Abstract

Wheat has a great potency to be cultivated in Timor Tengah Utara (TTU). However, disease is one of the risks for  introduction of wheat. The existence of wheat disease in TTU is unknown because wheat has never been grown  in this area. This study aims to determine the potential risks of major diseases that will infect wheat plants when introduced to the TTU. The methods used in this study consisted of  wheat cultivation in the field, followed by observation and identification of the main diseases on wheat. Wheat varieties grown in field were Dewata, Selayar and Nias. Koch’s postulate were performed to identify the suspected microbes as pathogens. Morphological-based identification was applied on the isolated pathogens. The main diseases on wheat cultivated in TTU were leaf blight and stem rot. The results of Koch’s postulate showed that Helminthosporium gramineum was the causal of leaf blight and Rhizoctonia sp. was the causal of stem rot. Disease incidence reached 82–93% and 11–22% for leaf bligh and stem rot disease, respectively. Disease severity of leaf blight and stem rot disease on var. Dewata  was the lowest compared to var. Selayar and var. Nias.

Abstract

Selection of Compositions of Growth Medium and Carriers for Formulation of Biological Agents of Non-Pathogenic Fusarium oxysporum P21aThe potency of non-pathogenic Fusarium oxysporum P21a (NPFo P21a) as a biological control agent has prospect to be developed commercially. The growth medium and carrier are the critical factor to formulate biological control. This study aimed to obtain the best solid medium dan lighting type for mycelium growth and sporulation as well as to determine the carrier and storage temperature which is suitable for NPFo P21a toward the survival of propagule and germination of shallot. The results showed that M2 medium treatment (rice grain:rice bran; 20:1 w/w) was the best medium for mycelium growth. The M3 medium treatment (20:2 w/w) was the best medium for sporulation of NPFo P21a. The near-UV lighting treatment was able to stimulate the macroconidium production of NPFo P21a. Talc-based formulation and the storage at 20 °C showed the best shelf-life for NPFo P21a with density of viable propagule and shallot germination better.

Abstract

Anthracnose caused by several species of Colletotrichum is one of limiting factors for chilli production. Up to now the control of antrachnose disease still rely on regular and intensive application of synthetic fungicides. Application of similar active ingredients of synthetic fungicides may create resistance of the pathogens. This study was conducted to evaluate the sensitivity of three species of Colletotrichum against four commercial fungicides with different active ingredients, i.e. benomyl, chlorotalonil, mancozeb, and propineb. The sensitivity level and the potential occurrence of resistance of Colletotrichum were observed based on relative inhibition rate using poisoning food method. The results showed that all isolates were highly resistant to chlorotalonil, even up to ten times of the highest recommended concentrations. The tested fungal isolates were resistant or highly resistant to the recommended concentrations of mancozeb and propineb, but they were still sensitive when the concentrations were increased up to five times.  Isolates of Colletotrichum spp. showed sensitive to highly sensitive response to benomyl.

Abstract

Stem rot disease caused by Phytophthora capsici is an important disease on chilli. Eight endophytic fungi that had been isolated and screened based on pathogenecity test were further tested for their potential as the biological control agent of the stem rot disease of chilli. The endophytic fungi suspension was applied twice during the trial. The first application was on 100 seed lot, by soaking them in 100 mL of suspension. The second application was on the 3 weeks-old chili seedlings by drenching them with 10 mL suspension per plant. The concentration of endophytic fungi  in the suspension was 2.8 × 106 cfu mL-1. The disease intensity and AUDPC value were measured for 4 weeks after the pathogen inoculation. The growth inhibition test of P. capsici  was performed in vitro and the colonization abilities of endophytic fungi were observed at 4 weeks-old chilli seedlings. Eight endophytic fungi  inhibited the growth of the P. capsici, and two of those isolates namely Penicillium strain MAG1 and Penicillium strain PAB2 showed antibiosis mechanism. Endophytic fungi has the ability more to colonize at the root (26–60%) than in the stem (20–40%). Fusarium strain MAGR1 has the highest level of endophytic colonization i.e. 60% compared to others.  Based on in vivo assay, six endophytic fungi isolates, i.e. Fusarium strain MAGR1, Penicillium strain MAG1, Penicillium strain PAB2, sterile hyphae HAJ1, sterile hyphae HAJ2, and  sterile hyphae PBG7, showed the potency to control stem rot disease with inhibition level of 25.5–35.5%

Abstract

Colletotrichum spp. is known as the causal agent of anthracnose and considered as an important limiting factors on papaya production. The objective of this study was to determine the species of Colletotrichum spp. from various plant parts of papaya (Carica papaya) showing anthracnose symptom. Twenty isolates of Colletotrichum spp. was isolated and were grouped into 3 morphological groups based on colony similarity, conidial morphology, and setae formation. A total of 12 isolates of group I and 3 isolates of group II were identified morphologically as C. gloeosporioides sensu lato based on cylindrical conidia with rounded on both ends and colony morphology.  Meanwhile, 1 isolate of group II was molecularly identified as C. magnum. Four isolates of group III with a curved and pointed-end conidia, and produced setae on artificial medium were identified molecularly as C. truncatum. C. gloeosporioides was obtained on stem, leaf petiole, and fruit. C. magnum and C. truncatum were only obtained on leaf petiole and fruit, respectively.  In addition to morphological and molecular characters, differences in colony growth responses to temperature can be used to distinguish the species of Colletotrichum. To our knowledge the existence of anthracnose symptom on the stems and leaf petioles of papaya in the fields and the discovery of C. magnum and C. truncatum isolated from papaya was the first report in Indonesia.

Abstract

Fusarium wilt disease caused by pathogenic fungus F. oxysporum f. sp. cubense is an important disease in banana plants. This research was aimed to identifiy Fusarium spp. collected from banana plants showing wilt disease based on morphology and molecular characters.  The fungi isolates obtained was further examined for its pathogenicity and distribution in various parts of banana plants.  Based on morphological characters, i.e. growth rate, total conidia, colony color, macroconidium, microconidium, and chlamydospores, F. oxysporum f. sp. cubense can be differentiated from other species of Fusarium, but not the strain. Molecular analysis using specific primers VCG 01213/16 and Foc1/Foc 2 successfully identified 13 and 7 isolates of F. oxysporum f. sp. cubense TR4 and F. oxysporum f. sp. cubense non TR4, respectively.  Analysis using universal primer ITS4/ITS5 identified 7 isolates of F. solani, 1 isolate of F. verticillioides, and 2 isolates non Fusarium.  Infection of F. oxysporum f. sp. cubense TR4 and non-TR4 were found mostly in pseudo stems, whereas F. solani and F. verticillioides dominantly colonized banana corm. Pathogenicity test showed that F. oxysporum f. sp. cubense TR4 and non-TR4 caused necrosis on corm of cv. Ambon Kuning.  Similar necrosis symptom was also observed on infection of F. solani but with less severity.

Abstract

Moleculer Characterization of Papaya ringspot virus type P on Cucumber in JavaInfection of Papaya ringspot virus (PRSV) on cucumber plants showing mosaic symptom was detected using specific antibody.  Further investigation was conducted to determine molecular characters and status of PRSV infecting cucumber in Java.  Infection of PRSV was detected from leaf samples collected from the field using dot immunobinding assay (DIBA).  Disease frequency caused by PRSV infection reached 81.11%, 95.86%, 91.66%, and 92.3% in East Java, Central Java, Yogyakarta, and West Java, respectively.  Characterization of PRSV isolates was conducted by reverse transcription polymerase chain reaction (RT-PCR) using specific primers for PRSV-P and PRSV-W, followed by cloning, and DNA sequencing.  DNA fragment of 470 bp was successfully amplified using specific primers for PRSV-P from several samples from Nganjuk, Brebes, Kulon Progo, and Subang; but no amplification was achieved using specific primers for PRSV-W.  Nucleotide and amino acid analysis showed high homology among PRSV-P isolates from Nganjuk, Brebes, Kulon Progo, and Subang, i.e. 98.6%-99.7% and 99.3%-100%, respectively.  This is an indication of a low genetic variation among PRSV-P from Java. Further phylogenetic analysis indicated that PRSV-P isolate cucumber is in the same cluster with PRSV-P isolate papaya from Bali, Indonesia.  This is the first report of PRSV-P infecting cucumber in Indonesia.

Abstract

Stem Canker: A New Disease of coffee in LampungStem cancer is a new disease that has attacked smallholder coffee plantations in Lampung since 2010. The cause of the disease was unknown. This study aims to describe the symptoms of the disease, the incidence of the disease in the affected plantation, and identify morphologically and molecularly the canker pathogens of the coffee stem canker diseases. All stages of Koch’s postulate were carried out in laboratories and greenhouses. The isolated pathogens were morphologically characterized by colony shape and color as well as the conidia shape and size. Molecular identification was carried out by using a general primer (ITS1 and ITS4) and followed by sequencing. The main symptoms of the disease are stem cancer and dieback, as well as more infecting older plants. Pathogen of the coffee stem canker disease that attacks coffee plants in Lampung has been identified as Fusarium solani which has 99% homology with F. solani KY245947.1.

Abstract

Layu Fusarium merupakan penyakit penting pada bawang merah (Allium cepa var. aggregatum) yang disebabkan oleh Fusarium oxysporum f.sp. cepae. Pada penelitian ini, isolat biokontrol potensial cendawan endofit dan mikoriza diuji kemampuan kolonisasinya secara tunggal dan kombinasi untuk menekan perkembangan penyakit layu Fusarium. Percobaan dilakukan menggunakan rancangan acak lengkap dengan 8 perlakuan dan 5 ulangan. Kemampuan menekan patogen diamati dari persentase kolonisasi agens biokontrol, periode laten penyakit, insidensi penyakit, laju infeksi, dan area under the disease progress curve (AUDPC). Agens biokontrol yang diujikan berhasil bersimbiosis dengan akar tanaman bawang merah. Aplikasi tunggal mikoriza arbuskular paling baik dalam memengaruhi pertumbuhan tanaman dan menekan perkembangan penyakit layu fusarium pada bawang merah dengan periode laten penyakit terpanjang 21 hari dan menurunkan insidensi penyakit hingga 40%.

Abstract

Activity of Fungal Endophyte Secondary Metabolites against Colletotrichum acutatum on Chili PepperColletotrichum acutatum is one of anthracnose causal agents on chili pepper that has been reported to be predominant species in a some regions of Java Island. Secondary metabolites of endophytic fungi have been reported to have a potency as antifungal agents against plant pathogen. However, its antifungal activity against C. acutatum has not been reported yet. This study was aimed to evaluate the antifungal activity of fungal endophyte secondary metabolite against C. acutatum at in vitro and in vivo assay. In vitro assay was conducted to evaluate antifungal activity of fungal endophyte CBR1D14 isolate culture filtrate (FCBR) and mycelia extract (MCBR) in inhibiting conidial germination of C. acutatum. The results of in vitro assay showed that ethyl acetate extract of FCBR (EA FCBR) had the highest activity in inhibiting C. acutatum conidial germination. Methanol fraction from the partition of EA FCBR (FM FCBR) and from the partition of MCBR ethyl acetate extract (FM MCBR) showed the ability in inhibiting C. acutatum conidial germination. In vivo assay to chili pepper fruit showed that the treatment of FM FCBR (IC95 609.9 µg mL-1) and FM MCBR (IC95 1178.27 µg mL-1) decreased anthracnose disease incidence and lesion diameter. The efficacy rate of FM FCBR and FM MCBR treatments against anthracnose was 36.72 and 48.68%, respectively. Bioautography test was done on silica gel thin layer chromatogram. Methanol fraction of FCBR and MCBR were separated into 3 bioautographic spots respectively (Rf 0.04, 0.07, 0.7 for FM FCBR and Rf 0.06, 0.52, 0.7 for FM MCBR).
