Abstract

Abstract

Kidney damage caused by high-dose paracetamol leads to elevated Blood Urea Nitrogen (BUN) and creatinine levels as indicators of impaired renal function. This study aimed to evaluate the dose-dependent nephroprotective effects of ethanolic Carica papaya seed extract on BUN and creatinine levels in paracetamol-induced albino rats. A posttest-only control group design was applied using 25 male Wistar rats divided into five groups: negative control (1% CMC-Na), positive control (paracetamol 1000 mg/kgBW), and three treatment groups receiving Carica papaya seed extract at doses of 100, 200, and 400 mg/kgBW. The extract was administered orally for seven consecutive days before paracetamol induction on day 8 and continued until day 11. Blood samples were collected on day 11, and serum was stored at -20 °C before BUN and creatinine concentrations were analyzed spectrophotometrically. Data were statistically analyzed using one-way ANOVA followed by Duncan’s Multiple Range Test (DMRT) at a significance level of p < 0.05. The results showed a significant and dose-dependent reduction in both parameters in all treated groups compared to the positive control, with the 400 mg/kgBW dose showing the strongest effect and restoring values close to physiological levels. The nephroprotective activity was associated with the antioxidant and anti-inflammatory mechanisms of the bioactive compounds that counteract oxidative stress and preserve renal cellular integrity. In conclusion, the ethanolic extract of Carica papaya seeds demonstrates significant dose-dependent nephroprotection against paracetamol-induced kidney injury in rats.

Abstract

Non-steroidal anti-inflammatory drugs (NSAIDs) can cause significant gastrointestinal mucosal injury, particularly in the duodenum. This damage is often initiated by localized erosion of the duodenal mucosa, which triggers a subsequent inflammatory cascade. The porang tuber (Amorphophallus oncophyllus) is a promising candidate for mitigating this injury due to its rich composition of bioactive compounds, including flavonoids, alkaloids, tannins, and glucomannan, all of which possess known anti-inflammatory properties. This study investigated the protective effect of an ethanolic A. oncophyllus extract on the duodenal mucosa in an indomethacin-induced rat model. Thirty Wistar white rats (Rattus norvegicus) were randomly assigned to five groups: a vehicle control, a model group receiving indomethacin, and three treatment groups that were pretreated with A. oncophyllus extract at doses of 50, 100, and 200 mg/kg, respectively, for seven days before a single dose of indomethacin. Our findings demonstrated that the A. oncophyllus extract, particularly at a dose of 200 mg/kg, effectively reduced duodenal mucosal injury caused by indomethacin. This protective effect is likely attributable to the extract's phytochemical profile, which contains flavonoids, tannins, saponins, terpenoids, and alkaloids. In conclusion, this research confirms that A. oncophyllus extract has the potential to protect the duodenal mucosa from NSAID-induced damage.

Abstract

This research aims to find out the administration effect of Ambon banana stem extract (Musa paradisiaca var. sapientum) to prevent gastric damage and observe histopathology of rat's gastric induced by indomethacin. This research used 30 male rats age 8-12 week having 150 gram weight. They were selected randomly and divided into five groups. Negative control (K-) was given 0.5 ml CMC Na 0.5 % for 9 days and 0.5 ml corn oil was given on 10th. Positive control (K+) was given 0.5 ml CMC Na 0.5% for 9 days and then given Indomethacin emultion 5 mg/0.5 ml/150 g BW once on 10th day, and the other groups were given Ambon banana stem extract for (P1) 20 mg/150 g BW, (P2) 40 mg/150 g BW and (P3) 80 mg/150 g BW for 9 days. Afterwards, they were given Indometachin emultion 5 mg/150 g BW once on 10th day. Each gastric specimen was processed and histopathological changes were observed. Scoring of mucosa epithelium erosion and hemorrhagic, as qualitative data, was analyzed by Kruskall-Wallis test and continued by using Z test. The result shows that Pisang Ambon's stem extract reduce significantly in gastric mucosa epithelium erosion and hemorrhagic induced by Indomethacin (p<0.05).

Abstract

Streptomyces sp. is a bacterium that can live in many kind of habitations e.i. marine, sea sponge, coastal area, soil, desert soil, river bank etc. Many antibiotics and secondary metabolites are produced by this bacterium. Several secondary metabolites of the bacterium can be used as an insecticide against insects including mosquito larva. The purpose of this study was to search and isolate until species of Streptomyces sp. from Sidoarjo Lapindo mud soil in East Java province that can be used as a larvicide against Aedes aegypti larva, the dengue haemorrhagic fever vector. The method of study was to collect several mud soil samples from Sidoarjo Lapindo. The collected samples were cultured in ISP-4 media for producing mix-cultures. Then, to isolate the suspected colony of Streptomyces sp. by culturing again in several replications on ISP-4 media in petri-dish. The pure isolates were cultured in ISP-4 slant media. There were could be obtained eight pure isolates of Streptomyces sp.The characterization of 16S rRNA of Streptomyces sp. was done in order to determine the species. DNA isolation was done and followed by DNA sequencing, then compared to Gene Bank with BLAST program. The results showed that the sequence nucleotide bases of Streptomyces Sp-D6 had high similarity to Streptomyces sp. 171524, beside the sequence nucleotide bases of Streptomyces Sp-D7 and Sp-D9 had high similarity to Strepto-myces sp. ACT-01578 and ACT-175695. These three species can be used as larvicide candidate against Ae. aegypti.
