Abstract

ABSTRACTThis experiment was carried out in order to study the effect of thyroxine hormone addition at different stages of giant gouramy larvae on their development, growth, and survival rate.  Treatments were conducted by dipping giant gouramy larvae at different stages (1, 5, and 1 0 days after hatching) in 0. 1 ppm thyroxine hormone solution for 24 hours.  The result of this experiment showed that dipping of giant gouramy larvae at different stages in thyroxine hormone solution did not affect their development, growth, and survival rate.Key words :  Giant gouramy larvae, thyroxine, development, growth, survival rate ABSTRAKPercobaan ini dilakukan untuk mengetahui pengaruh perbedaan umur larva ikan gurami yang direndam dalam hormon tiroksin terhadap perkembangan, pertumbuhan, dan kelangsungan hidup.  Larva ikan gurami yang digunakan diperoleh dari hasil penetasan dan kemudian dipelihara sebagai stok.  Periakuan dilakukan melalui perendaman larva ikan gurami pada umur yang berbeda (1, 5, dan 10 hari setelah menetas) dalam larutan tiroksin 0,1 ppm selama 24 jam.  Hasil percobaan menujukkan bahwa setelah 56 hari pemeliharaan, pemberian tiroksin yang dilakukan tidak berpengaruh nyata terhadap perkembangan, pertumbuhan, dan kelangsungan hidup ikan gurami.Kata kunci :  Larva ikan gurami, tiroksin, perkembangan, pertumbuhan, kelangsungan hidup

Abstract

ABSTRACTThis experiment was carried out to study the effect of female broodstocks immersion in 17amethyltestosterone (MT) solution on sex ratio of their offspring, Three-months old males and females were paired to mate in aquaria for four days.  Broodstocks were fed with frozen blood worm and water flea 2-3 tii-nes daily.  Twelve days after mating, female broodstock were treated by immersing in 2 mg/1 MT solution for 0. 6, 12, 24 and 48 hours.  Birthed guppy babies were collected and reared separately from their parents.  During the rearing period, the babies were fed with artemia nauplius and silkworm until identification for male and female.  Percentage of female offspring in control group were higher than those of male.  Exposure of pregnant females to MT solution for 0, 6, 12, 24, and 48 hours resulted in 42, 1'/o, 51%, 84,6%, 1 00%, dan 100% of males offspring, respectively.  The best result was obtained from 24 hours treatment.  Longer treatment duration tend to shorten time interval between treatment and birth. Key words :  Guppy, 17(x-methyltestosterone, exposure time, broodstock immersion, sex ratio   ABSTRAK Penefitian ini bertuiuan untuk mengetahui pengaruh lama waktu perendaman induk di dalam larutan hormon 17a-metiltestosteron (MT) terhadap nisbah kelamin ikan gapi.  Induk Ikan gapi berumur tiga bulan dikawinkan berpasangan di dalam akuarium selama empat hari Induk diberi pakan cuk merah beku dan kutu air dengan frekuensi 2-3 kali sehari.  Dua belas hari setelah masa perkawinan, induk betina diberi perlakuan berupa perendaman di dalam larutan MT 2 mg/1 selama 0 (kontrol), 6, 12, 24, dan 48 jam.  Anak-anak ikan gapi yang baru lahir dipelihara terpisah dari induknya.  Selama masa pemeliharaan, anak ikan gapi diberi pakan nauplius artemia dan cacing rambut.  Pemeliharaan berlangsung sampai jenis kelamin anak ikan gapi dapat diidentifikasi.  Persentase anak ikan gapi betina pada semua ulangan kontrol lebih tinggi daripada .iantan dengan perbandingan persentase rata-rata 57,9%: 42,1%.  Adapun pada lama waktu perendaman 0, 6, 12, 24, dan 48 jam, menghasilkan persentase anak ikan gapi berfenotip jantan berturut-turut sebesar 42,1%, 51%, 84,6%, 100%, dan 100%.  Lama waktu perendaman terbaik adalah 24 jam.  Selain itu terdapat kecenderungan bahwa semakin lama waktu perendaman semakin cepat kelahirannya. Abstract

ABSTRACTThe objectives of this research were carried out to determine exposure time of giant gouramy larvae in triiodothyronine (T3) hormone solution on development, growth and survival rate. One-day old larvae were immersed in 0,1 ppm T3 hormone solution for 0, 2, 4, 6, 8, 16, and 24 hours. Results showed that treated larvae developed faster than control larvae. At first, second, fifth, sixth and seventh week, larvae that immersed in T3 hormon solution had total length longer than that of control. Treated larvae had average body weight heavier than that of control until seven weeks of experiment. Immersion of larvae for 16 hours gave the best result in term of length and average body weight. The best survival rate of larvae were obtained from 8 hours treatment.Key words :  Giant gouramy larvae, triiodothyronine, exposure time, growth and survival rate.    ABSTRAKPenelitian ini dilakukan untuk menentukan lama perendaman di dalam larutan hormon triiodotironin (T3) terhadap perkembangan, pertumbuhan dan kelangsungan hidup larva ikan gurame. Larva ikan yang berumur satu hari direndam dalam larutan hormon triiodotironin 0,1 ppm dengan lama perendaman  0, 2, 4, 6, 8, 16, dan 24 jam. Perkembangan larva yang diberi perlakuan T3 lebih cepat daripada perkembangan larva kontrol. Larva yang direndam dalam larutan hormon T3 lebih panjang daripada kontrol pada minggu ke-5, 6 dan 7. Bobot rata-rata larva perlakuan lebih besar daripada bobot rata-rata kontrol dari minggu awal sampai minggu ke-7. Perendaman larva selama 16 jam memberikan hasil terbaik dari segi panjang total dan bobot rata-rata. Nilai kelangsungan hidup terbaik di akhir penelitian diperoleh pada perendaman selama 8 jam. At the end of experiment, there was no significant difference in yolk sac absorption between treated larvae and control. Abstract

Thyroxine is known as a hormone that can affect growth of fish by increasing the metabolic rate, feed efficiency and protein retention.  In this study, effect of thyroxine administration through feed on growth and survival of coral platy fish (Xiphophorus maculatus) was observed.  Dosage of thyroxine used was 0.2, 2 and 20 mg/kg of feed, and were combined with duration time of treatment i.e. 1, 2 and 3 weeks.  All treatments were performed triplicate.  The results of study show that higher growth in length (21.3 mm) and weight (244.5 mg) was obtained by administration of 20 mg thyroxine per kilogram of feed for two weeks and this treatment had no effect on the survival rate of coral platy fish Keywords: coral platy, Xiphophorus maculatus, tyroxine, growth   ABSTRAK Tiroksin diketahui sebagai hormon yang dapat mempengaruhi pertumbuhan dengan jalan meningkatkan laju metabolisme tubuh, efisiensi makanan dan retensi protein. Pada penelitian ini dipelajari pengaruh pemberian hormon tiroksin melalui pakan terhadap pertumbuhan dan kelangsungan hidup ikan plati koral (Xiphophorus maculatus). Dosis tiroksin yang digunakan yaitu 0,2; 2 dan 20 mg/kg pakan, dan dikombinasikan dengan lama waktu perlakuan satu, dua dan tiga pekan. Semua perlakuan diulang tiga kali. Hasil penelitian menunjukkan bahwa pemberian hormon tiroksin sebesar 20 mg/kg pakan selama dua pekan memberikan pertumbuhan panjang dan pertambahan berat terbaik masing-masing 21,3 mm dan 244,5 mg dan tidak berpengaruh terhadap kelangsungan hidup ikan platy koral. Abstract

This study was carried out to determine the effect of induction using common carp as a trigger on spawning of Puntius javanicus.  Mature male and female of Puntius javanicus were reared in outer of the hapa that contains mature male common carp, mature female common carp, or mature male and female common carp as a control.  Common carp broodstock was injected or not injected by ovaprim.  The result of study show that the use of male common carp injected or not injected by ovaprim could induced spawning of Puntius javanicus, without the release of sperm and eggs of common carp.   Fertilization rate of Puntius javanicus eggs was high, reached of 91.4%, by using ovaprim-injected male common carp as trigger. Keywords: Puntius javanicus, common carp, Cyprinus carpio, spawning, induction system   ABSTRAK Penelitian ini dilakukan untuk mengetahui pengaruh sistem imbas menggunakan ikan mas (Cyprinus carpio) sebagai pemicu terhadap pemijahan ikan tawes (Puntius javanicus). Induk ikan tawes jantan dan betina yang telah matang gonad ditempatkan di luar hapa tempat ikan mas jantan matang gonad, ikan mas betina matang gonad, atau sepasang ikan mas matang gonad sebagai perlakuan kontrol. Induk ikan mas disuntik atau tidak dengan ovaprim.  Hasil penelitian menunjukkan bahwa penggunaan ikan mas yang disuntik ovaprim maupun tidak, dapat mengimbas ikan tawes untuk memijah meskipun tidak terjadi pengeluaran sperma dan telur ikan mas. Derajat pembuahan telur ikan tawes cukup tinggi, mencapai 91,4%, pada perlakuan induksi ikan mas jantan yang disuntik ovaprim. Abstract

This experiment was conducted to determine the dietary Vitamin E requirement for reproduction of broodstock zebrafish Danio rerio.  Four isonitrogenous (39% crude protein) and isocaloric (3,260 kcal digestible energy/kg diet) practical diets, namely diets A, B, C, and D with different levels of Vitamin E were fed to zebrafish broodstock.  The broodstock were cultivated in aquaria.  Diet A contained low dosage of Vitamin E (5 mg Vitamin E /kg diet), while diets B (62 mg Vitamin E /kg diet), C (125 mg Vitamin E /kg diet), and diet D (187 mg Vitamin E /kg diet), combined respectively with 1% n-3 fatty acids and 2% n-6 fatty acids.  Fish were fed ad satiation for 60 days using these diets.  During feeding period, gonad maturation stages were examined.  The dietary with different level of Vitamin E affected egg size, chemical content, total number of larvae, normal larvae, and survival rate of larvae produced, fecundity, fertilization rate, and hatching rate.  On the other hand, fish fed on experimental diets did not show any significance differences in the gonad somatic index, fecundity, fertilization rate, and hatching rate.  Fish fed on diet D produced the highest fecundity (633.65 ± 71.86 eggs/grams of fish), fertilization rate (92.3 ± 7.7%), and hatching rate (80.31 ± 9.8%).  The total lipid content of eggs were significant, ranging from D (39.7%), C (33.5%), B (32.1%), and A (29.5%).  At a dosage of 1% of dietary n-3 fatty acids and 2% n-6 fatty acids, zebrafish require 187 mg Vitamin E/kg feed in the diet for reproduction.  . Keywords:  vitamin E, reproductive performance, Danio rerio   ABSTRAK Penelitian ini ditujukan untuk mengetahui kebutuhan Vitamin E pada pakan induk ikan zebra (Danio rerio) untuk reproduksi.  Empat pakan perlakuan yang iso-protein (39%) dan iso-kalori (3.260 kcal digestible energy/kg pakan), dinamakan pakan A, B, C, dan pakan D dengan kandungan Vitamin E yang berbeda diberikan kepada induk ikan zebra.  Induk ikan dipelihara pada akuarium.  Pakan A mengandung Vitamin E terendah (5 mg Vitamin E/kg pakan), sedangkan pakan B mengandung Vitamin E 62 mg Vitamin E/kg pakan, pakan C asam mengandung Vitamin E 125 mg /kg pakan, dan pakan D mengandung Vitamin E 187 mg /kg pakan.  Semua pakan perlakuan mempunyai kombinasi asam lemak n-3 berbanding n-6 sebesar 1:2.  Ikan diberi pakan secara ad satiation selama 60 hari pemeliharaan.  Selama masa pemberian pakan, tingkat kematangan gonad diperiksa secara teratur.  Perbedaan kandungan vitamin E dalam pakan mempengaruhi fekunditas, derajat pembuahan telur, serta derajat penetasan telur.  Pakan perlakuan tidak mempengaruhi nilai gonad somatik indeks dan kelangsungan hidup larva umur 3 hari.  Pemberian pakan D menghasilkan fekunditas (633.65 ± 71.86  telur/gram induk), derajat pembuahan telur (92.3 ± 7.7%), serta derajat penetasan telur (80.31 ± 9,8%) terbaik.  Total kandungan lemak pada telur berbeda nyata, yaitu pada pakan D (39.7%), C(33.5%), B(32.1%), dan pakan A(29.5%).  Pada kandungan 1% asam lemak n-3 dan 2% asam lemak n-6, ikan zebra membutuhkan 187 mg Vitamin E/kg pakan untuk reproduksi yang normal.  Kata kunci:  Vitamin E, penampilan reproduksi, Danio rerio

Abstract

Uncontrolled and excessive spawning of red tilapia (Oreochromis sp.) hinders fish growth to reach marketable size.  Several techniques have been developed to produce monosex population to overcome the problem, such as sex reversal by hormonal administration.  Androgen hormone is generally employed to meet the goal, although the use of this hormone had been restricted and its price is relatively high. Acriflavine is an alternative chemical and its effect on sex development toward male fish has been proved.  In this study, acriflavine at dose of 0, 15, 25 and 35 mg/kg feed were fed to 10-day-old larvae for 6 weeks.  Feeding of larvae by diet containing 17a-methyltestosterone (MT) at dose of 50 mg/kg was also observed as a comparison of acriflavin treatment.  The results indicated that feeding of larvae by diet containing acriflavine 25 mg/kg (78.3%) and 35 mg/kg (79.0%) produced significantly higher percentage of male fish compared to 15 mg/kg and control.  However, the result is still lower compared to that of MT treatment (99.2%).  Acriflavine treatment had no effect on survival of red tilapia and survival rate was similar among treatments, ranged from 91.7% to 95.0%. Keywords: red tilapia, Orechromis sp., monosex,  acriflavine   ABSTRAK Pemijahan tak terkontrol dan berlebih pada ikan nila merah (Oreochromis sp.) menghambat pertumbuhan ikan untuk mencapai ukuran konsumsi. Beberapa teknik yang telah dikembangkan untuk menghasilkan ikan monosek sebagai solusi permasalahan tersebut antara lain dengan seks reversal menggunakan hormon. Secara umum, hormon golongan androgen digunakan untuk mencapai tujuan, namun penggunaannya mulai dibatasi dan harganya relatif mahal. Akriflavin merupakan bahan alternatif yang terbukti dapat mempengaruhi perkembangan kelamin ke arah jantan.  Pada penelitian ini dilakukan pemberian akriflavin dengan dosis 0, 15, 25 dan 35 mg/kg pakan ke larva umur 10 hari selama 6 minggu. Sebagai pembanding juga dilakukan pemberian pakan yang mengandung hormon 17a-metiltestosteron dosis 50 mg/kg pakan.  Hasil penelitian menunjukkan bahwa pemberian pakan yang mengandung akriflavin 25 mg/kg pakan (78,3%) dan 35 mg/kg pakan (79,0%) menghasilkan ikan jantan dengan persentase lebih banyak dibandingkan dengan perlakuan 15 mg/kg (72,46%) dan kontrol (58,37%).   Namun demikian, hasil tersebut masih lebih rendah dibandingkan dengan perlakuan MT (99,2%).  Perlakuan akriflavin tidak mempengaruhi kelangsungan hidup ikan nila merah dan tidak berbeda  nyata dengan kontrol, berkisar antara 91,7-95,0%. Abstract

Attempt to produce a lot number of Tor soro is difficult if only obtained by natural spawning since they often get mature in dissimilar time. Stock of sperm kept in enough number for fertilization in anytime is needed to overcome the problem.  Cryopreservation of semen is one of sperm cryopreservation methods using liquid nitrogen (-196oC) to quickly freeze the sperm. Several cryoprotectants can be utilized in cryopreservation.  In this study, quality by means of motility of cryopreserved sperm using dimethylsulfoxide or glycerol as cryoprotectant in the dose of 5, 10 and 15% were analyzed. The result of study show that percentage of sperm motile after cryopreservation using dimethylsulfoxide and glycerol was similar ranged from 76.7 to 83.3%.  Therefore, both cryoprotectant used in this study could be employed for cryopreservation of Tor soro sperm. Keywords: cryopreservation, dimethylsulfoxide, glycerol, sperm, Tor soro   ABSTRAK Upaya produksi ikan batak (Tor soro) dalam jumlah besar relatif sulit jika hanya mengandalkan hasil pemijahan secara alami karena kematangan gonad antara ikan jantan dan betina sering tidak bersamaan. Untuk mengatasinya diperlukan stok sperma yang disimpan dalam jumlah yang cukup banyak untuk pembuahan yang dapat dipakai setiap saat. Kriopreservasi semen merupakan salah satu cara penyimpanan sperma yang menggunakan larutan nitrogen cair (-196°C) untuk membekukan sperma secara cepat. Beberapa jenis protektan dapat digunakan dalam pembekuan sperma. Pada penelitian ini, dilakukan analisis kualitas dalam arti motilitas sperma hasil kriopreservasi menggunakan dimetilsulfoksida dan gliserol sebagai krioprotektan dengan dosis 5, 10 dan 15%.  Hasil penelitian menunjukkan bahwa persentase sperma motil setelah dilakukan kriopreservasi adalah sama, berkisar antara 76,7% hingga 83,35%.  Dengan demikian, kedua jenis krioprotektan yang digunakan dalam penelitian ini bisa dipakai dalam kriopreservasi sperma ikan batak. One of methods could be applied to continuously meet the need of fish fry including catfish (Clarias sp.) is artificial propagation by inducing ovulation and spawning.   As an alternative of existing method, in this study, ginger extract was intramuscularly injected to induce development of catfish eggs.  Ginger is known as an important regulator of the balance of arachidonat cycle.  The dose of ginger extract injected was 0, 0.5, 1.0 and 1.5 mL/kg broodstock.  The results of study showed that injection 100% of ginger extracts in all doses was insignificantly inducing development of egg diameter and its nucleus position of catfish.  Other chemicals exist in ginger extract might be functions as an obstacle for egg development of catfish. Keywords: catfish, Clarias sp., ginger, ovulation, egg   ABSTRAK Salah satu cara untuk memenuhi kebutuhan benih ikan termasuk lele (Clarias sp.) secara kontinyu adalah penggunaan teknologi pembiakan buatan melalui perangsangan ovulasi dan pemijahan. Sebagai alternatif teknik yang sudah ada, dicobakan perangsangan perkembangan telur ikan lele menggunakan bahan ekstrak jahe yang dilakukan melalui penyuntikan secara intramuskular. Jahe telah dikenal sebagai suatu pengatur penting atas keseimbangan siklus arakidonat. Dosis ekstrak jahe yang disuntikkan adalah 0, 0,5, 1 dan 1,5 mL/kg induk. Penyuntikan 100% ekstrak jahe pada semua dosis perlakuan belum dapat merangsang perkembangan diameter dan posisi inti sel telur ikan lele sangkuriang. Adanya bahan lain yang terdapat pada jahe diduga sebagai penghambat bagi perkembangan telur ikan lele. Kata kunci: Ikan lele, Clarias sp., jahe, ovulasi,  sel telur
