Abstract

Infection or tissue injury induces inflammation, which causes pain. Non-Steroidal Anti-Inflammatory Drugs (NSAIDs) are commonly used anti-inflammatory agents but can cause adverse effects on the stomach, kidneys, and liver when used long-term. As a result, efforts have been made to discover new drugs from natural sources to minimize these risks. With advances in technology, the search for new drug candidates using computational modeling, such as molecular docking, has become increasingly common. The aim of this systematic literature review is to predict and evaluate the potential of natural compounds as anti-inflammatory agents through in silico analysis. A systematic literature review of 10 recent articles, selected from an initial 376 articles on the topic of natural plant compounds' anti-inflammatory potential in silico, published between 2020 and 2023, was conducted using the PubMed and Springer databases. The conclusion from the review of the structural analysis of secondary plant metabolites shows that all 45 compounds tested demonstrated the ability to interact with the COX-2 enzyme, which plays a key role in inflammation and pain. Among these compounds, catechin, a member of the flavonoid group, was identified as the best candidate based on in silico analysis.

Abstract

Hydroquinone is a compound that is often used in cosmetic products because of its ability to brighten the skin by inhibiting melanin production. However, its use can cause dangerous side effects such as irritation, hyperpigmentation and the risk of cancer, so its use in cosmetics has been limited by BPOM. High Performance Liquid Chromatography (HPLC) is an analytical method commonly used to detect hydroquinone in cosmetic preparations because of its sensitivity and ability to separate compounds with high complexity. This article is a literature review which aims to determine the effect of mobile phase comparison on hydroquinone analysis using the HPLC method. The research results show that selecting the right mobile phase is very important to guarantee accurate, precise and efficient analysis results. The combination of a polarity-based mobile phase that matches the properties of hydroquinone provides good resolution, high sensitivity, and optimal retention time. This research is expected to provide guidance in optimizing the HPLC method for hydroquinone analysis, and is expected to support cosmetic safety monitoring.

Abstract

Background: The rising consumption of coffee in Indonesia necessitates reliable methods for caffeine analysis to ensure product quality and safety. Thin Layer Chromatography (TLC) is a simple, effective, and polarity-based technique widely used for the qualitative identification of compounds like caffeine in coffee. However, the results of TLC analysis are highly dependent on several methodological parameters. Objectives: This literature review aims to identify and analyze the critical parameters of the TLC method—including the mobile phase, stationary phase, visualization techniques, and sample preparation—that influence the effectiveness of caffeine identification in coffee. Methods: A systematic literature review was conducted using online databases (Google Scholar, PubMed, Semantic Scholar, and Crossref). Articles from the last ten years were selected based on inclusion and exclusion criteria, resulting in nine primary studies for in-depth analysis. Results: The review confirms that silica gel 60 F254 is the most effective stationary phase. The composition of the mobile phase is crucial; a mixture of dichloromethane:methanol (9.5:0.5) was found to be optimal for separating caffeine, yielding Rf values within the ideal range of 0.3–0.7. Visualization under UV light at 254 nm successfully detected caffeine as dark spots. Sample preparation techniques, particularly reflux and ultrasonic extraction, significantly enhanced extraction efficiency and detection sensitivity. Factors such as mobile phase stability, spotting technique, and sample matrix were also identified as key influencers on the chromatographic results. Conclusion: TLC is an efficient and cost-effective method for the qualitative identification of caffeine in coffee. Its success is determined by the optimization of critical parameters, primarily the mobile phase composition. While excellent for initial screening, TLC requires complementary techniques for precise quantitative analysis. Future work should focus on standardizing these parameters to improve the reproducibility and reliability of caffeine analysis using TLC.

Abstract

The increasing prevalence of antibiotic-resistant bacteria poses a critical global health concern, necessitating the exploration of novel antibacterial solutions. Endophytic bacteria, which colonize plant tissues without causing harm, have gained attention as potential sources of bioactive metabolites. This study aimed to isolate and characterize endophytic bacteria from Bambusa vulgaris leaves and evaluate their antibacterial potential against pathogenic microorganisms. Leaf samples were collected from Bekasi, West Java, and subjected to a surface sterilization process prior to bacterial isolation. A total of 12 bacterial strains were successfully obtained and screened for antibacterial activity against Bacillus subtilis, Propionibacterium acnes, Pseudomonas aeruginosa, and Staphylococcus epidermidis using the agar well diffusion assay. Three isolates exhibited notable inhibitory activity, with P8 demonstrating the strongest antibacterial effects against B. subtilis, P. acnes, and S. epidermidis. The two most potent isolates, P8 and K3, were characterized via 16S rRNA gene sequencing. Genomic DNA extraction was performed, followed by Polymerase Chain Reaction (PCR) amplification using the universal primers 27F (5′-AGAGTTTGATYMTGGCTCAG-3′) and 1492R (5′-GGTTACCTTGTTACGACTT-3′). Sequencing and The Basic Local Alignment Search Tool (BLAST) analysis confirmed that isolate P8 exhibited 100% similarity to B. subtilis strain LZH-H1, whereas isolate K3 shared 99.85% similarity with Pantoea stewartii subsp. indologenes strain SR2-12. These findings suggest that endophytic bacteria from B. vulgaris endohytic bacteria hold promise as potential sources of antibacterial compounds. Further research is necessary to purify and characterize these bioactive metabolites for potential pharmaceutical applications.

Abstract

NSAIDs (Non-Steroid Anti-Inflammatory Drugs) are drugs that can treat pain and inflammation. The therapeutic effect of NSAIDs as anti-inflammatory analgesics is that they have the ability to inhibit the biosynthesis of prostaglandins which are pain mediators. This prostaglandin has a major role in inhibiting the COX enzyme (cyclooxygenase) in which COX has two isoforms, namely COX1 isoenzyme (non-selective) and COX2 isoenzyme (selective). Tricyclic COX-2 selective inhibitors work by blocking COX-2. COX-2 binds to PD-L1 via the PGE2 pathway and affects macrophages in suppressing myeloid. COX-2 has functions including producing reactive oxygen species that are responsible for DNA damage, activating substances that deviate from intracellular pathways such as the MAPK and PI3 K/AKT pathways, activating STAT3, inducing Bcl-2, and producing growth factors including growth factors. epidermal (EGF)) and fibroblast growth factor (FGF).
