Abstract

Obesity has become an epidemic around the world. High fat diet (HFD) have been implemented as one of intervention to battle obesity. Uncouple protein 1 (UCP1) is one of the key factor on energy expenditure. The aim of this experiment is to see the macronutrients composition on weight loss and UCP1 expression in the visceral fat. Fifty male mice, 2-3 months old, 18-30 grams, were put in five different groups. K1 were fed (20% protein, 62.0% carbohydrate, 12% fat), K2 (60% protein, 0% carbohydrate, 30% fat), K3 (45% protein, 0% carbohydrate, 45% fat), K4 (30% protein, 0% carbohydrate, 60% fat), K5 (15% protein, 0% carbohydrate, 75% fat). The experiment was done in four weeks, mice body weight was measured every week. UCP1 expression seen using immunohistochemistry staining was measured at the end of the fourth week. Significant weight loss was achieved by K4 (-9.60±3.81) gram by the end of week four (p<0.05). K4 had the least amount of visceral fat. The result was that K4 achieved a significant visceral fat mass (0.02±0.06) gram compared to K1 (0.53±0.08) gram. Compared to other groups K5 expressed UCP1 more than the others (3.78±3.72) cphfp. HFD fed groups produced significant weight loss, group that had the greatest weight loss is K4. Meanwhile, each group had a variety of UCP1 expression.

Abstract

Diabetes mellitus (DM) is one of the global health problems and in the top 4th ranks as the main cause of death in developing countries (IDF, 2015). The pathogenesis of type II DM involves abnormalities in insulin secretion and activity that leads to insulin resistance. This research aims to study the efficacy of celery (Apiumgraveolens) as a prevention of insulin resistance. In this study, the samples were 45 Wistar rats (Rattus norvegicus), male sex, aged 4-6 weeks, weight 150-175 grams, and had normal fasting blood glucose levels by tested before treatment. The experimental animals were divided into 5 groups, K1 was negative control group (insulin resistance by given 20% fructose 1,86 g/kg BW PO qDay);K2 was positive control group (those given 20% fructose 1,86 g/kg BW PO qDayand standard insulin resistance therapy metformin 500 mg/kg BW PO qDay); the K3 treatment group was given 20% fructose 1,86 g/kg BW PO qDay and celery extract 200 mg/kg BW PO qDay; the K4 treatment group was given fructose 20% 1,86 mg/kg BW PO qDay and celery extract 400 mg/kg BW PO qDay; and the K5 treatment group was given fructose 20% 1,86 mg/kg BW PO qDay and celery extract 600 mg/kg BW PO qDay. The treatment had been given every day for 60 days.Fasting blood glucose levels were measured using a Glucometer. Fasting blood insulin levels were measured using ELISA, HOMA-IR was calculated using a standardized formula, and GLUT4 protein expression was measured using immunohistochemistry. It the end of the intervention, there was a significant decreased in fasting blood glucose (FBG) in K4 group compared with K1 (p <0.05), insulin resistance in K1 was characterized by a higher HOMA-IR value compared to the therapy group, especially K4 and K5 (p <0.05). There was an increase in GLUT-4 expression on K4 and K5 compared with K1 (p <0.05). It can be concluded that celery extract has antihyperglycemia effect and furthermore it can prevent insulin resistance condition.

Abstract

Introduction Student-centered learning (SCL) is based on students' needs, capacities, and other characteristics, making the method more personalized. This study aimed to analyze new questionnaires to categorize 4th-semester medical faculty students based on their characteristics. Understanding these characteristics could make the teaching and learning process more effective, potentially reducing the number of students retaking exams. Material and Methods: Subjects were 4th semester students who agreed to enroll and self-fulfilled the questionnaire. High achievers are defined as high expectations and consistent high performance (GPAs). Validity and reliability of each item were analyzed by statistical calculations accordingly. GPA in the next semester was used as threshold to confirm the participant's categorization. Results: The study commenced in March 2022, with 107 participants with Grade Point Averages (GPAs) available. The study demonstrated that the questionnaires had high validity and reliability (p-value < 0.005; Pearson Correlation Coefficient and Cronbach alpha). Among the participants, 3 (2.8%) were classified as high achievers, while 3 (28%) were categorized as low achievers. Notably, 44.86% of participants reported their passions as unrelated to medicine, while 5.61% were uncertain of their passions. Regarding vocation, 8.2% cited parental influence, and 10.3% felt their educational progress was not within their control. Conclusion: The questionnaires had high validity and reliability in item analysis. There were 2,8% of participants predicted as High Achievers and Low Achievers. Refinements of items and score presentation are needed. Follow-up activities should be given to the low achievers to prevent becoming retakers.
