Abstract

This study aims to determine the immunity response of  Carassius auratus infected with Aeromonas hydrophila bacteria and post-treatment with propolis solution by measuring total leukocytes and phagocytosis index. The method used is experimental by applying a completely randomized design (CRD) with five treatments and three replications. The treatments were Kn (not infected with A. hydrophila and not treated with propolis), Kp (infected with A. hydrophila but not treated with propolis), while fish infected with A. hydrophila were treated with propolis doses P1 (700 ppm), P2 (800 ppm), and P3 (900 ppm). Treatment was done by injecting 0.1 ml of propolis into fish infected with A. hydrophila intramuscularly. The test material was 150 fish of 8-10 cm in size. The results showed that propolis significantly gave an immune response to the C. auratus infected with A. hydrophila (p<0.05). The 800 ppm propolis dose was the most effective, as indicated by a total leukocyte count of 3.70×104 cells/mm3, a phagocytosis index value of 28.33%, and a survival rate of 83.33%. This study highlights that propolis has potential as a natural immunostimulatory agent in enhancing non-specific defense mechanisms in C. auratus infected with pathogenic bacteria

Abstract

Antimicrobial resistance has become one of the most serious global health threats, prompting the search for new natural sources of antibiotics. Marine biota, such as algae, sponges, and bacteria, are known to produce a variety of secondary metabolites with potential biological activity, particularly as antimicrobial compounds. This review article examines the bioactive potential of these marine biota as an alternative source of natural antibacterial compounds. The study was conducted by reviewing various international and national literature related to the bioactive metabolites produced, their mechanisms of action against pathogenic bacteria, and the potential for their use in developing new antibacterial agents. The results of the review indicate that sulfated polysaccharides, terpenoids, alkaloids, and phenolic compounds from algae; alkaloids, cyclic peptides, and terpenoids from sponges; and secondary metabolites of marine symbiont bacteria have significant antibacterial activity, including against antibiotic-resistant bacteria. However, the development of marine bioactive compounds still faces various obstacles, particularly related to isolation techniques, mass production, and environmental sustainability issues. Therefore, modern biotechnological approaches, such as genomics, metabolomics, and synthetic biology, are needed to optimize the utilization of these marine resources. This article confirms that marine biota are important candidates for the development of new, sustainable antibacterial agents and have the potential to support the strengthening of the blue economy concept.

Abstract

Epiphytic macroalgae are phototrophic organisms that attach to rigid substrates in the intertidal zone and serve as key components of coastal ecosystem structure due to their roles in providing habitat, stabilizing substrates, and supporting nutrient cycling. This study aimed to inventory the types of epiphytic macroalgae present in the rocky intertidal zone of Padang Unoi Village, Salang District, Simeulue Regency. Observations were conducted in July 2025 along a 150-m transect, dividing the area into two tidal zones: the lower littoral and the mid-littoral. The inventory revealed seven species of epiphytic macroalgae, namely Turbinaria sp, Sargassum sp, Padina sp, Chaetomorpha aerea, Caulerpa taxifolia, C. racemosa, and Amphiroa subcylindrica. Species from the Phaeophyta division dominated the mid-littoral zone, while Chlorophyta were more abundant in the lower littoral zone, and Rhodophyta were observed on coarse-textured rocky substrates. These three major divisions indicate habitat heterogeneity that supports the growth of various types of epiphytic macroalgae. The results of this study highlight the importance of the rocky intertidal zone of Simeulue as a productive habitat and provide a basis for sustainable coastal resource management

Abstract

Chaetomorpha sp is a type of green marcoalgae that is rich in bioactive compound that function as antibacterial, anti-inflammatory and can increase fish immunity. This study aims to determine the immunostimulating effect of Chaetomorpha sp macroalgae extract on the leukocyte profile of Tilapia infected with A. hydrophila. The experimental method used a completely randomized design (CRD) with four extract dose treatments (0, 25, 50, 75 mL/kg feed) and three replicates. The test fish used were 2.63 0.26g, reared in black tanks with a size of 60x30x30 cm, a volume of 80 L with a stocking density of 1 fish/4 L, and a recirculation system. Maintenance was carried out for 60 days with a frequency of feeding three times a day, namely 08.00 AM, 01.00 PM, and 05.00 PM, as much as 5% of body weight. The parameters observed were total leukocytes, leukocyte differentiation, and phagocytosis index. The results showed that the administration of Chaetomorpha sp extract significantly increased total leukocytes, lymphocyte differentiation, and phagocytosis index (p0.05). The dose of 50 mL/kg feed is the optimal dose that affects the leukocyte profile of Tilapia. T0 (without extract) experienced 100% mortality, while the treatment group showed increased resistance to infection. Bioactive compounds such as sulfated polysaccharides and flavonoids in Chaetomorpha sp are thought to play a role in stimulating the innate immune system. This study highlights the potential of Chaetomorpha sp as an alternative to antibiotics in sustainable tilapia aquaculture.

Abstract

Carp (Cyprinus carpio) is one of the freshwater fishery commodities with high economic value. Keeping carp in saline media can affect the osmoregulation system, impacting the performance of vital organs such as the intestines and kidneys. Such disturbances can potentially reduce fish's health and survival rate. One strategy that can be used to increase the fish's immune system is through feed supplementation with natural immunostimulants, one of which is guava leaf flour (Psidium guajava), which is known to contain bioactive compounds such as flavonoids, tannins, and saponins. This study aims to evaluate the effectiveness of guava leaf flour-enriched feed on the histological structure of the intestines and kidneys of carp raised in saline media and exposed to Aeromonas hydrophila bacterial infection. The study was conducted from August to October 2024 at the Biotechnology Laboratory, Faculty of Fisheries and Marine, Universitas Riau. The method used was an experiment with a completely randomized design (CRD) consisting of five guava leaf flour dosage treatments with three replicates. The results showed that the histological structure of the intestine in all treatments was relatively normal, with neatly arranged villi and round to oval goblet cells. However, in the guava leaf powder supplementation treatment, several abnormalities were found in the kidney tissue, including inflammation, hemorrhage, and necrosis. This indicates that the salinity factor has a greater effect on kidney damage than the effect of immunostimulant administration. Nevertheless, the administration of guava leaf powder still positively improved the immune response and survival rate of fish. The optimal dose was obtained in treatment P2 (15 g/kg feed) with a survival rate of 83.33–93.33%. The water quality parameters during the study were still within the acceptable range for aquaculture, namely temperature 28–31°C, DO 3.7–7.4 mg/L, and pH 5.5–7. Thus, it can be concluded that feeding fish with guava leaf flour-enriched feed is effective in maintaining intestinal histology and increasing the survival rate of carp in saline media, although it cannot completely prevent kidney damage caused by environmental factors
