Abstract

Rheumatoid arthritis (RA) is a systemic, progressive, chronic autoimmune inflammatory disease that affects tissues, organs, and damages synovial joints. The RA can be treated with DMARD (disease modifying anti rheumatic drugs), such as methotrexate. However, the use of this drug long-term can cause side effects. Soft coral Lobophytum sp. has secondary metabolite compounds such as flavonoids, alkaloids, and terpenoids which has an anti-inflammatory activity, which beneficial for anti-rheumatoid arthritis agent. This study aims to determine the potential of anti RA in Lobophytum sp. using in vitro and in vivo. The anti-inflammatory assay for anti RA in vitro was performed by stabilization of human red blood cell (HRBC) membrane. In addition, anti RA was performed in arthritis model mice by induction of complete Freund’s adjuvant (CFA) by administering LEA once daily orally for 15 days. It was found that the ethyl acetate of Lobophytum sp. (LEA) reduced the haemolysis and increased the stability of HRBC membrane. Furthermore, LEA also showed anti RA by decreasing the edema in mouse paw at dose of 50 mg.kg-1 BW (LEA50), 100 mg.kg-1 BW (LEA100), and 200 mg.kg-1 BW (LEA200), respectively with 2.5 mg.kg-1 BW of methotrexate as positive control (C+) with P<0.05. Moreover, LEA200 demonstrated highest efficacy and showed no significant different with C+ (P>0.05). In conclusion, our research has shown that LEA has anti-inflammatory for treatment of rheumatoid arthritis from in vitro and in vivo studies.

Abstract

Meistera chinensis merupakan generasi baru famili Zingiberaceae yang tumbuh di Sulawesi Tenggara. Ekstrak buah mengandung metabolit sekunder seperti flavonoid, terpenoid, alkaloid, steroid, dan tanin yang memiliki efektivitas sebagai antioksidan dan antibakteri. Namun, belum ada laporan tentang antibakteri pada bagian rimpang. Penelitian ini bertujuan untuk mengetahui kandungan metabolit sekunder dan aktivitas antibakteri ekstrak etanol rimpang M. chinensis. Serbuk kering rimpang M. chinensis diekstraksi dengan maserasi menggunakan pelarut etanol 96%. Ekstrak dipekatkan menggunakan rotary vacuum evaporator. Metode analisis aktivitas antibakteri menggunakan difusi agar dengan konsentrasi ekstrak 10%, 20%, 30%, dan kontrol positif (Ciprofloxacin 30µg) terhadap Staphylococcus aureus ACTT 25023 dan Escherichia coli ACTT 35218 selama 24 jam. Hasil penelitian menunjukkan bahwa ekstrak etanol rimpang M. chinensis mengandung flavonoid, saponin, alkaloid, steroid, dan fenol, Ekstrak terbukti menghambat pertumbuhan E. coli dengan diameter zona hambat rata-rata secara berturut-turut pada konsentrasi 10%, 20%, dan 30% sebesar 6,08±1,79; 8,16±0,11 dan  10,57±1,34 mm. Pada S. aureus, zona hambat masing-masing konsentrasi 10%, 20%, dan  30% sebesar 5,02±0,79; 6,01±0,69; 8,03±0,76. Analisis One Way ANOVA menunjukkan perbedaan aktivitas antibakteri yang signifikan antar konsentrasi ekstrak pada E. coli dan S. aureus. Dapat disimpulkan rimpang M. chinensis memiliki aktivitas sebagai antibakteri.

Abstract

The wundu watu plant (Alpinia monopleura), is one of the endemic plants typical of Southeast Sulawesi which is commonly found in South Konawe Regency. The results of previous research on the essential oil content of the leaves and fruit of wundu watu are α-caryophyllene, β-pinene, limonene, α-pinene, β-caryophyllene and caryophyllene oxide. The essential oils of the leaves and fruit show antibacterial, antioxidant and anti-inflammatory properties. Pharmacological studies of this plant are not widely known, especially its efficacy as an anti-inflammatory. The purpose of this study was to determine the anti-inflammatory potential of extracts and fractions of wundu watu stem against inhibition of protein denaturation in Vitro. Wundu watu stem extract was extracted by maceration method using methanol solvent and ethyl acetate fraction. Anti-inflammatory testing using the protein denaturation inhibitor method using bovine and albumin and measured on a UV-Vis Spectrophotometer at a wavelength of 660 nm. The results showed that the extract and ethyl acetate fraction of wundu watu stem had anti-inflammatory activity with IC50 values of 10.59 mg/L and 2.02 mg/L. Based on the test results, it can be concluded that the stem extract of wundu watu has anti-inflammatory potential against protein denaturation with a very strong category.

Abstract

Alpinia monopleura is one of the endemic plants of Sulawesi, and it has an extensive distribution in the region. Research on chemical compounds and biological activities of A. monopleura is essential to continue as an effort to support the utilization of native plants for medicine. The extract was obtained using the maceration method. The chemical compounds in the extract were identified using Liquid Chromatography Mass Spectrometry (LCMS). Bovine Serum Albumin (BSA) and molecular docking methods were used to evaluate the anti-inflammatory activity. Ten compounds contained in the extract were successfully identified, E-para-coumaric acid (1), trans-ferulaldehyde (2), 3,5,6-trihydroxy-4',7-dimethoxyflavone (3), nevadensin (4), malvalic acid (5), ent-16α,17-hydroxy-19-kauranoic acid (6), 3′,5-dihydroxy-7,4'-dimethoxy flavone (7), saurufuran B (8), 5-hydroxy-7,8,2'-trimethoxyflavanone (9) and dehydroabietic acid (10). The anti-inflammatory activity of extracts from rhizomes and stems of A. monopleura were 8.62 and 10.59 mg/L, respectively. Some flavonoids (9 and 7) can bind strongly to specific residues around the COX-2 active site, such as Ser530, thereby interfering with the function of the COX-2 enzyme and reducing the production of pro-inflammatory prostaglandins. Thus, A. monopleura extract has the potential to inhibit inflammatory responses through molecular regulation of the COX-2 enzyme.

Abstract

Diarylheptanoids compounds from the genus Etlingera have attracted widespread attention due to their various structural diversity and potential pharmacological effects. This is certainly an important part to continue to study, especially from Sulawesi endemic species, one of which is Etlingera calophrys. The fruit of E. calophrys is widely utilized in the Sulawesi community. The lack of information on the content of chemical compounds and their pharmacological activities, so the study is very important. Diarylheptanoid compounds were obtained by chromatography technique, molecular structure interpretation by spectroscopy technique while pharmacological activity was tested against radical scavenging as antioxidant and inhibition of protein denaturation as anti-inflammatory. Two diarylheptanoid compounds were successfully isolated from E. calophrys fruit namely 1-(3'-methoxy-4'- hydroxyphenyl)-7-phenyl- 3-heptanone and 1-(3'-methoxy-4'- hydroxyphenyl)-7-(4"-hydroxyphenyl)- 3-heptanone. Antioxidant properties showed very strong ability with IC50 values of 32.21 ± 0.42 and 21.64 ± 0.63 µg/mL, respectively, while anti-inflammatory properties were strongly shown by the compound with IC50 values of 25.71 ± 0.54 and 15.28 ± 0.66 µg/mL, respectively. The presence of diarylheptanoid content that has good pharmacological activity can support the used of E. calophrys as a nutraceutical source. Abstract

Marine sponges display significant potencies as an anti-inflammatory agent. Thus this study aims to investigate the effect of anti-inflammatory of Melophlus sp. and Callyspongia sp. by decreasing plasma IL-1β levels of rats. This study was conducted by detecting chemical constituents of extracts and their anti-inflammatory by measuring the plasma IL-1β level of animals.  Animals were acclimatized for seven days, followed at day-8 animals were induced by 1% carrageenan injection intraplantar. Animals were divided into ten groups (n=4) and treated orally according to groups which were C+ and K+ (diclofenac sodium 3598 ppm), C- and K- (Na CMC 0.5%), M50 (Melophlus sp 50 ppm), M100 (Melophlus sp 100 ppm), M200 ((Melophlus sp 200 ppm), C50 (Callyspongia sp. 50 ppm), C100 (Callyspongia sp. 100 ppm), and C200 (Callyspongia sp. 200 ppm). After being left out for 1 hour, blood was collected at the first, second, and third hour. Blood samples were then centrifugated and assayed with ELISA kit Rat IL-1β. Data collected were statistically analyzed. Both marine sponges contain flavonoid, saponin, alkaloid, and terpenoid. Tannin was only detected in Melophlus sp. Both marine sponges provided an effect in decreasing plasma IL-1β at the concentration of 100 ppm and 200 ppm. In conclusion, both extracts of Melophlus sp. and Callyspongia sp. have anti-inflammatory activity with effective concentration are 100 ppm. Keywords: Anti-Inflammatory; Plasma IL-1β; Melophlus sp.; Callyspongia sp.

Abstract

Kanker merupakan suatu penyakit yang bersifat ganas dan ditandai dengan pertumbuhan sel abnormal secara cepat dan terus-menerus yang dapat merusak jaringan dan organ di sekitarnya. Ciri khas dari kanker yaitu mampu menghindari mekanisme apoptosis. Salah satu gen penanda apoptosis yaitu Caspase-3. Penggunaan kemoterapi untuk mengobati kanker masih memiliki banyak efek samping yang merugikan. Oleh karena itu dibutuhkan alternatif pengobatan yang lebih efektif dan efek samping yang lebih kecil seperti terapi target. Tumbuhan E. alba diketahui memiliki kandungan metabolit sekunder yang memiliki potensi sebagai antikanker. Tujuan penelitian ini untuk melihat potensi penekanan ekspresi gen dari E. alba tehadap penanda gen apoptosis Caspase-3 pada lini sel kanker T47D. Pengujian ini dilakukan menggunakan sampel batang E. alba yang dimaserasi dengan pelarut metanol. Ekstrak metanol batang E. alba dibuat dalam tiga konsentrasi yaitu 50 ppm; 100 ppm; dan 200 ppm. Selanjutnya dilakukan pengujian ekspresi gen secara in vitro dengan menggunakan metode RT-PCR, lalu diukur nilai CT. Hasil penelitian yang telah dilakukan yaitu pada konsentrasi ekstrak 50 ppm mampu menekan ekspresi gen 1,677 kali, ekstrak 100 ppm mampu menekan ekspresi gen 1,859 kali, sedangkan ekstrak 200 ppm mampu menekan ekspresi gen 1,601 kali. Berdasarkan hasil penelitian yang dilakukan maka dapat disimpulkan bahwa ekstrak metanol batang E. alba mampu menekan ekspresi gen Caspase-3 pada lini sel kanker payudara T47D, dengan konsentrasi ekstrak yang paling baik yaitu 100 ppm dengan hasil pengujian ekspresi gen 1,859 kali dari DMSO 2%.

Abstract

Penelitian ini bertujuan untuk mengevaluasi aktivitas antioksidan menggunakan metode DPPH dan ABTS, serta mengkaji potensi tabir surya alami dari ekstrak metanol dan fraksinya dari daun jambu mete (Anacardium occidentale L.) pada konsentrasi 12,5; 25; 50; dan 100 ppm. Hasil penelitian menunjukkan bahwa ekstrak metanol memiliki aktivitasantioksidan yang kuat dengan nilai IC 50 sebesar 71,94 ± 5,41 ppm (DPPH) dan 89,24 ± 1,49 ppm (ABTS). Fraksi n-heksana menunjukkan aktivitas antioksidan kategori sedang dengan nilai IC 50 sebesar 131,64 ± 5,39 ppm (DPPH) dan 124,20 ± 2,08 ppm (ABTS). Fraksi etil asetat juga menunjukkan aktivitas sedang, dengan nilai IC 50 sebesar 193,65 ± 3,86 ppm (DPPH) dan 128,82 ± 1,59 ppm (ABTS). Fraksi air menunjukkan aktivitas sedang dengan nilai IC 50 sebesar 115,53 ± 12,63 ppm (DPPH) dan 107,98 ± 1,22 ppm (ABTS). Sebagai perbandingan, vitamin C sebagai kontrol positif menunjukkan aktivitas antioksidan yang sangat kuat, dengan nilai IC 50 sebesar 11,68 ± 5,92 ppm (DPPH) dan 15,40 ± 3,09 ppm (ABTS). Nilai Sun Protection Factor (SPF) dari ekstrak metanol sebesar 4,24 ± 0,00, dikategorikan sebagai perlindungan sedang. Persentase penghambatan eritema menunjukkan bahwa ekstrak dan fraksi etil asetat memiliki potensi sebagai tabir surya (sunblock), sedangkan fraksi n-heksana dan fraksi air memberikan perlindungan ekstra. Namun, tidak ada ekstrak maupun fraksi yang memenuhi kriteria perlindungan terhadap pigmentasi kulit.

Abstract

Madu hutan dikenal sebagai produk alami yang kaya akan berbagai senyawa bioaktif, seperti flavonoid dan fenolik, yang memiliki aktivitas antioksidan dan antiinflamasi. Kabupaten Konawe, yang terletak di Sulawesi Tenggara, merupakan salah satu wilayah potensial penghasil madu hutan. Namun, informasi ilmiah mengenai aktivitas biologisnya masih terbatas. Penelitian ini bertujuan untuk mengevaluasi potensi aktivitas penangkap radikal bebas dan efek antiinflamasi dari madu hutan asal Konawe melalui uji in vitro. Aktivitas antioksidan dianalisis menggunakan metode DPPH dan ABTS, sedangkan aktivitas antiinflamasi dievaluasi melalui uji stabilisasi membran eritrosit (HRBC), inhibisi denaturasi protein albumin serum sapi (BSA), dan inhibisi produksi nitric oxide (NO). Hasil penelitian menunjukkan bahwa madu hutan Konawe memiliki nilai IC50 sebesar 71,69 ± 2,73 mg/L pada uji DPPH dan 58,90 ± 9,11 mg/L pada uji ABTS, yang mengindikasikan aktivitas antioksidan sedang hingga kuat. Pada uji stabilisasi membran, madu ini menunjukkan tingkat stabilisasi sebesar 90,31 ± 0,03% pada konsentrasi 100 mg/L, sementara pada uji denaturasi protein menghasilkan nilai IC50 sebesar 48,37 ± 0,42 mg/L. Uji inhibisi produksi NO menunjukkan nilai IC50 sebesar 6,65 ± 2,38 mg/L, yang dikategorikan sangat kuat. Analisis statistik menggunakan uji t independen menunjukkan perbedaan yang signifikan (p < 0,05) antara madu hutan Konawe dan kontrol positif pada seluruh parameter yang diuji. Secara keseluruhan, hasil ini mendukung potensi madu hutan Konawe sebagai sumber alami antioksidan dan antiinflamasi yang menjanjikan.

Abstract

Xestospongia sp. is one of marine sponge belongs to demosponges class that mainly found in Southeast Sulawesi and the secondary metabolites contained in Xestospongia sp. suspected to have immunomodulatory activity. A previous study exhibited the immunomodulatory of Xestospongia sp. ethanolic extract (XEE) at dose of 300 and 400 mg/Kg BW by affecting the phagocytic activity of macrophages. Thus, this study aims to investigate the effect of XEE towards interferon-gamma (IFN-γ) and tumor necrosis factor-alpha (TNF-α) at dose of 300 and 400 mg/Kg BW. Wistar male rats are divided into 4 groups (n=6) randomly and treated for 7 days orally each as follow: group I (XEE dose of 300 mg/KgBW); group II (XEE dose of 400 mg/KgBW); group III (0.5% NaCMC); and group IV (commercial phylantii extract). On day 8, animals were infected with Staphylococcus aureus and left for 1 hour. Animals were sacrificed and the blood was drawn by cardiac puncture (3 mL), followed by analyzed under ELISA Kit for IFN-γ and TNF-α. Collected data were analyzed statistically using SPSS®. The IFN-γ levels obtained were 350.113; 392.970; 118.416; and 61.958 ρg/mL, respectively and the TNF-α were 2808; 1308; 778; and 845.5 ρg/mL, respectively. According to results obtained, both doses of XEE are affecting the IFN-γ and TNF-α levels (p<0.05) compared to group III as negative control, and group IV as positive control. As conclusion, XEE of both doses is increasing IFN-γ and TNF-α levels of animals that respond to phagocytic activity
