Abstract

Spons merupakan fauna laut yang diketahui memiliki berbagai senyawa bioaktif. Senyawa tersebut berpotensi sebagai antibakteri, antivirus, dan antikanker. Penelitian ini bertujuan mempelajari aktivitas sitotoksik dan apoptosis ekstrak spons spesies A anggota ordo Astrophorida terhadap sel HeLa. Pada penelitian ini pengujian aktivitas sitotoksik ekstrak etanolik, metanolik, dan kloroform spons spesies A terhadap sel HeLa dilakukan menggunakan MTT assay dan uji apoptosis menggunakan double staining, yaitu etidium bromida-acridine orange. Deteksi golongan senyawa yang terkandung di dalam spons spesies A dilakukan menggunakan Kromatografi Lapis Tipis (KLT). Hasil uji sitotoksisitas menunjukkan bahwa ekstrak etanolik, metanolik, dan kloroform spons spesies A masing-masing memiliki nilai IC50 sebesar 18,25; 27,87; dan 13,87 µg/mL. Ekstrak etanolik, metanolik, dan kloroform spons spesies A pada konsentrasi 31,25 µg/mL dapat menginduksi kematian sel melalui apoptosis masing-masing sebesar 35,3 ± 11,16%; 82,64 ± 16,21%; dan 86,76 ± 9,27%. Berdasarkan uji menggunakan KLT diketahui bahwa spons spesies A menggandung golongan senyawa alkaloid, flavonoid, fenol, dan terpenoid. Oleh karena itu, dapat disimpulkan bahwa ekstrak spons spesies A berpotensi untuk dikembangkan sebagai obat antikanker.Kata kunci: ekstrak spons spesies A, sitotoksik, apoptosis, sel HeLa

Abstract

Angina pektoris merupakan salah satu gejala awal yang menjadi penanda masalah kardiovaskuler berat seperti penyakit jantung koroner. Sementara gen PLA2G10 dipercaya memiliki peran, baik dalam progresi aterosklerotik yang menyebabkan deposit plak dan penyumbatan pada arteri koroner, atau justru sebaliknya yaitu antiaterogenik. Polimorfisme PLA2G10 pada titik SNP T512C (rs36072688) dan T-123/in1C (rs4003232) mampu mengubah level ekspresi gen PLA2G10 sebagai gen pengode enzim sPLA2-GX. Penelitian ini bertujuan untuk mengetahui genotip dari PLA2G10 pada titik SNP T512C (rs36072688) and T-123/in1C (rs4003232) para penderita angina pektoris dan mengkaji apakah penyakit disebabkan karena mutasi. Sampel penelitian berupa peripheral blood mononuclear cell koleksi Divisi Litbang Rumah Sakit Jantung dan Pembuluh Darah Harapan Kita. Sampel berjumlah 113, berasal dari penderita angina pektoris dengan dan tanpa plak pada arteri koroner. Hasil penelitian menunjukkan bahwa gen PLA2G10 pada titik SNP T512C (rs36072688) memiliki genotip homozigot wildtype (TT) dan T-123/In1C (rs4003232) memiliki genotip heterozigot (TC) melalui metode TaqMan® SNP Genotyping Assay, dan tidak ada polimorfisme yang ditemukan pada kedua grup sampel tersebut.

Abstract

Dengue fever is caused by the mosquito-borne virus termed (DENV). However, DENV-2 has been identified as the most prevalent amongst the Indonesian pediatric urban population, in contrast with the other four serotypes. Therefore, it is important to reduce severe infection risk by adopting preventive measures, including through vaccine development. The aim of this study, therefore is to use various in silico tools in the design of epitope-based peptide vaccines (T-cell and B-cell types), based on the DENV-2 envelope glycoprotein sequences available. Therefore, in silico methods were adopted in the analysis of the retrieved protein sequences. This technique was required to determine the most immunogenic protein, and is achieved through conservancy analysis, epitope identification, molecular simulation, and allergenicity assessment. Furthermore, B4XPM1, and KAWLVHRQW were identified from positions 204-212, while the 77 to 85 peptide region was considered the most potent T-cell and B-cell epitopes. The interaction between KAWLVHRQW and HLA-C*12:03 occurs with maximum population coverage, alongside high conservancy (96.98%) and binding affinity. These results indicated a potential for the designed epitopes to demonstrate high immunity against DENV-2.

Abstract

Indonesia merupakan negara penghasil kakao terbesar ketiga di dunia. Penelitian-penelitian yang telah dilakukan menunjukan bahwa kakao (Theobroma cacao L) mengandung senyawa polifenol antara lain katekin, epikatekin, proantosianidin, dan antosianin yang berpotensi sebagai senyawa antioksidan. Penelitian ini bertujuan untuk mempelajari potensi antioksidan bubuk kakao (Theobroma cacao L) fermentasi dan non fermentasi terhadap kadar malondialdehid (MDA) tikus putih. Penelitian menggunakan Rancangan Acak Lengkap faktorial dengan delapan perlakuan dan tiga ulangan. Perlakuan terdiri dari kelompok normal, kontrol positif, 3 kelompok uji bubuk kakao fermentasi dan 3 kelompok uji bubuk kakao non fermentasi masing-masing dengan dosis 0,2, 0,4, dan 0,8 gram /200 gram BB. Tikus diberi diet tinggi lemak selama 8 minggu, dan larutan bubuk kakao diberikan 4 minggu terakhir masa penelitian. Pengukuran kadar Malondialdehid (MDA) diukur pada minggu ke-4 dan 8 masa penelitian. Hasil penelitian menunjukan bahwa kandungan total fenol bubuk kakao fermentasi lebih besar dari pada bubuk kakao non fermentasi. Pemberian bubuk kakao fermentasi secara oral mampu menurunkan kadar MDA, sedangkan bubuk kakao non fermentasi belum dapat menurunkan kadar MDA.

Abstract

Tikus diberi diet tinggi lemak selama 8 minggu, dan larutan bubuk kakao diberikan 4 minggu terakhir masa penelitian. Pengukuran uji profil lipid darah dilakukan pada minggu ke-0, 4 dan 8. Hasil penelitian menunjukan bahwa pemberian bubuk kakao fermentasi secara oral mampu menurunkan kadar kolesterol total, trigliserida, LDL, serta meningkatkan kadar HDL.

Abstract

Head and neck cancer is one of the deadly types of cancer in Indonesia. The main cause of this cancer is the consumption of alcohol and cigarettes. Head and neck cancer attacks the lips, mouth, palate, pharynx and larynx. Studies about head and neck cancer have been carried out in westerncountries, while in Indonesia is still limited. Earlier studies in western countries expressed that head and neck cancer is more common in men than women. Purpose of research were to determine the epidemiology of head and neck cancer in Indonesia related to patient ratio of men and women andthe correlation of stage and differentiation level of head and neck cancer. Data were obtained from the Rumah Sakit Kanker Dharmais-Pusat Kanker Nasional Jakarta and then analyzed descriptively. The data were analyzed came from 36 patients with head and neck cancer. The results showed, menwith head and neck cancer as much as 52,77% and females 47,22%. There were 16 cases with stage IV cancer, 9 with stage III, 8 with stage II and 2 with stage I. There were 6 cases of stage IV cancer with better differentiation, and there were 2 cases of stage II cancer with a poor differentiation. Head and neck cancer is more common in men than women. There was no correlation between the degree of differentiation-stage head and neck cancer. Abstract

This study was to determinte of development ovarian wader pari fish (Rasbora lateristriata) as part of the process of reproduction, which is the basic for information to R. lateristriata management. This study was done in october 2014 - april 2015 at Histology and Embryology Animal Laboratory of Gadjah Mada University. The object of the research was larva R. lateristriata with average weight of 0.02 grams. They were kept during 3 months. The result data of histology were analized and observed discussed descriptively. The results of observation showed that ovarian R. lateristriata indicated asincronous development patterns. The development of ovarian in the 1st month was at the chromatin nucleolar phase, while perinucleolar phase appeared in the 2nd and 3rd month.

Abstract

Marine actinomycetes is a group of bacteria that is highly potential in producing novel bioactivecompound. It has unique characteristics and is different from other terrestrial ones. Extreme environmentalcondition is suspected to lead marine actinomycetes produce different types of bioactive compoundfound previously. The aim of this study was to explore the presence and diversity of NRPS genes in 14anticancer-producing actinomycetes isolated from marine sediment in Indonesia. PCR amplificationand restriction fragment analysis of NRPS genes with HaeIII from 14 marine actinomycetes were doneto assess the diversity of NRPS genes. Genome mining of one species of marine actinomycetes (strainGMY01) also was employed towards this goal. The result showed that NRPS gene sequence diversity in 14marine actinomycetes could be divided into 4 groups based on NRPS gene restriction patterns. Analysisof 16S rRNA gene sequences of representatives from each group showed that all isolates belong to genusof Streptomyces. Genome mining result showed that strain GMY01 harboring 10 different NRPS geneclusters that encode secondary metabolites, as pure NRPS or hybrid between NRPS and other compounds.These results indicated that marine actinomycetes having a high potential to be developed as source ofanticancer drugs development.

Abstract

White spot syndrome virus (WSSV) is a devastating viral disease in shrimp aquaculture. Infection ofWSSV in penaeid shrimps affects immune defense and changes gene expression. PmVRP15 has been reported as a part of the WSSV propagation pathway that is highly up-regulated in hemocytes at the acute phase of WSSV infection. This study analyzed the expression of PmVRP15 in local populations of giant tiger shrimp (Penaeus monodon) to be associated with susceptibility to WSSV. Tested populations consisted of an inbreeding population (G8) and outbreeding population (G8iA) from Jepara, Indonesia. Susceptibility was determined by cumulative mortality, median lethal time (LT50), and severity of infection at time of death. Though all populations were susceptible to WSSV, the frst mortality in G8 occurred at 18 hours post-infection (hpi) with mild infection, while frst mortality of G8iA occurred at 30 hpi with severe infection. The LT50 of G8 was signifcantly lower than that of G8iA, indicating that G8iA was less susceptible to WSSV than G8. Relative PmVRP15 transcripts of G8iA were insignifcantly down-regulated, whereas relative PmVRP15 transcripts of G8were insignifcantly upregulated. Although it’s still not conclusive, the results of this study suggest that PmVRP15 has weak potentialas a WSSV susceptibility marker in G8 and G8iA broodstock selection.

Abstract

Acacia auriculiformis is a plant which often found in Indonesia. Acacia leaves have been known to have high antioxidant content. The phenolic compound of A. auriculiformis is the largest of these plants and phenolic compound have potential as anticancer protective agents. The aims of this study were to determine the phenolic compound, antioxidant activity, cytotoxic and antiproliferative activity of acacia leaves extract against T47D and MCF-7 breast cancer cell lines. Extraction was carried out by maceration method using methanol as a solvent. Phenolic compounds were examined by Thin Layer Chromatography (TLC) and antioxidant activity was measured using DPPH assay, followed by cytotoxicity and antiproliferation test measured by MTT Assay to obtain the IC50 value and percent of antiproliferation. Based on TLC, A. auriculiformis methanol leaves extract contain a phenolic compound with Rf value of 0.85 compared to gallic acid.  Antioxidant activity A. auriculiformis with IC50 value of 9 ppm is classified as high antioxidant activity and MTT assay results for cytotoxicity with IC50 value of 273.8 and 31.26 ppm for T47D and MCF-7 cells, respectively. Antiproliferative analysis also showed high values for MCF-7 cells. These data have shown that methanol leaves extract of A. auriculiformis has high antioxidant activity and inhibited MCF-7 cell proliferation but not T47D breast cancer cell lines.
