Abstract

Litsea firma (Blume) Hook. f., known locally as Medang Piawas, is a Lauraceae plant found in West Kalimantan, which was traditionally used as medicine and reported to contain alkaloids. This study determined the total alkaloid content in ethanol extracts of the roots, stems, and leaves. Phytochemical tests with Dragendorff, Mayer, and Wagner reagents confirmed alkaloid presence, with the highest intensity in stems, followed by leaves and roots. Quantitative analysis by UV-Vis spectrophotometry using caffeine and quinine standards showed the stem extract contained the highest alkaloid levels (18.289 mg CE/g and 2.299 mg QE/g). Cytotoxic activity was evaluated against cancer cell lines (4T1, MCF-7, T47D, WiDr, HeLa, Hep G2) and normal Vero cells using the MTT assay. The root extract exhibited weak cytotoxicity against WiDr cells (IC50 201"“500 µg/mL) but was inactive against other cancer and normal cells. Stem and leaf extracts showed no cytotoxic effects (IC50 >500 µg/mL). These findings indicate that L. firma contains notable alkaloids, predominantly in stems, although its ethanol extracts demonstrate limited cytotoxic activity.

Abstract

This study aims to determine and compare antioxidant activity, total phenolics, flavonoids, and the relationship of phenolic and total flavonoids with the antioxidant activity of the three kratom variants. The ethanol extract of the third variant of kratom leaves was obtained by maceration. Through the antioxidant activity test using DPPH method, all three extracts showed strong antioxidant activity with IC50 values of 26.39, 30.25, and 30.59 g/mL. The relationship of total phenolics and flavonoids with antioxidant activity was determined by the Pearson correlation test. Examination of total phenolic content using the Folin-Ciocalteu method with successive results for green, red, and white kratom are 6.11, 8.67, and 9.09 mg GAE/g extract. Examination of total flavonoid content using the colorimetric method for green, red, and white kratom were 0.86, 0.68, and 1.13 mg QE/g extract, respectively. The total phenolic content and antioxidant activity showed a correlation coefficient value of -0.32 (P0.05), and the total flavonoid content to antioxidant activity showed a correlation coefficient value of 0.81 (P0.05). Thus, all three variants of kratom have potency as natural antioxidants, but their total phenolic and flavonoid content does not influence their antioxidant activity.

Abstract

ABSTRACTImplementing entrepreneurship as part of the Merdeka Curriculum is considered suboptimal due to the numerous challenges schools face. This is consistent with the survey results among chemistry educators in the Singkawang, which showed that while most have implemented entrepreneurship in their teaching, it has not yet incorporated local wisdom and has not been included in their teaching tools. The purpose of this community service program (PKM) is to enhance educators' ability to develop entrepreneurship-based learning that incorporates local wisdom and to describe educators' responses to the training activities conducted. The method used in this PKM is a training method for 24 chemistry educators in Singkawang, consisting of three stages: preparation, implementation, and evaluation. The results of the PKM activities showed increased knowledge and helped Singkawang's chemistry educators integrate these two concepts into their teaching. Abstract

The enzyme tyrosinase (EC 1.14.18.1) plays a role in the browning reaction of peels, fruits and vegetables, by catalyzing the process of melanin production. Therefore, tyrosinase inhibitors are needed to prevent excess melanin production from causing adverse browning reactions. Kratom leaves (Mitragyna speciosa Korth) contain secondary metabolites of alkaloids and flavonoids which are thought to have better and safer activity as tyrosinase inhibitors than kojic acid. Based on the color of the leaf veins, kratom leaves are divided into 3 variants, namely red, green and white kratom veins. This study aims to examine the activity of the ethanol extract of three variants of kratom leaves as an inhibitor of the tyrosinase enzyme. The results showed that the IC50 value of the three kratom leaf variants was >700 µg/mL while the IC50 value of kojic acid was 32.85 µg/mL. From the results of the study, it can be concluded that the ethanol extract of kratom leaf variants of red, green and white veins did not have activity as an inhibitor of the tyrosinase enzyme because the IC50 values of all three were >700 µg/mL. Keywords:          Tyrosinase enzyme, Kratom, Mitragyna specios, melanin   Abstrak Enzim tirosinase (EC 1.14.18.1) berperan dalam reaksi pencoklatan kulit, buah, dan sayur, dengan mengkatalisis proses produksi melanin. Abstract

Liver, colon, and cervical cancer are the top three types of cancer which rank in the top five worldwide in terms of incidence and mortality. Simpur air (Dillenia suffruticosa)plant has traditionally been used for the treatment of various diseases, one of which is cancer. This study aims to prove cytotoxic effect and selectivity of simpur air wood bark fractions against liver HepG2, colon WiDr, and cervical HeLa cancer cells. Simpur air wood bark is macerated using 96% methanol. The methanolic crude extract was fractionated with the liquid-liquid partition method using n-hexane and ethyl acetate. Cytotoxicity test using MTT assay method. The result showed that the methanol extract, n-hexane fraction, ethyl acetate fraction, and methanol fraction in liver HepG2 cancer cells had IC50 values 179,32±13,55; 85,14±8,58; 140,08±22,63; >500 µg/mL and in colon WiDr cancer cells had IC50 values 133,02±27,64; 41,54±37,58; 124,76±17,22; >500 µg/mL. Meanwhile, all fractions had IC50 values >500 µg/mL in HeLa cervical cancer cells. These data indicate that the n-hexane fraction in WiDr cells is potentially cytotoxic and selective with a selectivity index value greater than 3. Thus, simpur air wood bark has the potential to be developed as an anticancer drug candidate.
