Abstract

Plant genetic diversity is an emerging variation in a crop group caused by its genetic factors. Local corn germplasm as a source of plant genes that are able to adapt to the local environment. The purpose of this research is to obtain information on genetic variation of Tana Toraja local maize germ plasm using SSR (Simple Sequence Repeat) marker. This research was conducted at Balitsereal Molecular Biology Laboratory, Agricultural Research Agency in Maros Regency, South Sulawesi. A total of 4 local maize populations were analyzed by laboratory experimental method with observation with NTSYS pc 2.1 program. The results showed that the average number of alleles was 3.72 alleles per locus and the polymorphism rate of 0.53 with the genetic similarity coefficient was in the range of 0.47 to 0.85. 2 main clusters formed in the genetic similarity coefficient 0.47. Klaster I is Local DallePondan and Local Purple. Klaster II is Local Bebo and Kandora. The genetic distance is in the range of 0.15 to 0.74 with an average genetic distance of 0.46. From the data obtained shows that the 4th germplasm of the population of Tana Toraja Local maize diteleti has a very informative level of genetic diversity. Genetic diversity of local maize germplasm of Tana Toraja, can be used as a source of genes in the assembly of improved varieties in the future.

Abstract

This study aimed to determine the optimum temperature and pH of the phytase activity produced by isolate of thermophilic bacteria from Sulili hot springs in the district Pinrang south sulawesi. This is a descriptive research, that measure of phytase activity by treated the variation of temperature and pH. The various of temperature has eight levels ie 20; 30; 40; 50; 60; 70; 80; and 90oC, the pH treatments consists of eight levels ie 2; 3; 4; 5; 6; 7; 8 and 9. Phytase activity was determined by converting the Na-phytate absorbance values produced by phytase enzyme against Na-phytate standard curve. One unit of phytate enzyme activity is defined as the amount of enzyme capable of hydrolysed Na-phytate and produce a peptide which is equivalent to one mole of tyrosine per minute under the conditions of analysis. The results of studies have shown that there is a significant effect of temperature on the phytase activity of Bacillus coagulans, thermophilic bacteria from Sulili hot springs, whereas no significant effect of pH treatment. Optimum phytase activity of B. coagulans are at 60°C and pH 7 ie 6.11 units/ mL.

Abstract

Penelitian ini bertujuan untuk mengetahui bobot molekul protein OMP S. Typhiasal Makassar dengan menggunakan metode SDS-PAGE. Penelitian ini dilakukanpada bulan Mei sampai Juni 2014 di Laboratorium Imunologi dan BiologiMolekuler Fakultas Kedokteran. Penelitian ini menggunakan antigen OMP hasilekstraksi kultur bakteri S. Typhi dari serum darah penderita demam tifoid yangdiperoleh dari Rumah Sakit di Kota Makassar. Monitoring bobot molekuldikerjakan menggunakan SDS-PAGE metode Laemmli yang telah dimodifikasiyaitu separating gel 12%, stacking gel 3%, dan pewarnaan gel (staining)menggunakan coomassie blue. Proses elektroforesis menggunakan teganganlistrik 120 volt 40 mA selama 3,5 jam. Gel hasil SDS-PAGE memperlihatkanadanya tiga band protein mayor OMP S. Thyphi yaitu pada 25 kda, 36 kda, 55kda, dikarenakan pada ekstrak kasar masih banyak debris dari protein lain

Abstract

Leptospirosis is a zoonotic disease caused by the bacteria Leptospira which is transmitted by mice. This disease is found in tropical and subtropical regions that have high rainfall. Leptospira bacteria can infect humans through skin wounds or mucous membranes such as the nose, mouth and eyes. The aim is to determine the factors that influence the incidence of leptospirosis and how to prevent it. The factors that influence the incidence of leptospirosis are the presence of standing water, the condition of the gutters, the presence of rats, activity in water and a history of wounds. Prevention of leptospirosis can be done by providing counseling, protecting workers, recognizing potentially contaminated water and soil, eradicating rodents (rats), separating infected pets, vaccinating livestock and pets, vaccines and covering wounds and abrasions with waterproof dressings.

Abstract

Budidaya tanaman hidroponik tanpa menggunakan media tanah dapat membantu masyarakat bercocok tanam pada lahan yang sempit, baik dalam skala kecil dan skala industri. Tanaman sayur hidroponik menjadi pilihan yang banyak ditanam dan dikonsumsi oleh masyarakat. Salah satu di antaranya adalah sawi hijau (Brassica rapa var. parachinensis) yang kaya akan antioksidan dan polifenol. Penelitian ini bertujuan untuk mengetahui kemampuan Bio-Catharantin sebagai agen poliploidi terhadap fenotipe B. rapa var. parachinensis yang dibudidayakan di Sulawesi Selatan. Penelitian dilaksanakan di Laboratorium Genetika dan Biologi Molekuler Prodi Biologi Universitas Islam Negeri Alauddin Makassar dengan menggunakan rancangan acak kelompok lengkap terdiri dari dua kondisi yaitu konsentrasi Bio-Catharantin meliputi C1 = Bio-Catharantin 0,05%; C2 = Bio-Catharantin 0,1%; C3 = Bio-Catharantin 0,15% dan waktu perendaman T1 = 3 jam; T2 = 6 jam. Pengamatan fenotipik meliputi tinggi batang, diameter batang, panjang daun, lebar daun, panjang buku, dan panjang akar. Hasil penelitian menunjukkan bahwa perendaman Bio-Catharantin 0,05-0,15% selama 6 jam pada B. rapa var. parachinensis berpengaruh nyata terhadap karakter fenotipiknya.

Abstract

Penelitian ini mengeksplorasi peran Lembaga Sertifikasi Halal (LSH) dalam membangun ekosistem halal yang kredibel dan berkelanjutan, serta mengidentifikasi tantangan dan peluang yang dihadapinya. LSH berfungsi sebagai penjaga standar kehalalan, memastikan produk dan layanan yang beredar memenuhi kriteria yang ketat. Tantangan utama yang dihadapi meliputi perlunya standarisasi dan harmonisasi internasional, menjaga transparansi dan kepercayaan publik, mengatasi regulasi yang berbeda di berbagai negara, serta keterbatasan sumber daya dan kapasitas. Di sisi lain, terdapat peluang besar dalam pasar halal global yang terus berkembang, penggunaan teknologi untuk meningkatkan efisiensi dan transparansi, peningkatan kerjasama internasional untuk pengakuan sertifikasi, edukasi yang lebih luas kepada produsen dan konsumen, serta diversifikasi layanan sertifikasi ke sektor-sektor lain seperti kosmetik, farmasi, dan keuangan syariah. Hasil penelitian menunjukkan bahwa dengan strategi yang tepat, LSH dapat memperkuat ekosistem halal, meningkatkan kepercayaan publik, dan mendorong pertumbuhan ekonomi halal di tingkat global. Penelitian ini memberikan wawasan penting bagi pemangku kepentingan dalam upaya memperbaiki dan mengoptimalkan peran LSH dalam ekosistem halal.

Abstract

This article discusses the opportunities for implementing halal certification to increase their economic growth. It is said that new opportunities for business actors who are benchmarks are the guarantee of halalness of products sold to consumers in attracting the business opportunities they are undergoing. Therefore, the Sambas Regency Government is always optimal and strives to develop the halal food and beverage industry in each region in order to encourage the growth of the halal industry for culinary business actors. In implementing the acceleration of the implementation of halal standards where one of the things needed is halal products, article 4 states that halal certification of a product is mandatory. To support one of the developments of the halal industry, an in-depth study is needed in this study to find out the extent of application in providing halal product assurance through halal certification for MSME food and beverage products in Sambas district. The research method used is a descriptive qualitative approach research method. Based on the results of observations that show that superior food and beverage products, culinary business actors have great potential in realizing halal product guarantees so that they have optimal potential and are found to be quite strategic and appropriate in realizing the design of the halal food and beverage industry.

Abstract

Industri pangan di Indonesia telah mengalami perubahan besar dengan implementasi Undang-Undang Pangan Halal. Kebijakan ini bertujuan untuk memastikan semua produk pangan yang tersedia di pasar domestik maupun internasional memenuhi standar halal yang ditetapkan. Sistem Jaminan Halal (SJH) menjadi elemen kunci dalam manajemen transformasi ini, mencakup seluruh proses dari bahan baku hingga distribusi produk akhir. Penelitian ini meneliti efektivitas manajemen SJH dalam mendukung transformasi industri pangan, dengan fokus pada regulasi, implementasi, dan pengawasan. Melalui pendekatan kualitatif, penelitian ini mengidentifikasi produsen dalam memenuhi persyaratan halal, serta peran pemerintah dan lembaga sertifikasi dalam memastikan kepatuhan dan konsistensi standar halal. Hasil penelitian menunjukkan bahwa kesuksesan transformasi industri pangan melalui UU Pangan Halal sangat bergantung pada manajemen SJH yang efektif, diharapkan industri pangan Indonesia dapat meningkatkan daya saing dan kepercayaan konsumen, baik di pasar domestik maupun internasional

Abstract

Genetic diseases are known to be difficult and could not be avoided. The disease is obtained from mutations and changes in genetic traits that are passed on from one or both genes from parent to child. Genes carrying traits or diseases can be dominant and recessive, their location can be on the autosomal chromosome or sex chromosome X or Y. This study was conducted on premarital female respondents with defined inclusion criteria. Data were collected through a google form questionnaire. A total of 110 respondents with several parameters of genetic counseling. The result was obtained that all respondents have known that certain diseases can be inherited. However, from all respondents, it was found that they had low knowledge of genetic counseling and premarital screening. From the respondent's data, it was also found that only 17% of respondents knew the history of the disease of the prospective partner and their family. In this case, it can be said that respondents might be carry the sick gene which means the probability that their child will carry the same gene is 50% normal: 50% sick. By screening, it might be predicted that the couple will be able to find out the potential diseases that will be passed on to their offspring later. Latest 48% decided to cancel and 26% said they did not agree and stated to continue the plan of marriage. Lack of knowledge about risk of genetic disorders need education about premarital screening to increase this information and premarital screening to the public.

Abstract

Indonesia is an archipelagic country with a high level of population diversity such as religion, ethnicity, culture and also genetic diversity. Environmental factors also have an impact on the genetic variation and diversity of the population, including high mobility and migration triggering the tendency for inter-ethnic marriages to occur. This environmental factor is one of the reasons for research based on DNA analysis on ethnicity in Indonesia for the purpose of forensic identification. One of the DNA analyzes for forensic identification is the microsatellite DNA analysis method with a high sensitivity level.
