Abstract

Radikal bebas merupakan satu faktor kontributor terjadinya penyakit degenerative. Antioksidan merupakan senyawa yang menghambat efek radikal bebas tersebut. Pada penelitian sebelumnya, ekstrak methanol daun Daun salam mempunyai efek antioksidan. Penelitian ini bertujuan menguji efek antioksidan ekstrak ethanol 70% daun salam (Syzygium polyanthum (Wight)). Metode yang digunakan adalah uji invitro dengan metode DPPH (Diphenyl picrylhydrazyl). Ekstrak dilarutkan dalam ethanol, kemudian dibuat dalam seri konsentrasi 10, 30, 50 dan 70 ug/mL. Masing-masing seri sebanyak 10 ml. Dalam masing-masing larutan tersebut ditambahkan 1 ml larutan DPPH 1 mM. larutan kemudian diinkubasi pada suhu 37°C selama 30 menit. Panjang gelombang yang digunakan adalah 515 nm. Metanol dan DPPH 1mM digunakan sebagai blanko.Untuk pembanding digunakan butylated hydroxytoluene (BHT) konsentrasi 2, 4, 6, 8 ug/mL. Hasil penelitian menunjukkan bahwa ekstrak ethanol 70% daun salam mempunyai efek antioksidan dengan IC 50 27,80 ug/mL.Kata kunci: daun salam (Syzygium polyanthum (Wight) , antioksidan, IC 50

Abstract

Muhammadiyah has contributed greatly to the maintenance of public health. Up to 2015 Data of muhamaditah hospitals/ KBIA / BP / maternity hospital owned by Muhammadiyah reached 457. The number of Muhammadiyah hospitals is because of the hard work MPKU in running programs above. But the number of hospitals and clinics does not necessarily bring health implications of muhammadiyah citizens. Most people have not seen a real contribution of Muhammadiyah (Muhammadiyah hospital).Muhammadiyah Hospital as one of the Muhammadiyah Health Charity expected to be more oriented to charity without forgetting their business, so residents can take muhammadiyah real benefit in health of the existence of the hospital, and the future expected Muhammadiyah became a pioneer in e-hospital, health tourism and travel medicine to face global challenges of health as part of the da’wah of Muhammadiyah in health.

Abstract

Seeing is crucial aspect in life includes in the field of education. Ifit is not cured the reduction of  sharpness of seeing/visus can affect thestudent ability to study. This action is done to detect  this reduction early.Therefore, the eyes will not be getting worse. The research is done bytraining and demonstrating how to check visus. The location of the researchis SD Gedongan I, Gedongan, Colomadu, Karanganayar. The 31 studentsare checked. The result is that there are 3 students have visus with miop.All the students can participate in this research well.

Abstract

Tempuyung (Sonchus arvensis) consisted of fiavonoid were effect as obstruct xantine oksidase enzyme, antioxidant captur superoxsidase radical. The fiavonoid total in leave of tempuyung is 0,1044%, in its root have 0,5% fiavonoid and the more is apigenin-7-O-glikosida (3,4,5). This research aim to know effect of infusa root of tempuyung (Sonchus arvensis) to lower the uric acid level at blood serum and infusa root of Tempuyung (Sonchus arvensis) to lower the uric acid level at mouse blood serum compared to allopurinol.This research is laboratory experimental method. The object were 25 Wistar male mice 2-3 months old, 150-200 gr divided into 5 groups. Negative Control group given potassium oxonate by intraperitoneal, Positive Control group given potassium oxonate by intraperitoneal, added by allopurinol 18mg/kgBB, Infusa Concentrated Group 1 given potassium oxonate by intraperitoneal added by infusa root of tempuyung 1,25g/kgBB, Infusa Concentrated Group 2 given potassium oxonate by intraperitoneal added by infusa root of tempuyung 2,5g/kgBB, Infusa Concentrated Group 3 given potassium oxonate by intraperitoneal added by infuse root of tempuyung 5g/kgBB. Executed until one day, where measurement of uric acid of mouse blood serum done before and after treatment. Measurement of uric acid level is done by using spectrophotometer. Obtained to be data to be analysed with Kolgomorov-Smirnov test, One-Way ANOVA and continued with LSD (Least Significant Difference) test with 95% confidence interval (CI). Result of statistical test of research shoe that infusa root of Tempuyung (Sonchus arvensis) dose 1,25g/kgBB, 2,5g/ kgBB, 5g/kgBB have effect to lower the uric acid level at mouse blood serum. Infusa root of Tempuyung (Sonchus arvensis) concentrated 5g/kgBB proportional wih dose allopurinol 18mg/kgBB to lower the uric acid level at mouse blood serum.Keyword: Infusa, root of Tempuyung (Sonchus arvensis), uric acid

Abstract

Daun kembang sepatu (Hibiscus rosa-sinensis L.) mengandung senyawa tannin dan flavonoid yang berefek sebagai agen hemostatik/penghentian perdarahan melalui mekanisme vasokonstriksi dan astringent. Penelitian ini dilakukan untuk mengetahui efek ekstrak topikal kembang sepatu terhadap penghentian perdarahan luar pada mencit. Penelitian bersifat eksperimental laboratoris dengan rancangan post test only with control group design. Subyek penelitian adalah daun kembang sepatu (Hibiscus rosa-sinensis L.) dan menggunakan 24 ekor mencit jantan galur Swiss yang dibagi dalam 6 kelompok yaitu : kontrol negatif (aquadest), kontrol positif (Epinefrin), dan kelompok III, IV, V, VI : diberikan ekstrak daun kembang sepatu dengan konsentrasi 5%, 10%, 20% dan 40% diberikan secara topikal. Ekor tikus diberi perlukaan kemudian dicelupkan pada tiap-tiap kelompok untuk diamati dan dihitung waktu perdarahan dan waktu pembekuan. Data yang diperoleh dianalisis dengan uji statistic Oneway Anova dan uji statistic LSD (Least significant Difference). Terdapat perbedaan bermakna dari bleeding timeantara kontrol negatif dengan perlakuan (p.0,000, sementara tidak terdapat perbedaan dalam cotting time  (p,0,313.Kesimpulan: ekstrak etanol 70% daun kembang sepatu (Hibiscus rosa-sinensis L.) konsentrasi 5%, 10%, 20% dan 40% mampu memperpendek waktu perdarahan, namun, tidak berefek memperpendek waktu pembekuan.Kata kunci : ekstrak daun kembang sepatu (Hibiscus rosa-sinensis L.), waktu perdarahan, waktu pembekuan

Abstract

Buah anggur merah diduga memiliki kandungan pterostilbene, resveratrol, proantosianidin dan likopen yang memiliki efek terhadap penurunan kadar trigliserida. Penelitian ini bersifat eksperimental laboratorium dengan metode pre and post test with control group design. Objek penelitian 25 ekor tikus putih jantan, Rattus Novergicus, berat badan 150-200 gram, berumur 3-4 bulan yang dibagi menjadi 5 kelompok dengan teknik simple random sampling, kontrol negatif (aquadest), kontrol positif (Simvastatin 0,2mg/200gramBB/hari), kelompok perlakuan dosis I (100mg/200gramBB/hari), dosis II (250mg/200gramBB/hari), dosis III (500mg/200gramBB/ hari). Ekstrak etanol anggur merah dosis I, dosis II, dosis IIIdapat menurunkan kadar trigliserida darah dengan rerata penurunan secara berturut-turut adalah 147,4mg/dL, 135,2mg/dL, 97,2mg/dL. Pada uji statistic menggunakan one-way ANOVA didapatkan nilai p=0,000 (p<0,05), sehingga terdapat perbedaan signifikan kadar trigliserida darah tikus putih antar kelompok. Ekstrak etanol 96% anggur merah dosis 100mg; 250 dan 500 /200 gramBB/hari dapat menurunkan kadar trigliserida darah tikus putih. Kata kunci :Ekstrak Anggur Merah, Trigliserida, Rattus Novergicus

Abstract

Digoksin merupakan salah satu obat digitalis. Pemakaian yang luas untuk gagal jantung dan aritmia (supraventrikuler) memerlukan pengawasan yang cukup ketat karena obat tersebut merupakan obat dengan indeks terapi sempit. Digoksin merupakan substrat P-glikoprotein (P-gp) yang dikode oleh multi drugs resistance-1 (MDR1). Gen MDR-1 terletak di kromosom 7q21.1. Gen tersebut mengandung 28 exon yang mengkode protein dari 1280 asam amino. Gen ini memegang peranan penting dalam absorbsi, distribusi, dan eliminasi beberapa obat. Polimorfisme gen MDR1C3435T terjadi pada exon 26. Terdapat tiga tipe gen MDR1C3435T yaitu MDR1C3435T CC, MDR1C3435T CT dan MDR1C3435T TT. Polimorfisme ini akan memengaruhi pembentukan PGP dan ini berpengaruh pada profil kinetika digoksin. Perubahan profil kinetika ini menyebabkan perubahan kadar digoksin dalam darah. Metode yang digunakan dalam review ini adalah penelusuran data berbasis pubmed, medline, dan embase dengan kata kunci MDR, digoxin. Terdapat beberapa studi yang berbeda mengenai pengaruh polimorfisme MDR1C3435T terhadap kadar digoksin dalam darah. Peningkatan kadar digoksin dalam darah akibat polimorfisme gen MDR1C3435T akan berisiko terjadinya intoksikasi digitalis. Pengobatan digoksin dalam jangka panjang atau dosis besar seyogyanya mulai mempertimbangkan profil genetika pasien. Distribusi polimorfisme MDR1C3435T penduduk suku Jawa adalah TT (0,10), CT (0,52), dan CC(0, 38).Kata kunci: Digoksin, MDR1C3435T, polimorfisme, P-gp Personalized Medicine Digoxin Theraphy in Individuals with MDR Gene PolymorphismDigoxin is one of digitalis drugs. Wider applicability to heart failure and arrhythmias (supraventricular) requires fairly strict scrutiny because of its narrow therapeutic index. Digoxin is a substrate of P-glycoprotein (P-gp) encoded by multi drugs resistance-1 (MDR1). MDR-1 gen located on chromosome 7q21.1. This gene contains 28 exons that encoded a protein of 1280 amino acids. This gene plays an important role in the absorption, distribution and elimination of many drugs. MDR1C3435T polymorphism occurs in exon 26. There are three types of MDR1C3435T gene namely MDR1C3435T CC, MDR1C3435T CT and MDR1C3435T TT. These polymorphisms will affect to the formation of P-gp and consequently to change the kinetic profile of digoxin. The change of kinetic profile causes changes in the digoxin blood levels. The method used in this review is data search based on pubmed, medline, and embase with keywords MDR and digoxin. There are several different studies of the influence of polymorphisms MDR1C3435T on blood digoxin levels. Increased levels of digoxin in the blood due to polymorphism of MDR1C3435T will be at risk of digitalis intoxication. Long-term digoxin treatment or large dose should consider the patient’s genetic profile. Distribution of polymorphism of MDR1C3435T in Javanese population is approximately TT (0,10), CT (0,52), and CC(0, 38).Key words: Digoxin, MDR1C3435T , polymorphism, P-gp

Abstract

The ethanolic extracts of the Zingiber zerumbet rhizome contains flavonoid and zerumbon compounds, the fish meat powder from Channa striata has an albumin content. The active compounds was contained in both materials had activity that can lower blood sugar levels in diabetic rats. This study aims to determine the hypoglycemic effect of each ingredients and the combination of Z. zerumbet rhizome extract and C. striata powder in alloxan-induced diabetic rats. This research uses pre and post test method with the control group design. The treatment was consisted of 5 groups, the first group was given aquades (as negative control), the second group was given glibenclamide with dose 5 mg/kgBW (as positive control), the third group was given 300 kg/kgBW of Z. zerumbet rhizome extract, the fourth group was given of C. striata fish powder 300 mg/kgBW, and the last group was given a combination materials with dosesrespectively, 300 mg/kgBW. The ingredient was given for 12 days and blood was taken on days 3, 6, and 12. The results shows the percentage of decrease in blood sugar of combination materials of 70.4% is greater than the single content of Z. zerumbet and C. striata, with the value of successive activity of 62.9% and 34.4%, respectively. The antidiabetic activity of combinations of materials had produced not different significant (p> 0.05) compared with single of C. striata powder as well as the Z. zerumbet rhizome extract.

Abstract

Hasil penelitian menunjukkan bahwa kanker masih menjadi penyebab utama kematian di Indonesia, demikian juga di dunia. Oleh karena itu, penelitian untuk mengembangkan obat baru sangat penting untuk dilakukan. Ceplukan (Physallis angulata L.) adalah salah satu tanaman yang potensial untuk dikembangkan sebagai cancer chemoprevention agent. Tujuan dari penelitian ini adalah untuk mengetahui efek sitotoksik tanaman ceplukan serta efek pada proliferasi dan apoptosis pada sel HeLa. Tanaman ceplukan diekstraksi dengan etanol 96%, selanjutnya ekstrak difraksinasi menggunakan pelarut yang berbeda kepolarannya yaitu petroleum eter, kloroform dan etil asetat. Penentuan efek sitotoksik dan pengaruh terhadap proliferasi sel HeLa dilakukan dengan metode MTT. Pengaruh sampel terhadap apoptosis pada sel HeLa dilakukan dengan doublestanning menggunakan ethidium bromide-acridyne orange. Protein markers pada apoptosis diamati dengan immunocytochemistry. Hasil penelitian menunjukkan bahwa fraksi petroleum eter mempunyai efek sitotoksik pada sel HeLa dengan nilai IC50 sebesar 120,198 µµg/ml. Fraksi petroleum eter menghambat proliferasi sel HeLa dan memacu terjadinya apoptosis dengan meningkatkan ekspresi protein p53 dan Bax.
