Abstract

 The Eurycoma longifoliais bitter therefore, to address this problem, it is prepared in a capsule form. The capsule dosage form  must has  the parameters of good quality, thus expected to increase the potential use of biodiversity and the interest of the community in consuming the medicine from the natural materials. The purpose of this research is to get the capsule product of ethanol extract of the Eurycoma longifolia root of quality through the test of total plate value and yeast yeast contamination, the determination of the metal content of Pb, Hg, Cd and the level of flavonoids released in the capsule dissolution test of ethanol extract Eurycoma longifolia root. The result of this research shows that the dissolution efficiency (DE120) capability of ethanol extract of Eurycoma longifolia root is 81,04±2,38%. Test of heavy metal content in capsule of  of ethanol extract of Eurycoma longifolia root found level of Pb equal to 4,132 ppm and not detected content of Cd and Hg in capsule. This indicates that the capsule meets the requirements set by BPOM RI. Tests for total plate numbers (ALT) and yeast capsule ethanol extract of the E. longifolia root were each obtained data of 70 x 102 colonies / g and 4 x 102 colonies / g. It also shows that the capsules meet the requirements by BPOM RI.

Abstract

The present study tried to investigate potency of L. sarmentosa root extract on quality and viability of mice sperm.This study used 25 Swiss male mice were randomly divided into five experimental groups of 5. The ethanolic extract of L. Sarmentosa (100, 200, 400 mg/kg b.w/days), control and X-gra were administered orally for 14 days. The animals were evaluated for sperma quality (sperm count and sperm motility) and viability at 14 days treatment. The results showed that administration of L.sarmentosa roots extract significantly increasing the sperm count (970x106cell/mL) at 400 mg/kg b.w/days (P 0.0040). The extract dose of 200 mg/Kg b.w and 400 mg/Kg b.w had the highest motility values (95%). Viability of sperm are 83.8%; 96.0%; 87.8% respectively at a dose of 100 mg/kg b.w, 200 mg/kg b.w and 400 mg/kg b.w. Present finding oral administration of L. sarmentosa roots extract has a potency of the quality and viability of mice sperm.

Abstract

Pasak Bumi roots contain quasinoid compounds that have carcinogenesis inhibitory, imunomudulator, antiulcer, and antimalaria effects. This study aims to determine the effect of a combination of ethyl acetate fraction of Pasak Bumi roots and doxorubicin on DMBA-induced white mice by weight and palpation observation. The mice were divided into 8 groups. Each group consisted of 7 female Sprague Dawley mice. Group 1 (normal) was only given food and drink, Group 2 was given fraction of ethyl acetate of Pasak Bumi roots (100 mg/kg body weight, Group 3 (positive control) was given doxorubicin (1.17 mg/kg body weight) 1 times a week for 5 weeks, Group 4 was given DMBA (20 mg/kg) two times a week for 5 weeks, Groups 5 was given DMBA (20 mg/kg) + doxorubicin (1.17 mg/kg), Group 6 was given DMBA (20 mg / kg) + ethyl acetate fractions of Pasak Bumi roots (100 mg / kg), Group 7 was given DMBA (20 mg / kg) + doxorubicin (1.17 mg / kg) + ethyl acetate fraction of Pasak Bumi roots (100 mg / kg) and Group 8 doxorubicin ( 1.17 mg / kg body weight) + ethyl acetate fraction of Pasak Bumi roots (100 mg / kg). Observations were made on the basis of body weight of mice and tumor incidence by palpation at week 6 of treatment. The result of  this research shows that the administration of ethyl acetate fraction of Pasak Bumi roots and doxorubicin combination can increase body weight of the mice and decrease the number of tumors compared to single doxorubicin.

Abstract

Background: A research has been done to test the In vitro antibacterial activities of sirsak leaf(Annona muricata L.) infuse toward Staphylococcus aureus ATCC 25923 and Escherichia coliATCC 35218 to analyze its thin layer chromatography profile. This research done to observe theantibacterial activity from sirsak leaf infuse.Method: A test on the antibacterial activity was done by using liquid dilution method. Theconcentration infuse in sterile destilate water using to test the antibacterial activity toward S.aureus were 100%, 95%, 90%, 85%, 80%, and 75% w/v, while toward E. coli were 100%, 90%,80%, 70%, 60%, and 50% w/v. To detect Chemical contains of the sirsak leaf infuse wereidentified using Screening method of Phytochemistry and Thin Layer Chromatography (TLC).Result: The result showed that the Minimum Bacterial Concentration (MBC) of sirsak leaf infuseS. aureus were 85% w/v, while E. coli could not be show antibacterial activity until 100% w/vconcentration. The Minimum Inhibition Concentration (MIC) could not be identified by the liquiddilution because the mixture were turbid and the colour is dark brown. Screening method ofPhytochemistry use tube-test and Thin Layer Chromatography showed that the infuse containflavonoid, poliphenol, and alkaloid.Conclusion: Infuse of sirsak leaf (Annona muricata L.) has antibacterial activity againstStaphylococcus aureus ATCC 25923 and Escherichia coli ATCC 35218 profil infuse. Thin LayerChromatography showed that the infuse contain flavonoid, poliphenol, and alkaloid.Keyword: Antibacterial, sirsak leaf, chromatography

Abstract

Kelopak bunga rosella (Hibiscus sabdariffa L.) memiliki kandungan flavonoid terutama antosianin dan quersetin yang berpotensi sebagai antioksidan, hepatoprotektor, diuretik, antikolesterol serta imunostimulator. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian serta keamanan ekstrak etanol kelopak bunga rosella merah selama 30 hari dan selama 15 hari setelah tidak mengkonsumsi rosella terhadap nilai hematokrit, trombosit dan eritrosit. Penelitian ini menggunakan rancangan pre-post treatment design study. Dua puluh satu sukarelawan sehat digunakan dalam penelitian yang telah memenuhi kriteria inklusi dan eksklusi serta bersedia mengisi informed consent. Sukarelawan diberikan kapsul dengan dosis 500 mg sebanyak 1 kali sehari, 10-15 menit setelah makan pada malam hari selama 30 hari. Pemeriksaan hematologi dilakukan pada hari ke-0; 31; dan 45 yang meliputi parameter hematokrit, trombosit, dan eritrosit menggunakan hematology analyzer. Hasil yang diperoleh dibandingkan secara statistik menggunakan paired t test dan wilcoxon test. Hasil rerata hematokrit, trombosit, dan eritrosit pada sukarelawan laki-laki dan perempuan hari ke-0, hari ke-31 dan hari ke-45 masuk dalam rentang normal. Hasil penelitian menunjukkan tidak terdapat perbedaan bermakna (p>0,05) nilai hematokrit, trombosit dan eritrosit perbandingan hari ke-0 dengan hari ke-31, hari ke-31 dan hari ke-45, serta hari ke-0 dan hari ke-45. Pemberian kapsul ekstrak etanol rosella merah 500 mg selama 30 hari dan setelah 30 hari tidak mengkonsumsi kapsul ekstrak etanol rosella tidak mempengaruhi semua parameter hematologi pada sukarelawan sehat.

Abstract

Infeksi kandidiasis meningkat selama dekade terakhir di dunia. Peningkatan ini menandai munculnya isolat Candida albicans resisten Flukonazol. Diperlukan obat baru untuk mengatasi permasalahan resistensi ini.  Daun pacar kuku (L. inermis L.) secara empiris mempunyai aktivitas sebagai antijamur. Kandungan kimia daun pacar kuku larut dalam etil asetat. Penelitian ini bertujuan untuk mengetahui aktivitas antifungi dari fraksi etil asetat ekstrak daun pacar kuku (L. inermis L.) terhadap strain C. albicans sensitif Flukonazol dan C. albicans resisten Flukonazol. Fraksi etil asetat ekstrak daun pacar kuku (L. inermis L.)dibuat dengan memfraksinasi ekstrak etanol dengan  pelarut etil asetat.Konsentrasi fraksi etil asetat ekstrak daun pacar kuku yang digunakan terhadap C. albicans sensitif Flukonazol dan C. albicans resisten Flukonazol sebesar 10%; 5%; 2,5%; 1,25% dan 0,625%. Pengujian aktivitas antifungi fraksi etil asetat ekstrak daun pacar kuku menggunakan metode mikrodilusi cair dengan parameter pengukuran Minimum Inhibitory Concentration (MIC) dan Minimum Fungicidal Concentration (MFC). Nilai MIC dan MFC yang diperoleh sama yaitu sebesar 1,25% untuk C. albicans resisten Flukonazol dan 0,625% untuk C. albicans sensitif Flukonazol. Rasio MFC terhadap MIC yang diperoleh sebesar 1 yang menyatakan bahwa fraksi etil asetat ekstrak daun pacar kuku memiliki daya fungicide. Skrining fitokimia fraksi etil asetat ekstrak daun pacar kuku mengandung fenolik dan kuinon.Kata kunci : Kandidiasis, pacar kuku, mikrodilusi cair, MIC dan MFC

Abstract

Validasi metode analisis (VMA) penentuan kandungan β-karoten dalam Spirulina maxima perlu dilakukan agar didapatkan hasil yang valid dan dapat dipertanggung jawabkan secara ilmiah. Tujuan penelitian adalah membuktikan metode analisis penetapan kadar β-karoten dalam ekstrak etanol 96% Spirulina maxima menggunakan spektrofotometer visibel memenuhi syarat validitas. Parameter yang digunakan dalam VMA diantaranya spesifisitas, linieritas, akurasi, presisi, limit deteksi dan limit kuantitas. Syarat yang harus dipenuhi ditiap parameter yaitu nilai korelasi (r) ≥ 0,98 untuk linieritas, persen perolehan kembali masuk dalam range 98-102% dan nilai RSD ≤ 2% untuk akurasi, nilai RSD ≤ 2% untuk presisi, profil spektragram spektrofotometri visibel yang sama antara sampel dan standar untuk uji spesifisitas. Hasil penelitian dengan parameter validasi diantaranya linieritas dengan nilai r=0,998, persen perolehan kembali dalam rentang 98-101%, presisi keberulangan sistem dengan RSD 1,14 %, presisi antara 0,99 % dan uji spesifisitas sesuai profil spektragram spektrofotometri visibel yang sama antara sampel dan standar. Nilai LOD yang diperoleh yaitu 1,656 ug/mL dan LOQ 5,017 ug/mL. Berdasarkan hasil tersebut maka analisis penetapan kadar β-karoten dalam ekstrak etanol 96% Spirulina maxima dengan metode spektrofotometri visibel memenuhi syarat validitas menurut Petunjuk Operasional Penerapan CPOB tahun 2013.

Abstract

Akar pasak bumi mengandung senyawa kuasinoid diduga mempunyai efek penghambatan karsinogenesis, antiulcer, dan antimalaria. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian eksktrak etanol akar pasak bumi terhadap ekspresi p53 pada kanker payudara tikus betina yang diinduksi DMBA.Tikus dibagi menjadi 6 kelompok. Kelompok I, II dan III diberi ekstrak etanol akar pasak bumi dosis berturut-turut 100; 200 dan 400 mg/kg BB. Kelompok IV diberi larutan DMBA 20 mg/kg BB. Kelompok V diberi corn oil. Kelompok VI sebagai baseline. Tikus yang mengalami kanker, dihitung jumlah nodul tumornya dan pada minggu ke-23 semua tikus dikorbankan dan diambil jaringannya untuk pengamatan secara mikroskopik menggunakan metode H&E dan imunohistokimia. Hasil penelitian menunjukkan bahwa rerata ekpresi p53 mutant pada pemberian ekstrak berturut-turut 100, 200 dan 400 mg/kg BB adalah 6,375 ± 4,07; 11,25 ± 16,53; dan 0,875 ± 1,75; kelompok  IV (DMBA) sebesar 13,125 ± 14,4; kelompok V (corn oil) adalah 1,375 ± 1,55; kelompok VI (baseline) adalah 0,375 ± 0,75. Hasil penelitian disimpulkan bahwa ekstrak etanol akar pasak bumi (Eurycoma longifolia Jack) dosis 400 mg/kg BB yang diberikan sebelum dan selama induksi DMBA mampu menurunkan ekspresi protein p53 mutant (proapotosis) dan mampu menghambat pertumbuhan kanker payudara tikus betina Sprague Dawley yang diinduksi 7,12-dimetilbenz(?)antrasen (DMBA).Kata kunci:  akar pasak bumi (Eurycoma longifolia Jack), kemopreventif, imunohistokimia, ekspresi p53

Abstract

The continued increase in the number of people with Diabetes Mellitus (DM) in Indonesia is a serious problem. One of the big problems for people with Diabetes Mellitus (DM) is the emergence of complications of diabetic wounds. To date the strategy for treatment of diabetic wounds has been limited to the use of wound dressing, cell therapy and oxygen therapy. The problem is that the strategy is not fully successful. Thus, it is very important to look for new strategies to improve the quality of diabetic wound healing, such as by applying a combination of plasma medicine and local natural product, like the extraction of Daun sirih (Piper betle) leaves. Plasma medicine is a relatively new and multidisciplinary study involving plasma science, biomedical, pharmaceutical and other health sciences aimed at applying plasma to therapeutic health. Plasma is the fourth phase of matter, after the solid, liquid and gas phase. The medical aspects of plasma are related to the ability of plasma to produce biological molecules Reactive Oxygen and Nitrogen Species (RONS). If RONS is controlled in the right dosage it can be efficacious for health therapy. This study intends to examine the effects of combinative treatment of plasma medicine and Piper betel leaf extract for proliferation phase of wound healing in diabetic small animal model. This study used male Balb c mice with acute wounds which were divided into 5 groups, namely groups of untreated normal mice (ND-TP), groups of untreated diabetic mice (D-TP), groups of diabetic mice wounds with Piper betel leaf extract (DS ), the wound group of diabetic mice with plasma medicine (DP) and the wound group of diabetic mice with plasma medicine and Piper betel leaf (DPS). The plasma medicine was treated on wound with condition non-contact style (the plasma jet did not touch the wound) with a distance of plasma jet reactor nozzle to the surface of wound about 20 mm, for 2 minutes, every day. Macroscopic observation of wounds is carried out every day from day 0 to 7. On day 7 it was seen that the size of the wound area for D-P-S was smaller than the other groups. The results of this study indicated that Piper betel leaf extract can potentially be used to optimize the performance of plasma medicine in accelerating diabetic wound healing during the proliferation phase. Further investigation, however, is important to be conducted to study the effect for all phases of wound healing and its mechanism histo-pathologically.
