Abstract

Raja Ampat (Southwest Papua Province) and Kepulauan Seribu (Jakarta Raya Province) are part of Indonesia#39;s territory, which includes the mangrove forest area. This habitat supports a diverse array of animals, plants, and microalgae, including Aurantiochytrium sp. It has antioxidant activity related to lipid content. This study aims to identify the lipid content and the antioxidant effects of the microalgae Aurantiochytrium sp.’s extract isolated from the mangrove forest areas of Raja Ampat and Kepulauan Seribu. The extraction process was conducted using the Folch method, which involves a 2:1 (v/v) chloroform-methanol solvent ratio. The microalgae Aurantiochytrium sp. extract of Raja Ampat (MAERA) and microalgae Aurantiochytrium sp. extract of Kepulauan Seribu (MAEKS) were analyzed using GC-MS, antioxidant potential tested in silico by target proteins (5ZLG, 2A1X, 3VLN, and 1EEM) using Autodock Tools, and in vitro by DPPH and ABTS radicals and the carotenoid bleaching method. The results showed that total squalene and fatty acids in MAERA and MAEKS were at different levels. Squalene as a ligand had the lowest binding energy (-9.4, -6.6, -7.7, and -6.5) compared to the native ligand (-5.6, -4.4, -5.2, and -5.2) and ascorbic acid (-5.6, -4.8, -5.3, and -4.9) by in silico antioxidant activity. The in vitro antioxidant activities by DPPH, ABTS, and ß-caroteen bleaching were 103.00 ± 3.75, 216.39 ± 367, 214.62 ± 7,07 (MAERA); and 131.85 ± 23.48, 225.50 ± 3.36, 251.69 ± 5.39 (MAEKS) were not significantly different (pgt;0.05) by all methods. These findings showed that the Aurantiochytrium sp. strains Raja Ampat and Kepulauan Seribu contained diverse levels of fatty acids and squalene. In the in silico study, squalene was evaluated and in vitro, the MAERA and MAEKS extracts showed moderate antioxidant activity.

Abstract

Hypertension is one of the Cardiovascular Diseases (CVD) which is the highest cause of death, and current treatment has many side effects that can be life-threatening, so researchers are interested in providing safe treatment solutions. One of them is traditional medicine such as aromatherapy. Therefore, it is necessary to carry out narrative research in the literature reviewing the effectiveness of lavender aroma therapy in reducing blood pressure in hypertensive patients. This research uses a Narrative Review study approach which focuses on evaluating several previous research results related to the topic. Based on data used by Google Scholar, Pubmed, and Spinger between 2019 and 2024. A total of 1,394 articles were found in the initial search stage, after filtering based on title, keywords and abstract this was reduced to 10 articles. And finally only 4 articles were selected that fit the criteria. There were 2 types of lavender aromatherapy applications, namely topical and inhalation. As well as two anti-hypertension active substances, namely linalool and linalool acetate. Lavender aromatherapy is effective in lowering blood pressure in hypertensive patients. The mechanism of reduction can be explained by the active anti-hypertensive substance (linalool & linalool acetate) and its application (inhalation & topical). However, further research with better designs is still needed.

Abstract

Spirulina (Spirulina platensis) and golden sea cucumber (Stichopus variegatus) are known to have antioxidant activity that has the potential to prevent neurodegeneration disease. The aim of this study was to examine the effect of the combination of spirulina and golden sea cucumber on Bcl2 gene expression in pyramidal hippocampus cells of trimethyltin-induced dementia (TMT) rats. The study used Sprague Dawley rats  which were divided into 6 groups, namely the normal control group (CMC-Na and NaCl 0.9%), pain control (CMC-Na and TMT), positive control (citicoline dose 200 mg / kg and TMT) and test control injected with TMT and given a combination of spirulina and golden sea cucumber dose 200 mg/KgBB with three ratios namely 3: 1, 1: 1 and 1: 3. Extract and citicoline were given on day 1 to day 28, while TMT injection was given a single dose of 8 mg/KgBB on day 8. On the 36th day the rats were sacrificed, brains were removed and the right hemispherium cerebri was fed to 10% formalin in pbs. After 6 days the hippocampus was separated for immunohistochemical observation. The test result data was statistically analyzed with a one-way ANOVA test then followed by post hoc tukey to see the differences between groups. Results showed the combination of spirulina and golden sea cucumber can increase the expression of the Bcl2 gene in the hippocampus. The combination of spirulina and golden sea cucumber (ST1: 3) dose of 200 mg / kg body weight was able to increase hippocampus Bcl2 expression with the number of Bcl2 cell expression almost the same as citicoline in both CA1 and CA2-CA3 regions.

Abstract

Abstract

The death of cells in the brain, especially the hippocampus, manifests in a decrease in memory, language, and behavior. Golden sea cucumber (Stichopus variegatus) andblue-green algae (Spirulina platensis) are reported to have high antioxidant content which has the potential to prevent cell death in the brain due to oxidative stress thereby preventing memory decline. This study aimed to investigate the effect of the combination of Stichopus variegatus and Spirulina platensis on preventing caspase 3 gene expression in pyramidal hippocampal cells in a rat model of trimethlytin-induced dementia (TMT). This study used Sprague Dawley rats, about 2 months old, weighing 180-200 g, divided into 6 treatment groups, with each group consisting of 8 rats. The hippocampus was taken from the right cerebral hemisphere for histological observations of pyramidal caspase-3 gene expression in the CA1 and CA2-CA3 regions. The combination of Spirulina platensis and Stichopus variegatus with a dose 200 mg/kg BB can prevent the expression of caspase-3 in the CA1 and CA2-CA3 areas of the hippocampus. Conclusion: The combination of spirulina and golden sea cucumber extracts has the potential to prevent caspase 3 gene expression in pyramidal cells of TMT-induced dementia rat models.
