Abstract

ABSTRAKKucing hutan (Felis chaus) dan kucing domesik (Felis domesticus) termasuk famili Felidae, namun  secara kasat mata terdapat perbedaan morfologi tubuh antara kedua spesies tersebut. Penelitian ini bertujuan mengidentifikasi dan membandingkan morfologi dan morfometri tulang-tulang pembentuk skeleton axiale kucing hutan dan domestik. Penelitian ini menggunakan preparat kucing hutan dan kucing domestik masing-masing 1 ekor dan berjenis kelamin jantan yang telah diawetkan dalam larutan formalin. Kucing hutan dan domestik dipreparir untuk mendapatkan tulang-tulang pembentuk skeleton axiale lalu diawetkan dalam larutan formalin 5 % dan dikeringkan pada suhu ruang (27°C). Selanjutnya dilakukan pengamatan morfologi dan morfometri setiap tulang. Hasil pengamatan dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa secara morfologi tulang-tulang pembentuk skeleton axiale kucing hutan dan domestik memiliki bentuk yang hampir sama, kecuali pada ala atlantis dari os atlas (os vertebrae cervicalis I) kucing hutan lebih sempit dibandingkan ala atlantis kucing domestik. Selain itu bentuk os axis (os vertebrae cervicalis II) kucing hutan lebih ramping dibandingkan kucing domestik. Perbedaan morfologi lainnya ditemukan pada ossa costales dan os sternum yang didukung dengan perbedaan morfometrinya. Dapat disimpulkan bahwa perbedaan morfologi dan morfomteri tulang-tulang penyusun skeleton axiale antara kucing hutan dan domestik ditemukan pada os atlas, os axis, ossa costales, dan os sternum.    Kata kunci: skeleton axiale, morfologi, morfometri, kucing hutan, dan kucing domestikABSTRACTWild cat (Felis chaus) and domestic cat (felis domesticus) belong to the Felidae family, but morphologically there are differences in the posture of both species. This study aims to identify and compare the morphology and morphometry of bones forming the skeleton of wild and domestic cats. This study used the preparation of a male wild cat and a male domestic cat that have been preserved in formalin solution.  Furthermore, cats were prepared for obtaining skeleton-forming bones and then preserved in 5% formalin and dried at room temperature. After drying, morphology and morphometry were observed and data were analyzed descriptively. The results showed that morphologically, the skeleton-forming bones of wild cat and domestic cat axiale had almost the same in shape, except in ala atlantis of os atlas (os vertebrae cervicalis I), where in wild cat the size was narrower than in domestic cat. Additionally, the size of os axis (os vertebrae cervicalis II) in wild cat was slimmer than the bone size in domestic cat. Other morphological differences were found in ossa costales and os sternum which were supported by differences in morphometry. It can be concluded that specifically, the difference of axial skeleton-forming bones between wild and domestic cats were found in os atlas, os axis, ossa costales, and os sternum.  Keywords: skeleton axiale, morphology, morphometry, wild cat, and domestic cat

Abstract

The aim of this research was to determine the anatomy and histology of pancreas of water monitor lizard (Varanus salvator). Two pancreas were used in this research (a male and a female). Water monitor lizard was euthanized using chloroform and subsequently necropsied to collect the pancreas. The pancreas was washed in 0.9 % NaCl solution. Anatomical observations were carried out including location, shape, and color of pancreas, also the length and weight measurements. Then, pancreas was fixed with 4% paraformaldehyde for 7 days to proceed to histological preparation and hematoxylin eosin (HE) staining. The results of the anatomical observations showed that pancreas of water monitor lizard have two lobes, dorsal lobe (splenic lobe) and ventral lobe (duodenal lobe). Dorsal lobe which was in oval form adhered with the spleen, and ventral lobe was located in the cranial of intestines with the form of long spherical. Both of pancreas lobes were white-yellow color with the length of 1.45±0.64 cm (dorsal lobe) and 7.00±0.42 cm (ventral lobe). Weight of dorsal lobe was 0.15±0.01 g and ventral lobe was 6.35±2.30 g. The histological observations found the acinar cells, ducts, and blood vessels in both of pancreas lobes. Additionally, the similar structure of Langerhans islet only found at the ventral lobe of pancreas. In conclusion, pancreas of water monitor lizard consists of two lobes which located differently. Ventral lobe is larger than the dorsal lobe. Both lobes have similar histological structure except the Langerhans islets are only appeared in the ventral lobe.

Abstract

The presence of glycoconjugates in caput, corpus, and cauda epididymis of the Javan muntjacs was performed using lectin histochemical method. The epididymis organs were obtained from two adult male Javan muntjacs in hard antler stage and then immediately fixed in Bouin’s solution and processed in histological manner. Six types of biotinylated lectins namely Peanut agglutinin (PNA), Soybean agglutinin (SBA), Wheat germ agglutinin (WGA), Ricinus communis agglutinin (RCA), Concanavalin A (Con A), and Ulex europaeus agglutinin I (UEA I) were applied to detect the specific glycoconjugates in the epididymal tissue of muntjacs. The result showed the presence of glycoconjugates were detected by five types of lectins except UEA I in caput, corpus, and cauda epididymis with specific distribution and also the intensity of lectins binding. Glycoconjugates β-galactose, β-glucose, mannose, N-acetyl-galactosamine, N-acetyl-glucosamine, and sialic acid were stained with various intensity in epithelial cells, basal cells, stereocilia, spermatozoa, and stroma of caput, corpus, and cauda epididymis. The presence of sialic acid on the surface of spermatozoa detected by WGA are thought to be play a role on sperm protection during their maturation and transit along epididymal lumen. In conclusion, the difference of glycoconjugates distribution pattern and lectin binding in the epididymal caput, corpus, and cauda of the Javan muntjac indicates the regional differences in epididymal function in spermatozoa maturation.

Abstract

Penelitian ini bertujuan mengetahui distribusi glycoconjugate yang terekspresi pada sel epitelium oviduk kancil (Tragulus javanicus). Dalam penelitian ini digunakan satu oviduk kancil yang berasal dari satu ekor kancil b etina dewasa berumur lebih dari satu tahun. Sampel difiksasi dengan larutan Bouin dan diproses menurut standar histologi sampai menjadi blok parafin dan dipotong dengan ketebalan 5 µm. Jenis lektin yang digunakan adalah biotinylated (Con A, PNA, RCA, UEA I, dan WGA) dengan dosis masing-masing sebanyak 15 µg/ml. Hasil penelitian diketahui bahwa glycoconjugate dengan residu gula galaktosa, glukosa, manosa, N-asetilgalaktosamin, N-asetilglukosamin, fukosa, dan asam sialat ditemukan pada bagian apikal sel epitel dan di dalam sitoplasma. Glycoconjugate dengan residu gula N-asetilgalaktosamin merupakan glycoconjugate yang paling banyak ditemukan di bagian apikal sel epitel dan di dalam sitoplasma dibandingkan dengan glycoconjugate dengan residu gula lainnya.

Abstract

Penelitian ini bertujuan mengetahui pengaruh pemberian ekstrak epididimis (EE) terhadap peningkatan kualitas spermatozoa kambing jantan  lokal. Dalam penelitian ini digunakan 12 ekor kambing jantan lokal, berumur 1,5 tahun dengan bobot badan 10-15 kg dan dibagi atas empat kelompok (K0, KP1, KP2, dan KP3). Kelompok K0, hanya diinjeksi dengan NaCl fisiologis sedangkan kelompok KP1, KP2, dan KP3 diinjeksi EE masing-masing 1, 2, dan 3 ml/ekor selama 13 hari berturut-turut. Pada hari ke-14, dilakukan pengambilan semen kambing dengan elektroejakulator dan selanjutnya dilakukan pemeriksaan kualitas spermatozoa. Hasil penelitian menunjukkan bahwa pemberian EE dengan dosis 1 dan 3 ml/ekor EE selama 13 hari berturut-turut menyebabkan peningkatan kualitas spermatozoa dibanding kelompok kontrol. Berdasarkan hal tersebut dapat disimpulkan bahwa EE berpotensi meningkatkan kualitas spermatozoa pada kambing jantan lokal.

Abstract

The objective of this study was to identify the type of specific glycoconjugates and its distribution in testicular spermatogenic cells in muntjak (Muntiacus muntjak muntjak) based on lectins histochemistry. An adult male muntjak aged 4-5 years old in hard antler period was used in this study. Testicular tissue was fixed in Bouin solution and processed histologically. Histochemistry method was performed using six types biotinylated lectins such as peanut agglutinin (PNA), soybean agglutinin (SBA), wheat germ agglutinin (WGA), ricinus communis agglutinin (RCA), concanavalin A (Con A), and ulex europaeus agglutinin I (UEA I) with 20 µg/ml of concentration for PNA lectins and 15µg/ml for other type of lectins. The results showed that glycoconjugates were detected by all type of lectins except UEA I in testicular spermatogenic cells with variation in distribution pattern and also the intensity of lectins binding. Glycoconjugates β-galactose, β-glucose, mannose, Nacetylgalactosamine, N-acetylglucosamine and sialic acid were stained intensely by lectins in golgy-cap phase and acrosomal phase of spermatids. Glycoconjugate N-acetylgalactosamine was the sugar residues which distributed abundantly that marked by positive reaction with PNA, SBA, and RCA lectins. In conclusion, glycoconjugates are detected in testicular spermatids cells of muntjak indicated that glycoconjugates have an important role in spermatogenesis particularly in spermiogenesis. Key words: glycoconjugates, lectins, spermatid, spermatozoa, muntjak

Abstract

This study aimed to analyze the muscle microstructure of longissimus dorsi Brahman cross (BX) cattle slaughtered at the slaughterhouse of North Aceh District. The meat samples for the study were taken from three male BX cattle aged 18-30 months with body weights ranging from 400 kg to 500 kg and the average body condition scores (BCS) of 3-4. The muscle used was the longissimus dorsi muscle. The mean (±SE) diameter and cross-sectional area (CSA) of muscle fibers and the percentage of fast fibers in the longissimus dorsi BX muscle were 71.69±1.23 µm, 4263.43±305.69 µm2 , and 48.17±8.81%, respectively. This study concluded that the longissimus dorsi muscle of the BX has relatively large muscle fibers, some of which in each fascicle are composed of fast muscle (fast fiber/type II).

Abstract

ABSTRAK Penelitian ini bertujuan mengidentifikasi karakteristik morfologi dan morfometri tulang-tulang pembentuk skeleton appendiculare antara itik (Anas platyrhynchos) dan entok (Cairina moschata). Sampel yang digunakan dalam penelitian ini berupa itik jantan (n=3) berumur 1 tahun dengan bobot badan 2-3 kg dan entok jantan (n=3) berumur 1 tahun dengan bobot badan 3-4 kg. Itik dan entok disembelih lalu dilakukan pemisahan (preparir) bulu, kulit, otot-otot dan organ-organ visceral. Setelah proses preparir, preparat direndam dalam larutan deterjen selama enam hari. Tulang-tulang penyusun skeleton appendiculare itik dan entok dipisahkan dari tulang tubuh lainnya. Proses pengawetan tulang dilakukan dengan merendam tulang-tulang dalam larutan formalin 5 % selama tiga hari kemudian dikeringkan pada suhu ruang. Setelah kering dilakukan pengamatan secara morfologi dan morfometri. Data morfologi dianalisis secara deskriptif sedangkan data morfometri dianalisis dengan uji t (P0,05). Berdasarkan pengamatan tulang-tulang pembentuk skeleton appendiculare terbagi atas dua kelompok tulang, yaitu ossa membri thoracici dan ossa membri pelvinae. Secara morfologi tulang-tulang pembentuk skeleton appendiculare itik dan entok memiliki bentuk yang sama, akan tetapi beberapa tulang entok lebih besar dibandingkan tulang-tulang itik. Secara morfometri ukuran beberapa tulang entok berbeda nyata dengan tulang-tulang itik (P0,05). Kesimpulan dari penelitian ini adalah tulang-tulang pembentuk skeleton appendiculare itik dan entok memiliki bentuk yang sama namun beberapa tulang berbeda ukurannya.  Kata kunci:  Itik, entok, skeleton appendiculare, morfologi dan morfometri. ABSTRACT This study aims to identify the morphological characteristics and morphometry of skeleton appendiculare ducks (Anas platyrhynchos) and muscovy ducks (Cairina moschata). The sample used in this study was drakers (n = 3) aged 1 year with a body weight of 2-3 kg and male muscovy (n = 3) aged 1 year with a body weight of 3-4 kg. Each of duck was slaughtered and then separated (for preparir section) to removed feathers, muscles, and visceral organs. After the preparation process the sample was soaked in detergent solution for six days. The skeletal bones that form ducks and muscovy ducks skeleton appendiculare were separated from other bones and subsquently. Soaked in 5% formalin solution for three days then drying it at room temperature. After drying, morphology and morphometry of bones were observed. Morphological data were analyzed descriptively while morphometric data were analyzed by student t test. Based on observations, the bones forming the appendicular skeleton were divided into two groups of bones, namely the thoracic bones and the pelvinae membrane. Morphologically the bones forming the appendiculare skeleton of ducks and Muscovy ducks have the same shape, but some Muscovy duck bones were larger than the bones of ducks. Morphometrically, the sizes of some Muscovy duck bones were significantly different from those of ducks (P0.05). The conclusion of this study is that the bones forming the appendiculare skeleton of ducks and Muscovy ducks have the same shape but several bones differ in size.Keyword:  Ducks, muscovy duck, skeleton appendiculare, morphology, and morphometry.

Abstract

Tanjong Selamat is one of the villages directly adjacent to Universitas Syiah Kuala, Banda Aceh. Its strategic location makes Tanjong Selamat Village a priority to be developed into an assisted village of Universitas Syiah Kuala. The main occupations of local Tanjong Selamat villagers are farmers and breeders. Efforts to increase the community income can be done through the cultivation of laying ducks and post-harvest management in the form of making salted eggs. This community service activity was carried out in the form of extension, demonstration, and training. The activity materials are knowledge about the prospects for laying ducks cultivation, prevention and treatment of diseases that often attack ducks, good cage systems, management of breeding, and the making of salted eggs. The cage system and breeding management were carried out by making a pilot cage consisting of the main cage of 15 m2 and a breeding area of 50 m2. The cage was filled with 50 domestic ducks (peking ducks and tegal ducks) aged 4 months. During the community service activity, the participants were very enthusiastic and able to implement the technology package that had been given. Based on the activity that has been conducted, the villagers of Tanjong Selamat Village, especially the members of the livestock group, can understand well the extension material and have high enthusiasm in improving the management of laying ducks as observed during the demonstration of the cage plot as well as better understand the making of salted eggs that are in accordance with consumer desires and have a long shelf life.

Abstract

The objective of this study was to identified the development of the immunoreactive follicle stimulating hormone (ir-FSH) cells in pituitary glands of rats (Rattus norvegicus)in different age using immunohistochemical (IHC) methods. The pituitary organ that used in this study was collected from eight white female rats aged 2, 4, 6, and 12 months. Pituitary glands were processed histologically and stained with IHC methods to detect the ir-FSH cells. The result showed that the ir-FSH cells were observed in pars distalis (PD), pars tuberalis (PT), and pars intermedia (PI) of adenohypophysis with different number of cells. Furthermore, ir-FSH cells were found abundantly (+++) in the PT and a few (+) in PD of rats aged two months. In rats aged 4 months those cells were distributed in slight number (+) in PT and abundant (+++) in PD. In addition, the cells were also found in a great quantities in rats whose ages were 6 months, (+++) in both PT and PD, but these cells decreased (++) in rats aged 12 months in PD and absent (-) in PT. In conclusion, the devalopment and distribution of ir-FSH cells were found in pars tuberalis and pars distalis adenohypophysis in the pituitary glands of rats aged 2 up to 12 months with different number and distribution patterns.
