Abstract

Most of drugs are metabolized by cytochrome P 450 (CYP) enzyme. Cytochrome P450 3A4 is thecytochrome that is involved in metabolizing more than 60% of all medicine used in human. The variationof this CYP3A4 gene will affect the catalytic activity of this enzyme. Recently, CYP3A4*1G in intron 10 wasfound in Chinese and Japanese population. There is a substitution of G to A at position 82266 in intron 10. Thepurpose of this research was to investigate the frequency of allele and genotype CYP3A4*1G. Samples weretaken from bloods of the subjects of the research. The examination of CYP3A4*1G was conducted by RTLP-PCRmethod.As the results of this research, the frequency of CYP3A4*1G in Javanese people is CYP3A4*1/*1 0.25,CYP3A4*1/*1G 0.55 and CYP3A4*1G/*1G 0.20. Frequency of allele G: 0.53, allele A: 0.47. The Fisher’s exact- testshows that the allele and genotype frequencyis p. 1.000. The allele and genotype frequency of Javanese peopleisstill in Hardy-Weinberg equilibrium.

Abstract

Hasil penelitian menunjukkan bahwa kanker masih menjadi penyebab utama kematian di Indonesia, demikian juga di dunia. Oleh karena itu, penelitian untuk mengembangkan obat baru sangat penting untuk dilakukan. Ceplukan (Physallis angulata L.) adalah salah satu tanaman yang potensial untuk dikembangkan sebagai cancer chemoprevention agent. Tujuan dari penelitian ini adalah untuk mengetahui efek sitotoksik tanaman ceplukan serta efek pada proliferasi dan apoptosis pada sel HeLa. Tanaman ceplukan diekstraksi dengan etanol 96%, selanjutnya ekstrak difraksinasi menggunakan pelarut yang berbeda kepolarannya yaitu petroleum eter, kloroform dan etil asetat. Penentuan efek sitotoksik dan pengaruh terhadap proliferasi sel HeLa dilakukan dengan metode MTT. Pengaruh sampel terhadap apoptosis pada sel HeLa dilakukan dengan doublestanning menggunakan ethidium bromide-acridyne orange. Protein markers pada apoptosis diamati dengan immunocytochemistry. Hasil penelitian menunjukkan bahwa fraksi petroleum eter mempunyai efek sitotoksik pada sel HeLa dengan nilai IC50 sebesar 120,198 µµg/ml. Fraksi petroleum eter menghambat proliferasi sel HeLa dan memacu terjadinya apoptosis dengan meningkatkan ekspresi protein p53 dan Bax.

Abstract

Seeing is crucial aspect in life includes in the field of education. Ifit is not cured the reduction of  sharpness of seeing/visus can affect thestudent ability to study. This action is done to detect  this reduction early.Therefore, the eyes will not be getting worse. The research is done bytraining and demonstrating how to check visus. The location of the researchis SD Gedongan I, Gedongan, Colomadu, Karanganayar. The 31 studentsare checked. The result is that there are 3 students have visus with miop.All the students can participate in this research well.

Abstract

Muhammadiyah has contributed greatly to the maintenance of public health. Up to 2015 Data of muhamaditah hospitals/ KBIA / BP / maternity hospital owned by Muhammadiyah reached 457. The number of Muhammadiyah hospitals is because of the hard work MPKU in running programs above. But the number of hospitals and clinics does not necessarily bring health implications of muhammadiyah citizens. Most people have not seen a real contribution of Muhammadiyah (Muhammadiyah hospital).Muhammadiyah Hospital as one of the Muhammadiyah Health Charity expected to be more oriented to charity without forgetting their business, so residents can take muhammadiyah real benefit in health of the existence of the hospital, and the future expected Muhammadiyah became a pioneer in e-hospital, health tourism and travel medicine to face global challenges of health as part of the da’wah of Muhammadiyah in health.

Abstract

In the worldwide about 300 million people have asthma. β-2 agonists and steroids are widely prescribed drugs in the treatment of asthma. Different responses raises the suspicion of a genetic infiuence on the host’s response to the treatment of this disease. The purpose of this review is to provide a comprehensive understanding of the infiuence of genetic factors on the treatment of asthma. This review is done by the method in Pubmed search using keywords Pharmacogenetic asthma; polymorphism beta agonist AND asthma. The analysis shows that there are several genes that affect treatment response. ADRB2 gene affects the bronchodilatation response for asthmatics treated with β-2 agonist. CRHR1 gene polymorphism, Glucocorticoid-complex genes, and CER2 genes and TBX21 genes affect the response to corticosteroids inhalers.Keywords: Asthma, Pharmacogenetics, Treatment response

Abstract

Extract of eggplant fruit (Solanum melongena L.) seed is a plant that has anti-hypercholesterolemia effect on the state of dyslipidemia. One of the compounds contained in them are fl avonoids nasunin which can increase levels of HDL cholesterol (high density lipoprotein). HDL cholesterol is considered as a compound have the ability to remove excess cholesterol from the arteries, and therefore called good cholesterol. This study is a laboratory experimental research using pretest and posttest control group. The sample consisted of 25 rats were divided into 5 groups. Group I is a negative control (-), Group II is a positive control (+), group III was treated 1 (dose 1), group IV was treated 2 (dose 2), and V is the treatment group 3 (dose 3). Each group was given a high-cholesterol diet. Negative and positive control group were not given the extract of fruit seed of eggplant, the treatment groups III, IV, and V were given the extract of fruit seed of eggplant with each dose of 20 mg/kg, 40 mg/kg, 80 mg/kg were given for 4 weeks. HDL cholesterol levels will be measured before and after cholesterol diet and calculated each week after giving of the extract. The data obtained tested with statistical tests One-Way Anova. HDL cholesterol positive control group and the treatment group showed signifi cant differences between groups. Treatment groups 1 and 2 levels of HDL cholesterol lower than the positive control group and the treatment group 3. Extract of eggplant fruit seed dose of 80 mg/kg show an increase in HDL cholesterol levels were signifi cantly (p 0.05). The 70% ethanol extract of eggplant fruit seed at a dose of 80 mg/kg can increase HDL cholesterol levels signifi cantly.Keywords : extract of eggplant fruit seed, hypercholesterol, HDL cholesterol

Abstract

This research aim to know the descent of blood glucose rate by extract etanol 70% of bean leaf at male rabbit which induced glucose. Research using the method test glucose tolerance of oral was including category research of sham eksperimental with complete random attempt device of unidirectional pattern. This research use 20 male rabbits which divided in 5 treatment group. Group I given by CMC Na 1% as negative control, group II given by acarbose suspensi 2,33 mg/kgBB as positive control, group III, IV and V given by extract etanol 70% bean leaf with each dose 200, 300 and 450 mg/kgBB. Treatment given at once after test animal induced glucose 50% ( 5 ml/1,5 kgBB). Glucose blood rate to define every 30 minute, 0 minute until 300 minutes by using method of enzimatik GOD PAP (Glucose Oxidase Phenol Aminoantipyrina Peroxidase), what was later then analysed with one track Anava and continued by test of LSD (Least Significant Diffeerence) with trust level 95%. Extract etanol 70% bean leaf with dose 200, 300 and 450 mg/kgBB have effect descent of blood glucose rate to negative control of CMC Na 1% (p = 0,000) with each percentage descent of blood glucose rate (% PKGD) 80,88 ± 0,93 ; 82,73 ± 0,67 and 85,32 ± 0,21.Keyword : Extract etanol 70%, bean leaf (Phaseolus vulgaris L), degradation of glucose rate

Abstract

Antihyperuricemia activity in vivo testing has been done of single and combined extracts of Salam (Syzigium polyanthum Walp) and Belimbing Wuluh leaves (Averrhoa bilimbi L.), and also standardized extracts. Salam leaves extraction and Belimbing Wuluh leaves carried by the water solvent. Dried extract of each ingredient had been done antihyperuricemia preclinical in vivo testing against male white mice strain Balb-C induced by potassium oxonate dose of 250 mg / kg b.w. Antihyperuricemia preclinical in vivo testing results showed that the single extracts of Salam and Belimbing Wuluh at a dose of 200 mg / kg b.w. had lower activity levels of uric acid in the blood serum of male white mice, respectively 0.640 and 0.680. Abstract

Tempuyung (Sonchus arvensis) consisted of fiavonoid were effect as obstruct xantine oksidase enzyme, antioxidant captur superoxsidase radical. The fiavonoid total in leave of tempuyung is 0,1044%, in its root have 0,5% fiavonoid and the more is apigenin-7-O-glikosida (3,4,5). This research aim to know effect of infusa root of tempuyung (Sonchus arvensis) to lower the uric acid level at blood serum and infusa root of Tempuyung (Sonchus arvensis) to lower the uric acid level at mouse blood serum compared to allopurinol.This research is laboratory experimental method. The object were 25 Wistar male mice 2-3 months old, 150-200 gr divided into 5 groups. Negative Control group given potassium oxonate by intraperitoneal, Positive Control group given potassium oxonate by intraperitoneal, added by allopurinol 18mg/kgBB, Infusa Concentrated Group 1 given potassium oxonate by intraperitoneal added by infusa root of tempuyung 1,25g/kgBB, Infusa Concentrated Group 2 given potassium oxonate by intraperitoneal added by infusa root of tempuyung 2,5g/kgBB, Infusa Concentrated Group 3 given potassium oxonate by intraperitoneal added by infuse root of tempuyung 5g/kgBB. Executed until one day, where measurement of uric acid of mouse blood serum done before and after treatment. Measurement of uric acid level is done by using spectrophotometer. Obtained to be data to be analysed with Kolgomorov-Smirnov test, One-Way ANOVA and continued with LSD (Least Significant Difference) test with 95% confidence interval (CI). Result of statistical test of research shoe that infusa root of Tempuyung (Sonchus arvensis) dose 1,25g/kgBB, 2,5g/ kgBB, 5g/kgBB have effect to lower the uric acid level at mouse blood serum. Infusa root of Tempuyung (Sonchus arvensis) concentrated 5g/kgBB proportional wih dose allopurinol 18mg/kgBB to lower the uric acid level at mouse blood serum.Keyword: Infusa, root of Tempuyung (Sonchus arvensis), uric acid

Abstract

Meniran is one of traditional plants that contains several compounds such as terpenoid, flavonoid, and alkaloid that role as antibactrial agent. The aims of the research are to determine the antibacteria effect of extract aethyl acetat and chloroform of meniran. This research was conducted as a laboratory experimental by using post test control group design only. The concentration of extract aethyl acetat and chloroform meniran are 5%, 10%, 20%, 40%, and 80%. The result indicate that extract aethyl acetat of meniran have not effect to Escherichia coli. Meanwhile, to Staphyloccus aureus have an effect (p=0,003). The concentration of the extract chloroform meniran are effective to inhibite the growth of Escherichia coli (p=0,029) and Staphylococcus aureus (p=0,004). The extract aethyl acetat of meniran have an antibacteria effect to Staphylococcus aureus but not to Escherichia coli. Whereas, the extract chloroform of meniran have an antibacteria effect to Staphylococcus aureus and Escherichia coli.Keyword : The aethyl acetat and chloroform extract of Meniran (Phyllanthus niruri L.), antibacteria effect, Staphylococcus aureus and Escherichia coli
