Abstract

Radikal bebas bersifat reaktif yang menyebabkan penyakit degeneratif sehingga tubuh memerlukan antioksidan eksogen. Salah satu contohnya adalah buah Jamblang (Syzygium cumini (L)) yang memiliki aktivitas antioksidan tinggi karena antosianin alaminya.. Antosianin bersifat lebih stabil dalam suasana asam sehingga cocok jika ekstrak buah jamblang difortifikasi dalam sediaan yoghurt. Yoghurt merupakan produk fermentasi susu yang menggunakan bakteri asam laktat yaitu: Streptococcus thermophilus, Lactobacillus bulgaricus, Lactobacillus acidophilus. Tujuan penelitian ini untuk mengetahui kadar antosianin ekstrak buah jamblang pada formulasi yoghurt dengan konsentrasi 0%, 5%, 7,5% dan 10% serta syarat mutu SNI. Pengujian meliputi uji sifat fisika, kimia dan mikrobiologi yaitu sensoris, viskositas, kadar asam, kadar lemak, dan jumlah starter bakteri, serta uji stabilitas yoghurt dengan penyimpanan pada suhu 4°C dan uji total antosianin pada hari ke-1, ke-7, ke-14, 21 dan hari ke-28. Hasil penelitian menunjukkan bahwa kadar antosianin total pada formula yoghurt terfortifikasi ekstrak buah jamblang pada konsentrasi 10% (60.87±2.24 mg/L) menghasilkan stabilitas antosianin lebih baik, sesuai dengan persyaratan SNI.

Abstract

Soap is obtained from the saponification reaction that occurs between oil and alkaline solution. Various kinds of oils used in soap making also affect the quality of soap. The purpose of this study is to determine the effect of oil type on the physical properties of liquid bath soap. This research was divided into 3 groups, coconut oil group (A), VCO oil group (B), and olive oil group (C). Liquid bath soap obtained from each type of oil is tested based on SNI provisions including a soap quality test (oil characteristics test, organoleptic test, pH test, specific gravity, total plate count), viscosity test, and antioxidant test. Liquid bath soap of the three types of oil has the same shape, color, and odor. Coconut liquid soap, VCO, and olives respectively have a pH value of 6.64; 6.9; 6.52. (P = 0,000) Specific weight 1.0511; 1.0427; 1.0425. Viscosity 558cps; 686; 420. (p = 0.003) IC50 3547.36; 4523.46; 3542.02. (p = 0.035). Bath soaps that best meet SNI requirements are those made with olive oil.

Abstract

 The Eurycoma longifoliais bitter therefore, to address this problem, it is prepared in a capsule form. The capsule dosage form  must has  the parameters of good quality, thus expected to increase the potential use of biodiversity and the interest of the community in consuming the medicine from the natural materials. The purpose of this research is to get the capsule product of ethanol extract of the Eurycoma longifolia root of quality through the test of total plate value and yeast yeast contamination, the determination of the metal content of Pb, Hg, Cd and the level of flavonoids released in the capsule dissolution test of ethanol extract Eurycoma longifolia root. The result of this research shows that the dissolution efficiency (DE120) capability of ethanol extract of Eurycoma longifolia root is 81,04±2,38%. Test of heavy metal content in capsule of  of ethanol extract of Eurycoma longifolia root found level of Pb equal to 4,132 ppm and not detected content of Cd and Hg in capsule. This indicates that the capsule meets the requirements set by BPOM RI. Tests for total plate numbers (ALT) and yeast capsule ethanol extract of the E. longifolia root were each obtained data of 70 x 102 colonies / g and 4 x 102 colonies / g. It also shows that the capsules meet the requirements by BPOM RI.

Abstract

The present study tried to investigate potency of L. sarmentosa root extract on quality and viability of mice sperm.This study used 25 Swiss male mice were randomly divided into five experimental groups of 5. The ethanolic extract of L. Sarmentosa (100, 200, 400 mg/kg b.w/days), control and X-gra were administered orally for 14 days. The animals were evaluated for sperma quality (sperm count and sperm motility) and viability at 14 days treatment. The results showed that administration of L.sarmentosa roots extract significantly increasing the sperm count (970x106cell/mL) at 400 mg/kg b.w/days (P 0.0040). The extract dose of 200 mg/Kg b.w and 400 mg/Kg b.w had the highest motility values (95%). Viability of sperm are 83.8%; 96.0%; 87.8% respectively at a dose of 100 mg/kg b.w, 200 mg/kg b.w and 400 mg/kg b.w. Present finding oral administration of L. sarmentosa roots extract has a potency of the quality and viability of mice sperm.

Abstract

Pasak Bumi roots contain quasinoid compounds that have carcinogenesis inhibitory, imunomudulator, antiulcer, and antimalaria effects. This study aims to determine the effect of a combination of ethyl acetate fraction of Pasak Bumi roots and doxorubicin on DMBA-induced white mice by weight and palpation observation. The mice were divided into 8 groups. Each group consisted of 7 female Sprague Dawley mice. Group 1 (normal) was only given food and drink, Group 2 was given fraction of ethyl acetate of Pasak Bumi roots (100 mg/kg body weight, Group 3 (positive control) was given doxorubicin (1.17 mg/kg body weight) 1 times a week for 5 weeks, Group 4 was given DMBA (20 mg/kg) two times a week for 5 weeks, Groups 5 was given DMBA (20 mg/kg) + doxorubicin (1.17 mg/kg), Group 6 was given DMBA (20 mg / kg) + ethyl acetate fractions of Pasak Bumi roots (100 mg / kg), Group 7 was given DMBA (20 mg / kg) + doxorubicin (1.17 mg / kg) + ethyl acetate fraction of Pasak Bumi roots (100 mg / kg) and Group 8 doxorubicin ( 1.17 mg / kg body weight) + ethyl acetate fraction of Pasak Bumi roots (100 mg / kg). Observations were made on the basis of body weight of mice and tumor incidence by palpation at week 6 of treatment. The result of  this research shows that the administration of ethyl acetate fraction of Pasak Bumi roots and doxorubicin combination can increase body weight of the mice and decrease the number of tumors compared to single doxorubicin.

Abstract

Kencur (Kempferia galanga, L) is one of the rhizomes that is often used as a constituent component in traditional medicinal formulas. One of the kencur's pharmacological activities is anti-inflammatory. The active compound as an anti-inflammatory is ethyl p-methoxycinnamate (EPMC). This study aims to determine the content and ascertainment of EPMC compounds and the anti-inflammatory activity of kencur rhizome extract. Kencur extract can be obtained by the maceration method using 96% ethanol solvent. The EPMC content determination was held using the TLC-densitometry method, while the EPMC compounds were confirmed using the GC MS method. The anti-inflammatory activity test was done using inhibiting protein denaturation methods. The extract results obtained 12.67% yield. The result of EPMC content in kencur extract was 10.05 ± 4.57. The GCMS kencur extract results showed an abundance of EPMC compounds at a retention time of 7.088 with a peak area of 72290779. The results of IC50 anti-inflammatory for the standard substance (EPMC) were 5.306 ± 5.028. The results of IC50 anti-inflammatory the sample (kencur extract) was 303.487 ± 1.201. Ethanol extract of kencur contains ethyl p-methoxycinnamate (EPMC) and has anti-inflammatory activity by inhibiting protein denaturation.

Abstract

Black cumin seeds contain oil classes atisiri, terpenes, and alkaloids that can beused for traditional medicine as anticancer. The purpose of this study was to assess thecytotoxic effects of ether extract, ethanol, and infusa seed Nigella sativa, L. (Blackcumin) to inhibit the growth of T47D and normal (Vero) cells and its effect on thekinetics of T47D breast cancer cells proliferation. This study were used extracts ofether, ethanol, and black cumin seeds infusa that obtained by maceration method andinfundasi. Cytotoxicity test was performed by incubating T47D breast cancer cells at a2 x 104 density with concentrations series of 2000; 1000; 500: 250: 125; 62.5; 31.25and 15.625 microg/ml for 24 hours. Vero cell with a density of 2 x 104 withconcentrations series of 4000; 2000; 1000; 500; 250; 125 and 62.5 microg/ml. Thenumber of cells was calculated by direct counting method and the calculated the deathpercentage. The LC50 values calculated using probit analysis. Observations on thenature of the growth inhibition were done by observing kinetics of cell proliferationwith the addition of trypan blue at-24, 48 and 72 to determine its doubling time. Theresults showed that the ether extract, ethanol, and black cumin seeds infusa arecytotoxic to T47D breast cancer cells with successive LC50 of 32.63: 10.02, and 23.82mg mL. Vero cell cytotoxicity test to produce successive LC50 of 300.6; 328.41, and778.64 g/ml. Antiproliferative test results showed that in 62.5 ug/ml and 31.625microg/ml prolong the doubling time. Ethanol extract of cumin seeds have a higherpotential due to the highest security index compared to ether extract and infusa.

Abstract

Cancer is the formation of new tissue which is abnormal and malignant. A group of cells suddenly become disorganized and reduplicate themselves rigorously (hyperproliferation). Nigella sativa L. is one of the herbs which have an anticancer effect. This research aims to assess the cytotoxic and antiproliferative effect of Nigella sativa L. ethanol extract of Myeloma cells. Ethanolic extract was produced from Nigella sativa L. powder with maseration method. The cytotoxicity test was done by incubating Myeloma cells with the treatment concentration group of N. sativa L. ethyl acetic fraction of ethanolic extract 2000; 1000; 500; 250; and 62.5 µg/ml, respectively. The test was done with an MTT method and then with a calculation of its death percentage. The LC50 is calculated using a probit analysis method. The test was then continued with the antiproliferative test to assess the doubling time at treatment concentration 125; 62.5 µg/ml and cellular control at hours 24, 48, and 72. The results showed that Nigella sativa L. ethanolic extract had cytotoxic activity towards the Mieloma cells with an LC50 value 177.01 µg/ml. The antiproliferative test showed that there was a growth inhibition, even cell death at the extract treatments. The doubling time was 253 hours at 62.5 µg/ml concentration, 298.4 hours at 125 ug/ml, while the cell control had 54.52 hours

Abstract

Roselle calyx (Hibiscus sabdariffa L) is widely used as an antioxidant, antihepatotoxic, diuretic, anti-cholesterol, and immunostimulant. Roselle contains anthocyanin and quercetin that have antioxidant activity. This study aimed to determine the effects of the ethanolic extract of roselle calyx on hematocrit, platelets, and erythrocytes in healthy volunteers during the 30-day administration and 15 days after it ceased. This study employed pre- and post-treatment design on 21 healthy volunteers, comprising 11 males and 10 females who fulfilled the inclusion criteria and filled out the informed consent form willingly. Volunteers were provided with roselle capsules for 30 days with a dosage of 500 mg a day taken 10-15 minutes after dinner. The hematologic examination was performed on Day 0, 31, and 45, including the measurement of hematocrit, platelet, and erythrocyte levels using a hematology analyzer. The obtained data were analyzed in SPSS with paired t-test and Wilcoxon test. The results showed that the hematocrit, platelets, and erythrocytes of all male and female volunteers on Day 0, 31, and 45 were within the normal range. The comparison analysis affirmed that there was no significant difference between these three parameters from Day 0 to 31, Day 31 to 45, and Day 0 to 45 (p>0.05). As a conclusion, the administration of the ethanolic extract of roselle calyx with a dosage of one 500mg capsule per day for 30 days did not affect the hematocrit, platelets, and erythrocytes in healthy volunteers. There was also no delayed effect on these three hematological parameters 15 days after the administration stopped.

Abstract

The free radical was produced when undertaking processes metabolit or opposedinfection and when the body dissolved food. The unstable free radical could beneutralised with antioxidant. The root of the pasak bumi (Eurycoma longifolia Jack)was known contained the compound flavonoid, that it was known had the activity asantioxidant. This research aimed at knowing the activity capacity of the capture of thefree radical the water fraction and the faction ethyl acetate fraction the extract ethanolof the root of the pasak bumi by using the DPPH method.The root of the pasak bumi was extracted with ethanol 70% used maceration. Theethanolic extract was dissolved in ethyl acetate, fractination with ethyl acetate so as tobe received by the fraction ethyl acetate. The concentration of the ethanolic extract andthe fraction of ethyl acetate that was used were 2; 4; 8; 16 ug/mL and isolat-1 that is:0.8; 1.6; 3.2; and 6.4 ug/mL.Results of the research showed that all the treatments had the activity asscavenging the free radical. Results of the analysis statistika with the Kruskal Wallismethod in the level of the confident 95% that was followed by the test Mann Whitneyshowed the existence of the difference of the activity of the capture of the free radicalwho was significant between respectively the treatment group. The conclusion from thisresearch was the value ES50 ethanalic extract (15.64 ug/mL) bigger than the factionethyl acetate fraction and higher (13.948 ug/mL) value of ethanalic extract was thanisolat 1 (3.961 g/mL).
