Abstract

Using leaf explants, this research aimed at identifying the type of growth response that can be created and the optimal mix of media for the growth of Cattleya sp orchid. This was an experimental investigation employing the RAL method (completely randomized design) with a combination of 2.4-D (0;1; 2; 3 ppm) and Kinetin  (0;0.3;0.6 ppm) repeated three times. According to the results of this study, the growth regulators 2.4-D and kinetin  were unable to promote the development of all explants. Explants transferred to media containing polyvinylpyrrolidone (PVP) to prevent browning after 12 weeks of observation. Then the results of the study also found the emergence of shoots in the treatment P0 (control), P2 (2,4-D 0 + Kinetin  0.6 ppm), P8 (2,4-D 2 + Kinetin  0.6) and P11 (2.4- D 3 + K 0.6 ppm) in the 3rd week of observation, the appearance of callus at P5 (2.4-D 1 + Kinetin  0.6 ppm) in the 4th week of observation

Abstract

 Penelitian ini bertujuan untuk Untuk mendapatkan teknik sterilisasi yang terbaik untuk eksplan batang anggrek Cattleya sp. muda. Penelitian ini dilakukan di Laboratorium Kultur Jaringan YAHDI Perum Pelabuhan Jl. Lambung No. 8 Tanah 600 Medan Marelan. Rancangan penelitian yang digunakan dalam penelitian ini adalah metode eksperimental laboratorium dalam mencobakan beberapa teknik sterilisasi. Untuk mengetahui teknik sterilisasi yang tepat, penelitian ini menggunakan eksplan lapang anggrek Cattleya Sp. dengan jenis pendekatan deskriptif kuantitatif. Data dianalisis dan ditabulasi menggunakan parameter penelitian hari terkontaminasi, persentase eksplan yang terkontaminasi, persentase eksplan browning, persentase eksplan yang membengkak, persentase eksplan yang membentuk bakal tunas dan persentase eksplan hidup. Hasil penelitian ini menunjukkan bahwa terdapat 2 teknik sterilisasi yang optimal dalam sterilisasi eksplan lapang Anggrek Cattleya Sp. yaitu teknik sterilisasi 2 dan 4. Teknik sterilisasi 2 menggunakan zat sterilan deterjen, akuades steril, Tween-20, benomil 50%, agrimicin, NaOCl 15% 10% dan alcohol 70% kemudian teknik sterilisasi 4 dengan zat sterilan Clorox 1,05%, HgCl 0,02% dan akuades steril merupakan teknik sterilisasi yang tersebut. Hal ini ditunjukkan dengan rendahnya tingkat kontaminasi eksplan dan tampaknya spot-spot hijau pada eksplan dimana eksplan akan mengalami pertumbuhan tunas. Faktor-faktor yang menyebabkan kontaminasi pada eksplan Anggrek Cattleya Sp. adalah kurangnya kehati-hatian praktikan pada saat melakukan sterilisasi eksplan lapang, kontaminasi endogenus yang tinggi dan penggunaan zat steril yang kurang tepat

Abstract

Revolusi Industri 4.0 dan era society 5.0 saat ini sedang kita hadapi. Seiring dengan hal tersebut, maka riset biologi juga berkembang pesat. STEM berperan penting dalam menggali potensi Biodiversitas dan riset biologi melalui berbagai model pembelajaran yang mengasah kemampuan berfikir kritis, berfikir kreatif, berkomunikasi dengan baik, berfikir tingkat tinggi (HOTS), berkolaborasi dalam proses pendidikan. Pelaksanaan Project base learning, Problem Base Learning, inquiry learning, Discovery Learning, Case Methode menjadi terintegrasi dengan proses pendidikan melalui STEM. Metaverse menawarkan cara baru bagi pengguna khusunya pada bidang pendidikan  untuk berkolaborasi secara virtual dan berperan didalamnya. Di dunia riset dan edukasi, metaverse membawa dampak, khusunya pada Biodiversitas Indonesia yang dapat divisualisasikan secara virtual.Kata kunci: Revolusi Industri 4.0, STEM, Metaverse

Abstract

This study develops and tests (LKPD) based on search, solve, create, and share (SSCS) in biology learning for class X MIA Semester I at SMA Negeri 5 Medan. To actively assess students' learning interests, science process skills, and learning outcomes at school because this process helps students search, find, and build their knowledge to solve problems. This research is development. Uses 4D development research (Define, Design, Develop, Disseminate). Interviews, material expert validation questionnaires, design validation questionnaires, language validation questionnaires, study field teacher responses to SSCS-based LKPD, student learning interest questionnaires, student science process skills questionnaires, and pretest-posttest assessment sheets were used. Material experts rate material validity as "Very Good/Decent" with an average of 91.75%. Design expert product quality results averaged 87.5% "Good/Decent". Teacher responses on the SSCS-based LKPD averaged 86.25% "Good/Decent". Students are 71% "Interested" in learning in small groups and 88% "Very Interested" in large groups. Science students' process skills averaged 86.67 in the big group and 76.67 in the small group in the "Good/Decent" category. Out of 37 students, 4 had a "Medium" N-gain of 0.7 on the biodiversity and virus meters, whereas 33 had a "High" N-gain of 0.8.

Abstract

ABSTRACT This study aims to determine the effect of soaking Adenanthera pavonina L with various concentrations of coconut water to improve the quality of germinated seeds and determine the best concentration of soaking Adenanthera pavonina in coconut water to maintain the quality of germinated seeds. The research method used was a completely randomized design (CRD) with 4 treatments, namely 1 control and 5 repetitions. The treatments consisted of: K0 = saga seeds without soaking coconut water; K1 = soaking saga seeds in 10% concentration coconut water; K2 = soaking saga seeds in 20% concentration coconut water; K3 = soaking saga seeds in 30% concentration coconut water; and K4 = soaking saga seeds in 40% concentration coconut water. The study showed that soaking Adenanthera pavonina seeds in 10% coconut water had a significant effect on the height of seed germination (3.36 cm) and soaking Adenanthera pavonina seeds in coconut water had no significant difference on seed germination power and seed germination time.

Abstract

This study aims to determine the effect of plant growth regulators and seed cutting patterns on the induction of mangosteen shoot growth. The research was conducted at the YAHDI Tissue Culture Laboratory in Medan, North Sumatra, Indonesia, in 20245. The method used in this study was a Completely Randomized Design (CRD) with two factors: BAP concentration and seed cutting pattern. The data collected were analyzed using statistical analysis of variance (ANOVA) with Duncan's Multiple Range Test (DMRT) at a 5% significance level using SPSS 26. The results of the study show that the interaction between BAP concentration and seed cutting pattern significantly affected the time of shoot emergence, the number of leaves, and the shoot height. The best shoot emergence time was obtained with a BAP treatment of 4 mg/L using whole seed explants, which occurred at 1 MST (Month After Sowing). A BAP concentration of 2 mg/L with whole seed explants produced the highest number of leaves, with 8 leaves. The highest number of shoots was obtained from explants planted in a medium with 6 mg/L BAP, which resulted in 5.67 shoots. Whole seeds planted in a medium with 0 mg/L BAP produced the tallest plants, measuring 8.9 cm. The combination of BAP concentration and seed cutting pattern did not significantly affect the number of roots.

Abstract

The research on the development of this textbook was carried out based on the results of the analysis of student needs, namely that there are no teaching materials that can improve students' science process skills in the Tissue Culture course at the Department of Biology, State University of Medan. This textbook is compiled based on the learning syntax of Project-Based Learning (PjBL). This study aims to develop a Project-Based Learning-based tissue culture textbook to improve students' science process skills. This research method uses the Research and Development (R&D) with Thiagarajan's 4D (Define, Design, Develop, and Disseminate). The population of this study was students of the Biology Department of Universitas Negeri Medan with a sample of 47 people who were given a PjBL-based tissue culture textbook. The results showed that (1) the textbooks have been tested for feasibility by material experts (87,73) (2) instructional design experts (92) with a very feasible category, (3) the response of tissue culture lecturers (92) and (4) students based on individual tests (95.1), small group tests (93,75), and limited group tests (90.02) with a very good category and (5) the results of the calculation of N-Gain for science process skills were obtained at 0.66 which is in the medium category.

Abstract

Orchids are ornamental plants that have high aesthetic value and beauty. Many human activities use this plant, including weddings, Christmas, New Year and birthdays. Orchid propagation can be done conventionally, but it requires a large area of land and takes a long time to grow. However, it can also be multiplied using tissue culture. Plant tissue culture techniques have better prospects than conventional propagation because tissue culture can produce large numbers of new plants without being influenced by time or season. This research aims to determine the effect of IAA and leaf order on shoot growth of Cattleya sp orchid plants. This type of research is experimental research with a Factorial Completely Randomized Design (CRD) with 12 treatments and 3 replications. There are 2 factors in this study, namely factor I PGR IAA (0, 0.5 mg/l, 1 mg/l, and 1.5 mg/l) and factor II Leaf order (1st leaf, 2nd leaf, and 3rd leaf). This research is a quantitative descriptive study with the parameters of the time of shoot emergence and using 2-way Statistics with Analysis of Variance (ANOVA) with the parameters of number of shoots, number of leaves (strands), and plantlet height. From the research results, it was found that the administration of IAA (Indole Acetic Acid) and the order of the leaves did not have a real effect on the number of shoots, number of leaves (strands) and plantlet height. However, the best treatment is the I0.5D2 treatment (IAA 0.5 mg/l and 2nd leaf).

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh model pembelajaran Project Based Learning terhadap hasil belajar siswa pada materi sistem peredaran Darah di kelas XI IPA di SMA Negeri 1 Barumun Selatan. Desain penelitian yang digunakan adalah pretest-posttest control group design. Pengambilan sampel diambil secara purposive sampling. Penelitian ini menggunakan 1 kelas eksperimen dan 1 kelas kontrol . Setelah diberikan perlakuan, kelas eksperimen dan kelas kontrol akan diberikan posttest untuk mengetahui hasil belajar pasca perlakuan. Teknik analisis data yang digunakan pada penelitian ini terdiri dari uji prasyarat dan uji hipotesis. Uji prasyarat dilakukan dengan menggunakan uji normalitas Shapiro- Wilk dan uji homogenitas Levene Pada penelitian ini didapatkan hasil rata-rata nilai pretest kelas eksperimen adalah 47,92 dan kelas kontrol adalah 46,91. Nilai postest yang diperoleh dari kelas eksperimen (80,00) lebih tinggi dibandingkan pada kelas kontrol (75,00). Rata-rata nilai pretest dan posttest kelas eksperimen dan kontrol dilakukan uji normalitas Shapiro-Wilk dan uji homogenitas Levene Uji normalitas diperoleh hasil dengan pretest 0,398 dan nilai posttest 0,322. Berdasarkan hal tersebut, dapat dikatakan rata- rata nilai pretest dan posttest kelas eksperimen dan kontrol berdistribusi normal. Uji homogenitas diperoleh nilai pretest 0,340 dan nilai posttest 0,278, dimana pada perhitungan uji-t pada data nilai postest diperoleh 0,003 < 0,05 dimana hal ini menunjukkan bahwasanya model pembelajaran Project Based Learning memberikan pengaruh terhadap hasil belajar.

Abstract

Penggunaan pupuk organik, baik dalam bentuk padat maupun cair, mampu meningkatkan kualitas tanaman secara keseluruhan. Tanaman yang dipupuk dengan organik cenderung menghasilkan panen yang lebih melimpah, dengan bobot yang lebih berat, kesegaran yang lebih lama, serta cita rasa yang lebih lezat dibandingkan dengan tanaman yang dipupuk dengan bahan kimia. Kandungan mineral, zat-zat penting, dan hormon pertumbuhan tanaman yang terdapat dalam pupuk organik cair turut berkontribusi dalam memaksimalkan potensi pertumbuhan tanaman. Lebih lanjut, dibandingkan dengan pupuk kimia, pupuk organik, khususnya dalam bentuk cair, lebih efektif dalam merangsang pertumbuhan vegetatif tanaman dan meningkatkan kapasitas tanah dalam menyerap serta menahan unsur hara yang dibutuhkan tanaman. Penelitian ini secara mendalam mengeksplorasi bagaimana variasi konsentrasi pupuk organik cair yang berasal dari bekas limbah beras dapat memberikan pengaruh yang berbeda-beda bagi proses penumbuhan bayam. Melalui pendekatan kuantitatif yang teliti, penelitian eksperimental ini dirancang untuk mengumpulkan data empiris yang dapat diuji secara statistik. Dengan menggunakan rancangan acak lengkap, penelitian ini membandingkan pertumbuhan tanaman bayam yang diberi perlakuan pupuk dengan dua tingkat konsentrasi yang berbeda, yaitu 80% dan 100%. Setelah dilakukan pengamatan lapangan selama 28 hari di Dusun II, Gang Buntu, Kecamatan Tanjung Morawa, data pertumbuhan tanaman bayam yang diperoleh kemudian diolah secara komprehensif menggunakan aplikasi perangkat lunak Excel. Analisis deskriptif yang dilakukan bertujuan untuk memberikan gambaran yang jelas mengenai pola pertumbuhan tanaman sebagai respons terhadap variasi konsentrasi pupuk organik cair yang diberikan. Penelitian ini memberikan bukti empiris mengenai potensi pemanfaatan limbah rumah tangga, yaitu air cucian beras, sebagai sumber nutrisi tanaman. Hasil penelitian menunjukkan bahwa pemberian pupuk organik cair dengan konsentrasi 100% memberikan pengaruh yang signifikan terhadap pertumbuhan tanaman bayam. Namun demikian, penelitian ini juga menyarankan perlunya penelitian lebih lanjut untuk mengoptimalkan penggunaan pupuk organik cair dalam sistem budidaya tanaman.
