The objective of this study was to determine geneticdiversity of the soursop (Annona muricata L.) populations from Java (Indonesia) using RAPD markers. A total of 70 individuals werecollected from 7 soursop populations, distributed along the geographical range of natural distribution in Java. Genetic diversity wasestimated by RAPD technique using 6 arbitrary selected primers. Those primers produced 151 polymorphic bands with the percentageof polymorphism for each primer ranged from 95% to 100%. The genetic diversity value (h) within each population ranged from 0.0418to 0.0525. The highest h value (0.0525) was found in the KRA population whereas the lowest h value was observed in the PCTpopulation. The highest genetic distance value (0.0410) was observed in SKH-GKD populations pair whereas the lowest geneticdistance value (0.02448) was estimated in KRA-PCT populations pair. Based on the dendogram, the seven soursop populations weresegregated into three major clusters. The first cluster consisted of SKH, KRA, PCT, NGW, and KPG populations. The GKD populationwas then grouped into second cluster. In the third cluster, the BGR population was grouped separately and more genetically distant thanthe others. However, the relationship dendrogram showed that the grouping did not always indicate the geographical origins similarity,but possibly showed the genetic similarity.

Abstract

Monascus-nata complex was made up from fermented nata which was grown in the medium containing Monascus purpureus fungi, that could be used as coloration of nata. Rice flour usually used as medium to produce pigments by M. purpureus. Tofu solid waste and rice bran could be used to produce pigments although the colour intensity of M. purpureus pigment was lower than rice. The aims of these research were to study colour, pH medium and weight of miselium Monascus-nata complex from the combination of rice flour with tofu solid waste extracts and rice flour with rice bran extracts as the medium. In these research, 5% (b/v) rice flour extracts, combination of rice flour extract : tofu solid waste extract in the ratio of 1:1, 1:2, 1:3 and rice flour : rice bran extracts in the ratio of 1:1, 1:2, 1:3 were used as the medium. Ten nata de coco (1x1x1 cm3) were put in to the extracts of medium (100mL) in bottle, then inoculated with 10% (v/v) starter of M. purpureus containing 1.3x107 cfu/mL and incubated at room temperature on an orbital shaker at 100 rpm for 16 days. Parameters which were measured, i.e. colour intensity, preferable test, pH medium, weight of miselium Monascus-nata complex. Data were analyzed using analysis of variant and followed by DMRT in 5% significations except preferable test which were analyzed using hedonic test. Combination of rice flour extract : tofu solid waste extracts in the ratio of 1:1 could be used to replace rice flour extracts on the making of Monascus-nata complex. Orange colour intensity from rice flour extract : tofu solid waste extracts in the ratio of 1:1 was higher than from rice flour extracts. Orange colour intensity from rice flour extract : tofu solid waste extracts in the ratio of 1:1 and rice flour were 0.156 and 0.123 respectively. Red colour intensity from medium rice flour extract : tofu solid waste extracts in the ratio of 1:1 approach the red colour intensity of rice flour. Red colour intensity from medium rice flour extract : tofu solid waste extracts in the ratio of 1:1 and rice flour extract were 0.088 and 0.145 respectively. All of pH value decreased from 7 to 6.82. Abstract

The aims of this research were to study about the growth and diversity of moulds in the cucumber pickle and the influencesof acetic acid concentration and long storage as well as additional of onion on the growth of moulds. Cucumber pickle was treated in different acetic acid concentrations (0,8%, 1,4%, 2,0%), long storage and with or without additional of onion. Theused method to isolate of mould was dilution planting. For enumerating moulds, 0,1 ml samples were blended and dilutedafter that inoculated on to potato sucrose agar (PSA) medium and then incubated at 270C for 7 days. To identify the moulds, the colony was transferred on to CDA and PSA media, and then incubated at 270C for 7 days. The moulds were identified according to their macroscopic and microscopic characters. To know the degrees of taste, the cucumber pickle was tested to 25 people with hedonic scale method. The results showed that, there was 12 different kinds of moulds can be found in cucumber pickle without additional of onion. There were Aspergillus fumigatus, A. niger, A. ochraceus, A. tamarii Kita, Cochliobolus sp, Botrytis, Penicillium chrysogenum, P. expansum, P. citrinum, Rhizopus oligosporus, Trichoderma sp,and Monilia. On the other hand, experiment with onion, P. expansum and A. niger could not be observed. The highestvarieties of moulds can be found in cucumber pickle with acetic acid concentration 1,4%. In experiment without onion, on nil(0) day, with acetic acid concentration of 1,4%, the amount of moulds is lower than in the pickle with acetic acid concentration 0,8%. The diversity of moulds in the cucumber pickle with additional of onion is lower than that of without onion. The additional of onion showed decreasing the diversity and growth of moulds in cucumber pickle with acetic acid concentration of 0,8% on nil (0) day. Abstract

The aims of this research are firstly, to study the advantages of electrophoretic techniques. Secondly, to look at the usefulness of a few mediums support of electrophoretic proteins especially the acrylamide gel. Thirdly, to examine thenumber of plant organs which could be used as the sources of plant proteins, and how these plants protein should be applied in the medium support that has been selected. Besides, the staining and detection procedures would be described, while the application of electrophoretic approach in higher plant taxonomy will also be evaluated. In this study we recorded that a number of taxonomic problems usually caused by morphological complexity within species can be solved using this experimental approach of electrophoresis. Abstract

The aim of the experiment was to examine whether the morphological characters of eleven species of Ranunculuscollected from a number of populations were in agreement with the genetic data (isozyme). The method used in this study was polyacrilamide gel electrophoresis using peroxides, estarase, malate dehydrogenase, and acid phosphatase enzymes. The results showed that cluster analysis based on isozyme data have given a good support to classification of eleven species based on morphological groups. Abstract

Species of Ranunculus is small herb grows at quite high altitudes, ranging from several hundreds to more than a thousand meter above sea level. They can occupy a variety of habitats such as moist soils or can even grow sub merged or floating in stream. A few numbers of species from different populations have been recorded to have morphological complexity, which could cause a problem for the work of taxonomists in making decisions. In order to support taxonomists in solving their problem, a new experimental method using SDS-PAGE will be used to explore the isozyme data. The main purpose of this research was to investigate whether or not isozyme data can be used to clarify the morphological complexity of Ranunculus species. In this study, nine species of Ranunculus from different populations were used. Five to twenty plants were sampled for electrophoresis studies. Four enzyme systems: peroxidase, esterase, malate dehydrogenase and acid phosphatase were chosen. The results showed that every enzyme gave its specific pattern in each species and common band always found in nine species tested. Abstract

The main purposes of this research were to investigate whether the two distinct types of the morphological characterof Ranunculus triplodontus were genetically controlled or environmental influence. In order to prove the above,electrophoretic examinations were carried out employing for four enzyme systems. The medium support of polyacrilamide was chosen. The samples were collected from seven populations around central plateau and the leaves were used as the isozyme data. The result indicated that variation occurred in certained populations. However, this isozyme data were not able to separate the two types of R. triplodontus into different species. Based on the cluster analysis showed that three groups of seven populations of R. triplodontus were appeared. This research confirms that morphologically distinct species was not supported by the isozyme data, thus the variation found in certain population was mainly influenced by the environmental conditions, and therefore could not be considered as taxonomically distinct.© 2001 Jurusan Biologi FMIPA UNS SurakartaKey words: Ranunculus triplodontus, isozyme.

Suranto, Syahidah AT, Mahadjoeno E. 2018. Variation of morphology, anatomy and nutrition contents of local cultivar mentik rice based on the altitudes at Ngawi District, East Java, Indonesia. Biodiversitas 19: 652-659. The purposes of this research were to examine the variation in morphology, anatomy and nutrition contents of rice plants grown at three different altitudes at Ngawi District, East Java, Indonesia. The plant samples were collected from three different altitudes (200, 500 and 900 m asl. respectively). The morphological characters of leaf, root and stem were used in this examination, while Paraffin method was employed to look at the anatomical appearances. To look at the content of lipids and proteins, Soxhlet and Kjeldahl methods were used, while the Iodocolorymetre method was chosen to test the amylose content. Randomized Complete Block Design (RCBD) with one factor, namely altitude was used in this research. Morphological and anatomical characters and nutrition contents of rice plant were analyzed using one way of ANOVA. The results showed that the length and diameter of roots, plant height and also stem diameter were declining due to a higher altitude. On the other hand, the length of leaf, the number of roots, the total number of stomata and also the ratio of length per diameter of stomata increased, and so did the number of aerenchyma space roots and stems. In general, there was a tendency of decreasing amylose and protein contents due to higher altitudes.

Hidayati NR, Suranto, Sajidan. 2018. Morphological characteristics and isozyme banding patterns of Cucurbita moschata at different altitudes. Biodiversitas 19: 1683-1689. Aims of this research were to investigate the morphological character and isozyme banding patterns of Cucurbita moschata plants grown at three different altitudes. Samples in this study consisted of leaf, stem, and flowers. The morphological characters were conducted by direct observation in the field and analyzed descriptively as well as statically by one way ANOVA. The isozyme bands appearance of esterase and peroxidase of leaf samples were conducted using polyacrylamide gel electrophoresis (PAGE). Qualitative approach was used to analyze the presence and the absence of isozyme bands, while Retardation factor (Rf) was used to analyze quantitatively. The results showed that most plants grown at middle altitude (351-750 m asl.) were well-developed in terms of length of leaves, stems and flowers. Accordingly, the isozyme banding pattern of peroxidase was also found varied in plants grown at middle altitudes from which the presence of very unique bands was detected. Conversely, the band detected in plants grown at the lower and the highest altitudes was similar in term of band's number but it was different in the quality of the bands. Meanwhile, esterase isozyme banding pattern of plants grown at the lower and higher altitude had more bands than the middle altitude. Based on this result it is obvious that the isozyme data could be used to support in understanding the diversity morphological characters of plants grown in three different altitudes. This early result suggests that altitudes as a crucial factor in contributing the expression of isozyme appearance, which is useful for further pumpkin characterizations.

Morphological, anatomical and isozyme variability among taro (Colocasia esculenta) accessions from southeastern part of Central Java, Indonesia. Biodiversitas 19: 1811-1819. The objective of this study was to evaluate morphological, anatomical and isozyme variability among taro accessions from southeastern part of Central Java (Indonesia). A total of 20 taro accessions were collected from a wide range of sites during field surveys. Morphological characters measurements were taken on vegetative structures such as roots, stems, leaves, and corms. Anatomical characters were observed from both paradermal and transverse sections of leaf. Identification of biochemical markers was done by using peroxidase and esterase isozyme system. A UPGMA dendrogram among accessions was constructed based on the genetic similarity matrix by applying a cluster analysis using a computer programme, NTSYS Version 2.00. The analysis of variance for morphological and anatomical characters revealed that there was significant difference for majority of the tested traits indicating that there was a variability among the taro accessions. Polymorphism was observed using isozymes of esterase (12 banding pattern) and peroxidase (8 banding pattern). Based on the dendrogram at a level of 62 % similarity, taro accessions were segregated into two major clusters. In Cluster I, the closest relationship was shown between SKH and SKA accessions that had 96 % coefficient of similarity. The ten accessions from Klaten, Sragen, and Karanganyar were then clustered separately as Cluster II with coefficient of similarity 73.52 %.
