Abstract

The use of pocket book as a learning media is still limited. This will make it difficult for students to understand the material. The purpose of this research was to determine the feasibility results of tissue culture pocket book, the level of product feasibility through responses from lecturers, students and to see the effectiveness of tissue culture pocket book products especially in tool sterilization and in vitro propagation topics. The research method uses R&D with 4D models from Thiagrajaan. The results of the needs analysis showed that 66,7% of students had difficulty in understanding the material of tool sterilization and in vitro propagation. In curriculum analysis using the KKNI curriculum. The feasibility level of the assessment product from media experts obtained the criteria "Very worthy" with percentage of 91,67%, assessment of material experts with the criteria "worhty" with percentage of 84,74% and assessment results of learning design experts with the criteria "Very worthy" with percentage of 91,67%. Then the lecturer's response obtained results with the criteria "Worthy" with a percentage of 83,75%. In small group trials with scores of 89.4% and limited field trials of 90%. The disseminate stage obtained an N-Gain score of 0,56 with the "medium" criteria. ABSTRAK: Penggunaan buku saku sebagai media pembelajaran masih terbatas. Hal ini akan menyulitkan mahasiswa dalam memahami materi pembelajaran. Tujuan dari penelitian ini adalah untuk mengetahui hasil kelayakan buku saku kultur jaringan, tingkat kelayakan produk melalui tanggapan dari dosen, mahasiswa dan untuk melihat efektivitas produk buku saku kultur jaringan khususnya pada topik sterilisasi alat dan perbanyakan in vitro. Dalam penelitian ini menggunakan R&D dengan model 4D Thiagrajaan. Hasil analisis kebutuhan menunjukkan bahwa 66,7% mahasiswa mengalami kesulitan dalam memahami materi sterilisasi alat dan propagasi in vitro. Kurikulum yang digunakan adalah KKNI. Tingkat kelayakan produk penilaian dari ahli media diperoleh hasil dengan kriteria "Sangat layak" dengan persentase 91,67%, penilaian ahli materi dengan kriteria "Layak" dengan persentase 84,74% dan hasil penilaian ahli desain pembelajaran dengan kriteria "Sangat layak" dengan persentase 91,67%. Kemudian respon dosen diperoleh hasil dengan kriteria "Layak" dengan persentase 83,75%. Dalam uji coba kelompok kecil diperoleh skor 89,4% dan uji coba lapangan terbatas 90%. Tahap disseminate memperoleh skor N-Gain 0,56 dengan kriteria "sedang".

Abstract

This research aims to develop a digital comic by determining the feasibility and the effectiveness of using material in the plant tissue culture to help students understand the material presented in the course at the Department of Biology, Universitas Negeri Medan. Data collection techniques include observation, giving questionnaires, and giving instruments to instructional experts, material experts, and media experts. The research method used is Research and Development (RD), which uses the ADDIE development model. This development research resulted in the creation of a digital comic learning media based on mobile learning focused on sterilization and planting materials, developed using the ADDIE (Analysis, Design, Development, Implementation, Evaluation) model.  Feasibility test results indicated high approval ratings: 93.75% from instructional experts in the “highly feasible”, 84.5% from material experts in the “highly feasible”, and 95.4% from media experts in the “highly feasible”. Lecturer responses were also favorable, with scores of 79.9. Student responses were also highly favorable, with scores of 91 from an individual group, 90.2 from a small group, and 94.2 from a limited group. Based on the data analysis, digital comic learning media based on mobile learning is effective enough for use in plant tissue culture courses.

Abstract

This study aims to obtain a good concentration of IBA and plantain and is suitable for root induction in Dendrobium sp. The benefit of this study is to provide information about the successful root induction of orchid plantlets Dendrobium sp. In vitro culture and creating formulations using IBA hormones and organic plantain ingredients in in vitro culture media optimally. This research has been carried out at the YAHDI Plant Tissue Culture Laboratory. Rooting orchids Dendrobium sp. In vitro using MS basic media which was added with IBA (Indole 3 Butyric Acid) 0 ppm, 2 ppm, 4 ppm, and 6 ppm, and added with plantain pulp with levels of 0 g/l, 100 g/l. and 200 g/l. This study was a factorial experiment with 2 factors compiled based on a 4 x 3 Complete Random Design (RAL) with 12 treatment combinations. The data obtained was analyzed for variance analysis (ANOVA). If there are results that have a real effect, it is followed by the DMRT (Duncan Multiple Range Test) test at the level of 5%. (1) The results of the study showed that the treatment of the variables of the number of roots, plant height, and root length was influenced by IBA. (2) The treatment of the variables of the number of buds, the number of leaves, the number of roots, the height of the plant, and the length of the roots is influenced by plantains. (3) The treatment of the variables on the number of roots, root length, and leaf width were influenced by the interaction of IBA and banana pulp.

Abstract

This study aimed to determine the effect and identify the optimal sterilization pattern for suppressing contaminant growth in the in vitro culture of Sidikalang Arabica coffee (Coffea arabica L.) leaf explants. The experiment was conducted using a completely randomized design (CRD) with five treatment levels and 15 replications. Data were analyzed using the Kruskal Wallis test, and if significant differences were found, the Mann Whitney test was applied at a 5% significance level. The results showed that sterilization patterns significantly affected the time to contamination onset, the percentage of contaminated explants, the percentage of browning explants, and the percentage of explants forming callus. However, no significant effect was observed on the time of callus initiation. Sterilization pattern 5 was identified as the most optimal in suppressing contamination, with the lowest contamination percentage (6,6%), browning (6,6%), and the highest callus formation (93,3%). This pattern involved washing with detergent; soaking in Agrept (30 minutes) and Benlate (30 minutes); followed by sterilization in a laminar airflow cabinet (LAFC) using Tween 20 (20 minutes), graded Bayclin at 10%, 20%, and 30% (for 20, 15, and 5 minutes, respectively), 70% alcohol (20 seconds), and ascorbic acid (20 minutes).

Abstract

Revolusi Industri 4.0 dan era society 5.0 saat ini sedang kita hadapi. Seiring dengan hal tersebut, maka riset biologi juga berkembang pesat. STEM berperan penting dalam menggali potensi Biodiversitas dan riset biologi melalui berbagai model pembelajaran yang mengasah kemampuan berfikir kritis, berfikir kreatif, berkomunikasi dengan baik, berfikir tingkat tinggi (HOTS), berkolaborasi dalam proses pendidikan. Pelaksanaan Project base learning, Problem Base Learning, inquiry learning, Discovery Learning, Case Methode menjadi terintegrasi dengan proses pendidikan melalui STEM. Metaverse menawarkan cara baru bagi pengguna khusunya pada bidang pendidikan  untuk berkolaborasi secara virtual dan berperan didalamnya. Di dunia riset dan edukasi, metaverse membawa dampak, khusunya pada Biodiversitas Indonesia yang dapat divisualisasikan secara virtual.Kata kunci: Revolusi Industri 4.0, STEM, Metaverse

Abstract

Penelitian ini menelusuri makna falsafah kepemimpinan dalam budaya Minangkabau dan relevansinya bagi praktik kepemimpinan di Indonesia masa kini. Metode yang digunakan ialah pendekatan kualitatif dengan dukungan kuantitatif korelasional, melalui wawancara mendalam dan penyebaran angket kepada informan terpilih secara purposive. Analisis data dilakukan menggunakan uji korelasi Pearson untuk melihat hubungan antara tingkat pemahaman terhadap nilai-nilai kepemimpinan adat dan sikap penerapannya dalam konteks nasional. Hasil penelitian memperlihatkan bahwa pemahaman responden terhadap nilai-nilai kepemimpinan Minangkabau tergolong sangat tinggi (rata-rata skor 4,87 dari 5), sedangkan nilai korelasi r = 0,97 menunjukkan hubungan positif yang kuat antara kedua variabel tersebut. Temuan ini membuktikan bahwa semakin mendalam pemahaman seseorang terhadap nilai-nilai seperti adat basandi syarak, syarak basandi Kitabullah, bajanjang naik batanggo turun, serta duduak samo randah tagak samo tinggi, semakin bijak pula sikap kepemimpinannya. Nilai-nilai falsafah Minangkabau ini terbukti tetap relevan sebagai pedoman moral dan etis untuk membentuk karakter pemimpin yang adil, berintegritas, serta demokratis di Indonesia

Abstract

Orchids are ornamental plants that have high aesthetic value and beauty. Many human activities use this plant, including weddings, Christmas, New Year and birthdays. Orchid propagation can be done conventionally, but it requires a large area of land and takes a long time to grow. However, it can also be multiplied using tissue culture. Plant tissue culture techniques have better prospects than conventional propagation because tissue culture can produce large numbers of new plants without being influenced by time or season. This research aims to determine the effect of IAA and leaf order on shoot growth of Cattleya sp orchid plants. This type of research is experimental research with a Factorial Completely Randomized Design (CRD) with 12 treatments and 3 replications. There are 2 factors in this study, namely factor I PGR IAA (0, 0.5 mg/l, 1 mg/l, and 1.5 mg/l) and factor II Leaf order (1st leaf, 2nd leaf, and 3rd leaf). This research is a quantitative descriptive study with the parameters of the time of shoot emergence and using 2-way Statistics with Analysis of Variance (ANOVA) with the parameters of number of shoots, number of leaves (strands), and plantlet height. From the research results, it was found that the administration of IAA (Indole Acetic Acid) and the order of the leaves did not have a real effect on the number of shoots, number of leaves (strands) and plantlet height. However, the best treatment is the I0.5D2 treatment (IAA 0.5 mg/l and 2nd leaf).

Abstract

This study aims to determine the effect of plant growth regulators and seed cutting patterns on the induction of mangosteen shoot growth. The research was conducted at the YAHDI Tissue Culture Laboratory in Medan, North Sumatra, Indonesia, in 20245. The method used in this study was a Completely Randomized Design (CRD) with two factors: BAP concentration and seed cutting pattern. The data collected were analyzed using statistical analysis of variance (ANOVA) with Duncan's Multiple Range Test (DMRT) at a 5% significance level using SPSS 26. The results of the study show that the interaction between BAP concentration and seed cutting pattern significantly affected the time of shoot emergence, the number of leaves, and the shoot height. The best shoot emergence time was obtained with a BAP treatment of 4 mg/L using whole seed explants, which occurred at 1 MST (Month After Sowing). A BAP concentration of 2 mg/L with whole seed explants produced the highest number of leaves, with 8 leaves. The highest number of shoots was obtained from explants planted in a medium with 6 mg/L BAP, which resulted in 5.67 shoots. Whole seeds planted in a medium with 0 mg/L BAP produced the tallest plants, measuring 8.9 cm. The combination of BAP concentration and seed cutting pattern did not significantly affect the number of roots.

Abstract

The research on the development of this textbook was carried out based on the results of the analysis of student needs, namely that there are no teaching materials that can improve students' science process skills in the Tissue Culture course at the Department of Biology, State University of Medan. This textbook is compiled based on the learning syntax of Project-Based Learning (PjBL). This study aims to develop a Project-Based Learning-based tissue culture textbook to improve students' science process skills. This research method uses the Research and Development (R&D) with Thiagarajan's 4D (Define, Design, Develop, and Disseminate). The population of this study was students of the Biology Department of Universitas Negeri Medan with a sample of 47 people who were given a PjBL-based tissue culture textbook. The results showed that (1) the textbooks have been tested for feasibility by material experts (87,73) (2) instructional design experts (92) with a very feasible category, (3) the response of tissue culture lecturers (92) and (4) students based on individual tests (95.1), small group tests (93,75), and limited group tests (90.02) with a very good category and (5) the results of the calculation of N-Gain for science process skills were obtained at 0.66 which is in the medium category.

Abstract

This study evaluates the effects of NaOCl concentration and immersion time on contamination levels and viability of in vitro mangosteen seed explants, aiming to identify the optimal sterilization protocol. Conducted at YAHDI Tissue Culture Laboratory, Medan, Indonesia, a Randomized Complete Block Design tested three treatment combinations: (1) 15% and 10% NaOCl for 5 minutes, (2) 20% and 15% NaOCl for 15 minutes, and (3) 30% and 20% NaOCl for 25 minutes. After immersion, explants were rinsed three times with sterile distilled water. Observations spanned 12 MST, with data analyzed using ANOVA and Games-Howell post hoc tests; contamination sources were qualitatively described. Results showed the first combination had the lowest effectiveness, the second was better but suboptimal, while the third significantly delayed contamination and yielded the highest explant survival rate of 86.1%. The third treatment also prolonged the contamination-free period compared to others. Fungal and bacterial contaminants were predominant, highlighting the need for effective disinfectants against both. These findings indicate that the third combination is a promising sterilization protocol for mangosteen seed explants, especially for hard endocarp tissues prone to contamination.
