Abstract

Adhesi merupakan tahap inisiasi dari proses kolonisasi bakteri. Kemampuan adhesi ini diperantarai oleh keberadaan hemaglutinin pada permukaan bakteri. Tujuan penelitian ini adalah melihat kemampuan hemaglutinin pada Escherichia coli dalam proses adhesi. Uji adhesi pada sel epitel pipi dilakukan dengan menggunakan metode Velentine-Weiggand. Hasil penelitian menunjukkan bahwa Escherichia coli yang memiliki hemaglutinin menunjukkan kemampuan adhesi yang jauh lebih baik yakni rata-rata 67,6 bakteri/sel dibandingkan dengan   yang tidak memiliki hemaglutinin rata-rata 7,2 bakteri/sel. Kemampuan adhesi ini berkaitan dengan keberadaan hemaglutinin di permukaan bakteri.

Abstract

Dalam penelitian ini isolasi dan karakterisasi hemaglutinin Staphylococcus aureus dilakukan dengan teknik afinitas kromatograf Karakterisasi hemaglutinin yang dihasilkan dilajutkan dengan teknik elektroforesis menggunakan metode sodium dodecyl sulphate gel electrophoresis (SDS-PAGE) dan dilanjutkan untuk melihat pengaruh suhu dan enzim terhadap aktivitas hemaglutinin. Hasil penelitia menunjukkan bahwa komponen hemaglutinin Staphylococcus aureus yang telah diisolasi memiliki berat molekul 46 kDa. Aktivit Staphylococcus aureus dalam meghemaglutinasi hilang pada pemanasan 60° C dan pengaruh enzim proteolitik. Hasil ini mengindikasika bahwa hemaglutinin Staphylococcus aureus adalah protein.

Abstract

The objective of this research is to see the influence of weather on the prevalence of Newcastle Disease (ND) in chicken in Kabupaten Aceh Utara (North Aceh). Data used in this research were obtained from Dinas Peternakan North Aceh for the number of chicken suffered ND and from Badan Meteorologi dan Geofisika Lhokseumawe, North Aceh for the form of weather. Multiple Linear Regression Model with five independent variables (the average of rainfall per month, the average of maximum temperature, the average of minimum temperature, the velocity of the wind, and the average of humidity per month) was used to see the influence of wheather to the prevalence of Newcastle Disease. Proportion the number of chicken suffered from ND which is the ratio of the number of chicken suffered from ND to the total number of chicken was used as dependent variables. The result shows that the best model is Ŷ= 120.529278 – 1.33 x wind humidity + 1.907 x wind velocity.

Abstract

Tujuan penelitian ini adalah mengetahui kemampuan hemaglutinin pada Staphylococcus aureus (S. aureus) dalam proses adhesi. Uji adhesi pada sel epitel ambing dilakukan dengan menggunakan metode Velentine-Weiggand. Hasil penelitian ini menunjukkan bahwa S. aureus yang memiliki hemaglutinin menunjukkan kemampuan adhesi yang jauh lebih baik (±380 bakteri/20 sel) dibandingkan dengan yang tidak memiliki hemaglutinin (±56 bakteri/20 sel).

Abstract

The objective of this study was to evaluate the invasiveness and persistence of Pasteurella multocida after intratracheal inoculation on turkeys. Four strains of Pasteurella multocida, P-1059, T-325, CU, and M-9 were used in this study and intratracheally (IT) inoculated into turkeys. The number of viable bacteria in the trachea, lung, liver, spleen, and the blood were enumerated during periods of 6 and 9 hrs post inoculation (PI). The result showed that four strains of organism were present in the organ and the tissues observed. In the liver, spleen, and blood, the virulent P-1059 strain showed significant increases within 6 or 9 hrs PI. Strain CU showed significant increase only in the spleen. Two other strains did not show significant increase in any organs; strain M-9, on the other hand, showed significant decrease in the trachea. These results indicated that P-1059 and three other strains showed their ability to invade the tissues or organs when they are intratracheally inoculated.

Abstract

The objective of this research is to know Aceh cattle follicle dynamic under environmental heat stress condition. This research was conducted on December 2012 until August 2013. Animals used in this research were 20 Aceh cattle aged 5-8 years old, weighing of 150-250 kg, and had at least 2 regular cycles. All cattle used were clinically in good body condition score. The cattle were divided into two groups of 10, used in two different period of times (December-January and July-August) to examine the effect of environmental heat to their follicle dynamic. Each group is separated into two different keeping management; one group was kept in pens while the others in the pasture, 5 cattle in each management group. Research timing was based on information acquired from Indrapuri Agency for Meteorology, Climatology, and Geophysics (BMKG) which predicted that extreme weather would last from July to August while December to January would be relatively normal in Aceh region. All cattle’s estrous cycle were synchronized by 5 mg/mL of PGF2α intramuscularly (Lutalyse™, Pharmacia Upjohn Company, Pfizer Inc.). Ultrasonography examination was performed to monitor ovary follicle’s growth and dynamic during one cycle. Days during ovulation marked by standing heat was regarded as Day 0 of estrous cycle. Follicular dynamic examination during estrous cycle on December-January and also on July-August, both penned or pastured cattle showed the follicle waves that was 3 follicle growth waves. The size of follicle growth on first wave (1st DF) on cattle kept in pen on July-August and December-January were relatively similar. The size of follicle growth on first wave for cattle kept in pasture on July-August and December-January were relatively similar. To conclude, Aceh cattle’s follicle dynamic is not change during environment heat stress condition and is not affected by different cattle-keeping management.

Abstract

This study aimed to investigate the correlation between progesterone hormone concentration and number of fetus as well as the difference in progesterone hormone concentration from different gestation periods in Etawah crossbred (EC). Five EC goats used in this study were injected with 2 mL of 5.5 mg prostaglandin F2α (PGF2α, Capriglandin) intramuscularly, with an interval of 10 days. Goats that showed symptoms of estrus were mated naturally with male goats. Blood samples were taken on the 7th, 14th, 21st, and 75th days of gestation. Progesterone hormone concentration was determined using enzyme linked immunosorbent assay (ELISA). The number of fetus was determined at 35th gestational day using ultrasonography (USG). Four out of 5 goats were found to have 1 fetus and 1 goat had 2 fetuses. The progesterone concentration on the 7th, 14th, 21st, and 75th days of gestation in goats with single fetus were 5.21, 7.78, 11.97, and 18.78 ng/mL, respectively, while in goat with two fetuses were 8.44, 14.53, 16.81, and 22.73 ng/mL, respectively. The correlation (r) between progesterone hormone concentration and number of fetus on the 7th, 14th, 21st, and 75th days of gestation were 0.442, 0.854, 0.592, and 0.757, respectively. It is concluded that progesterone concentrations are highly correlated to the number of fetus in each gestation period in EC goats.

Abstract

This study was aimed to determine the effect of epididymis extract (EE) on the testosterone and dihydrotestosterone (DHT) level of local male goat. An experimental study was performed using a completely randomized design (CRD) pattern of one-way analysis of variance (ANOVA). 15 local male goats aged 1.5 years with body weight 14-16 kg were used in this study. The K0 group as a control group, injected with only 1 ml physiological saline, while each KP1, KP2, KP3, and KP4 groups treated with multilevel EE dose, ie 1, 2, 3, and 4 ml / goat for 13 consecutive days. At the end of treatment (day 14th), testes, epididymis (caput, corpus, and cauda) and ductus deferens samples were taken through the close-castration method for examining the testosterone and DHT concentration by using enzyme-linked immunosorbent assay (ELISA) technique. Data gathered were later analyzed using ANOVA followed by Tukey’s HSD in SPSS 16.0 for Windows. The result showed that the average concentration of testosterone on K0, KP1, KP2, KP3, and KP 4 in testis respectively were 10.00±2.64 ng/ml; 7.66±2.51 ng/ml; 10.00±6.55 ng/ml; 0.66±0.57 ng/ml; 11.66±7.37 ng/ml; caput epididymis; 5.00±1.73 ng/ml; 2.33±1.52 ng/ml; 5.00±2.64 ng/ml; 1.33±0.57 ng/ml; 5.66±1.15 ng/ml; corpus epididymis; 1.33±0.57 ng/ml; 0.66±0.57 ng/ml; 4.00±2.64 ng/ml; 0.66±0.57 ng/ml; 4.33±2.30 ng/ml; cauda epididymis: 1.00±0.00 ng/ml; 0.66±0.57 ng/ml; 1.66±0.57 ng/ml; 1.00 ± 0.00 ng/ml; 2.00±1.73 ng/ml; ductus deferens: 3.66±2.51 ng/ml; 0.66±0.57 ng/ml; 3.00±1.00 ng/ml; 1.00±0.00 ng/ml and 3.66±1.15 ng/ml. Statistical analysis showed that the administration of EE only increased testosterone concentration in testes had significant effect (P 0.05). From this study, it can be concluded that the EE has the potential to improve spermatogenesis and sperm quality through increasing the testosterone concentration in the local male goats.

Abstract

Escherichia coli (E. coli) is a normal flora and a facultative anaerobic bacterium that plays an essential role in the humoral nutritional metabolism of broiler intestines. This study aims to determine the effect of salix (Salix tetrasperma roxb) extract 50 and 100 mg/litre in drinking water of E. coli colonies in the broiler intestines experiencing heat stress. The sample is 12 intestines of broiler strain MB-90 aged 28 days. The method used a completely randomized design. The study by user three treatments: P0 was not given salix extract in drinking water; P1 and P2 were given salix extract 50 and 100 mg/litre in drinking water. The data were analyzed using One-way ANOVA and were continued with the Duncan test. We can conclude that the salix extract given to broiler chickens with heat stress could affect many E. coli colonies. This result can be seen from the doses given, indicating that the number of E. coli colonies in chicken intestines that experience heat stress depends on the dose of salix extract. The higher dose is given, the fewer colonies in the chicken intestine
