Abstract

Penelitian ini bertujuan melakukan isolasi dan identifikasi serotipe lokal Escherichia coli (E. coli) O157:H7 dan uji profil hemolisisnya pada media agar darah. Isolasi bakteri dilakukan dengan media eosin methylene blue agar (EMBA), dilanjutkan dengan identifikasi pada media selektif sorbitol MacConkey agar (SMAC) dan uji konfirmasi menggunakan uji aglutinasi lateks O157 serta uji antiserum H7 sebagai konfirmasi akhir dari E. coli O157:H7. Gambaran hemolisis diuji dengan menumbuhkan isolat pada media agar darah domba. Hasil penelitian menunjukkan bahwa 7 isolat (8,54%) dari 82 sampel feses ayam teridentifikasi E. coli O157:H7 dan memperlihatkan profil enterohemolisis seperti halnya isolat kontrol ATCC 43894. Dari hasil penelitian disimpulkan bahwa isolat lokal E. coli O157:H7 hasil isolasi dari feses ayam diketahui memiliki patogenitas yang tinggi terkait dengan dihasilkannya enterohemolisin yang merupakan marka/penanda kemampuan dari isolat untuk menghasilkan faktor virulensi Shiga like toxin.

Abstract

Reform of the National Criminal Law is absolutely necessary with political, practical and sociological considerations. The amend of Criminal Code also include adjustments of the punishment aims, which leads to restorative justice. Likewise the sanctions for criminal offense in adat law, the punishment is less than optimal if only imposed the freedom deprivation to the perpetrator's, it is necessary to make efforts in restoring the situation (balance).This type of research is normative research, by analyzing norms and legislation related to the aim of punishment in customary offenses. The results of the research that is the aim of customary sentencing is not specifically regulated in positive law, but customary criminal law have the power to apply, based: Article 18 B paragraph (2) of The 1945 State Constitution of the Republic of Indonesia, Emergency Law No. 1 of 1951 on Temporary Measure in Organizing the Unity of the powers and procedure of Civil Courts, Law No. 48 of 2009 on Judicial Power, Law No.39 of 1999 on Human Rights, and the Supreme Court Decisions, among others: (1) Decision No. 1644K / Pid / 1988 dated May 15, 1991; (2) Decision No. 984 K / Pid / 1996 dated January 30, 1996.In the positive law, there is a reflection of restorative justice as a concept that developed in modern criminal law in line with the purpose of sentencing the customary offenses. The aim of sentencing as ius constituendum is formulated in Article 55 paragraph (1) of the 2017 Criminal Code Concept which includes resolving conflicts that arise, restoring balance, and bringing sense of peace and secure in the community, which is in line with customary criminal sanctions because of the visible values of customary law; resolve conflicts, restore balance, and bring a sense of peace in society.

Abstract

A research has conducted, in order to identify the genetic markers of Bali cattle that can be used as an indicator of their qualitative superior. The genetic marker identified was the growth hormone gen. As many as 50 whole blood samples which were collected from 50 individual cattle which were certificated as good breed or had qualitative superior, were used in this study. The DNA fragments were identified by using Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) method with primers, GH5 (5’-CCC ACG GGC AAG AAT GAG GC-3’ and GH6 (5’TGA GGA ACT GCA GGG GCC CA-3), respectively. Furthermore, the PCR products were restricted with MspI endonucleic restriction enzyme, before electrophoresis in 2% agarose The result showed that the growth hormone gene of bali cattle with qualitative superior has polymorphism which characterized by the formation of two fragments i.e. 230 and 329 bp that categorized as: allele +/+ and allele -/-.

Abstract

Abstract. Spit roast pork or commonly called “be guling” is food from pigs of approximately 3-4-month-olds, varied between female or male pigs, filled with spices and vegetables such as cassava and grilled (rolled) to be cooked, marked by color transformation from skin color to brownish read and crispy skin. In Bali Island, Balinese and Landrace pigs are commonly used for cooking spit roast pork. The pig selection is determined by the market price and customer needs. A high market price and demand such as ceremonies lead people to select small-sized pigs; however, a low market price and demand (sold for consumption) lead people to select large-sized pigs. This study aimed to examine Balinese and Landrace pig body sizes used for spit roast pork. The study was performed by measuring the pig's length, height, chest circumference, and abdomen circumference. Data collected were then analyzed using the T-test to discover the mean difference. Meanwhile, the variation difference was tested using the F-test. The study demonstrated that Balinese pig’s body size was insignificantly (P>0,05) smaller than the Landrace pig for spit roast pork on 3-4-month-olds. Balinese pig’s body size had a significantly larger variation (P<0.05) (more varied) than the Landrace pig on 3-4-month-olds. Keywords: Body size; Balinese pig; Landrace pig; Spit roast pork

Abstract

Escherichia coli O157:H7 has been identified as one of the most devastating microorganism causing diseases in human. The infection by this bacteria causes a wide spectrum of clinical manifestations ranging from asymptomatic, diarrhea or bloody diarrhea, up to serious clinical conditions such as hemorrhagic colitis, and hemolytic uremic syndrome. Poultry is known as one of its reservoir besides cattle and pig. Moreover, the use of antibiotic in animal feed as growth promoters is common worldwide. Due to its potential threat to public health, sensitivity test against various antibiotics and studying the plasmid DNA profiles are necessary. The study was initiated by cultivation of isolates, followed by test of antimicrobial susceptibility against various antibiotics and then by analyzing the plasmid DNA profiles. Results of study showed that among of 7 E. coli O157:H7 local isolates originated from chicken feces, as many as 42.9%; 14.3%; 14.3%; and 14.3% were resistant to 2; 3; 4; and 5 various of antibiotics, respectively. Base on the type of antibiotic, 85.7%; 71.4%; and 42.9% showed resistance to Methicillin, Penicillin G, Doxycycline and Streptomycin. Plasmid DNA of each isolates showed characters variation too, ranging from 2 to 4 bands with molecular weight ranging from 19.457 bp; 15.137 bp, 5.749 bp, 4.118 bp, 3.500 bp, 2.957 bp and 2.024 bp. Results of study indicated that local isolates of E. coli O157:H7 isolated from chicken showed multiple antibiotic resistance against various antibiotics, as well as its plasmid DNA profiles.

Abstract

Lactic acid bacteria SR4 strain is known to have potential as a probiotic source according to the previous study. The isolate derived from the gastric’s juice of bali cattle has characteristics such as its resistance to growth at low pH, namely pH 2, 3, and 4, its tolerance to bile concentration of 0.6 mM, and its inhibition against Gram-positive and negative bacteria. On the other hand, a complete molecular analysis of the isolate has never been carried out. Based on these considerations, the molecular analysis of the LAB SR4 strain becomes interesting to present. The study was initiated by cultivating LAB SR4 isolate on an MRS broth medium. The isolate was further confirmed molecularly through analysis of the 16S rRNA gene which was initiated by isolating genomic DNA followed by PCR reaction using universal primers 27F and 1492R. The PCR product of the 16S rRNA gene was sequenced and further analyzed using the MEGA 5.2 software. The data were then analyzed descriptively and presented in the form of tables or figures. The results showed that the LAB strain SR4 was identified as a Pediococcus pentosaceus. This strain potentially to develops as a source of bacteriocin and it can be applied as a bio preservative, improvement of the meat taste, inhibition of inflammation, and also as antagonism of cancer.

Abstract

Examination of the blood profile is crucial because blood functioned as an important role in the circulatory system and can be used to evaluate the emergence of stress in animals. The purposes of this research are to examine the complete blood profile and examine the possibility of stress symptoms by measuring the ratio of neutrophils-lymphocytes as the indicator of stress in electrical stunned and non-stunned swine. There were 40 six-month life swine with a weight of 80-100kg. The swine grouped into stunning and non-stunning group. A total of 3 ml of blood sample occupied and placed to a contained anticoagulant Ethylene Diamine Tetraacetic Acid (EDTA) vacutainer. The total of erythrocytes, hemoglobin, PCV, MCV, MCH, leukocytes and differential leukocytes were calculated using routine hematological examination methods with autoanalyzer machines. Based on the results of the research, the complete blood profile of electrical stunning slaughtered swine was healthier and more reliable than non-stunning. The ratio of neutrophils-lymphocytes in electrical stunning slaughtered swine was lower than non-stunning. This condition indicated a lesser amount of stress on the use of electric stunning.

Abstract

The tourism industry has experienced a decline due to the Covid-19 pandemic. The Indonesia Central Statistics (BPS) noted that the number of foreign tourist arrivals to Indonesia between January and November 2020 was only 3.89 million. In line with the implementation of the health protocol, the Ministry of Tourism and Creative Economy in 2020 issued a guide book and certification program for the implementation of Cleanliness, Healthy, Safety and Environment (CHSE) in the tourism sector. On September 25, 2020 Tanah Lot already pocketed the CHSE certificate. This study was designed to assess the relationship between the implementation of the CHSE program on satisfaction and revisit intention of tourists visiting the Tanah Lot Tourist Attraction, Tabanan. This study uses quantitative methods. The sample in this study was 30-100 samples using accidental sampling technique. The data collection technique in this study was done by distributing online and offline to visitors visiting Tanah Lot. Data analysis in this study used two analytical techniques, namely Structural Equation Modeling - Partial Least Square (SEM-PLS) and descriptive qualitative analysis. The results of this study indicate that CHSE, both directly and through mediation, has a positive and significant effect on the satisfaction and intention to revisit visitors who travel to Tanah Lot. Satisfaction has a positive and significant effect on the intention to revisit tourists who travel to Tanah Lot.

Abstract

This research aims to combine the advantages of LMS technology with the potential of two-dimensional animated learning media in an educational context. This research explores the basic concept of a Learning Management System (LMS) and its advantages in providing a structured platform for delivering learning material, student-teacher interaction, and evaluating learning progress. LMS offers broad accessibility, student progress tracking, and the ability to facilitate collaborative learning. The research focus shifted to the potential of two-dimensional animation-based classification learning media. Two-dimensional animation offers interesting and engaging visualizations and can help students understand complex concepts better. In this context, this research integrates the visual power of animation with the learning structure provided by the LMS. This integration is expected to increase student engagement, facilitate a deeper understanding of concepts, and increase information retention. In addition, this combination is also expected to improve the overall quality of teaching by enabling the use of more innovative and interactive teaching methods. This research aims to contribute to our understanding of how learning technology can be integrated effectively to improve student's learning experiences. Thus, it is hoped that the results of this research can provide valuable insight into the development of better learning practices.

Abstract

Gerhana merupakan proses tertutupnya bulan atau matahari oleh benda langit yang melintas di depannya. Perubahan warna bulan dan matahari saat terjadinya gerhana ini merupakan ilusi warna. Ilusi warna tidak berbicara tentang kebenaran, hal ini dipengaruhi oleh beberapa faktor seperti intensitas cahaya, intensitas warna, dan warna benda di sekitarnya. Dari hal tersebut, pencipta terinspirasi menciptakan karya busana dengan konsep ilusi warna yang terinspirasi dari gerhana dengan judul Kapangan. Penciptaan ini bertujuan untuk menciptakan karya seni yang memadukan pengetahuan ilmiah dan fashion yang bergaya edgy dramatic dan street dramatic. Penciptaan ini menggunakan metode Hawkins, yaitu eksplorasi, improvisasi, dan pembentukan. Pada tahap eksplorasi proses penciptaan dimulai dengan membuat mindmap, menentukan kata kunci, dan membuat moodboard. Tahap improvisasi dibagi menjadi dua yaitu improvisasi medium dan improvisasi visual. Improvisasi media merupakan tahap menentukan alat, material, dan teknik. Sedangkan improvisasi visual merupakan tahap eksperimen dan mengembangkan konsep menjadi desain dan gambar kerja. Tahap pembentukan, proses terumit dalam menciptakan karya, pencipta melakukan pewujudan karya yang dilanjutkan dengan konsep hasil karya berupa photoshoot, publikasi, dan fashion show. Kapangan terdiri atas koleksi busana Candra Kapangan sebagai pemvisualisasian gerhana bulan dan Surya Kapangan sebagai pemvisualisasian gerhana matahari. Koleksi Candra Kapangan terdiri atas tiga look busana, yaitu Gerhana Bulan Total, Gerhana Bulan Sebagian, dan Gerhana Bulan Penumbra. Koleksi Surya Kapangan terdiri atas empat look busana, yaitu Gerhana Matahari Total, Gerhana Matahari Sebagian, Gerhana Matahari Cincin, dan Gerhana Matahari Hibrida. Penciptaan ini merupakan penggabungan antara pengetahuan ilmiah dan fashion dengan mengadopsi teori warna Bezolt Effect dan Chubb Illusion. Candra Kapangan terdiri dari warna hitam, abu tua, abu muda, dan abu kebiruan. Sedangkan Surya Kapangan terdiri atas warna kuning, biru, dan hitam.Ilusi warna direpresentasikan dengan menggunakan bentuk lingkaran yang menghasilkan perubahan warna berdasarkan intensitasnya. Kata Kunci: Gerhana; Bezolt Effect; Chubb Illusion; Fashion KontemporerABSTRACTAn eclipse is the process by which the moon or sun is covered by a celestial body that passes in front of it. The change in the color of the moon and sun at the time of this disaster is an illusion of color. Color illusion does not speak the truth, it is influenced by several factors such as light intensity, color intensity, and the color of the objects around it. From this, the creators were inspired to create fashion works with the concept of color illusion inspired by the title Kapangan. This creation aims to create works of art that combine scientific knowledge and fashion in an edgy dramatic and street dramatic style. This creation uses the Hawkins method, namely exploration, improvisation, and shaping. In the exploration stage, the creation process starts with creating a mindmap, determining keywords, and creating a moodboard. The stages of improvisation are divided into two, namely media improvisation and visual improvisation. Improvised media is the determining stage of tools, materials, and techniques. Meanwhile, visual improvisation is the experimental stage and develops concepts into designs and working drawings. The formation stage, the most complicated process in creating a work, the creator carries out the embodiment of the work followed by the conceptualization of the work in the form of photoshoots, publications and fashion shows. Kapangan consists of the Candra Kapangan fashion collection as a visualization of the cold weather of the moon and Surya Kapangan as a visualization of the sun's weather. The Candra Kapangan collection consists of three fashion looks, namely a total lunar eclipse, a partial lunar eclipse, and a penumbral lunar eclipse. The Surya Kapangan collection consists of four fashion looks, namely Total Solar Eclipse, Partial Solar Eclipse, Ring Solar Eclipse, and Hybrid Solar Eclipse. This creation is a combination of scientific knowledge and the art of Fashion by adopting the Bezolt Effect and Chubb Illusion color theories. Candra Kapangan consists of black, dark gray, light gray, and bluish gray. While Surya Kapangan consists of yellow, blue and black. The illusion of color is represented by using a circle shape which produces a color change based on its intensity.Keywords: Eclipse; Bezolt Effect; Chubb Illusion; Contemporary Fashion
