Abstract

The research aimed to know the effect of total protein extracted from black sea cucumbers (Holothuria leucospilota) on agglutination of mouse sperms (Mus musculus). The flesh of black sea cucumber  was homogenized in Tris Buffer Saline (TBS) at pH 7.4. The homogenate was added with ammonium sulphate 50%(w/v) for presipitating protein. The presipated protein was purified with 12.000 MWCO dialysis bag to remove ammonium sulphate. The purified protein was quantitated with Biuret Method. The concentration of purified protein was 4,97g/dL. There were five concentration proteins used mouse sperm agglutination test, i.e. 4.97, 3.52, 2.48, 1.17 g/dL and 0% (control). Mouse sperm suspension was prepared by soaking an epypydimal duct in 0,9% (w/v) NaCl solution. The purified protein in varied concentration was then used for agglutination of suspension of mouse sperm. This research used Completely Randomized Design (CRD) with 5 concentrations of protein  and 5 replications. The data of this research were analyzed by using ANOVA. The result showed that total protein extract of black sea cucumberwas significantly different on mouse sperm agglutination. The fastest time of agglutination of was indicated by concentrations of  4.97 g/dL. total proten of black sea cucumber. Keywords: Black Sea Cucumber (Holothuria leucospilota); mouse sperm suspension; sperm agglutination; total protein extract

Abstract

Agricultural practice is suspected to influence the availability of spiders in cashew plantation. The aim of this research was to study the diversity of spider in two different agricultural practices: monoculture and polyculture. The research was conducted in cashew plantations in Desa Kayangan (monoculture) and Desa Salut (polyculture), Lombok Utara. Two trapping techniques were used to sample the spiders: sweep net and pitfall trap. In each study area, 10 sampling sites were selected along line transect that was 5000 meters long. The study found 36 species of spiders from 12 families. The diversity and richness of spiders were higher in the polyculture cashew plantation than that in monoculture. Habitat structure was more complex in the polyculture cashew plantation; hence many species of spiders were able to coexist there.

Abstract

The purpose of this study was to measure the service quality of the implementing unit of higher education institutions, to measure the level of external customer satisfaction, and to analyze the relationship between service quality and external customer satisfaction levels. To achieve these objectives, research was carried out by combining observation methods, distributing questionnaires to 200 external customers, focus group discussions, and public consultations. As the unit of analysis in this study, external customers consist of parents of students, alumni, graduate users, and cooperation partners. The collected data were analyzed using descriptive statistical analysis, and non-parametric statistical analysis, namely Spearman rank correlation. The results showed: the service quality of higher education implementing units is identical to the level of satisfaction of external customers; achievement of the level of external customer satisfaction is from good to very good; external customer satisfaction of alumni and graduate users is in a good position, while external customers of students' parents and cooperation partners are in a very good position.

Abstract

The application of non-precoated Indirect Enzyme-Linked Immunosorbent Assay (ELISA) and Dot Immunobinding Assay (DIBA) employing monoclonal antibodies (MCA) against Yogyakarta isolate of Baculovirus oryctes Huger. was described. The MCA-Bv-4 having subclass of IgG2a and titer in vitro of 10^4 - 10^5 proved to be useful antibody for virus detection. The great potential of I-ELISA using MCA-Bv-4 has been it's specificity being able to discriminate between healthy an virus-infected coconut beetle (Oryctes rhinoceros L.). The assay could also differentiate between B. oryctes and Monodon baculovirus, the pathogen of shrimp disease. The best tissue for preparing virus antigen from crude extract was found to be the midgut of the beetle. Head, thorax, abdomen and tibia were not suitable for preparing the test antigen. The crude extract of beetle showed high endogenous enzyme activity to the substrate of DIBA, which precluded the detection of B. oryctes using DIBA. The MCA-Bv-4 could be used to improve the monitoring of the virus to support the program of biological control of coconut beetle using B. oryctes.

Abstract

Diversity and parasitization of egg parasitoid of Leptocorisa acuta on various rice cropping pattern. Study on parasitoid diversity and their parasitization on various rice cropping pattern was conducted during April - October 2012. Exploration of egg parasitoids was conducted by collecting host eggs from rice field in Lombok Island. Sample of Insect were collected by hand. Two species have been identified, namely Ooencyrtus malayensis and Hadronotus leptocorisae. The result showed that eggs of both parasitoid were distributed in all locations with composition 50%. Generally, three times rice cropping pattern showed higher rate of parasitization (31,45%) compared to two times (22,69%) and once cropping pattern (18,03%). Parasitoid and diversity index were similar in different time, although the tend to increase linearly with the rice growth. The Shannon diversity index H’ between 0,24-0,30. Domination index 0,62-0,51, evenness index (E) 0,35-0,43. The result suggest that parasitoid diversity and parasitization is depend on many factors, including rice cropping pattern.

Abstract

Sanurus sp., Acrocercops sp. and Helopeltis sp. are important pests of cashews in Nusa Tenggara Barat. Pesticide usage to control these pests has so far not been successful, and thus the usage of natural enemy as an alternative control methods should be developed. Spider represents one of the natural enemies which are found in cashew plantation. The aims of the research is to study spider population and its predatory ability in cashew plantation. The study has been conducted in an area of cashew plantation at Dusun Jugil, Desa Sambik Bangkol, Lombok Utara. Spider predation was directly observed for 78 days, starting from May to August 2009. Edgar formula was used to measure spider predation per day. The study found 19 species of spider consisting from eight families. The abundance of Neoscona theisi is higher than Tetragnatha maxillosa, but the proportion of N. theisi with prey is less than T. maxillosa. This phenomenon indicates that spider’s abundance, does not directly show its potential as natural enemy in that habitat. N. nautica consumed at least 3.84 preys per day, but not all of its prey represents the pest of cashew. N. theisi consumed 1.67 preys per day and all of its prey represents pests of cashew. This study show that the amount of prey which can be consumed by spider per day does not directly show the potential of spiders as natural enemey of cashew pest.

Abstract

ABSTRAK Pengamatan (monitoring) lalat buah pada daerah lahan kering merupakan kegiatan yang sangat penting dan mendasar dalam penerapan PHT, karena dari pengamatan lalat buah di lapangan dapat diperoleh informasi tentang spesies (jenis), kepadatan populasi, luas dan intensitas serangan, perkembangan populasi serta faktor-faktor iklim yang mempengaruhi perkembangan lalat buah. Hal lain yang sangat penting adalah adanya persamaan persepsi/pemahaman tentang kelimpahan, komposisi spesies lalat buah yang ada pada suatu wilayah/lokasi, terutama daerah lahan kering Lombok Barat. Data dan informasi tentang kelimpahan, spesies lalat buah di lahan kering masih kurang, oleh karena itu telah dilakukan penelitian tentang bagaimana kelimpahan dan komposisi spesies lalat buah pada lahan kering di Lombok Barat. Hasil penelitian menunjukkan bahwa Bactrocera philippinensis merupakan spesies lalat buah yang paling dominan dengan nilai kelimpahan paling tinggi 64,76 % di wilayah lahan kering Kabupaten Lombok Barat, dan kelimpahan paling rendah adalah B. badius 0,02 %. Komposisi spesies lalat buah Bactrocera spp. di wilayah lahan kering Kabupaten Lombok Barat berturut-turut dari yang paling besar sampai yang paling kecil adalah B. Philippinensis, B. Limbifera, B. cucurbitae, B. pepayae, B. caudata, B. beckerae, B. albistrigata, B. Umbrosa, B. calumniata, dan B. Badius. ABSTRACT Monitoring of fruit flies in dry land is a very important and fundamental activities related to the implementation of Integrated Pest management that will provide the crusial information of the species including abundance of population, attack intensity, development of population, as well as the climate factors affect the fruit flies population. In addition, from those monitoring would achieve the same perseption /undersanding of abundance and compisition of fruit flies in the specific location , particul;arly in dry land of West Lombok. Currently, there is a lack of data and information on the abundance and composition of fruit flies species. Therefor, the invantigation on the abundance and composition of fruit flies spesies on dry land of West Lombok has been conducted. Ther result of this investigation shows that the most dominat of fruit flies species in dry land of West Lombok is Bactrocera philippinensis with the highest abundance value about 64,76 % and the lowest one is B. badius about 0,02 %. The highest composition of Bactrocera spp is B. Philippinensis, followed by B. Limbifera, B. cucurbitae, B. pepayae, B. caudata, B. beckerae, B. albistrigata, B. umbrosa, B. calumniata, and B. Badius., respectively.

Abstract

ABSTRAK Hasil penelitian ini menunjukkan bahwa hama-hama yang menyerang tanaman sayuran tomat organik relatif sama dengan jenis hama yang menyerang sayuran secara umum yaitu Aphis sp, Thrips, Aulocophora similis, pengorok daun (Liriomyza sp.) pada stadia vegetaif, dan hama Bemisia tabacci, Heliothis armigera, dan lalat buah pada fase reproduktif. Sebagian besar pengaruh dosis dan saat perlakuan insektisida nabati (nimba) maupun hayati (Bacillus thuringiensis) secara sendiri-sendiri tidak berpengaruh terhadap penekanan populasi dan intensitas serangan hama penting tanaman sayuran tomat organik. Perlakuan kombinasi antara Nimba dan Bacillus thuringiensis cenderung memberikan hasil lebih baik dalam hal penekanan populasi dan intensitas serangan hama tanaman tomat yang dibudidayakan secara organik. ABSTRACT The rersult of this investigation shown that the insect pests in organic farming system system of vegetables were the same as generally found in vegetables, such as Aphis sp, Thrips, Aulocophora similis, leaf miner (Liriomyza sp.) at vegetative stage, and Bemisia tabacci Heliothis armigera, fruit fly at reproductive stage of tomatoes. Most impact of doses and time application of botanical (Neem) and biological insecticides (Bacillus thuringiensis) used in this experiment individually were not significant in suppressing the population and attack intensity of major pest on organic vegetables farming system. From the result of this investigation, it is suggested to use the non-chemical insecticides such as botanical insecticides (Neem) and Biological insecticides (Bacillus thuringiensis) in organic farming system of vegetables, but according to the result it will be better to combine both of them by scheduling rotation of aplication. However, it is needed to find out what combination would be the best other than scheduling rotation, for example mixturing of both agents and aplying them together . By using the correct doses, time application of botanical and biological insecticides will be useful in Pest Management practices on organic farming system. Therefore, to optimizes the outcome of this experiment, it is necessary to follow up the research on Nische management approaches. By doing this, it is expected to have the conprehenship model of Pest Management in Organic farming system.

Abstract

Interventions in the use of technology to farmers as an alternative in Pest Management in potato cultivation are highly expected by farmers in the Sembalun area. Therefore, it is worth trying its utilization on potato crops to control important pests that attack. The method used in this activity is the Action Research Method by applying the Participatory Action Program approach from participants through discussions, and group work on all activities. The results of the group discussion showed that the group members had largely not recognized the types and functions of natural enemies in Potato Pest Management. After joint observations accompanied by the team, participants actually knew the types of natural enemies, both predators, and parasitoids, but they did not know the role of natural enemies in the agroecosystem. After finishing the meeting, the group realized the importance of maintaining the presence of natural enemies of pests in the potato agroecosystem so that the use of pesticides, which had been the main control technique for pests, could be reduced. Thus, it is hoped that the potato farming business in Sembalun will increase profits and produce environmentally friendly products

Abstract

Pelaksanaan kegiatan pengabdian kepada masyarakat dilakukan di desa Puyung kecamatan Jonggat, Lombok Tengah. Kelompok sasaran adalah kelompok tani yang terbiasa menanam tanaman palawija, jagung dan padi. Kendala yang sering dialami oleh petani tersebut adalah serangan hama pada tanaman kedelai sejak fase vegetatif sampai generatif. Biasanya petani menyemprot dengan pestisida sehingga tidak disadari lama kelamaan akan menyebabkan serangga hama menjadi tahan dan merusak lingkungan. Selain itu, pestisida yang diaplikasi secara terus menerus dapat mengakibatkan matinya organisme bukan sasaran termasuk predator. Sehingga perlu dilakukan inovasi baru untuk mengendalikan hama selain menggunakan bahan kimia yang tidak ramah lingkungan. Inovasi baru yang dimaksud adalah menanam tanaman refugia sebagai perangkap musuh alami hama. Metode yang dilakukan adalah teknik survei dengan metode deskriptif eksploratif dilanjutkan dengan kaji tindak (Action Research) dengan menerapkan pendekatan program tindak partisipatif (Partisipatory Action Program) dari anggota kelompok tani melalui diskusi, dan kerja kelompok di seluruh kegiatan. Selanjutnya tahapan pelaksanaan yang diawali dengan pelatihan dengan teknik ceramah dan diskusi, dengan proporsi 30% teori dan 70% praktek lapang. Sebagai tindak lanjut dari kegiatan tersebut adalah dilakukan demonstrasi plot dengan menanam tanaman refugia seperti bunga matahari, kenikir, bunga kertas, di sekitar tanaman kedelai dari bulan Agustus sampai dengan bulan November 2021. Hasilnya kegiatan ini dapat disimpulkan bahwa, a) pengetahuan dan keterampilan anggota Kelompok tani meningkat dalam kaitannya dengan pengendalian hama, b) Kelompok tani mau menerapkan tanaman refugia sebagai perangkap musuh alami hama pada tanaman kedelai, c) terbentuknya paket teknologi tentang teknik pengendalian hama dengan memanfaatkan tanaman refugia sebagai tanaman perangkap musuh alami hama.
