Abstract

Research has been conducted to determine the effect of breed (bali cattle and wagyu) andperiod time at cold storage temperatures (4°C) of beef protein and amino acids charateristics. Thisstudy used the completely randomized factorial design. First factors were type of beef (bali cattle and wagyu) and the second factors were period storage at cold temperature 4°C (day of 0, 3, 7, and14). The bands of proteins were analyzed using SDS-PAGE method while the amino acids wereanalized using HPLC method. Data were analyzed by analyse of variance. The result showed thatthe storage time affected to the profil of protein bands of bali and wagyu beef after 7 days and 14days of storage, while the concentration of amino acids has been decreased. This means that theactivity of proteolytic enzymes in meat stored at 7 days was degrading proteins and affected thequality of meat. The conclusion indicated that bali and wagyu beef showed differencescharacteristic of protein (the thickness and number of bands) as well as significant differences ofthe amino acid Phenylalanine in wagyu beef compared with bali beef. The period time at coldstorage temperature 4°C showed real effect to the decline of the amino acid histidine, phenylalanineand isoleucine of bali and wagyu beef.

Abstract

Lactic acid bacteria can excrete antimicrobial compounds like bacteriocins. The study aimed to find out the characteristic of physic-chemical of bacteriocin producing Enterococcus durans isolate 18A isolated from colon’s bali cattle. The study initiated by Gram staining and catalase test, followed by isolation and purification of bacteriocin. The result of the research showed that bacteriocins of Enterococcus durans isolate 18A as a protein with it’s concentrationis 0,272?g/ml and it does not contain carbohydrate. On the other hand, the bacteriocins was not showed a band while tested on sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). The result showed, the bacteriocin producing Enterococcus durans isolate 18A has antimicrobial activity to Bacillus cereus as 23,88%.

Abstract

Lactic acid bacteria are the type of bacteria that has benefits in food and health industries as a biopreservative, fermentative, or probiotics. Bali cattle are known as potential host for specific lactic acid bacteria. The aim of this study is to identify phenotypically lactic acid bacteria 9A isolated from bali cattle’s colon, that producing a substance which known has potency as antimicrobial. In this study, phenotypic identification included conventional method and API 50 CHL. The result of this study showed that lactic acid bacteria isolate 9A was Streptococcus sp., whereas identification by kit API 50 CHL showed isolate 9A as Lactobacillus fermentum with 83% identity. The difference between the results of conventional method and kit API 50 CHL, may indicate the difference in sensitivity and specificity of the two methods, hence it needs further confirmation.

Abstract

Lactic acid bacteria (LAB) is a group of Gram-positive bacteria, in shape of coccus or bacilli and catalase-negative and also have the ability to produce lactic acid. LAB is also a microorganism which is not toxic and has the ability to produce antimicrobial compounds, known as bacteriocins. This research aimed to find out the physicochemical characteristic and the antimicrobial activities of bacteriocin isolated from isolates 15B originated from the bali cattle’colon. This study began by reculturing of isolates 15B and followed by cultivating the isolates, isolating, producing, and precipitating of bacteriocins. The physical and chemical properties of the bacteriocins were also tested before, antimicrobial activity test was conducted. The result of the cultivation showed that isolates 15B is a lactic acid bacteria characterized by the result of Gram staining as a Gram positive, catalase negative and grown on anaerobic condition. The result of chemical test, showed that bacteriocins producing isolates 15B were positive on Ninhydrin test, negative on Molisch test, and positive on Lowry test with 0,11 µg/ml concentration. Physical properties test using SDS PAGE showed negative result and antimicrobial activity test showed that the bacteriocin from isolates 15B has 35,82% effectiveness of inhibition.

Abstract

Lactic acid bacteria (LAB) is a microorganism that is not toxic and able to produce antimicrobial compounds, namely bacteriocins. This study aims to determine the physicochemical characterization of bacteriocins origin originated from lactic acid bacteria isolates 17B isolated from colon Bali cattle. The research was started by re-cultivation of isolates 17B and continued with production and precipitation of bacteriocins. Further tests were chemical and physical test that, followed by antimicrobial activity test of bacteriocin isolates 17B against Bacillus cereus FTCC 005. Data From this study are presented in tables or figure and analyzed descriptively. Results of study showed that LAB isolates 17B is a Gram-positive bacteria and catalase negative. The test also showed the bacteriocins produced isolates 17B as a protein compound indicated by the ninhydrin test positive, Molisch test negative and Lowry test positive with protein concentration was 0,11 µg/ml. Physical characterization tested using SDS PAGE showed that the test is negative. Antimicrobial activity test of bacteriocin isolates 17B showed that bacteriocin 17B has effectiveness against Bacillus cereus FTCC 005 as 18,92%.

Abstract

Streptococcosis is a disease caused by Streptococcus sp., with clinical manifestations such as polyarthritis, septicemia, and meningitis. ?-hemolytic Streptococcus strains are known as one of the important pathogenic bacteria that cause serious illness. The bacteria enter their host through wounds, abrasions, food, or the immune system's decrease. This study aimed to determine the presence of ?-hemolytic Streptococcus spp in pigs raised on the people's farms in Punggul and Bongkasa Villages Abiansemal, Badung, Bali. As many as 47 isolates isolated from nasal mucosa swabs and tonsils of pigs were cultured on blood agar media, continued by planted them on Brain Heart Infussion (BHI) agar. Gram staining, catalase, and oxidaase tests were performed as biochemical tests to complete the identification of isolates. Finally, the bacitracin test was conducted on several isolates characterized with ?- hemolytic Streptococcus to differentiate Streptococcus pyogenes strains from others. The results show, 8 out of 47 (17%) samples tested are presumptive ?-hemolytic Streptococcus. Furthermore, the bacitracin test clarifies 3 out of 8 (37.5%) ?-hemolytic isolates are S. pyogenes, and the remaining 5 out of 8 (62.5%) are non-pyogenes ?-hemolytic Streptococcus.

Abstract

Unggas diketahui merupakan salah satu reservoir penting dari agen bakterial zoonosis Escherichia coli O157:H7. Disisi lain, fenomena pemakaian obat obatan pada unggas terutama antibiotika melalui makanan, minuman maupun secara parenteral mengalami peningkatan dari waktu kewaktu. Antibiotika yang digunakan secara intensif ini cenderung tidak sesuai dengan dosis maupun waktu pemakaian sehingga berkontribusi terhadap munculnya peningkatan resistensi termasuk E. coli O157:H7. Penelitian untuk mengetahui  pola kepekaan isolat E. coli O157:H7  ini diawali dengan identifikasi E. coli dengan  penumbuhan  bakteri  asal feses ayam pada media eosin methylene blue agar (EMBA), dilanjutkan dengan uji indol, methyl red, voges-proskauer dan citrat (IMVIC). Identifikasi O157 dilakukan dengan penumbuhan isolat E. coli pada  media selektif  sorbitol MacConkey agar (SMAC),  dilanjutkan dengan uji konfirmasi menggunakan lateks O157. Uji pola kepekaan dilakukan mengacu pada metode Kirby Bauer  seperti yang direkomendasikan oleh national committee for clinical laboratory standard (NCCLS). Hasil penelitian dari 7 isolat  E. coli O157:H7 hasil isolasi 82 sampel feses ayam serta  satu isolat  kontrol ATCC 43894 menunjukan  85,7%  isolat  bersifat resisten terhadap antibiotika metisilin, 71,4% resisten terhadap antibiotika penisilin G, serta 42,9% resisten terhadap antibiotika doksisiklin hidroklorida dan streptomisin. Hasil kajian juga menemukan 42,9% isolat bersifat resisten  terhadap 2 jenis antibiotika,  14,3% resisten  terhadap 3 jenis antibiotika,  14,3%  resisten  terhadap 4 jenis antibiotika  dan 14,3% resisten terhadap 5 jenis antibiotika.  Berdasarkan hasil penelitian ini maka dapat disimpulkan telah terjadinya peningkatan pola resistensi dan bersifat multi-drug resistance dari agen E. coli O157:H7 sehingga perlu dipertimbangkan jenis-jenis antibiotika tertentu yang masih sensitif didalam penanganan infeksi oleh agen ini.

Abstract

Lactic Acid Bacteria (LAB) is a bacterium that has an ability to inhibit a pathogenic bacteria growth, including isolates 18A. The aim of this study is to determine the species of LAB isolates 18A by conventional tests and API 50 CH test. This study was a descriptive observational study, with lactic acid bacteria isolates 18A were obtained from bali cattle’s colon as an object. LAB isolates 18A were re-cultured on MRS media, catalase test, and Gram staining. Identification test were performed by CO2 production test, growth in the presence of 15% NaCl, growth at 10°C, and growth at pH 9.6. Confirmation test were performed using API 50 CH (Standard Analytical Profile Index). The results indicated that LAB isolate 18A were able to grow well on MRS broth media, catalase-negative, Gram-positive, and spherical-chain in shape. Identification test revealed that the bacteria is homofermentative, were able to grow at 10°C, and do not grow at pH 9.6. Fermentation test using API 50 CH showed that LAB isolates 18A were able to degrade 15 sugar components, consist of no: 5, 10, 11, 12, 13, 22, 24, 25, 26, 27, 28, 29, 30 , 31, and 39. Based on conventional identification results, it can be concluded that BAL isolates 18A is a Lactococcus. In addition, the result of API 50 CHL test showed that BAL isolates 18A is a Lactococcus lactis ssp lactis 1.

Abstract

Coliform are commonly found in livestock stool, including cattles and Escherichia coli (E. coli) is one of the most popular of Coliform groups. Various strains of E. coli are pathogenic which oneof them is serotype O157:H7 which known producing Shiga like toxin. The study initiated by collecting 58 samples of fresh cattle stool which were taken from seven villages in the district of Petang, followed by isolating and identificating of strain. The results of study presented indescriptive and the relationship between each vaiable was analyzed by Spearman 's rho and McNemar test. Among 58 samples, all of them were positive Coliform (100 %), and 29 samples positive E. coli (50 %). Among E. coli positive, that were found 7 samples were positive E. coli O157 ( 12.07% ) and 5 samples positive E. coli O157:H7 (8.62 %). The total Coliform and the number of E. coliwere highly significant difference, while the number of E. coli O157 and E. coli O157:H7 were not significant difference. Coliform bacteria in the stool of cattle could not be used as a predictable factor for the presence of E. coli, E. coli O157 and E. coli O157:H7, but the presence of E. coli would be enable to predict the presence of E. coli O157 as well as E. coli O157:H7. Futhermore, the study also showed the presence of E. coli O157 was as a good predictor to the presence of zoonotic agents E. coli O157:H7.

Abstract

Intervensi kesehatan ternak merupakan upaya untuk memenuhi salah satu panca usaha peternakan yaitu terciptanya kesehatan ternak. Tujuan kegiatan ini adalah meningkatkan kesehatan dan pertumbuhan ternak melalui intervensi vitamin, dan pemberian obat-obatan pada ternak di wilayah Desa Getasan. Metode yang digunakan adalah pendekatan pada kelompok peternak simantri, gapoktan, atau individu yang memiliki ternak. Ternak dilakukan pemeriksaan klinis selanjutnya diberikan intervensi sesuai dengan kondisi terknak yang bersangkutan. Hasil pelayanan kesehatan ternak menunjukkan bahwa jenis ternak yang telah diberi intervensi yaitu sapi sebanyak 108 ekor dengan rincian, sapi jantan 21 ekor, sapi betina 87 ekor. Ternak babi sebanyak 90 ekor dengan rincian, jantan 40 ekor dan babi betina 50 ekor. Berdasarkan jenis layanan, pemberian vaksin Septicemia epizootica (SE) pada sapi sebanyak 77 ekor, vitamin B kompleks sebanyak 198 ekor, obat ektoparasit sebanyak 198 ekor, dan antibiotik 14 ekor. Berdasarkan kelompok ternak telah berhasil diberi pelayanan kepada kelompok ternak Suka Nadi sebanyak 10 ekor, Tani Mas sebanyak 34 ekor, Gana Sari sebanyak 42 ekor, Simantri 086 sebanyak 6 ekor, Simantri 521 sebanyak 6 ekor, Br. Buangga 5 ekor, Br. Tengah 4 ekor, dan Br. Ubud 1 ekor. Dapat disimpulkan bahwa ternak sapi dan babi di desa Getasan dalam kondisi sehat. Intervensi dengan vitamin dan obat ektoparasit sangat dominan kemudian disusul dengan pemberian vaksin SE dan antibiotik.
