Abstract

Dyera polyphylla (Miq.) Steenis, or jelutung rawa, is an endemic tree species native to Indonesia’s tropical peat swamp forests. While related species such as Dyera costulata have been reported to contain various bioactive compounds particularly phenolics with antioxidant, anti-inflammatory, and antimicrobial properties, there has not been phytochemical studies on D. polyphylla. This study aimed to investigate the phenolic compound profile and total phenolic content in the stem wood, stem bark, and leaves of D. polyphylla. Extraction was carried out using total and stratified maceration methods with n-hexane, ethyl acetate, and methanol as solvents. Qualitative analysis involved phytochemical screening by using phenolic specific reagents, thin-layer chromatography (TLC) with FeCl₃ reagents, and high-performance liquid chromatography (HPLC). Total phenolic content was quantified using the Folin–Ciocalteu spectrophotometric method. Phenolic compounds were detected in all plant parts, particularly in the ethyl acetate and methanol extracts. HPLC analysis showed that the chromatographic profile of the methanol extract from stem wood closely resembled that of the leaves. The highest phenolic content was observed in the methanol extract of the leaves (29.02 mg GAE/g). These results suggested that D. polyphylla, particularly its leaves, is a promising natural source of phenolic compounds with potential antioxidant and therapeutic value.

Abstract

Indonesia, as an agrarian country rich in natural resources, holds enormous potential in the essential oil industry. There are approximately 150–200 species of essential oil-producing plants worldwide, 40 of which are found in Indonesia, including those from the Lauraceae family. The genus Litsea within the Lauraceae family is known for producing essential oils, with Litsea cubeba being the most popular. However, high demand for Litsea’s essential oils encourages the exploration of essential oils from other Litsea species, such as Litsea firma. This study used gas chromatography-mass spectrometry to look at the chemical profile of essential oils from the roots, stems, and leaves of L. firma grown in West Kalimantan. The chemical profile data is then compared with data from other studies. The analysis results indicated that the composition of essential oils from roots is similar to the stems but differs from the leaf. The stem and root essential oils primarily consist of 4,4-diethyl-spiro[2,3]hexan-5-on, whereas the leaf essential oils mostly contain 1-cyclohexyl-ethanone. This difference in chemical constituents suggests that different parts of L. firma may have different ecological roles or therapeutic benefits. Further research could explore the implications of these findings for the plant's potential use in traditional medicine or natural product development. Therefore, we should continue exploring L. firma's essential oil to maximize its potential.

Abstract

Colon cancer is one of the most deadly cancers in the world. Conventional treatments such as surgery, radiotherapy and chemotherapy are often limited, especially in metastatic cancer. Therefore, the development of alternative therapies, such as chemoprevention, becomes very important. This study aimed to test the anticancer potential of bengkal (Nauclea subdita) stem bark against WiDr colon cancer cells through cytotoxic, cell cycle, and apoptocic assays. The stem bark of bengkal was extracted using n-hexane and ethanol, then fractionated using dichloromethane (DCM). The results of the cytotoxic test using the MTT method showed that the DCM pH 3 fraction had the highest cytotoxic potential with an IC50 value of 112.81 ± 22.55 µg/mL against WiDr cells. However, the Selectivity Index (IS) value of the DCM pH 3 fraction showed low selectivity (<2). Cell cycle assay using flow cytometry indicated that DCM pH 3 fraction was able to inhibit cell cycle distribution. Furthermore, apoptocic test with Annexin V-FTIC/PI method showed that DCM fraction pH 3 triggered apoptocic mechanism in WiDr cancer cells. This study provides preliminary evidence that DCM fraction pH 3 of bengkal stem bark has anticancer potential against WiDr colon cancer cells, thus DCM fraction pH 3 of bengkal stem bark can be a source of WiDr colon anticancer agent.
