Abstract

This research aims to describe the kinship relationship of baronang fish species, based on morphological character analysis. The approach used is descriptive explorative. Research samples were obtained from the sea waters of Labean Village, Donggala Regency using purposive sampling techniques. The obtained baronang fish were then identified and measured based on 21 morphometric characters, 5 meristic characters, and 4 qualitative characters in each of the six species found. Data is processed using Microsoft Excel then analyzed using cluster analysis on MVSP (Multivariate Statistical Package) software. The similarity index uses the Simple Matching Coefficient method, the reconstruction of the dendogram using the UPGMA algorithm. The results of this research found six species of baronang fish, namely: Siganus canaliculatus, Siganus doliatus, Siganus vulpinus, Siganus vermiculatus, Siganus guttatus, and Siganus puellus. The kinship relationship between six types of baronang fish is formed into five clusters with various kinship categories, namely very close, close, and not close. The first cluster shows a very close kinship occupied by Siganus canaliculatus with Siganus doliatus with a similarity index of 0.93%. Meanwhile, the fifth cluster that shows not close kinship is the grouping between fish species on Node 4 with fish species on Node 3 with a similarity index of 0.43%. The results of this research, are expected to be the basis for further research related to kinship or the development of baronang fish species in Labean Village, Donggala Regency.

Abstract

Abstrak: Penelitian ini bertujuan untuk mendeskripsikan persepsi siswa tentang keterampilan mengajar guru IPA. Jenis penelitian ini menggunakan penelitian deskritif kuantitatif. Sampel penelitian ini adalah kelas VII dan VIII yang berjumlah 32 siswa. Instrumen penelitian menggunakan lembar angket untuk siswa. Hasil penelitian ini menunjukkan nilai persepsi siswa terhadap keterampilan mengajar guru IPA itu sebanyak 47% dengan kategori sangat baik, selanjutnya kategori baik persentasenya 38% dan kategori cukup yaitu 15%. Kemudian data hasil identifikasi penggunaan keterampilan mengajar guru IPA di SMP Negeri 7 Palu dalam kategori cukup terutama keterampilan membuka dan menutup pembelajaran memiliki nilai persentase 56%, keterampilan menjelaskan 56%, keterampilan bertanya 54%, keterampilan mengadakan variasi 56%, keterampilan memberikan penguatan 52%, keterampilan mengelola kelas 49%, keterampilan membimbing diskusi kelompok kecil 56% dan keterampilan mengajar kelompok kecil dan perorangan memiliki 50%. Sehingga dapat disimpulkan meskipun sebagian besar siswa memberikan persepsi positif, namun beberapa aspek keterampilan mengajar guru yang perlu ditingkatkan, terutama dalam pengelolaan kelas dan mengajar kelompok kecil maupun perorangan. Oleh karena itu, diperlukan upaya berkelanjutan dalam bentuk pelatihan, supervisi, dan pembinaan untuk meningkatkan kompetensi guru IPA dalam menerapkan keterampilan mengajar yang efektif dan menyeluruh.

Abstract

This study aims to identify the types of fungi and bacteria that play a role in the fermentation process of cocoa beans. This type of research uses an exploratory laboratory method. The samples in this study were fungi and bacteria during the fermentation process of cocoa beans that had been fermented for approximately 3-5 days. The tools used in this study were digital scales, clear plastic boxes, autoclaves, scissors, micropipettes, spoons, label paper, Petri dishes, incubators, glass covers, Bunsen burners, Erlenmeyer flasks, inoculation needles, stirring rods, microscopes, beakers, measuring cups, scales, hot plates, test tubes, spirit lamp, colony counter, aluminum foil, camera, and writing instruments. The materials used in this study were fermented cocoa bean waste samples colonized by fungi and bacteria, Nutrient Agar (NA) medium, Triple Sugar Iron Agar (TSIA) medium, Bacillus Agar Base, distilled water, crystal violet, eosin dye, iodine, alcohol, and safranin. There are five stages in this research procedure, namely sample collection and preparation, media preparation and sample dilution, sample cultivation on general and selective media, Gram staining, and biochemical testing. The results of this study show that the fungi Saccharomyces cerevisiae and Mucor Sp, as well as the bacteria Bacilllus Sp, are involved in the fermentation process of cacao beans.

Abstract

This study aims to describe the variation in haplotype distribution and phylogenetic relationships of the Philippine scrub fowl based on the EEF2 gene. Eggshell samples were obtained from Tolitoli and Luwuk, Central Sulawesi. DNA isolation followed the gSYNCTM Extraction Kit protocol, and the amplification process was carried out in 38 cycles, including the pre-denaturation stage (95°C, 5 minutes), denaturation stage (95°C, 45 seconds), annealing stage (53°C, 40 seconds), elongation stage (72°C, 1 minute), and post-elongation stage (72°C, 10 minutes), followed by sequencing. Sequence editing was performed using GeneStudio, sequence alignment and phylogenetic tree reconstruction using MEGA 11, and haplotype network reconstruction using DnaSP and PopART. The results showed that the haplotype network based on the EEF2 gene formed three haplotypes, with the population from Tolitoli having a widespread haplotype, while the population from Luwuk had a specific haplotype. In addition, based on the results of phylogenetic tree reconstruction, it was found that the Philippine white-eye birds from Tolitoli and Luwuk were in the same clade but there was genetic variation between populations.

Abstract

This study aims to determine the relationship between the intensity of playing online games and student learning outcomes in science subjects in grade VIII at SMP Negeri 18 Model Palu. This type of research uses quantitative research with correlation analysis. The sample of this study is class VIII which totals 105 students. The data collection technique in this study is a questionnaire. The results of this study showed that  the intensity of playing online games obtained in the medium category was 65 students (61.90%), the high category was 27 students (25.71%), and the low category was 13 students (12.38%). The number of hours used in one play was obtained more than 5 hours by 32 students (55.17%), up to 3 hours by 18 students (31.03%), and up to 1 hour by 8 students (13.79%). The results of bivariate analysis, it showed a correlation coefficient (Spearman's rho) of 0.106, there was a positive but very weak relationship between the intensity of playing online games and student learning outcomes. With a significance of 0.282, because it is 0.282>0.05, it is concluded that there is no significant relationship between the intensity of playing online games and student learning outcomes in science subjects in grade VIII at SMP Negeri 18 Model Palu.

Abstract

Science education in the digital era continues to face challenges in optimally developing students’ science process skills and information technology competencies. The lack of instructional models that effectively connect theoretical concepts to real-world contexts remains a barrier to achieving 21st-century competencies. This study aims to analyze the effectiveness of implementing the Case Based Learning (CBL) model in integrating students’ science process skills and information technology capabilities. This research employed a quantitative approach with a quasi-experimental design. The study involved 50 tenth-grade students divided into two groups: an experimental group and a control group. Data were collected using a science process skills test and an observation sheet to assess information technology skills. Data analysis was conducted using an independent sample t-test to determine significant differences between the two groups. The results revealed an increase in the average scores for both science process skills and information technology abilities. This improvement indicates that the CBL model encourages students to engage more actively, think critically, and relate learning to real-life situations. The findings conclude that the CBL model is effective in enhancing students’ science process skills and information technology competence. The implication is that CBL can be recommended as an innovative instructional strategy for science education that is contextual and aligned with the demands of the 21st century.

Abstract

Penelitian ini bertujuan mengetahui ekspresi protein AdhF36 strain Salmonella Typhi, sebagai kajian kandidat vaksin berbasis molekul adesin. Strain yang digunakan pada penelitian ini bersumber dari Rumah Sakit Hasan Sadikin Bandung, Laboratorium Kesehatan Sulawesi Tengah, Palu, Rumah Sakit Udayana Denpasar, dan Rumah Sakit Saiful Anwar, Malang. Hasil penelitian menunjukkan bahwa sekitar 12 strain Salmonella Typhi tersebut mengekspresikan dan memiliki protein AdhF36. Hal ini memberikan gambaran umum bahwa protein AdhF36 potensial dikembangkan sebagai kandidat vaksin.

Abstract

Penelitian ini bertujuan mengamati stabilitas ekspresi protein AdhF36 Salmonella typhi pada perlakuan osmolaritas dan pH dengan metodeeksperimen laboratorium-eksploratif. Hasil penelitian ini menunjukkan perbedaan ekspresi protein AdhF36 Salmonella typhi pada perlakuan osmolaritas dengan pH. Hasil SDS-PAGE menunjukkan bahwa pita protein AdhF36 tetap terekspresi pada perlakuan osmolaritas 50-350 mM, sebaliknya pita protein ini tidak terdeteksi pada perlakuan pH 4,5-6,0. Hal ini didukung oleh hasil uji Western Blot yang berhasil mendeteksi protein AdhF36 hanya pada perlakuan osmolaritas. Hasil ini membuktikan bahwa Salmonella typhi lebih toleran terhadap perubahan osmolaritas dibandingkan dengan pH.

Abstract

This study aims to identify the presence of Escherichia coli and Salmonella sp. in "ayam geprek" products sold at the canteens of Tadulako University. The type of research used is descriptive-exploratory with quantitative examination methods using the Most Probable Number (MPN), Total Plate Count (TPC), and further identification using the Microbact System and Microbact System 2000 Software. Samples were taken from three different faculty canteens using a stratified random sampling technique. The results showed that (1) Test using the Most Probable Number (MPN) method obtained the highest value of 150x102 CFU / ml came from the FMIPA canteen sample, 28x102 CFU / ml samples from the FKIP canteen and the lowest value of 27x102 CFU / ml samples from the FKIP canteen. (2) Test using the Total PlateCount (TPC) method obtained the highest value of 105x103 CFU / ml came from the FMIPA canteen sample, 217x102 CFU / ml FKIP canteen sample, and the lowest value of 120x102 CFU / ml FKIP canteen sample. (3) Validation identification of the percentage of samples showing contamination by Escherichia coli and Salmonella sp., with a validation level above 90%. Thus, all samples in this study were positive for the presence of Escherichia coli and Salmonella sp. bacteria.

Abstract

This research aimed to isolate and identify the types of bacteria present on the surface of biology education students' mobile phone screens. This was an exploratory study using a bacterial isolation method.Bacteria found on the mobile phone screens were cultivated using MacConkey agar, a selective medium specifically designed to detect and identify Gram-negative bacteria, particularly Enterobacteriaceae like Escherichia coli. The accuracy of the Escherichia coli data from the phone screen samples, after processing with the Microbact System, was 96.39%.Malt Extract Agar (MEA) was also used as a selective medium to detect and identify Gram-positive fungi, such as Candida albicans.Based on these findings, the developed module was deemed highly feasible. A learning medium is considered highly feasible for use if it achieves a presentation rate between 60-100%. With an average score of 82.39%, the module meets this criterion and can be effectively used in learning.
