Abstract

The study was conducted to investigate the clinico-pathological features of highly contagious disease occurred in chicken located in East and West Java during the outbreak in September-October 2003. Six farms located in Districts of Surabaya, Malang and Blitar of East Java had been visited. They were mainly commercial layer, breeder layer and breeder broiler, which the population was between 14.000, 80.000, and aged 17-70 weeks. Where as five farms in West Java (Districts of Bogor, Bekasi and their surrounding areas) were visited and consisted of commercial layer and breeder broiler, having population of 3000-16.000 and aged 11-53 weeks. Observation was made according to clinical, gross pathological and histopathological changes. Clinically, most of them had cyanotic wattle and comb and subcutaneous petechiation of non-feathered part of the legs. These were also seen in necropsy, accompanied by general circulatory disturbances in most organs: namely pectoral and thigh muscle, trachea, lungs, epicard, myocard, proventriculus, liver, kidney and ovary. In addition, the liver was congested, friable and necrotic in some parts. Histologically, hemorrhage and non suppurative inflammatory reaction were observed in the brain, skin (comb, wattle and non feathered leg), skeletal muscle, trachea, lung, heart, proventriculus, liver, kidney and ovary whereas vasculitis was found especially in the skin of the wattle and comb, brain and kidney. It is concluded that based on the clinicopathological findings the outbreak of poultry disease in East and West Java were attributed to highly pathogenic avian influenza.   Key words: Highly pathogenic avian influenza (HPAI), chicken, clinico-pathology, outbreak, East Java, West Java

Abstract

Ram spermatozoa are sensitive to extreme changes in temperature during the freeze-thawed process. The present study was conducted to examine the effects of addition of various sugars in Tris extender on sperm cryosurvival of Garut ram. Semen was collected using an artificial vagina from three mature rams once a week. Immediately after initial evaluation, semen was divided into five parts and diluted with Tris extender (control), Tris extender + 0.4% dextrose, Tris extender + 0.4% raffinose, Tris extender + 0.4% trehalose, and Tris extender + 0.4% sucrose, respectively. Semen was loaded in to 0.25 ml mini straw with the concentration of 200 million or 800 million motile spermatozoa per ml. Semen was equilibrated at 5oC for three hours, then frozen and stored in liquid nitrogen for seven days. Quality of processed-semen including percentages of motile spermatozoa (MS), live spermatozoa (LS), intact acrosome cap (IAC), and intact plasma membrane (IPM) were evaluated after dilution, equilibration, and thawing, respectively. Data were analyzed using completely randomized design with five treatments and six replicates. Means were compared significant difference test at 0.05 significant level. Results of this research showed that there was no significantly difference (P>0.05) between treatments for all sperm quality parameters after dilution and equilibration. Mean percentages of post thawing MS, LS, IAC, and IPM for dextrose (54.00; 68.00; 66.60, and 57.83%), raffinose (50.00; 64.33; 61.80, and 61.75%), trehalose (50.83; 65.67; 61.40 and 57.75%), and sucrose (49.00; 66.80; 58.50 and 58.50%) were significantly (P<0.05) higher than control (40.83; 52.67; 54.60, and 49.40%) respectively. In conclusion, addition of 0.4% dextrose, raffinose, trehalose or sucrose in Tris extender are effective in improving frozen semen quality of Garut ram. Abstract

Cauda epididymal spermatozoa could be used as an alternative source of gamete in the application of various reproductive technologies, because the spermatozoa is motile and has ability of fertilizing the oocyte. The purpose of this research was to examine the effect of addition of seminal plasma on the quality of ram cauda epididymal spermatozoa preserved at 3–5oC. Collected-spermatozoa was divided into five parts then centrifuged at 3,000 rpm for 30 min and the supernatant was removed. One-fifth was diluted with Tris extender (Tris) and the remaining was added 0.5 ml ram seminal plasma then diluted with Tris extender (PS-Tris), 15% AndroMed + 85% distilled water (PS-AM15), 20% AndroMed + 80% distilled water (PS-AM20), and 25% AndroMed + 75% distilled water (PS-AM25), respectively. Quality of collected-spermatozoa including spermatozoa concentration (SC), percentages of motile spermatozoa (MS), live spermatozoa (LS), abnormal spermatozoa (AS), cytoplasmic droplet (CD), and intact plasma membrane (IPM) were evaluated. Percentages of MS, LS, and IPM of diluted-spermatozoa was evaluated every day during preserved at 3–5oC for three days. Results of this study showed that mean SC, MS, LS, AS, CD, and IPM of fresh spermatozoa were 11,660 million/ml, 65, 81, 7.6, 10.2, and 82.2%, respectively. Addition of seminal plasma in cauda epididymal spermatozoa prior to dilution could maintain the quality of spermatozoa during preserved at 3–5oC for three days. At day-4 of storage, percentages of MS, LS, and IPM for PS-Tris (43, 58, and 59.2%), PS-AM20 (40.5, 53.75, and 53.75%), and PS-AM25 (40, 54.8, and 55.2%) were significantly (P<0.05) higher than Tris (21, 34.8, and 33.6%) and PS-AM15 (20, 40, and 42.2%). In conclusion, addition of seminal plasma in ram cauda epididymal spermatozoa prior to dilution with Tris or 20 and 25% AndroMed extenders could maintain the quality of spermatozoa during preserved at 3–5oC for three days, and could be used for artificial insemination or in vitro embryo production programs. Abstract

Artificial insemination represents one of technologies in livestock reproduction that can be applied to cattle, sheep, goats and other livestock. Application of livestock reproduction technology includes artificial insemination to increase reproductive efficiency. Semen processing is one critical phase in an artificial insemination program. The use of animal origin ingredient for semen extenders, such as egg yolk and milk, presents a risk of microbial contamination, which lead to the search for alternatives. To increase standard of quality, researchers exploits phyto-lesitin for semen extender and the results showed no significant differences in motility, viability, and acrosomal status of spermatozoa with phyto-lesitin extender when compared to tris-egg yolk-containing extenders. Abstract

Penelitian ini bertujuan meningkatkan mutu genetik ayam lokal melalui pendekatan genetik dan karakterisasi fenotip untuk keperluan seleksi. Gen cGH (Chicken Growth Hormon) merupakan salah satu gen yang bertanggungjawab terhadap sifat pertumbuhan ayam lokal. Ayam Tolaki adalah ayam lokal yang berasal dari Sulawesi Tenggara. Penelitian ini menggunakan metode fenotyping sifat produksi dan genotyping dengan Teknik PCR. Sebanyak 50 tetua ayam Tolaki dipelihara 4 minggu. Hasil Penelitian menunjukkan Rataan bobot badan ayam Tolaki berkisar antara 1.395 kg – 1.910 kg, Rataan pertambahan bobot badan sebesar 58,78 (g/ekor/minggu),  konsumsi ransum sebesar 81,37 (g/ekor/hari) dan konversi ransum 9,68. Pertambahan bobot badan pada jantan sedikit lebih tinggi (60,30 g/ekor/mg) dibandingkan dengan betina (58,42 g/ekor/mg). Pada penelitian ini berhasil diidentifikasi keberadaan gen cGH ayam Tolaki melalui ekstraksi DNA dan amplifikasi melalui mesin PCR dengan panjang Ruas DNA sebesar 399 bp. Kata Kunci : gen cGH, ayam Tolaki,  PCR, fenotipe

Abstract

The population of Indonesia in 2021 is about 273 million people. The increasing population is followed by an increase in food needs, including animal food. The area of land as a base of food production does not increase, even tends to decrease due to conversion, abrasion, and submerged seawater as a result of global warming and climate change, as well as the declining quality of natural resources. Animal protein consumption in Indonesia is very low and is expected to increase sharply as the income of the population continues to increase. Anticipating this, it is necessary to prepare a medium- and long-term farm development strategy in a sustainable manner by utilizing the availability of local resources. Exploring the genetic resources of livestock that are efficient in the use of feed, should also be able to utilize feed materials in the form of crop byproducts and agricultural industries. This policy should be supported by technological innovation and sustainable productivity and livestock production. Can save natural resources while reducing greenhouse gas emissions in order to realize the concept of green economy.

Abstract

This research was conducted in January 2019 until February 2019, located in North Rorowatu District, Bombana Regency. The purpose of this study was to determine body weight with body size (chest circumference, body length, shoulder height and hip height). Male and female goats in North Rarowatu District Bombana Regency. The material used in this study is adult male and female goats aged 12-24 years as many as 201 animals. The instrument used is a 50 kg scale with a capacity of 50 kg to measure life weight, a measuring tape to measure chest circumference, body length and a measuring stick to measure shoulder height, hip height. The results of this study indicate that the average body weight of male goats is 29.06 ± 5.85 kg and female goats are 23.75 ± 3.00 kg, chest circumference of male goats is 69.60 ± 5.83 cm and on goats females at 67.38 ± 7.92 cm, body length on male goats at 62.05 ± 6.46 cm and on female goats at 60.50 ± 8.46 cm shoulder height on male goats at 58.03 ± 4, 54 and in females 54.49 ± 7.65 cm, hip height in male goats was 59.44 ± 4.85 and in female goats 57.49 ± 7.75 cm. Based on the results of the study concluded there was a relationship between body measurements with body weight of goat nuts in North Rarowatu District. Chest circumference has the most close and very strong relationship to body weight as indicated by the correlation coefficient of 0.92 in males and 0.86 in females. Sequentially the results of the analysis of the correlation coefficient and determination are body weight - chest circumference 0.92 and 84.09% male, body weight - chest circumference 0.86-74.46% female, body weight - body length 0.68 and 46, 79% male, body weight - body length 0.70.48.89% female, body weight - shoulder height 0.60 and 36.36% male, body weight - shoulder height 0.64 and 40.98% female body weight - hip height 0.66 and 43.01% male, body weight - hip height 0.59 and 35.09% female.
