristic of ovaries varied between right and left ovaries in which right ovaries showed bigger size and weight compared to the left ovaries (P0.05), whereas the number of follicles and follicular fluid did not significantly different (P0.05). The concentrations of estradiol in follicular fluid of large and medium follicles were significantly higher compared to small follicles (P0.05), whereas the concentration of estradiol between large and medium follicles, did not significant different (P0.05). In conclusion, the follicles from abattoir can be used as potential oocytes resources to support IVF development for Aceh cattle in the future. The follicular fluid provided from these follicles can be an alternative for in vitro maturation media of oocytes due to containing high estradiol concentrations.

Abstract

The semen quality of Gembrong goats is lower than other goats and may be related to the low concentration of testosterone hormone. Implementation of reproductive technology using prostaglandin F2α (PGF2α) hormone is beneficial to increase the testosterone hormone in Gembrong goats. This study aimed to determine the effect of PGF2α injection on increasing testosterone levels in Gembrong goats. Male Gembrong goats (n=4), aged 2.5-4 years with similar body condition scores (BCS=3) were used in this study. Goats were divided into two treatment groups (n=2). Goats in group 1 (G1) were injected intramuscularly with 1 ml PGF2α (75 μg), while those in group 2 (G2) were injected with 1 ml physiological NaCl. Semen collection was carried out 30 minutes after treatment using an artificial vagina. Testosterone levels were measured using the enzyme-linked immunosorbent assay (ELISA). The collected data was tabulated and analyzed descriptively. The results showed that the average testosterone concentration of G1 was higher than G2 with respective concentrations of 6.41 ± 0.70 and 2.81 ± 1.75 ng/ml. It was concluded that administration of PGF2α in vivo could increase testosterone concentration in Gembrong goats.

Abstract

Cedera kepala merupakan masalah pada struktur kepala akibat mengalami benturan yang berpotensi menimbulkan gangguan pada fungsi otak. Cedera kepala cenderung mengalami masalah yang komplit karena akan terjadi masalah pada otak dan saraf. Masalah ini dapat berupa luka ringan, memar di kulit kepala, bengkak, perdarahan, patah tulang tengkorak, atau gegar otak. Penyebab kematian atau kecacatan yang dapat terjadi apabila cedera kepala tidak mendapatkan pertolongan yang benar pada saat kegawatdaruratan. Melihat besarnya dampak yang dapat diakibatkan dari cedera kepala perlu adanya pencegahan dan sistem pendukung yang intensif, sehingga diharapkan masalah yang ada dapat teratasi dan komplikasi yang mungkin terjadi dapat dihindari secara dini, serta tindakan pencegahan yang komprehensif. Pencegahan yang dilakukan untuk mengurangi resiko cedera kepala di dalam masyarakat adalah dengan menggunakan alat pengaman saat berolahraga, selalu menggunakan alat keselamatan seperti helm atau pelindung kepala apabila bekerja di lingkungan yang berisiko menimbulkan cedera kepala. Memasang pegangan besi di kamar mandi dan di samping tangga untuk mengurangi risiko terpeleset Memastikan lantai selalu kering dan tidak licin. Memasang penerangan yang baik di seluruh bagian rumah. Memeriksa kondisi mata secara rutin, terutama jika mengalami gejala gangguan penglihatan, seperti buram atau penglihatan berbayang. Pada kegiatan pengabdian masyarakat ini, telah dilakukan penjelasan tentang pencegahan resiko cedera kepala pada 20 orang dan peserta mampu menjelaskan tentang pencegahan resiko cedera kepala.

Abstract

The aim of this study was to analyze the effect of spawnprim hormone induction with different doses on gonadal maturity of tinfoil barb fish broodstock (Barbonymus schwanenfeldii). A completely randomized design (CRD) was used in this study with 4 treatments and 3 replications. Fish broodstock in group A (control) was not given any treatment, while fish broodstocks in groups B, C, and D were injected with spawnprim hormone at doses of 0.3 ml/kg body weight, 0.6 ml/kg body weight, and 0.9 ml/kg body weight, respectively. The measured parameters were gonad maturity level, broodstock weight gain, egg diameter, and fecundity. Data were analyzed using one way analysis of variance (ANOVA). The result showed that the gonad maturity level (GML) in groups B, C, and D (GML III and IV) was better than group A (GML I). Statistical analysis showed that the induction of spawnprim hormone on tinfoil barb broodstocks significantly affect (P<0.05) broodstock weight gain, egg diameter, and fecundity. The optimal dose of spawnprim hormone for tinfoil barb was 0.6 ml/kg body weight, with the average broodstock weight gain, egg diameter, and fecundity were 0.011 kg, 1.55 mm, and 102.15 eggs, respectively. The induction of spawnprim hormone has a positive effect on gonad maturation of tinfoil barb fish (B. schwanenfeldii), with the optimum dose is 0.6 ml/kg fish Keywords: gonad maturity, tinfoil barb fish, spawnprim ABSTRAK Penelitian ini bertujuan untuk menganalisis pengaruh penggunaan hormon spawnprim dengan dosis yang berbeda terhadap kematangan gonad induk ikan lemeduk Barbonymus schwanenfeldii. Metode yang digunakan pada penelitian ini adalah metode rancangan acak lengkap (RAL) dengan 4 kelompok perlakuan dan 3 kali pengulangan. Ikan pada kelompok A (kontrol) tidak diberikan perlakuan, sedangkan ikan pada kelompok B, C, dan D masing-masing diinjeksi hormon spawnprim dengan dosis 0.3 ml/kg bobot badan; 0.6 ml/kg bobot badan; dan 0,9 ml/kg bobot badan. Parameter yang diamati adalah persentase induk matang gonad akhir, pertambahan bobot induk, pertambahan diameter telur, dan fekunditas. Data dianalisis dengan menggunakan analisis varians (ANAVA) pola satu arah. Hasil penelitian menunjukkan tingkat kematangan gonad (TKG) pada kelompok perlakuan B, C, dan D (TKG III dan IV) lebih baik dari kelompok A (TKG I). Hasil uji ANAVA menunjukkan bahwa pemberian hormon spawnprim pada induk ikan lemeduk berpengaruh nyata (P<0.05) terhadap pertambahan bobot induk, pertambahan diameter telur, dan fekunditas. Tingkat kematangan gonad terbaik pada penelitian ini diperoleh pada kelompok perlakuan C yang diinduksi hormon spawnprim dengan dosis 0.6 ml/kg bobot badan induk ikan dengan rata - rata pertambahan bobot induk, pertambahan diameter telur, dan fekunditas masing-masing adalah 0.011 kg, 1.55 mm, dan 102.15 butir telur. Induksi hormon spawnprim menimbulkan pengaruh yang positif terhadap pertambahan bobot induk, diameter telur dan fekunditas ikan lemeduk (B. schwanenfeldii), dengan dosis optimum 0.6 ml/kg bobot badan. Abstract

Kegiatan pengabdian kepada masyarakat bertujuan meningkatkan pengetahuan peternak tentang kerugian yang diakibatkan oleh ketiadaan pemeriksaan kebuntingan dan meningkatkan pengetahuan dan keterampilan inseminator tentang metode pemeriksaan kimia urin. Kelompok sasaran dari kegiatan ini adalah 15 (lima belas) orang masyarakat petani peternak sapi di Desa Lampuuk Kecamatan Kuta Baro Kabupaten Aceh Besar dan 2 (dua) orang inseminator di wilayah setempat. Pelaksanaan kegiatan ini dilakukan dengan 2 metode, yaitu (1) metode pendidikan dan penyuluhan dan (2) demonstrasi dan pelatihan di lapangan. Pada kegiatan penyuluhan, peternak akan diajarkan tentang kerugian akibat ketiadaan diagnosis kebuntingan. Kepada inseminator setempat diperkenalkan cara dan alat/bahan yang diperlukan untuk diagnosis kebuntingan. Pada kegiatan demonstrasi akan dilakukan pemeriksaan urin sapi yang telah diinseminasi buatan. Dari hasil kegiatan disimpulkan bahwa pengetahuan peternak tentang kerugian yang diakibatkan oleh ketiadaan pemeriksaan kebuntingan meningkat dan inseminator terampil melakukan diagnosis kebuntingan melalui pemeriksaan kimia urin.

Abstract

This study aimed to determine the correlation between ERVM scores and the levels of estrogen and progesterone in Aceh cows during estrus.This study utilized 8 clinically healthy Aceh cows aged 3-5 years. ERVM scores were determined using a DraminskiTMestrus detector probe insertedinto the vagina on the first day of estrus. Estrogen and progesterone concentrations were measured in blood samples collected on the first day of estrususing the enzyme-linked immunosorbent assay (ELISA) method. Data on the correlation between ERVM scores and the levels of estrogen andprogesterone concentrations were analyzed using the Pearson correlation test. The results showed that the average ERVM score during the estrusphase was 192.9216.94 , and the average concentrations of estrogen and progesterone were 103.1344.86 pg/mL and 0.960.60 ng/mL,respectively. Correlation analysis revealed that coefficient correlation (r) values between EVRM and estrogen was -0.607 and with progesterone was-0.149. In conclusion, the ERVM score in Aceh cattle indicates moderate estrus, and the ERVM score obtained in this study is negatively correlatedwith estrogen and progesterone concentrations.

Abstract

Background and Aim: This study aimed to evaluate the effect of prostaglandin F2 (PGF2) supplementation in semen extenders on the post-thaw sperm motility of Waringin rams.Materials and Methods: Semen was collected from Waringin rams, diluted with Andromed extender, and divided into three treatment groups: control (K0) without PGF2, P1 with 37.5 g PGF2, and P2 with 75 g PGF2.Results : The results showed that post-thaw sperm motility in P1 (49.33%) and P2 (54.00%) was significantly higher than in the control group K0 (25.33%) (P 0.05), although no significant difference was observed between P1 and P2 (P 0.05). Additionally, pre-freezing sperm motility also increased in P1 (75.00%) and P2 (82.33%) compared to K0 (43.67%).Conclusion: It is concluded that PGF2 supplementation in semen extenders can improve the post-thaw motility of Waringin ram spermatozoa, with a 75 g dosage yielding the most optimal effect.

Abstract

This study aimed to determine the length of the estrus period in Aceh cows based on changes in the characteristics of cervical mucus observed using a camera-equipped artificial insemination (AI) endoscope. This study used twelve healthy and reproductively sound cows aged 4-6 years. All cows were synchronized using prostaglandin F2α (PGF2α) injected intramuscularly twice, 11 days apart. Estrus detection was conducted 24 hours following the second injection of PGF2α three times daily for 30 minutes, respectively. Estrous cows were then subjected to the examination of their cervical mucus at 0, 6, 12, and 18 hours after the beginning of estrus. This study showed that out of 12 samples, only eight cows exhibited estrus signs. Characteristics of cervical mucus of cows at 0 and 6 hours of estrus were transparent. At 12 hours of estrus, five cows showed transparency, and the remaining three cows showed cloudy cervical mucus. Meanwhile, at 18 hours of estrus, all sampled cows showed cloudy cervical mucus then regarded as the end of the estrus. That means the estrus length of Aceh cows in this study was 15.75 ± 3.11 hours. It can be concluded that the determination of the duration of estrus in Aceh cows based on changes in the characteristics of the cervical mucus can be carried out using an AI endoscope with a camera. For further research, it is suggested to apply this estrus detection technique in the implementation of AI and determine the pregnancy rate.

Abstract

Accurate determination of estrus status in an animal based on an appropriate estrus detection method can guarantee the cows' pregnancy after mating. This study aims to determine the estrus status of Aceh cows using the van Eerdenburg estrus behavior scoring method. This study was conducted from 22 January to 12 February 2021, at the Technical Implementation Unit for Animals of the Faculty of Veterinary Medicine, Universitas Syiah Kuala, Banda Aceh, Indonesia. The sample used was eleven Aceh cows aged 4-6 years old. Parameters observed were the number of cows exhibiting signs of estrus using the estrus behavior scoring method. The accuracy of estrus detection was confirmed by palpation per rectum to detect the presence of uterine tone. By observing signs of estrus using the method studied, 90.9% (10/11) of the cows were determined to be in estrus. Confirmation by the presence of uterine tone showed a 100% accuracy rate. The most effective sign of estrus was the passivity of the cow when being ridden by another cow or bull. This study concluded that estrus status in local cows could be determined using the estrus behavior scoring of the van Eerdenburg method.

Abstract

This study aims to analyze the effect of administering Malacca leaves (Phyllanthus emblica) extract on the sperm quality of mice. This leaf extract was administered orally using gavage method once daily for 21 days. There were four treatment groups: the control group (P0) received distilled water while the first (P1), second (P2), and third (P3) treatment groups received 300 mg, 600 mg, and 1200 mg of Malacca leaf ethanol extract/kg body weight, respectively. Each groups consisted of four mice and was evaluated for sperrm motility, viability, morphology, and concentration. The results showed that sperm motility, viability, morphology and sperm concentration tended to decrease after administration of Malacca leaf ethanol extract for all parameters, although there was no significant difference (P0.05). It was concluded that the administration of Malacca leaf ethanol extract tends to reduce the sperm quality of mice. ___________________________________________________________
