Abstract

Background: Murraya koenigii (Curry) leaves are herbal plants that have bioactive compounds such as phenolics, flavonoids, and alkaloids that function as antioxidants and anti-cancers. Aims: This study aimed to quantitatively determine the bioactive compounds in curry leaves by calculating phenolic, flavonoid, and alkaloid content. Its potential as an antioxidant and anti-proliferative compound in T47D breast cancer cell lines was also analyzed. Methods: Murraya koenigii leaves extract was obtained by maceration using 80% methanol (1:5 w/v), then the antioxidant and anti-proliferative test was carried out using the 1,1-Diphenyl-2-Pycrylhidracyl (DPPH) and MTT (-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide) assay, respectively, followed by flow cytometry to determine the apoptotic activity of the extract on T47D breast cancer cell lines. Results: The results for the bioactive compounds in the methanol extract of Murraya koenigii leaves were 156.62±1.49 mg/g phenolics, 99.19±0.25 mg/g flavonoids, and 2.90±0.01 mg/g alkaloids. The IC50 value for antioxidant activity was 25.058±2.2 μg/ml and showed an anti-proliferative effect on T47D cells in a dose-dependent manner with IC50 74.71±5.45 μg/ml for cytotoxicity. Furthermore, flow cytometry showed that a concentration of 1/16 IC50 has the best results for apoptosis. Conclusion: The methanol extract of Murraya koenigii leaves has the potential as an antioxidant and anti-proliferative agent and can induce apoptosis of T47D cells.

Abstract

Leptospirosis presents with nonspecific clinical features and requires time‐consuming laboratory tests for gold standard diagnosis. This study aims to design and characterize the recombinant LipL32 from synthetic gene and assess its performance as an antigen for detecting leptospirosis. The antigen was developed by cloning the LipL32 gene conserved portion of Leptospira interrogans serovar Icterohaemorrhagiae strain Langkawi. The immunoinformatic was used to characterize the developed rLipL32. Western blot results using anti‐histidine revealed a band of rLipL32 protein at ~40 kDa. Subsequently, it was used to examine the IgM antibody on human sera by using ELISA. The IgM‐LipL32 ELISA was evaluated using 67‐positive and 25‐negative sera and compared with a commercial ELISA. With a cut‐off value of 0.8, it showed 85.7% sensitivity, 83.3% specificity, a 48% positive prediction value (PPV), and 97% negative prediction value (NPV), indicating modest performance compared to existing commercial kits. The rLipL32 is a potential antigen for detecting IgM using ELISA; however, for use in low incidence areas, a confirmation test is crucial.

Abstract

Human papillomavirus (HPV) type 52 is among the top three high-risk oncogenic types that are associated with cervical cancer worldwide, especially in Indonesia. Designing a vaccine against HPV using an Lactococcus lactis expression system is a promising strategy, as this bacterium is well-known for its probiotic properties and can be safely utilized as a delivery carrier for oral vaccines. This study aimed to optimize the codon usage of the L1 protein of HPV type 52 for expression in L. lactis expression system and to identify potential epitopes using bioinformatics tools. To determine the codon optimization, the L1 protein sequence of HPV type 52 was retrieved from the National Center for Biotechnology Information (NCBI) database. Furthermore, codon optimization was conducted once the model had been established using OPTIMIZER, Clustal Omega, Restriction Mapper, and the ExPASy tool, based on L. lactis. The final construct was predicted to have a B-cell epitope by Ellipro tools, antigenicity by VaxiJen v.2.0, allergenicity by AllerTOP v.2.0, and toxicity by ToxIBTL. This resulted in a potential vaccine candidate with a Codon Adaptation Index (CAI) of 0.754 and GC content of 39.0%. Identification of epitopes from the optimized gene resulted in two peptide sequences, WRPSEATVYLPPVPVSKVVS and GTLGDPVPGDLYIKGSNSGNTATVQ, as components of the vaccine candidate.

Abstract

Paracetamol overdose is a well-documented cause of hepatic injury across vertebrate species, including teleost fish. This study aimed to evaluate the hepatoprotective potential of fucoidan, a sulfated polysaccharide derived from brown algae, against paracetamol-induced liver damage in Rasbora lateristriata. Fish were divided into five experimental groups and exposed to paracetamol (3 mg/L), either alone or in combination with fucoidan at concentrations of 50, 100, or 300 µg/mL, for seven days. Histopathological evaluation of liver tissues was performed using hematoxylin–eosin staining, with semi-quantitative scoring focused on hydropic degeneration, nuclear pyknosis, and necrosis. The results demonstrated that paracetamol exposure induced moderate hepatocellular injury, characterized by cytoplasmic vacuolization, apoptotic nuclear alterations, and necrotic lesions. Co-treatment with fucoidan at 300 µg/mL was associated with reduced severity across all histopathological parameters, indicating partial hepatoprotective effects. In contrast, the 50 µg/mL fucoidan group exhibited paradoxically severe hydropic degeneration despite the absence of pyknosis and necrosis, suggesting a delayed or altered injury profile. Intermediate outcomes were observed at 100 µg/mL. Overall, fucoidan exhibited dose-dependent but inconsistent hepatoprotective effects. The observed histological variability across concentrations suggests that protection may be influenced by factors such as bioavailability, cellular uptake, or interactions with intracellular stress pathways. These findings highlight the need for further mechanistic investigations before fucoidan can be considered a reliable hepatoprotective agent in aquatic toxicology.

Abstract

Butterfly pea flower (Clitoria ternatea) is an Indonesian native plant known for its medicinal properties due to its rich content of anthocyanins, flavonoids, and ternatins. Although widely recognized for its health benefits, the long-term effects of its bioactive compounds require careful evaluation. This study aimed to investigate the potential hepatotoxic effects of butterfly pea flower extract on the liver of Rasbora lateristriata fish. Fish were exposed to 0 (control), 50, 75, and 100 ppm of butterfly pea extract for four days, with four fish allocated per group. Histological markers, including necrosis, vacuolization, pyknosis, and hemorrhage, were assessed in liver tissues. Data were analyzed using the Kruskal-Wallis test and Mann-Whitney U test at a 95% confidence level. Results indicated that butterfly pea flower extract caused significant hepatic damage, with increased severity at higher concentrations. The most pronounced damage was observed at 75 ppm, suggesting an optimal dose for inducing oxidative stress. This study highlights the potential hepatotoxicity of butterfly pea flower extract in aquatic species and underscores the need for further research to evaluate its safety for therapeutic use.
