Abstract

The inhibitory activity of heme polymerization of (1)-N-(2-nitrobenzyl)-1,10- phenantrolinium iodide and (1)-N-(4-nitrobenzyl)-1,10-phenantrolinium iodide have been done. This study aims to analyse the (1)-N-(2-nitrobenzyl)-1,10-phenantrolinium iodide and (1)-N-(4-nitrobenzyl)-1,10-phenantrolinium iodide as inhibitory of polimerization heme. Analysis of heme inhibtory polimerization activity used the experimental in vitro method. The activity showed by IC50 (the capable concentration of extract to inhibiting polymerization heme by 50% ). The IC50 value acquired by probit analysis. Assess IC50 of (1)-N-(2-nitrobenzyl)- 1,10-phenantrolinium iodide not to be identified, (1)-N-(4-nitrobenzyl)-1,10-phenantrolinium iodide and chloroquine by successively are 0,571±0,071; 25,498±1,876 mg/mL. The result showed the (1)-N-(4-nitrobenzyl)-1,10-phenantrolinium iodide had the highest value of the heme polymerization inhibitory activity than chloroquin, (1)-N-(2-nitrobenzyl)-1,10- phenantrolinium iodide hadn’t the heme polymerization inhibitory activity.

Abstract

Leunca (Solanum nigrum,L.) is one of the medicinal plant which used society for traditional medication as antipyretic, hypotensive and anticancer. The aim of  this research was to know an anticancer activity of ethanol extract of leunca leaves, emphasized on an ability to inhibit the growth Raji cell. Ethanol extract took from leunca leaf powder that epitomized using ethanol solvent by Soxhlet instrument. Citotoxicity test was done by incubating Raji cell at a density of 2x104 with treatment using ethanol extract from leunca (Solanum nigrum L.) leaf in several concentration 500; 250; 125; 62,5; 31,25; 15,62; 7,81 and 3,90 µg/ml. A test was done by MTT method and the percent of cell mortality was calculated. The LC50 values were calculated using probit analysis. The research continued with antiproliferation test on treatment sample concentration 50; 25 µg/ml with cell control for 24, 48, and 72 hours.The result of research indicate that ethanol extract of leunca leaves had cytotoxic effect towards Raji cell with LC50 values 59,22 µg/ml. The result of  antiproliferation test showed that there were the growth of inhibitation on treatment sample with doubling time values 69,56 hour at concentration 50 µg/ml; 60,00 hour at concentration 25 µg/ml, and doubling time cell control is 44,98 hour.

Abstract

Infeksi kandidiasis meningkat selama dekade terakhir di dunia. Peningkatan ini menandai munculnya isolat Candida albicans resisten Flukonazol. Diperlukan obat baru untuk mengatasi permasalahan resistensi ini.  Daun pacar kuku (L. inermis L.) secara empiris mempunyai aktivitas sebagai antijamur. Kandungan kimia daun pacar kuku larut dalam etil asetat. Penelitian ini bertujuan untuk mengetahui aktivitas antifungi dari fraksi etil asetat ekstrak daun pacar kuku (L. inermis L.) terhadap strain C. albicans sensitif Flukonazol dan C. albicans resisten Flukonazol. Fraksi etil asetat ekstrak daun pacar kuku (L. inermis L.)dibuat dengan memfraksinasi ekstrak etanol dengan  pelarut etil asetat.Konsentrasi fraksi etil asetat ekstrak daun pacar kuku yang digunakan terhadap C. albicans sensitif Flukonazol dan C. albicans resisten Flukonazol sebesar 10%; 5%; 2,5%; 1,25% dan 0,625%. Pengujian aktivitas antifungi fraksi etil asetat ekstrak daun pacar kuku menggunakan metode mikrodilusi cair dengan parameter pengukuran Minimum Inhibitory Concentration (MIC) dan Minimum Fungicidal Concentration (MFC). Nilai MIC dan MFC yang diperoleh sama yaitu sebesar 1,25% untuk C. albicans resisten Flukonazol dan 0,625% untuk C. albicans sensitif Flukonazol. Rasio MFC terhadap MIC yang diperoleh sebesar 1 yang menyatakan bahwa fraksi etil asetat ekstrak daun pacar kuku memiliki daya fungicide. Skrining fitokimia fraksi etil asetat ekstrak daun pacar kuku mengandung fenolik dan kuinon.

Abstract

Validasi metode analisis (VMA) penentuan kandungan β-karoten dalam Spirulina maxima perlu dilakukan agar didapatkan hasil yang valid dan dapat dipertanggung jawabkan secara ilmiah. Tujuan penelitian adalah membuktikan metode analisis penetapan kadar β-karoten dalam ekstrak etanol 96% Spirulina maxima menggunakan spektrofotometer visibel memenuhi syarat validitas. Parameter yang digunakan dalam VMA diantaranya spesifisitas, linieritas, akurasi, presisi, limit deteksi dan limit kuantitas. Syarat yang harus dipenuhi ditiap parameter yaitu nilai korelasi (r) ≥ 0,98 untuk linieritas, persen perolehan kembali masuk dalam range 98-102% dan nilai RSD ≤ 2% untuk akurasi, nilai RSD ≤ 2% untuk presisi, profil spektragram spektrofotometri visibel yang sama antara sampel dan standar untuk uji spesifisitas. Hasil penelitian dengan parameter validasi diantaranya linieritas dengan nilai r=0,998, persen perolehan kembali dalam rentang 98-101%, presisi keberulangan sistem dengan RSD 1,14 %, presisi antara 0,99 % dan uji spesifisitas sesuai profil spektragram spektrofotometri visibel yang sama antara sampel dan standar. Nilai LOD yang diperoleh yaitu 1,656 ug/mL dan LOQ 5,017 ug/mL. Berdasarkan hasil tersebut maka analisis penetapan kadar β-karoten dalam ekstrak etanol 96% Spirulina maxima dengan metode spektrofotometri visibel memenuhi syarat validitas menurut Petunjuk Operasional Penerapan CPOB tahun 2013.

Abstract

Akar pasak bumi mengandung senyawa kuasinoid diduga mempunyai efek penghambatan karsinogenesis, antiulcer, dan antimalaria. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian eksktrak etanol akar pasak bumi terhadap ekspresi p53 pada kanker payudara tikus betina yang diinduksi DMBA.Tikus dibagi menjadi 6 kelompok. Kelompok I, II dan III diberi ekstrak etanol akar pasak bumi dosis berturut-turut 100; 200 dan 400 mg/kg BB. Kelompok IV diberi larutan DMBA 20 mg/kg BB. Kelompok V diberi corn oil. Kelompok VI sebagai baseline. Tikus yang mengalami kanker, dihitung jumlah nodul tumornya dan pada minggu ke-23 semua tikus dikorbankan dan diambil jaringannya untuk pengamatan secara mikroskopik menggunakan metode HE dan imunohistokimia. Hasil penelitian menunjukkan bahwa rerata ekpresi p53 mutant pada pemberian ekstrak berturut-turut 100, 200 dan 400 mg/kg BB adalah 6,375 ± 4,07; 11,25 ± 16,53; dan 0,875 ± 1,75; kelompok  IV (DMBA) sebesar 13,125 ± 14,4; kelompok V (corn oil) adalah 1,375 ± 1,55; kelompok VI (baseline) adalah 0,375 ± 0,75. Hasil penelitian disimpulkan bahwa ekstrak etanol akar pasak bumi (Eurycoma longifolia Jack) dosis 400 mg/kg BB yang diberikan sebelum dan selama induksi DMBA mampu menurunkan ekspresi protein p53 mutant (proapotosis) dan mampu menghambat pertumbuhan kanker payudara tikus betina Sprague Dawley yang diinduksi 7,12-dimetilbenz(α)antrasen (DMBA).Kata kunci:  akar pasak bumi (Eurycoma longifolia Jack), kemopreventif, imunohistokimia, ekspresi p53

Abstract

Tea (Camellia sinensis  (L.) O.K.)  is one of medical plant traditionally used by society as anticancer. The aim of this research is to evaluate the citotoxic and antiproliferative effect of ethanolic fraction of tea (Camellia sinensis  (L.) O.K.) and its effect on P53 and bcl-2. Tea were collected and extracted by infundation and fractioned with ethanol as solvent. Citotoxicity test was done by incubating HeLa cell at a density of 2.104 with treatment using extract in concentration of 250; 125 ; 62,5; 31,25; 15,63; and 7,81 µg/mL during 24 hours.  Antiproliferative test were done by calculating doubling time which was equaling a life cell on treatment sample concentration 31,25 µg/mL; 15,63 µg/mL; 7,81 µg/mL; and 3,91 µg/mL with cell control at 24, 48 and 72 hours.  Imunohistochemistry assay was used in LC50  with p53 and bcl-2 antibody. P53 and bcl-2 expression is compared to control. The result indicated that ethanol fraction of tea (Camellia sinensis  (L.) O.K.) leaf infuse has citotoxicity effect on HeLa cell with LC50 of 24,45 µg/mL. The result of  doubling time test indicated that doubling time value for a solvent  control is 74,11 hours and a cell control is 78,22 hours. While, with treatment 31,25 µg/mL; 15,63 µg/mL; 7,81 µg/mL; and 3,91 µg/mL resulted on negative slope then did not produce doubling time. The extract can induce p53 expression and inhibit bcl-2  expression compared to control.

Abstract

Pasak bumi (Eurycoma longifolia) has the potential to be developed as antihypertensive, antipyretic, aphrodisiacs and health supplements. The use of E. longifolia as a traditional medicine needs to be pursued in the form of more effective and appropriate formulation. The capsule preparations are easy to make and cancover the bitter taste of E. longifolia. Clinical trials in this study use design pre-post treatment in healthy humans. Subjects used were male - healthy men and healthy women who met inclusion criteria and were subjected with formulated capsule for 14 days. The study resulted capsule formula comprising of the ethanolic extract of E. longifolia 300 mg, vivapur 101 300 mg, 58 mg maydis starch, aerosil 3%, talc 2%, and Mg stearate 1%. The results showed that the capsule of E. longifolia did not affect the value of heart rate, respiration rate, body temperature and weight (P > 0.05), based on paired t-test, but they causes a decrease in blood pressure of healthy human. The ethanol extract of E. longifolia caused vasodilation of blood vessels that can be used in antihypertensive therapy.

Abstract

Pneumonia is an acute inflammation of the pulmonary parenchyma that can be caused by Klebsiella pneumoniae. This study aims to determine the active fraction of cayenne pepper leaves on the growth of K. pneumoniae. Cayenne pepper leaf which previously defatted using n-hexane was macerated with 95% ethanol, then fractionated successively with dichloromethane, ethyl acetate and methanol. Ethanol extract and each fraction with concentration of 40% were tested for their antibacterial activity against K. pneumoniae using disc diffusion method (Kirby-Bauer). 1% amoxicillin was used as positive control and Dimethyl sulfoxide (DMSO) as negative control. The Minimum Inhibitory Concentration (MIC) of the most active fraction was then determined. Determination of antibacterial compound in the most active fraction was carried out by TLC-bioautography and followed by Gass Chromatography Mass Spectrophotometry. Cell leakage analysis was performed using UV spectrophotometry to detect the release of protein and nucleic acid, as well as Atomic Absorption Spectrophotometry was used to detect ion release of K+ and Ca2+. The results showed that the most active fraction against K. pneumoniae was the ethyl acetate fraction with MIC value of 10% and inhibition zone of 7.25±0.25 mm. TLC-Bioautography of ethyl acetate fraction with eluen n-hexane: ethyl acetate (6:4) obtained an active stain at Rf 0.12. Compounds having 94% similarity with 1-propanol, 2-amino was predicted as the active compound.

Abstract

Type 2 diabetes mellitus is a non-contagious disease, can damage the glucose metabolic system in the body, and is characterized by hyperglycemic conditions. Moringa leaves (Moringa oleifera) and Andrographis herbs (Andrographis paniculata) have been studied to have antihyperglycemic activity in diabetic rats. The purpose of this study was to determine the effect of the combination of Moringa leaf ethanol extract (MLEE) and ethanol extract of Andrographis herbs (AHEE) in streptozotocin-induced rats (45 mg/kgBW). The subjects consisted of 32 rats, divided into 8 groups (1 healthy group and 7 type 2 DM groups). The study was conducted for 4 weeks by measuring blood glucose levels in pre-treatment, day 0, 14 and 28 with a single MLEE treatment dose of 300 mg/kgBW, single AHEE 300 mg/kgBW, combination of MLEE and AHEE 150+150 mg/kgBW, 200+100 mg/kgBW, 100 + 200 mg/kgBW, and gliclazide 5 mg/kgBW orally. The results of measurement of fasting blood glucose levels on day 28 showed that administration of gliclazide 5 mg/kg BW, single dose MLEE and AHEE, as well as its combination, had significant differences (p<0.05) compared to the hyperglycemic control group. Pancreatic organ histopathology features in the extract dose group showed that there was a change in the repair of insula Langerhans compared to the hyperglycemic control group which showed necrotic damage due to streptozotocin induction. Combination administration has the same antihyperglycemic effect by single dose extract in diabetic rats within 28 days, which also restore weight loss to normal.

Abstract

The roots of Eurycoma longifolia, Jack has a lot of beneficial activity on human health. The use of Eurycoma longifolia, Jack roots as a traditional medicine should be made in the form of a dosage that is more effective, safe, and had no side effects, especially on hematology. Easy preparations are capsule. This study aimed at identifying the effect of ethanol extract capsule of Eurycoma longifolia, Jack roots on hematology of healthy volunteers. Healthy volunteers were assigned in the study, each of which were 10 male and 10 female who met the inclusion criteria. Both groups were given ethanol extract capsule of Eurycoma longifolia, Jack roots for 14 days, with the dose of 300 mg extract once a day, after meal, at night. Hematology examination was performed on day 0; 14 and 42. Results were compared statistically using Repeated ANOVA and Friedman tests. The result of the study indicated that the average value of hematology at the examination day of 0; 14; and 42, there were significant differences in MCV and leukocyte parameters in healthy male volunteers (p<0.05). MCH, LED and monocyte parameters in healthy female volunteers had a significant difference of (p<0.05). However, the parameter values were still within the normal range. Monitoring results showed that some of the side effects were frequent urinary, dizziness, nausea, sweating, insomnia, and increasing appetite on healthy male volunteers. Meanwhile, on healthy female volunteers, there were frequent urinary, insomnia, constipation. Having ethanol extract capsule of the Eurycoma longifolia, Jack roots did not affect the hematology parameters of healthy male and female volunteers, but it caused some side effects.
