Abstract

The research was conducted using completely randomized design by divided 24 Texel sheeps into 4 treatments, each treatment consisted of 6 repetitions. The treatment groups consisted of: P0 (sheeps were given CMC-Na 0,5% solution), P1 (given meniran extracts at 5% concentration), P2 (given meniran extracts at 10% concentration) and P3 (given meniran extracts at 20% concentration). Administration of treatment between groups using a syringe (20 ml). The data were analyzed statistically using the Analysis of Variance (ANOVA) and continued with Duncan's test. The economic value of feed is obtained from the feed conversion value multiplied by the feed price, where the feed conversion is obtained from the ratio between the amount of feed consumption and body weight gain. The income over feed cost obtained through the difference between income and total feed costs, where the total feed price is obtained from the cost of Texel sheep feed plus the cost of meniran extract (Phyllanthus niruri Linn). P1 with meniran extract 5% gives the best result. It can be conclude that the addition of 5% meniran extract could reduce the economic value of feed and increase income over feed costs on Texel sheeps.

Abstract

Repeat breeding is a condition in which cows fail to conceive after being bred three or more times. This study aims to determine the quality of estrus, non-return rate (NRR) and conception rates obtained after cows were treated with 2% povidone iodine. The study involved 30 Holstein Friesian cows, each having calved three times and experiencing repeat breeding. Cows in estrus were treated with 10 mL of 2% povidone iodine intrauterinely, followed by an intramuscular injection of 2 mL (250 mg/head) of prostaglandin F2α (PGF2α, Luteosyl) five days later. Estrus quality was observed by examining changes in vulvar mucosa color, mucus discharge, and vulvar temperature. Pregnancy diagnosis was made at 60 days. Results showed that 43.3% of cows had estrus quality score 3 (cows showed dark red vulvar mucosa, transparent mucus, temperature 38.9 ± 0.5 ºC), 23.3% had score 2 (cows showed reddish vulvar mucosa, some cloudy mucus, temperature 38.7 ± 1.0 ºC), and 33.3% had score 1 (cows showed pink vulvar mucosa, cloudy mucus, temperature 38.6 ± 0.5 ºC). Of the 30 cows, 20 had a non-return rate (NRR) of 66.7%. Based on these results, it can be concluded that 2% povidone iodine has a positive effect on estrus quality, NRR, and conception rates in dairy cows experiencing repeat breeding.

Abstract

This study aimed to evaluate the association of factors such as nutrition, thermal humidity index (THI), age, and lactation period with the occurrence of repeat breeding of dairy cows. The sample consisted of Holstein Friesian cows aged 1-11 years, with lactation periods ranging from 1 to 7 months, currently in lactation, and experiencing repeat breeding (inseminated three or more times without conception, despite exhibiting normal estrous cycles and intervals). The results showed that feeding practices by farmers did not significantly differ (p >0.05) in relation to the incidence of repeat breeding. THI values recorded in several barns also showed no significant differences (p >0.05), with the cows experiencing light to moderate heat stress. Similarly, the frequency of artificial insemination and the estrous cycle intervals remained within the normal range. The highest incidence of repeat breeding was observed in cows aged 2.5 to 5.5 years, accounting for 78.13%, and during lactation periods of 1-3 months, accounting for 81.26%. It can be concluded that there were no significant differences in the provision of forage and leguminous feed between normal cows and those with repeat breeding, and the crude protein content of forage did not influence its occurrence. Additionally, there was no indirect relationship between THI and repeat breeding. However, the incidence of repeat breeding tended to increase in cows aged 3-4 years, and during the second and third lactation periods.

Abstract

Anti-Müllerian hormone (AMH) has been identified as a potential biomarker for assessing ovarian reserve and reproductive capacity in cattle, with high heritability and relatively stable expression. However, studies detecting AMH mRNA in bovine serum remain scarce due to the matrix’s inherently low RNA yield, susceptibility to degradation, and potential presence of PCR inhibitors. This study provides an exploratory validation of AMH mRNA detection in serum — a challenging matrix compared with ovarian tissue — using EvaGreen-based reverse transcriptase–quantitative PCR (RT–qPCR). Specific primers for the AMH and β-actin genes were designed in silico and validated through melting curve analysis and linearity testing. The results showed high amplification efficiency (AMH: 100.2%, R² = 0.994; β-actin: 109.1%, R² = 0.996), with specific amplification of both targets. AMH detection in serum samples was successful in some samples, while the β-actin gene was consistently amplified as a reference gene. Despite the low RNA quality from serum and the presence of organic contaminants, the method demonstrated its feasibility for detecting AMH transcripts in a minimally invasive manner. Physiologically, AMH levels positively correlate with antral follicle count, superovulation success, and embryo quality, and are sensitive to heat stress and other environmental factors. These findings provide a foundational basis for developing molecular diagnostic approaches based on AMH gene expression in cattle reproductive management programs and support the future development of efficient, accurate, and context-specific biomolecular-assisted selection technologies for tropical livestock systems.
