Abstract

Melanocortin-4 receptor (MC4R) is one of G protein-coupled receptors that plays an important role inregulation of energy homeostasis. MC4R mutations constitute the most common cause of human obesity.The study was conducted in order to investigate the variation of MC4R gene in Macaca fascicularis and itsassociation with obesity as a model of human obesity. Forty eight adult male macaques from Bali (Ubudand Uluwatu), East Java (Alas Purwo and Baluran), and Sumatera island (Palembang) were used in thisresearch. The animals had been anaesthetized using ketamine (10 mg/kg body weight) and xylazine (2mg/kg body weight) before collecting blood samples and phenotypic data (weight, crown rump length. Bloodsamples were used as source of DNA. To determine MC4R variation, coding region of this gene wasamplified and sequenced. The results showed that 20 variations sites were identified and 13 of them werenon-synonymous. Among the non-synonymous mutations, five mutations were only found in obese macaque;two mutations were found both in obese and non-obese macaque; and six mutations were only found in nonobesemacaque .

Abstract

This experiment was conducted to study the profile of total protein, albumin, and globulin concentrationson Ettawah crossbreed neonates after consuming various colostrums. Twenty four healthy neonatal kidswere used in this study. The neonates were divided into four groups. Each group received fresh maternal(goat) colostrum, frozen-thawed bovine colostrum, bovine spray dried colostrum, and bovine powdercommercial colostrum, respectively. Colostrums were given at 10% of body weight directly after birth andfollowed by the same amount every 12 hours, for three days. The blood was taken from jugular vein at 0, 12,24, 48, 72, and 168 hours after birth to determine total protein, albumin, and globulin concentrations.Results of this study indicated that the serum total protein and globulin concentration increased andreached the peak at 24 hours after birth. Compared to the concentration at birth, the increase of totalprotein concentration were 62.77%, 59.26%, 48.05%, and 66.67% in fresh maternal (goat), frozen-thawedbovine, bovine spray dried, and commercial bovine colostrum, respectively. Serum globulin concentrationincreased 4.9, 4.4, 4.8, and 14.6 times in fresh matermnal goat, frozen-thawed bovine, spray dried, andcommercial bovine colostrums respectively, compared to the concentration at birth. In conclusion, theconsumption of various colostrums i.e. fresh maternal goat colostrums, bovine colostrums (frozen-thawed,spray dried and commercial colostrums) would increase the concentration of blood total protein and globulin,which both reached the highest concentration at 24 h after birth.

Abstract

The effectiveness of atherogenic diet in raising plasma cholesterol level of cynomolgus macaques(Macaca fascicularis) can be predicted for population, but not for individuals.This study aimed to evaluatethe individual plasma cholesterol of cynomolgus macaques in response to IPB-1 atherogenic diet. Theanimals under study were 22 adult malemonkeys from the animal facility of Primate Research CenterBogor Agricultural University (PSSP IPB). All animals were intervened with the IPB-1 atherogenic diet forthree months and individual plasma cholesterol wasevaluated in a monthly basis.The results showedthat the monkeys’ plasma cholesterol had increased significantly (P <0.05) after one month of interventionperiod and the increases were very significantly different (P <0.01) after three months. Individually, increasesin plasma cholesterol varied among animals. Based on these variations, 2 animals were categorised ashypo-response, 18 animals as hyper-response and 2 animals as extreme.This study showed the variationof individual plasma cholesterol of cynomolgus macaques in response tothe IPB-1 atherogenic diet so thatthe animals might be selected based on the their responsiveness.

Abstract

The aim of this study was to compare the efficacy of trehalose and raffinose supplementation in Trisegg yolk (TEY) and Tris soya (TS) diluents in optimizing the quality of Etawah Crossbreed liquid semen.Semen were collected from three sexually mature bucks using artificial vagina. Semen were then evaluatedand divided into six aliquot tubes. Each of them was diluted in TEY and TS supplemented with 50 mMtrehalose or raffinose, respectively. The liquid semen were then stored in refrigerator (5°C). The motility,viability and plasma membrane integrity (PMI) of the spermatozoa were evaluated every 12 hours untilsperm motility remained up to 50%. The results showed that sperm motility in TEY supplemented withtrehalose or raffinose remained up to 50% for 72-84 hours, compared to in TS which was for 48-60 hours(P<0,05). The best diluent was demonstrated by TEY supplemented with trehalose where the spermmotility was 52,82±3,21% up to 84 h compared to raffinose supplementation (52,78±4,41%) and control(51,78±4,86%) which was up to 72 hours, respectively. Meanwhile, the spermatozoa motility in TS diluentsupplemented with raffinose was 52,78±4,41% for up to 60 hours compared to supplemented with trehalose(53,33±3,54%) and control (51,11±4,86%) which was up to 48 hours. In all diluents, the viability ofspermatozoa was 6-9% higher than the percentage of sperm motility, whilst the percentage of PMI wassimilar to the percentage of sperm motility. In conclusion, TEY supplemented with trehalose was the bestdiluents for preservation of Etawah Crossbreed buck liquid semen, and when using TS diluent it isrecommended to add raffinose  rather than trehalose.

Abstract

The objective of the research is determining the ICM isolation method to produce ESC. Blastocyst stage of DDy mice embryos were used in this study. Zona pellucida of blastocysts were removed by 0.25% pronase, the ICM isolation were done by enzimatic or immunosurgery method, and then they were cultured in DMEM-high glucose supplemented with mercaptoethanol, gentamycin, fetal bovine serum, and cumulus cells as feeder layer. The result of the research indicated that immunosurgery method yielding attachment rate and number ESC colony 93.85% and 43.08%, respectively, higher (P<0.05) than enzimatic method that weree 79.63% and 18.52%, respectively, but the viability of ICM cells were equal (P >0.05) that are 93.59% in enzymatic method and 98.56% in immunosurgery method. This research concluded that immunosurgery more effective method for isolation of ICM and ESC production than enzymatic method.

Abstract

Monyet ekor panjang (Macaca fascicularis) adalah salah satu hewan laboratorium dalam penelitian biomedis. Fasilitas penangkaran untuk hewan laboratorium diatur dan didukung oleh kaidah kesejahteraan hewan dan manajemen kesehatan yang baik. Salah satu masalah yang sering terjadi di fasilitas penangkaran adalah diare, termasuk di fasilitas penangkaran IPB Dramaga. Diare menyebabkan penurunan produktivitas karena tingginya morbiditas dan mortalitas. Bakteri enteropatogenik adalah salah satu penyebab diare yang dapat ditularkan antar hewan juga melalui air dan pakan. Oleh karena itu, tujuan dari penelitian ini adalah untuk mengidentifikasi prevalensi bakteri enteropatogen pada sampel feses, air dan pakan, menguji kepekanan bakteri enteropatogen terhadap antibiotik dari sampel feses di fasilitas penangkaran Dramaga. Sebanyak 30 sampel feses, enam sampel air dan dua sampel pakan dikoleksi kemudian diuji untuk mengetahui adanya bakteri enteropatogen. Metode mikrobiologis standar digunakan untuk mengisolasi dan mengidentifikasi bakteri enteropatogen dari sampel feses dan pengujian kepekaan antibiotik.. Air dan pakan diuji menggunakan metode angka paling mungkin. Hasil isolasi dan identifikasi bakteri dari sampel feses adalah E. coli (100%), Salmonella enteritidis (97%) dan Shigella sp. (60%). Kontaminasi E. coli dan coliform dalam sampel air dan pakan (pisang dan monkey chow) terdeteksi di atas batas minimum (0/100 mL/g). Semua bakteri enteropatogen sensitif terhadap siprofloksasin dan gentamisin tetapi resisten terhadap eritromisin.

Abstract

The purpose of this study was to investigate changes in cellular pathology of adrenal gland as impact of bone marrow mesenchymal stem cells (BMMSC) therapy in hypertensive rats. The research used ten male wistar rats with age ± 10-12 weeks with body weight ± 200-250 grams which is devided into two different groups ie BMMSc(-) hyprtention and BMMSc(+) hypertension. Rats are conditioned with hypertention using Hasbinoto et al method. Right nephrectomy and left carotid communic artery ligation are performed on all rats. Rats are injected with deoxycorticosterone acetate (DOCA), then 0.12 % of âaminopropionitrile fumarate (BAPN) is added into drinking water. 1% NaCl solution was administered as drinking water during the study. Rats were evaluated for particular indicator ie blood pressure then were euthanized for adrenal organs collections. The data that obtained then analysed using qualitative descriptive. The result of the study indicate hypertention therapy using BMMSC can repair cell damaged of the adrenal gland such as hyperplasia, nodules, cysts and cytoplasm vacuolization.

Abstract

Akhir-akhir ini banyak peneliti yang mempelajari terapi berbasis sel sebagai terapi regeneratif untuk melindungi atau memperbaiki jaringan yang rusak. Bone marrow mesenchymal stem cell (BMMSC) memiliki kemampuan yang menjanjikan dalam memperbaiki ginjal. Tujuan penelitian ini adalah untuk mengetahui apakah induksi yang dilakukan dapat menyebabkan kerusakan tubulus ginjal dan BMMSC mampu melindungi atau memperbaiki kerusakan yang terjadi. Penelitian ini menggunakan dua belas tikus jantan umur 10-12 minggu dengan rata-rata tekanan darah sistolik minimum mencapai 140-150 mmHg yang dibagi menjadi kelompok kontrol dan kelompok terapi. Tekanan darah yang tinggi diinduksi dengan mengangkat ginjal kanan, mengikat arteri karotis komunis kiri, serta memberikan NaCl 1%, DOCA 2,5 mg/100gBB, dan BAPN 0,12%. Bone marrow mesenchymal stem cell disuntikkan setelah 16 minggu induksi. Evaluasi histopatologi ginjal yang didukung dengan evaluasi darah dilakukan dua minggu setelah injeksi BMMSC. Berdasarkan evaluasi histopatologi, kerusakan tubulus ginjal pada dua kelompok tidak menunjukkan hasil yang berbeda. Demikian juga regenerasi pada kedua kelompok menunjukkan hasil sama. Kadar blood urea nitrogen (BUN) dan kreatinin pada kedua kelompok berada dalam kisaran kadar normal. Hasil yang didapat menunjukkan bahwa kerusakan yang terjadi tidak parah dan BMMSC memperbaiki tubulus ginjal akan tetapi belum dapat melindungi tubulus ginjal dari kerusakan.

Abstract

The purpose of the present study was to investigate the viability of mice and hamster embryos developed in Kalium Simplex Optimized Medium amino acid (KSOMaa) and Hamster Embryo Culture Medium-6 (HECM-6) medium. Female DDY mice were superovulated by injection i.p. of 5 IU Pregnant Mare Serum Gonadotropine (PMSG) and 5 IU Human Chorionic Gonadotropine (hCG) in 48 h interval, hamster (Phodopus campbelli) injected by 2.5 IU PMSG and 2.5 IU hCG 48 h later. Then females were mated with fertile males. Eight-cell embryos were recovered at day 3 after natural mating. The mice embryos were cultured in KSOMaa+5% NBCS (New Born Calf Serum) (T1) and HECM-6+5% NBCS (T2), the hamster embryos were cultured in KSOMaa+5% NBCS (T3) and HECM-6 + 5% NBCS (T4) for further development at 37oC in a humidified atmosphere of 5% CO2 in air for 48 h. The examinations were replicated five times. The T1 embryos developed to compact morulla and early blastocyst 100% (140/140), 92.1% (129/140) to blastocyst and expanded blastocyst, and 22.9% (32/140) became hatching/hatched. The T3 reached 100% (60/60) to compact morulla and early blastocyst, 85.0% (51/60) blastocyst, and 48.3% (29/60) expanded blastocyst, no embryo observed hatching/hatced. The T2 embryos had more expanded blastocyst than T3 (P<0.05), hatching/hatched rate higher than T1 and T3 but lower than T4 (P<0.05). Shortly, KSOMaa enable to support 8-cell stage mice and hamster embryo, but the hamster embryo developed lower at expanded blastocyst stage. HECM-6 is more appropriate than KSOMaa to support 8-cell mice embryos development and suitable to develop 8-cell stage hamster embryos.
