Abstract

Immunomodulator is a substance that increases or suppresses the immune response through the certain mechanism. The marine sponge Xestospongia sp. has immunomodulatory activity by increasing phagocytic activity. In addition, the phagocytic activity is affected by CD4+ and CD14 cells levels. Thus, this study aims to investigate the effect of Xestopongia sp. extract toward CD4+ and CD14 cells level in model rat. Animals were divided into 4 groups (n=5) and treated for 7 days, as follow: Group I (Ethanolic extract of Xestospongia sp. dose of 300 mg/KgBW); Group II (Ethanolic extract of Xestospongia sp. dose of 400 mg/KgBW); Group III (Phylantus niruri extract); and Group IV (0.5% Na CMC). On day 8, animals were infected with Staphylococcus aureus intraperitoneally. And the blood was collected by cardiac puncture and assayed with ELISA kit CD4 (elabscience®) and ELISA kit CD14 (elabscience®). Ethanolic extract of Xestospongia sp. provided high levels of both CD4+ and CD14 cells (Group II) compared to baseline (Group IV)  (p<0.05). Group I provided similar activity to group III (p>0.05) and Group II provided significant activity with higher levels of CD4+ and CD14 cells compared to group III (p<0.05). In conclusion, both doses of Xestospongia sp. extract provide activity as immunomodulator by increasing CD4+ and CD14 cells levels, yet dose of 400 mg/KgBw provides the higher immunomodulatory activity.

Abstract

Imunomodulator merupakan bahan yang dapat mengembalikan ketidakseimbangan sistem imun. Spons Xestospongia Sp. diduga mengandung senyawa-senyawa aktif yang berperan sebagai agen imunomodulator. Penelitian ini dilakukan untuk mengetahui pengaruh pemberian ekstrak etanol Spons Xestospongia Sp. terhadap aktivitas fagositosis makrofag. Sebanyak dua puluh empat ekor mencit jantan galur Balb/C umur 2-3 bulan dengan berat badan 20-30 gram dibagi ke dalam 6 kelompok. Kelompok pertama mendapat pemberian ekstrak etanol Spons Xestospongia Sp. 100 mg/kgBB, kelompok kedua mendapat pemberian ekstrak etanol Spons Xestospongia Sp. 200 mg/kgBB, kelompok ketiga mendapat pemberian ekstrak etanol Spons Xestospongia Sp. 300 mg/kgBB dan kelompok keempat mendapat pemberian ekstrak etanol Spons Xestospongia Sp. 400 mg/kgBB. Kelompok kontrol positif mendapat ekstrak Phyllanthus niruri Linn. (Stimuno®) 0,13 mg/gBB dan kelompok kontrol negatif mendapatkan Na-CMC 0,5%. Ekstrak diberikan secara peroral sejak hari pertama hingga ketujuh. Pada hari kedelapan masing-masing mencit diinjeksikan bakteri Staphylococcus aureus (SA) 0,5 mL secara intra peritoneal. Aktivitas sel makrofag dihitung dari apusan cairan peritoneum mencit. Peningkatan dosis ekstrak etanol Spons Xestospongia Sp. meningkatkan jumlah aktivitas fagositosis makrofag dari 24,25 % (Na-CMC), 34,25% (100 mg/kgBB), 47,00% (200 mg/kgBB), 59,50 % (300 mg/kgBB) dan 62,75% (400 mg/kgBB). Hasil penelitian menunjukan bahwa ekstrak etanol Spons Xestospongia Sp. memiliki potensi sebagai imunomodulator pada dosis 300 mg/kgBB dan 400 mg/kgBB dengan efektivitas yang tidak berbeda bermakna dengan kontrol positif (Stimuno®) dalam meningkatkan aktivitas fagositosis sel makrofag berdasarkan hasil uji statistik post hoc TUKEY (sig. > 0,05).

Abstract

Hyperlipidemia is an increase of lipid due to excessive fat consumption, that the intake and alteration of lipid is not balanced. Notika are plants that contain secondary metabolites in the form of flavonoids, tannins, saponins and triterpenoids which are antioxidant compounds that are thought to have the effect of reducing blood lipids. This study aims to determine the effect of ethanol extract of leaves of Notika on decreasing total cholesterol levels in rats induced by egg and propylthiouracil (PTU). This research was experimental in 6 groups, namely 4 groups given ethanol extract of leaves of Notika at a dose of 50 mg/kgBB, 100 mg/kgBB, 200 mg/kgBB, and 300 mg/kgBB, one group was given Na CMC 0.5% as a negative control, and one group was given simvastatin 10 mg as a positive control. The results showed that the ethanol extract of the leaves of Notika significantly reduced the total cholesterol level of rats with a significant value 0.05 and 300 mg/kgBB is an effective dose to reduce total cholesterol level.

Abstract

ABSTRAK Batang bambu-bambu (Polygonum pulchrum) adalah salah satu tanaman yang berkhasiat sebagai antidiabetik dimana kandungan kimia batang bambu-bambu yang berpotensi sebagai antidiabetik antara lain flavonoid dan fitosterol. Fitosterol berkhasiat untuk meluruhkan kencing (diuretik) dan menurunkan kadar glukosa darah (hipoglikemik), karena peran senyawa aktif diantaranya β-sitosterol dan stigmasterol dan senyawa flavonoid berperan secara signifikan meningkatkan aktivitas enzim antioksidan dan mampu meregenerasi sel-sel β-pankreas yang rusak sehingga defisiensi insulin dapat diatasi juga serta dapat memperbaiki sensitifitas reseptor insulin. Penelitian ini bertujuan untuk mengetahui efek ekstrak batang bambu-bambu terhadap antidiabetik pada mencit (Mus musculus) Galur Balb/C yang diinduksi Streptozotocin. Pada penelitian ini merupakan penelitian eksperimen dengan menggunakan metode RAL (Rancagan Acak Lengkap) dimana digunakan 20 ekor mencit dibuat model diabetes dengan menggunakan streptozotocin 150 mg/kgBB secara intraperitonial. Kemudaian dibagi menjadi 5 kelompok perlakuan, yaitu: Kontrol negatif Na.CMC 0,5%, kontrol positif glibenklamide 5 mg, ekstrak dosis 250 mg/kgBB, 350 mg/kgBB, 450 mg/kgBB. Hasil analisis statistik menunjukkan aktivitas antidiabetes pada mencit Balb/C yang diinduksi STZ dengan Fhit>Ftabel0.05 = 12.531>3.06 berpengaruh secara signifikan pada taraf kepercayaan 0.05 dimana ketiga dosis ekstrak memiliki efektivitas sebagai antidiabetes jika dibandingkan dengan kelompok kontrol positif (glibenklamid). Kata Kunci : Ekstrak batang bambu-bambu, antidiabetik, streptozotocin, Mencit Balb/C, glukosa dara ABSTRACT Stems of bambu-bambu (Polygonum pulchrum) is one of the plants are useful as anti diabetic which the chemical content of the bambu-bambu stems potentially as anti diabetic among others Flavonoids and Phytosterols. Fitosterol using to Shed urine (diuretic) and lower the blood glucose (hypoglycemic, because the role of active compounds such as β-sitosterol and stigmasterol and flavonoid compounds play a significant role in increasing antioxidant enzyme activity and able to regenerate damaged β-pancreatic cells so that, its creates insulin deficiency which can be overcome as well as to improve insulin receptor sensitivity. This study aimed to able to know the effect of antidiabetic in bambu-bambu stems extract toward mice (Mus musculus) Babl/C who inducted by streptozotocin (STZ). This study is kind of experimental study and using complete random design which is using twenty of mice as a diabetic model and its using streptozotocin 150 mg/kgBB intraperitonially. Then, it will be divided in to five groups of treatment namely negative control Na.CMC 0.5%, positive control glibenclamide 5 mg, extract dose 250 mg/kgBB, 350 mg/kgBB, and 450 mg/kgBB. The result of analysis statistics showing the lowering aktivity of antidiabetic at Fhit > F table0. 05 = 12.531>3.06 influential as significant 0,05. Then all three doses of extract have antidiabetic effectiveness when compared to the positive control group (glibenclamide). Abstract

Etlingera elatior (Wualae) termasuk dalam tumbuhan famili zingiberaceae yang memiliki aktivitas antiinflamasi. Namun, bioavailabilitas dan permeabilitas zat aktif yang rendah sehingga diperlukan suatu sistem penghantaran dalam komplek lipid-bahan alam. Sistem yang dikenal dengan fitofosfolipid ini merupakan penghantaran fitokonstituen dalam matriks fosfolipid yang dapat meningkatkan permeabilitas dan bioavailabilitas zat aktif melalui karakteristik ampifilik dan aksi emulsifikasi oleh fosfolipid. Penelitian ini bertujuan untuk melakukan formulasi dan karakterisasi sistem vesikel sehingga diperoleh sistem yang optimum antara ekstrak etanol buah wualae dan fosfatidilkolin yang memiliki karakteristik vesikel terbaik. Preparasi dilakukan    menggunakan    teknik    hidrasi    lapis   tipis dengan adanya variasi komposisi ekstrak dan fosfatidilkolin, yakni 0,5:0,5 (Formula 1); 0,5:1 (Formula 2); 1:0,5 (Formula 3); 1:1 (Formula 4); 1:2 (Formula 5); dan 2:1 (Formula 6). Hasil yang diperoleh adalah komposisi formula optimum antara ekstrak dan fosfatidilkolin yaitu pada F4 dengan komposisi 1:1 dengan karakterisasi meliputi pengamatan morfologi berbentuk bulat (sferis) menggunakan mikroskop optic, efisiensi penjerapan vesikel diperoleh 99,20%, ukuran partikel 898,2 nm dan indeks polidispersitas 0,573 dengan kategori vesikel jenis LUV (Large Unilamellar Vesicle) menggunakan Particel Size Analyzer.

Abstract

Fungsi sistem imun adalah untuk melindungi tubuh dari benda-benda asing, dimana sistem imun dapat ditingkatkan dengan pemberian imunomodulator. Pendekatan kemotaksonomi Genus Etlingera memungkinkan Etlingera rubroloba A.D. Poulsen memiliki kandungan metabolit sekunder sebagai agen imunomodulator. Penelitian dimaksudkan untuk mengetahui aktivitas imunomodulator ekstrak etanol buah Etlingera rubroloba A.D. Poulsen terhadap fagositosis sel makrofag pada tikus jantan galur wistar. Hewan uji tikus sebanyak dua puluh empat ekor dibagi dalam enam kelompok perlakuan yaitu kontrol negatif (Na-CMC 0,5%), kontrol positif (ekstrak meniran komersial), kontrol normal (tanpa perlakuan), perlakuan dosis 1 (200 mg/kgBB), perlakuan dosis 2 (300 mg/kgBB)  dan  perlakuan dosis 3  (400 mg/kgBB). Perlakuan diberikan selama tujuh hari berturut-turut secara per oral  dan diberikan injeksi bakteri Staphylococcus aureus 0,5 mL secara intraperitonial pada hari kedelapan masing-masing kelompok. Apusan cairan peritoneum digunakan untuk menentukan aktivitas sel makrofag. Hasil penelitian menunjukkan aktivitas fagositosis sel makrofag pada kelompok kontrol negatif, kontrol positif, perlakuan 1, perlakuan 2, perlakuan 3 berturut-turut 36,75 %, 74,25%, 71.75%, 80,50%, 70,75%. Hasil penelitian menunjukkan pemberian ekstrak etanol buah Etlingera rubroloba A.D. Poulsen memiliki aktivitas imunomodulator terhadap fagositosis sel makrofag.

Abstract

Plants Etlingera elatior (Wualae) is a family Zingiberaceae of ​​the genus Etlingera which has a large population in the world. The E. elatior plant is widely used empirically in the community to increase endurance and seasoning in fruit. In this study, ethanol extract was used to determine its potential and effectiveness as an antihyperuricemia (in vivo) was tested pharmacologically on Rattus novergicus L. In this study, twenty-four rats were grouped into six groups, namely the ethanol extract of wualae fruit at a dose (100 mg/kgBW, 200 mg/kgBW, 300 mg/kgBW, 400 mg/kgBW), and the negative control group Sodium CMC 0 .5%, and the positive control group used Allopurinol 10 mg/kgBW. Furthermore, To increase the value of uric acid levels, the rats were first induced with 250 mg/kgBW Potassium Oxonate via intraperitoneal (i.p). Furthermore, the preparation according to the treatment was given orally one hour later after the administration of potassium oxonate. At the hour (1, 2, 3), the blood was taken intravenously (i.v) through the rat's tail, and then the uric acid level was analyzed with a photometer. All doses used in this study, namely 100, 200, 300, and 400 mg/kgBW, showed that they could reduce uric acid overall. The effective dose used is the dose (300 mg/kgBW, 400 mg/kgBW). This research can be concluded that the ethanol extract of the fruit of Etlingera elatior (Jack) R.M. Smith (Wualae) has potential as an antihyperuricemia (uric acid) and can be developed as a traditional medicine

Abstract

Breast cancer is a type of cancer with a high prevalence in the world and causes death in women. Chemotherapy is one type of treatment that is widely used. However, the problems of chemotherapy drugs such as side effects, drug resistance and inadequate efficacy. So, to overcome these problems, many natural ingredients have been explored to find anticancer agents that are expected to have good efficacy with minor side effects. Kasumba turate flowers (Carthamus tinctorius Linn.) is a plant from the Asteraceae tribe which is known to contain secondary metabolites with pharmacological activity as anticancer. This study aims to determine the types of secondary metabolites and the cytotoxic activity of the ethanolic extract of Kasumba turate flower against the T47D breast cancer cell line using the MTT assay method. Kasumba turate flower extract was obtained by maceration using 96% ethanol solvent, so that a concentrated extract with a weight of 107.8 grams was obtained with a yield value of 10.81%. The extract obtained was then subjected to a phytochemical screening test using the tube method and the results are alkaloids, flavonoids, tannins, and terpenoids. In the cytotoxic activity test, the test samples were varied into 7 concentration series, namely 10, 50, 100, 200, 400, 800 and 1000 ppm. The positive control used was 5-Fu. Cytotoxic parameter (IC50 value) was determined using GraphPad Prism and obtained IC50 5-Fu value of 65.88 ppm with active category while IC50 of the test sample was 157.3 with moderately active category as anticancer breast.

Abstract

Etlingera is one of the plant genera from the Zingiberaceae family and is widely distributed in Asia to the Pacific Islands, with various species ranging from 150 to 200 species. The Etlingera are commonly used as spices, vegetables, and traditional medicines. Many pharmacological activities have been reported from this genus, including antioxidants, antibacterials, cholesterol-lowering, anticancer, and others. The phytochemical content of Etlingera reported are phenolics, diarylheptanoids, flavonoids, steroids, alkaloids, and terpenoids. Eight species from Etlingera, namely Etlingera elatior, E. pavieana, E. brevilabrum, E. pyramidosphaera, E. megalocheilos, E. coccinea, E. fimbriobracteata, and E. corneri are reported provided anticancer activity. This review article aims to review the phytochemical screening from Etlingera genus, and its anticancer activity.

Abstract

The utilization of the stems, leaves, and hulls of peanuts (Arachis hypogea) is not as popular as the seeds. This study aimed to investigate the chemical contents and pharmacological activities of A. hypogaea stems in-vitro and in-silico. This study was also completed with bibliometric analysis. The methanol extract (ME) was reextracted by ethylacetate to get ethyl acetate extract (EAE). The chemical contents of EAE were analyzed by phytochemical screening, Liquid Chromatography-Mass Spectroscopy (LC-MS/MS), TPC (Total Phenolic Content), and TFC (Total Flavonoid Content). In-vitro and in-silico studies evaluated antioxidant potency, toxicity, and cytotoxicity toward MCF-7 cell lines. The results showed that EAE contained terpenoids, flavonoids, alkaloids, and phenolics which were supported by LC-MS/MS data. The EAE was categorized as a very strong antioxidant and moderately active in both cytotoxicity and toxicity.
