Abstract

The objective of this study was to determine the anatomical morphology and histology of the intestineof water monitor (Varanussalvator). Intestine organ from an adult male water monito was used in thisstudy. Perfusion was performed to flow the 4% paraformaldehyde fixative solution through the heart.Intestine was collected, then divided into six sections, followed with dipping them into 4% paraformaldehydesolution for four days, continued histological preparations and stained with haematoxylin eosin (HE). Theanatomical morphology observation was performed before the intestine was processed into histologicalpreparations by observing the outer shape of the intestine and measuring it’slength. Morphologicalobservation was made †after processing theintestinal tissue by observing each portion of the intestinalwall structure. The results showed that transverse fold were found on the outer and inner surfaces of theintestine section I-IV, while in the V-VI part of the intestine, the folds were not observed anymore. Theintestine wall consisted of four layers, such as mucosa, submucosa, tunica muscularis, and serosa. Thevilli on intestine section I-IV were relatively longer and higher, while the intestine vili in section V-VI wererelatively short. The entire mucosa in water monitor was coated with absorptive columnar epitheliumwith goblet cells. Based on the result study, it can be concluded that the water monitor has more efficientdigestive process which can be seen morphologically from the relatively short intestinal, has transversefolds on the surface of the small intestine, and does not have cecum. The intestinal wall consists of layersi.e : mucosa, submucosa, tunica muscularis, and serosa.

Abstract

ABSTRAKSusu memiliki sumber gizi yang paling sempurna. Nilai gizinya yang lengkap menyebabkan susu dapat menjadi media yang sangat baik bagi mikroorganisme untuk pertumbuhan dan perkembangannya. Penelitian ini bertujuan untuk mengetahui jumlah cemaran Staphylococcus aureus pada susu kambing peranakan etawa (PE) di peternakan Adoe A Kabupaten Aceh Besar. Penelitian ini dilaksanakan di Laboratorium Kesmavet Fakultas Kedokteran Hewan Universitas Syiah Kuala, Darussalam Banda Aceh. Penelitian ini menggunakan 10 sampel susu kambing peranakan etawa. Pengambilan sampel dilakukan secara acak dan diencerkan dengan pengenceran desimal dari 101 sampai 106. Selanjutnya dilakukan penghitungan jumlah koloni dengan menggunakan Total Plate Count (TPC) untuk mengetahui jumlah cemaran Staphylococcus aureus. Hasil penelitian menunjukkan bahwa terdapat 30% sampel tercemar Staphylocoucus aureus. Nilai TPC tertinggi diperoleh dari sampel susu K3 yaitu 3,0 x 102 cfu/ml, dan nilai TPC terendah diperoleh dari sampel susu K1 yaitu 1,6 x 102 cfu/ml. Sehingga dapat disimpulkan bahwa nilai TPC yang ditemukan melebihi batas standar SNI untuk bakteri Staphylocoucus aureus pada susu yaitu 1 x 102 cfu/ml.ABSTRACTMilk has the most perfect source of nutrition its complete nutritional value causes milk to be an excellent for microorganisms for growth and development. This study aims to determine the amount of Staphylococcus aureus contamination in Etawa crossbreed’s milk on Adoe A farms in Aceh Besar Regency. This study was carried out at the Veterinary Public Health Laboratory, Faculty of Veterinary Medicine, Syiah Kuala University, Banda Aceh. This study use 10 samples of Etawa Crossbreed’s milk. Sampling was carried out randomly and diluted with a decimal dilution from 101 to 106. Furthermore, the number of colonies was counted using the Total Plate Count (TPC) to determine the amount of staphylococcus aureus contamination. The results of this study showed that 30% of the samples were contaminated with Staphylocoucus aureus. The highest TPC value was obtained from the K3 milk sample, which was 3.0 x 102 cfu/ml, and the lowest TPC value was obtained from the K1 milk sample, which was 1.6 x 102 cfu/ml. So it can be concluded that the TPC value found exceeds the SNI standard limit for Staphylocoucus aureus bacteria in milk, which is 1 x 102 cfu/ml.

Abstract

Changes in the genetic code can trigger changes in the function of the proteins it produces. Genetic changes that commonly occur in the form of single nucleotide polymorphism such as in the aquaporin group gene (AQPs) which play an important role in maintaining moisture and elasticity of the skin epidermis. This study aims to analyze the effect of mutations in one of the aquaporin protein groups (AQPs), namely aquaporin 3 (AQP3) by in silico. Research material in the form of data sequences and 3D structures of AQP3 wild-type dan AQP3 mutant downloaded from the protein databank (pdb id 1FX8 and 1LDF). These two sequences were aligned using the “ClustalW” method to observe the position of the mutation, while the 3D structures were also aligned using the “Alignment” method to observe changes in the native ligand’s “site binding” region. The impact of the conformational changes of mutant AQP3 was also observed on several compounds as test ligands which are known to work to support the function of AQP3 protein such as asiatic acid, madecassic acid, asiaticoside, and alpha-retinoic acid through molecular docking method. Sequence alignment results showed that mutations occurred in the 48th (tryptophan-phenylalanine) and 200th (phenylalanine-threonine) residue sequences. The mutation of the two residues caused a change in the interaction model of all test ligands compared to the normal type of AQP3. The mutation also causes a decrease in the stability of AQP3, but on the contrary does not change the function of AQP3 but only decreases the binding ability of the native ligand molecule which has implications for the decrease in the binding ability of water molecules, glycerol and the test ligand.

Abstract

ABSTRAKAnjing (Canis lupus familiaris) merupakan salah satu hewan peliharaan yang banyak dipelihara di dunia. Karakteristik anjing yang setia, mudah dilatih serta bersahabat dengan manusia menjadi nilai tambah hewan peliharaan satu ini, sejatinya anjing merupakan hewan karnivora yang menjadikan protein sebagai sumber energi utama. Namun, ditengah masyarakat masih banyak memberikan pakan berupa nasi kepada anjing peliharaannya untuk itu penelitian ini dilakukan guna mengetahui kadar gula darah anjing serta faktor-faktor yang memengaruhi kadar gula darah tersebut. Penelitian ini dilakukan secara observasi lapangan dan pemeriksaan sampel darah anjing sebanyak 30 sampel di Kota Bukittinggi, lalu dilakukan pengecekan menggunakan easy touch GCU dan didapatkan hasil bahwa terdapat perbedaan yang signifikan pada kadar gula darah anjing sebelum dan sesudah makan, pada saat sebelum makan kadar gula darah anjing sebesar 73,37±10,88 mg/dL dan dua jam sesudah pemberian pakan sebesar 112,06±14,148 mg/dL hal ini berhubungan dengan karakteristik dari karbohidrat yang cepat dicerna dan diedarkan ke seluruh tubuh. Hasil uji statistik menunjukkan bahwa terdapat perbedaan yang signifikan (p0.05) antara usia dan jenis kelamin terhadap kadar gula darah anjing kampung. Dapat disimpulkan bahwa rata- rata kadar gula arah anjing kampung di kota Bukittinggi masih berada pada kisaran normal.Kata kunci: domestikasi, easy touch GCU, gula darah, karbohidratABSTRACTDogs (Canis lupus familiaris) are one of the most kept pets in the world. The characteristics of being loyal, easy to train and friendly with humans are added values for this pet, in fact dogs are carnivorous animals that made protein be their main energy source. However, in the midst of society, there are still give rice to their dogs, so this research was conducted to determine the blood sugar levels of dogs and the factors that influence it. This research was carried out by field observation and examination of 30 samples of dog blood samples in Bukittinggi City, then checked the sample using the easy touch GCU and the results showed that there were significant differences in the blood sugar levels of dogs before and after eating, which at the time of eating the dog blood sugar was 73.37±10.88 mg/dL and two hours after feeding was 112.06±14.148 mg/dL this was related to the characteristics of carbohydrates that were quickly digested and circulated throughout the body. The results of statistical tests showed that there was a significant difference (p0.05) between age and sex on blood sugar levels of domestic dogs. Based on the research concluded that the average sugar content of domestic dogs in the Bukittinggi City is still in the normal range.Keywords: Blood sugar, carbohydrates, domestication, easy touch GCU

Abstract

ABSTRAKKucing hutan (Felis chaus) dan kucing domesik (Felis domesticus) termasuk famili Felidae, namun  secara kasat mata terdapat perbedaan morfologi tubuh antara kedua spesies tersebut. Penelitian ini bertujuan mengidentifikasi dan membandingkan morfologi dan morfometri tulang-tulang pembentuk skeleton axiale kucing hutan dan domestik. Penelitian ini menggunakan preparat kucing hutan dan kucing domestik masing-masing 1 ekor dan berjenis kelamin jantan yang telah diawetkan dalam larutan formalin. Kucing hutan dan domestik dipreparir untuk mendapatkan tulang-tulang pembentuk skeleton axiale lalu diawetkan dalam larutan formalin 5 % dan dikeringkan pada suhu ruang (27°C). Selanjutnya dilakukan pengamatan morfologi dan morfometri setiap tulang. Hasil pengamatan dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa secara morfologi tulang-tulang pembentuk skeleton axiale kucing hutan dan domestik memiliki bentuk yang hampir sama, kecuali pada ala atlantis dari os atlas (os vertebrae cervicalis I) kucing hutan lebih sempit dibandingkan ala atlantis kucing domestik. Selain itu bentuk os axis (os vertebrae cervicalis II) kucing hutan lebih ramping dibandingkan kucing domestik. Perbedaan morfologi lainnya ditemukan pada ossa costales dan os sternum yang didukung dengan perbedaan morfometrinya. Dapat disimpulkan bahwa perbedaan morfologi dan morfomteri tulang-tulang penyusun skeleton axiale antara kucing hutan dan domestik ditemukan pada os atlas, os axis, ossa costales, dan os sternum.    Kata kunci: skeleton axiale, morfologi, morfometri, kucing hutan, dan kucing domestikABSTRACTWild cat (Felis chaus) and domestic cat (felis domesticus) belong to the Felidae family, but morphologically there are differences in the posture of both species. This study aims to identify and compare the morphology and morphometry of bones forming the skeleton of wild and domestic cats. This study used the preparation of a male wild cat and a male domestic cat that have been preserved in formalin solution.  Furthermore, cats were prepared for obtaining skeleton-forming bones and then preserved in 5% formalin and dried at room temperature. After drying, morphology and morphometry were observed and data were analyzed descriptively. The results showed that morphologically, the skeleton-forming bones of wild cat and domestic cat axiale had almost the same in shape, except in ala atlantis of os atlas (os vertebrae cervicalis I), where in wild cat the size was narrower than in domestic cat. Additionally, the size of os axis (os vertebrae cervicalis II) in wild cat was slimmer than the bone size in domestic cat. Other morphological differences were found in ossa costales and os sternum which were supported by differences in morphometry. It can be concluded that specifically, the difference of axial skeleton-forming bones between wild and domestic cats were found in os atlas, os axis, ossa costales, and os sternum.  Keywords: skeleton axiale, morphology, morphometry, wild cat, and domestic cat

Abstract

The aim of this research was to determine the anatomy and histology of pancreas of water monitor lizard (Varanus salvator). Two pancreas were used in this research (a male and a female). Water monitor lizard was euthanized using chloroform and subsequently necropsied to collect the pancreas. The pancreas was washed in 0.9 % NaCl solution. Anatomical observations were carried out including location, shape, and color of pancreas, also the length and weight measurements. Then, pancreas was fixed with 4% paraformaldehyde for 7 days to proceed to histological preparation and hematoxylin eosin (HE) staining. The results of the anatomical observations showed that pancreas of water monitor lizard have two lobes, dorsal lobe (splenic lobe) and ventral lobe (duodenal lobe). Dorsal lobe which was in oval form adhered with the spleen, and ventral lobe was located in the cranial of intestines with the form of long spherical. Both of pancreas lobes were white-yellow color with the length of 1.45±0.64 cm (dorsal lobe) and 7.00±0.42 cm (ventral lobe). Weight of dorsal lobe was 0.15±0.01 g and ventral lobe was 6.35±2.30 g. The histological observations found the acinar cells, ducts, and blood vessels in both of pancreas lobes. Additionally, the similar structure of Langerhans islet only found at the ventral lobe of pancreas. In conclusion, pancreas of water monitor lizard consists of two lobes which located differently. Ventral lobe is larger than the dorsal lobe. Both lobes have similar histological structure except the Langerhans islets are only appeared in the ventral lobe.

Abstract

The presence of glycoconjugates in caput, corpus, and cauda epididymis of the Javan muntjacs was performed using lectin histochemical method. The epididymis organs were obtained from two adult male Javan muntjacs in hard antler stage and then immediately fixed in Bouin’s solution and processed in histological manner. Six types of biotinylated lectins namely Peanut agglutinin (PNA), Soybean agglutinin (SBA), Wheat germ agglutinin (WGA), Ricinus communis agglutinin (RCA), Concanavalin A (Con A), and Ulex europaeus agglutinin I (UEA I) were applied to detect the specific glycoconjugates in the epididymal tissue of muntjacs. The result showed the presence of glycoconjugates were detected by five types of lectins except UEA I in caput, corpus, and cauda epididymis with specific distribution and also the intensity of lectins binding. Glycoconjugates β-galactose, β-glucose, mannose, N-acetyl-galactosamine, N-acetyl-glucosamine, and sialic acid were stained with various intensity in epithelial cells, basal cells, stereocilia, spermatozoa, and stroma of caput, corpus, and cauda epididymis. The presence of sialic acid on the surface of spermatozoa detected by WGA are thought to be play a role on sperm protection during their maturation and transit along epididymal lumen. In conclusion, the difference of glycoconjugates distribution pattern and lectin binding in the epididymal caput, corpus, and cauda of the Javan muntjac indicates the regional differences in epididymal function in spermatozoa maturation.

Abstract

Penelitian ini bertujuan mengetahui distribusi glycoconjugate yang terekspresi pada sel epitelium oviduk kancil (Tragulus javanicus). Dalam penelitian ini digunakan satu oviduk kancil yang berasal dari satu ekor kancil b etina dewasa berumur lebih dari satu tahun. Sampel difiksasi dengan larutan Bouin dan diproses menurut standar histologi sampai menjadi blok parafin dan dipotong dengan ketebalan 5 µm. Jenis lektin yang digunakan adalah biotinylated (Con A, PNA, RCA, UEA I, dan WGA) dengan dosis masing-masing sebanyak 15 µg/ml. Hasil penelitian diketahui bahwa glycoconjugate dengan residu gula galaktosa, glukosa, manosa, N-asetilgalaktosamin, N-asetilglukosamin, fukosa, dan asam sialat ditemukan pada bagian apikal sel epitel dan di dalam sitoplasma. Glycoconjugate dengan residu gula N-asetilgalaktosamin merupakan glycoconjugate yang paling banyak ditemukan di bagian apikal sel epitel dan di dalam sitoplasma dibandingkan dengan glycoconjugate dengan residu gula lainnya.

Abstract

Penelitian ini bertujuan mengetahui pengaruh pemberian ekstrak epididimis (EE) terhadap peningkatan kualitas spermatozoa kambing jantan  lokal. Dalam penelitian ini digunakan 12 ekor kambing jantan lokal, berumur 1,5 tahun dengan bobot badan 10-15 kg dan dibagi atas empat kelompok (K0, KP1, KP2, dan KP3). Kelompok K0, hanya diinjeksi dengan NaCl fisiologis sedangkan kelompok KP1, KP2, dan KP3 diinjeksi EE masing-masing 1, 2, dan 3 ml/ekor selama 13 hari berturut-turut. Pada hari ke-14, dilakukan pengambilan semen kambing dengan elektroejakulator dan selanjutnya dilakukan pemeriksaan kualitas spermatozoa. Hasil penelitian menunjukkan bahwa pemberian EE dengan dosis 1 dan 3 ml/ekor EE selama 13 hari berturut-turut menyebabkan peningkatan kualitas spermatozoa dibanding kelompok kontrol. Berdasarkan hal tersebut dapat disimpulkan bahwa EE berpotensi meningkatkan kualitas spermatozoa pada kambing jantan lokal.

Abstract

The objective of this study was to identify the type of specific glycoconjugates and its distribution in testicular spermatogenic cells in muntjak (Muntiacus muntjak muntjak) based on lectins histochemistry. An adult male muntjak aged 4-5 years old in hard antler period was used in this study. Testicular tissue was fixed in Bouin solution and processed histologically. Histochemistry method was performed using six types biotinylated lectins such as peanut agglutinin (PNA), soybean agglutinin (SBA), wheat germ agglutinin (WGA), ricinus communis agglutinin (RCA), concanavalin A (Con A), and ulex europaeus agglutinin I (UEA I) with 20 µg/ml of concentration for PNA lectins and 15µg/ml for other type of lectins. The results showed that glycoconjugates were detected by all type of lectins except UEA I in testicular spermatogenic cells with variation in distribution pattern and also the intensity of lectins binding. Glycoconjugates β-galactose, β-glucose, mannose, Nacetylgalactosamine, N-acetylglucosamine and sialic acid were stained intensely by lectins in golgy-cap phase and acrosomal phase of spermatids. Glycoconjugate N-acetylgalactosamine was the sugar residues which distributed abundantly that marked by positive reaction with PNA, SBA, and RCA lectins. In conclusion, glycoconjugates are detected in testicular spermatids cells of muntjak indicated that glycoconjugates have an important role in spermatogenesis particularly in spermiogenesis. Key words: glycoconjugates, lectins, spermatid, spermatozoa, muntjak
