Abstract

An experiment was designed to study the effects of curcumin and thyroxine hormone supplementation in the diet to improve the reproductive performance of catfish broodstock by improving egg quality that eventually increases the production of seedlings. Catfish used in this experiment were supplemented with curcumin and thyroxine hormone through their feeds for 12 weeks. The results showed that there was no significant difference (p > 0.05) in the hepatosomatic index (HSI), gonadosomatic index (GSI), percentage of gonad maturity, total cholesterol concentration in the spawned eggs, high density lipoprotein (HDL) concentration in the spawned eggs, fertilization rate of spawned eggs, and hatching rate of fertilized eggs.  However, there were significant differences (p < 0.05) in the concentration of vitellogenin in the spawned eggs, egg diameter of the spawned eggs, and the triglycerides contents of spawned eggs. It was concluded that curcumin and thyroxine supplementations of African catfish increased vitellogenin concentrations and diameters of spawned eggs that have great potential to improve the reproductive performance.

Abstract

The purposes of this study were to determine oceanographic and water quality parameters and their suitability for white shrimp Litopenaeus vannamei culture. The measurements were carried out on dry season in Semak Daun Island, Karya Island, and Panggang Island waters of Thousand Islands, with areas of 315.0, 12.0, and 102.8 ha, water depth average of 4.6 m (0.5-28.1 m), 14.6 m (0.5-26.7 m), and 5.3 m (0.8-13.6 m), mean current water velocity of  12.9, 12.7, and 13.5 cm/second, respectively.  In the study areas, we found a diurnal tidal pattern with high wave in January and July-August.  Based on temperature, salinity, and water density in Semak Daun Island waters, there seemingly occurred a turn over indicating a good water circulation, while in Panggang Island and Karya Island waters tended to have a stratification. Generaly, water qualities in the study areas were in the op-timum range for white shrimp culture, i.e., temperature of 29.6-30.8oC, turbidity of 0.10-1.05 NTU, transparency of 5.8-9.7 m, total suspended solid of <8 mg/L, total dissolved solid of 20-164 mg/L, pH of 6.89-7.22, salinity of 32.2-32.3, dissolved oxygen of 5.8-10.8 mg/L, ammonia of 0.068-0.145 mg/L, nitrate 1.247-2.589 mg/L, and phosphate  of 1.021-2.352 mg/L. Moreover, in Semak Daun Island wa-ters, we found the highest suitability for white shrimp culture due to its better water circulation.Keywords: mariculture, coral reef waters, strait, water current, turnover, stratification.

Abstract

The aim of this study was to evaluate the effectiveness of vaccination based on gonad maturationstages on tilapia brood stocks in which the released antibodies was able to be transferred to the seed.Vaccine composed with whole cells and extracellular product (ECP) was injected at stage 2 and stage 3 ofthe gonad development stages at concentration of 109 CFU mL1 as much as 4 mL to 1 kg of brood fish.Control fish was unvaccinated treatment. Challenge study at seed was conducted by immersing S. agalactiaefor 30 minutes at 7, 14, 21, and 28 days post hatching (DPH) in 107 CFU/mL. Antibody levels on broodstocks, eggs, and body fluids of seed, and relative percentage survival (RPS) of seed post challenge studywere evaluated. The results showed that stage 2 of gonad developmental stages was found on 7 days postinitial spawning and stage 3 found on 14 days post initial spawning of brood fish. Vaccinated done in stage 2 of gonad developmental stages gave immunoglobulin serum in brood (0,166±0,001), egg (0,165±0,002),and seed aged 7, 14, 21, and 28 days post hatching (0,164±0,002, 0,162±0,005, 0,155±0,006, and 0,14±0,008respectively) were significantly higher (P<0,05) compared to other treatment. Challenged test that doneby immersing with S. agalactiae suspension on larval aged 7, 14, 21, and 28 days had highest RPS(95,24%, 83,33%, 72,22%, and 56,02% respectively) formed on seed from brood stock vaccination in gonaddevelopment stage 2. Vaccination in tilapia brood stocks at stage 2 of gonad developmental stages gavehighest protection by maternal immunity to the seed against S. agalactiae.

Abstract

Perairan laut tropis seperti di Indonesia yang memiliki variasi lingkungan yang hampir konstan– diduga periode pemijahan karang melebar sampai beberapa bulan dan pada fase bulan yang berbeda, sehingga sulit menentukan waktu pemijahannya dalam skala bulan, hari dan jam. Penelitian ini akan memberikan informasi pola reproduksi dan sinkronisasi pemijahan beberapa jenis karang Acropora spp di Kepulauan Spermonde, Makassar. Sebanyak tujuh jenis karang Acropora spp. diamati kematangan atau kemunculan gonad dan pemijahannya di alam (in situ) dan atau di laboratorium (ex situ) di Marine Station Universitas Hasanuddin. Pola reproduksi menunjukkan spawning berlangsung setiap musim hujan pada bulan Februari-Maret selama tiga tahun berturut-turut. Pemijahan berlangsung secara sinkron dan broadcasting pada awal bulan purnama (0 BP sampai +2 BP), pada jam 18:10–19:00). Bersamaan dengan itu, kondisi lingkungan pemijahan berlangsung  saat puncak pasang tinggi dengan suhu rata-rata harian perairan 30,3ºC dan curah hujan yang masih tinggi di bulan Maret. Informasi ilmiah ini akan bermanfaat untuk mengembangkan riset dan tehnik reproduksi karang di alam dan laboratorium sebagai upaya merestorasi dan merepopulasi jenis karang tertentu. Kata kunci : Acropora spp, reproduksi seksual, Kepulauan Spermonde It has been thought that Indonesian marine tropical waters have less environmental variability, so that spawning period of coral extend for several months and occured during different lunar phases. Therefore the timing of coral spawning in a year cannot be predicted especially for monthly, daily and hourly scales. This study was aimed to investigate the reproductive pattern, and the environmental cues of Acropora spp. in Spermonde Islands  reefs of Makassar. Spawning corals have been determined the presence of mature gonad and spawning event in their habitats (in situ) and in the laboratory (ex situ) of Marine Station of Barrang Lompo Island, Hasanuddin University. Here we showed that seven species of Acropora spp. spawned in February and March of rainy season for consecutive three years (2010, 2011, 2012). The multispecific broadcasting spawning took place in lunar period (0–2 AFM) at 06:10–07:00 pm). The spawning time occured in high tide and the temperature was 30,3oC. This study will be useful for development of coral reproduction research and technique in both field and laboratory as an effort to restore coral reef and enhance coral population in particular. Abstract

Permasalahan utama dalam kegiatan usaha budidaya perikanan antara lain adalah ketersediaan benih ikan yang tepat waktu, tepat jumlah dan berkualitas. Ketersediaan benih ikan sangat tergantung pada jumlah telur yang dapat dibuahi dan ditetaskan. Semakin tinggi presentase pembuahan dan penetasan maka akan semakin tinggi pula jumlah larva yang dapat dihasilkan. Penelitian ini menggunakan ikan lele sebagai hewan coba dengan tujuan untuk mengetahui pengaruh suplementasi kurkumin dan tiroksin pada induk ikan lele terhadap kualitas dan performa telur yang dihasilkan. Suplematasi kurkumin dan hormon tiroksin melalui pakan selama 12 minggu pemeliharaan. Parameter yang dianalisis berupa konsentrasi vitelogenin telur, diameter telur, koefisien keragaman diameter telur, konsentrasi lipid dalam telur (triglserida, kolesterol dan HDL), derajat pembuahan dan derajat penetasan. Hasil penelitian menunjukkan bahwa kelompok perlakuan yang diberikan kombinasi penambahan kurkumin dan hormon tiroksin memiliki nilai konsentrasi vitelogenin (8.17+2.74 µg/mL) (p<0,05), diameter telur (1.43+0.00 mm) (p<0,05), dan konsentrasi trigliserida (4.89+0.53 mg/g) (p<0,05) tertinggi diantara semua perlakuan, sementara itu kelompok perlakuan yang baik hanya diberikan penambahan kurkumin, maupun kombinasi penambahan kurkumin dan tiroksin cenderung memiliki nilai derajat pembuahan dan penetasan yang lebih tinggi dibandingkan dengan kelompok perlakuan yang tidak diberikan penambahan kurkumin.  The main problem in aquaculture is the availability of fish seedlings on time, in the right quantity and quality. The availability of fish seeds is determined by the number of eggs that can be fertilized and hatched. The higher the percentage of fertilization and hatching, the higher the number of larvae that can be produced. This study uses catfish as experimental animals because catfish are one of the most consumed cultivation commodities. The aim of this study is to determine the effect of curcumin and thyroxine supplementation on catfish brooders on the quality and performance of eggs produced. Catfish used in this experiment were given the addition of curcumin and thyroxine hormone through the feed for 12 weeks of rearing. Parameters analyzed were egg vitellogenin concentration, egg diameter, coefficient of diversity in egg diameter, lipid concentration in eggs (triglycerides, cholesterol, and HDL), degree of fertilization and degree of hatching. The results showed that the group given the combination of the supplementation curcumin and the thyroxine hormone had the values of vitellogenin concentration (8.17+2.74 µg/mL) (p <0.05), egg diameter (1.43+0.00 mm) (p <0.05), and triglyceride concentration (4.89+0.53 mg/g) (p <0.05) the highest among all treatments, meanwhile the group is only given the addition of curcumin, and the combination of supplementation of curcumin and thyroxine tends to have higher values of fertilization and hatching compared to the group which is not given the addition of curcumin.

Abstract

Synthetic steroids are commonly used to sex-reverse tilapia to produce male tilapia commercially, but gradually feared a negative impact on food security and environmental sustainability. The use of new natural compound is a potential alternative to be explored, one of which is bull testes meal extract (BTME). This study aimed to evaluate the effectiveness of BTME immersion and optimal concentrations for sex-reverse tilapia to produce monosex male tilapia, and its influence on survival and growth of fish. The study consisted of 5 treatments with 3 replications: immersion treatment with BTME concentration of 1 ml l-1 (A), 3 ml l-1 (B), 5 ml l-1 (C), immersion treatment in 17α-methyltestosterone of 500 µg l-1 (K+) and without hormone immersion treatment (K-). Hormone immersion was performed on tilapia larvae aged 4 and 7 days after hatching.The fish were then maintained for 60 days. The parameters observed were: the percentage of male tilapia and intersex tilapia, survival rates, and growth rate of fish. BTME immersion for sex-reverse the tilapia larvae have significant effect on masculinization of tilapia. The highest percentage of male fish was obtained on treatment C (85.56%) and was not significantly different from K (+) (81.11 %). Immersion of tilapia in BTME or 17α-MT did not affect the survival and growth of tilapia fish.Keywords: sex reversal, bull testes meal extract (BTME), tilapia

Abstract

The objective of the experiment was to describe the influence of estradiol (E2) and methyltestosterone (MT) in combination with hCG and anti-dopamine (AD) on the percentage of female and quality of ovary development of Indonesian short-finned eel (Anguilla bicolor bicolor). Eel with 200±15 g of body weight in the yellow phase injected with a estradiol solution of 3mg mL-1 + hCG 2mg mL-1 + AD 10 mg mL-1 (hEA), MT 3 mg mL-1 + hCG 2 mg mL-1 + AD 10 mg mL-1 (hTA), estradiol 3 mg mL-1 + AD 10 mg mL-1 (EA), MT 3 mg mL-1 + AD 10 mg mL-1 (TA), AD 10 mg mL-1 (A) and 9 mg mL-1 NaCl control (F). Injection frequency was six times with a period of two weeks. After six weeks of experiment, EA and hTA combination hormone treatments were effective to increase the female percentage 72% and 66%, respectively. The best ovary development was found in the hTA treatment i.e. attain the late-vitellogenesis with GSI value of 4.80%. Abstrak Penelitian ini bertujuan untuk menjelaskan pengaruh estradiol (E2) dan metil testosteron (MT) yang dikombinasikan dengan hCG dan anti dopamin (AD) terhadap persentase jenis kelamin betina dan kualitas perkembangan ovari sidat (Anguilla bicolor bicolor). Sidat dengan bobot 200±15g pada faseyellow diinjeksi dengan larutan estradiol 3 mg mL-1 + hCG, 2 mg mL-1 + AD 10 mg mL-1 (hEA), MT 3 mg mL-1 + hCG 2 mg mL-1 + AD 10 mg mL-1 (hTA), estradiol 3 mg mL-1 + AD 10 mg mL-1 (EA), MT 3 mg mL-1 + AD 10 mg mL-1 (TA), AD 10 mg mL-1 (A), dan kontrol 9 mg mL-1 NaCl (F). Frekuensi injeksi sebanyak enam kali dengan periode dua minggu sekali. Pemberian hormon kombinasi dengan perlakuan hEA dan hTA ternyata efektif meningkatkan persentase jenis kelamin betina berturut-turut sebesar 72% dan 66% setelah enam minggu. Perkembangan gonad betina terbaik ditemukan pada perlakuan hTA yaitu mencapai fase vitellogenesis akhir dengan nilai GSI 4,80%

Abstract

The objective of this study was to compare the growth response of 3 different sizes of humpback grouper juveniles, which were come from the same brood stock group and spawning season after “stop and go” experiment of recombinant Epinephelus lanceolatus growth hormone (rElGH); that are with, without and retreated with rElGH for 42 days respectively. Each size group was divided into two treatment groups. The first group was treated with 50 mg crude rElGH kg-1 in commercial diet (pC) and a second group as a control. Weight gain of pC compares to control for small size group, medium size group and large size groups of juveniles subsequently for first stage were 85.89%, 39.66% and 16.34%; second stage were -34.57%, -14.76%, and -5.27%; and third stage were 56.16%, 50.24% and 59.14%. Specific growth rate differences of small, medium and large size of pC compared to control in first stage were 41.6%, 19.06% and 7.52%; second stage were -44.81%, -27.23% and -14.66%; and third stage were 55.9%, 40.62% and 48.42%. No significant difference of condition factor among all sizes of pC and control fish. Protein content and retention, and liver glycogen content from pooled sample of all size fish groups pC treatment in the second stage were decreasing compared to the first stage, respectively, 11.49%, 35.14% and 84.73%. It can be concluded that rElGH treatment improved growth performance of all size fish groups, however small juveniles have highest growth response compared to medium and large juvenile groups. The ceasing of rElGH treatment on second experiment stage is most likely causing the loss of accelerating growth factor then decreasing growth performance, protein content and retention, and liver glycogen content. Abstrak Penelitian bertujuan membandingkan respons pertumbuhan tiga kelompok ukuran benih ikan kerapu bebek dari kelompok induk dan periode pemijahan yang sama terhadap hormon pertumbuhan rekombinan ikan kerapu kertang Epine-phelus lanceolatus (rElGH); melalui eksperimen “putus dan sambung” yaitu dengan, tanpa, dan perlakuan kembali rElGH masing-masing selama 42 hari. Setiap kelompok ukuran dibagi menjadi dua kelompok perlakuan, kelompok pertama diberi perlakuan rElGH dengan dosis 50 mg rElGH-HP55 kg-1 pakan (pC) sedangkan kelompok kedua sebagai kontrol. Pertambahan bobot badan kelompok pC dibandingkan dengan kontrol pada benih berukuran kecil, sedang dan besar berturut-turut pada eksperimen tahap pertama 85,89%, 39,66% dan 16,34%; tahap kedua -34,57%, -14,76%, dan -5,27%, dan tahap ketiga 56,16%, 50,24% dan 59,14%. Perbedaan laju pertumbuhan spesifik benih berukuran kecil, se-dang dan besar perlakuan pC terhadap kontrol pada eksperimen tahap pertama 41,6%, 19,06% dan 7,52%; tahap kedua -44,81%, -27,23% dan -14,66%; dan tahap ketiga 55,9%, 40,62% dan 48,42%. Faktor kondisi pC dan kontrol pada se-mua kelompok ukuran tidak berbeda nyata. Kandungan dan retensi protein, dan kandungan glikogen hati gabungan sampel dari semua kelompok ukuran ikan perlakuan pC pada eksperimen tahap kedua menurun dibandingkan eksperi-men tahap pertama, masing-masing sebesar 11,49%, 35,14% dan 84,73%. Dapat disimpulkan pemberian rElGH mema-cu pertumbuhan semua kelompok ukuran benih ikan, namun benih berukuran kecil mempunyai respons pertumbuhan lebih tinggi daripada kelompok benih berukuran sedang dan besar. Penghentian pemberian rElGH menyebabkan ber-hentinya faktor pemacu pertumbuhan, sehingga performa pertumbuhan, kandungan dan retensi protein, dan kandungan glikogen hati menurun.

Abstract

Good sperm quality and inactive condition during the electroporation process determines the success of transgenesis. The study aims to observe the morphology and physiology of sperm, and evaluate the effects of different voltage and number of electric shocks on motility and viability of climbing perch sperm. Observation of morphological characters by microscope with a magnification of 400X, physiological character with various parameters, such as the level of os-molarity, volume, pH, density and total of sperm. The climbing perch sperm consisted of three parts, namely the ovoid head, middle, and tail. Sperm viability in the water was 116±34 seconds. Sperm quantity, volume of semen, pH, and sperm density were increased after ovaprim injection (p<0.05). Osmolarity levels of body and testes of climbing perch ranged at 390-405 mOsmol kg-1. Sperm diluted in 13 g L-1 NaCl solution could be kept for a maximum of 60 minutes at room temperature (26°C) and cold temperature (4°C), while without NaCl solution could be 360 minutes. Fertilization and hatching rates of eegs fertilized by sperm form the minced testes, and larval survival up to 10 days post-hatching were similar with semi-natural spawning. Electroporation of climbing perch sperm diluted in 13 g/L NaCl solution was successfully performed using a program with a long shock of 0.5 milliseconds and the interval shock of 0.1 seconds. Increasing the voltage and number of shock decreased motility and viability of sperm (p<0.05). Those electric shock parameters can potentially be used to generate climbing perch transgenic by electroporation method. Abstrak Kualitas sperma yang baik dan dalam keadaan tak aktif selama proses elektroporasi sangat menentukan keberhasilan transgenesis. Penelitian ini bertujuan untuk mengeksplorasi karakter morfologis dan fisiologis sel sperma, serta menge-valuasi efek voltase dan jumlah kejut listrik berbeda terhadap motilitas dan viabilitas sperma ikan betok. Pengamatan karakter morfologi dilakukan pada mikroskop dengan pembesaran 400x, karakter fisiologi dengan berbagai parameter yakni tingkat osmolaritas, volume, pH, kepadatan dan total sperma. Hasil penelitian menunjukkan bahwa sperma ikan betok terdiri atas tiga bagian, yakni kepala yang berbentuk oval, bagian tengah, dan ekor sperma. Viabilitas sperma ikan betok dalam air adalah selama 116±34 detik. Induksi ovaprim dapat meningkatkan volume sperma, pH, kepadatan dan total sperma (p<0,05). Tingkat osmolaritas tubuh dan testis ikan betok (390-405 mOsmol kg-1). Penyimpanan sperma yang diencerkan dengan larutan NaCl 1,3% hanya dapat bertahan maksimum selama 60 menit pada suhu ruang (26°C) dan suhu dingin (4°C), sedangkan tanpa pengenceran dapat bertahan selama 360 menit. Derajat pembuahan dan kelangsungan hidup larva hingga 10 hari pascatetas menggunakan sperma dari testes cacah sama dengan pemijahan semi alami. Elektroporasi sperma ikan betok dengan pengencer larutan NaCl 1,3% berhasil dilakukan menggunakan lama kejut 0,5 milidetik, dan jeda antar kejut 0,1 detik. Peningkatan voltase dan jumlah kejut yang diberikan saling berinter-aksi menurunkan motilitas dan viabilitas sperma (p<0,05). Metode kejut listrik tersebut berpotensi digunakan untuk menghasilkan ikan betok transgenik dengan metode elektroporasi.

Abstract

This study aims to analyze the hormone profile of FSH, LH and E2 to the eel after exogenous hormone stimulation and blood glucose levels. Six formulated treatment applied with combination of dopamine antagonize 10 mg.mL–1 (A), estradiol (E2) 3 mg.mL–1 + A (EA), metyltestosteron (MT) 3 mg.mL–1 + A (MTA), hCG 2 mg.mL–1 + EA (hEA) and hCG 2 mg.mL–1 + MTA (hMTA), with (F) physiologis 0.9% NaCl to control. Six group eel (200±15g) reared in a concrete tank with a capacity of 3,400 liters and filled with sea water of 35 mg L–1 as much as 2000 liters. Eels injected 1 mL.kg–1 hormone by intramusculary, were feed to apparent satiation daily for 10 weeks. The study used Completely Randomized Design with one treatment factor, namely hormonal factor and its combination. Fish blood that ware directly concentration to FSH, LH and E2, the enhanced significantly high in the blood plasma on treatment hMTA and hEA P<0.05. Glucose concentration in the blood palsma is high enough in a row on a formula hMTA 67.33 mg.dL–1 and significantly different to P<0.05. The result indicates that induction of exogenous hormone (hMTA) improve FSH, LH and E2. FSH and LH profiles show permutation patterns during the development of eel gonad, beginning with the increase of FSH in the early phases of gonadal development. LH profile moves in line with the increase in E2 during gonadal maturation process eels (A. bicolor bicolor), with blood glucose levels in the normal range. Abstrak Penelitian ini bertujuan menganalisis profil hormon FSH, LH dan E2 ikan sidat yang dirangsang dengan hormon ekso-genous serta kadar glukosa darah. Enam perlakuan diterapkan dengan kombinasi anti dopamin 10 mg.mL–1 (A), estra-diol (E2) 3 mg.mL–1 + A (EA), metyltestosteron (MT) 3 mg.mL–1 + A (MTA), hormon hCG 2 mg.mL–1 + EA (hEA) dan hCG 2 mg.mL–1 + MTA (hMTA), dengan kontrol (F) fisiologis 0,9% NaCl. Enam kelompok sidat uji (200±15 g) dipelihara pada bak beton berkapasitas 3.400 liter dan di isi dengan air laut bersalinitas 35 mg L–1 sebanyak 2000 liter. Sidat diinjeksi dengan dosis hormon 1 mL.kg–1 secara intramuskular, hewan uji diberi pakan secara at satiation sekali sehari selama 10 minggu. Penelitian menggunakan Rancangan Acak Lengkap dengan satu faktor perlakuan, yaitu faktor hormon dan kombinasinya. Darah sidat terbukti mengandung FSH, LH, E2, peningkatannya terlihat nyata terjadi mela-lui plasma darah pada perlakuan hMTA dan hEA pada tingkat P<0,05. Konsentrasi glukosa tertinggi pada formula hMTA 67,33 mg.dL–1 dan berbeda nyata pada taraf P<0,05. Hasil ini menunjukkan bahwa penambahan hormon ekso-genous perlakuan hMTA meningkatkan konsentrasi FSH, LH, dan E2. Profil FSH dan LH menunjukkan pola permutasi selama proses perkembangan gonad sidat, di mulai dengan meningkatnya FSH di fase awal perkembangan gonad. Profil LH bergerak seiring dengan peningkatan E2 selama proses pematangan gonad sidat (A. bicolor bicolor), dengan kadar glukosa darah pada kisaran normal.
