Abstract

Pseudomonas aeruginosa is a gram negative bacterium that is pathogens opportunistic and the cause of nosocomial infections. P. aeruginosa is resistant to antibiotics that are often given. The aim of this study is to prove the presence of antibacterial activity of isolated glutathione from S. cerevisiae on P. aeruginosa infection. The design of this study is a completely randomized design. The variation concentration used was 100%, 75% and 50%. The parameters observed were inhibition zone diameter, turbidity level and the presence or absence of growth in MHA media. The results showed that the diameter of the largest inhibition zone was found at a concentration of 100% which was 7.88 mm and included a moderate inhibition criteria. The clearest solution after 24 hours incubation is a solution with an isolated glutathione concentration of S. cerevisiae 100%. While when inoculated into MHA media, all concentrations still produce P. aeruginosa growth. It means that the isolated glutathione from S. cerevisiae is bacteriostatic. Based on the results of these studies, it can be concluded that the isolation of glutathione from S. cerevisiae has antibacterial activity against P. aeruginosa so that can be used as an alternative treatment for P. aeruginosa infection. Pseudomonas aeruginosa merupakan bakteri gram negatif yang bersifat patogen oportunistik dan penyebab infeksi nosokomial. P. aeruginosa resisten terhadap antibiotik yang sering diberikan. penelitian ini bertujuan untuk membuktikan adanya aktivitas antibakteri dari glutathion hasil isolasi dari S. cerevisiae terhadap infeksi P. aeruginosa. Rancangan penelitian ini adalah rancangan acak lengkap. Variasi konsentrasi yang digunakan adalah 100%, 75% dan 50%. Parameter yang diamati adalah diameter zona hambat, tingkat kekeruhan dan ada tidaknya pertumbuhan pada media MHA. Hasil penelitian menunjukan bahwa diameter zona hambat paling luas terdapat pada konsentrasi 100% yaitu 7,88 mm dan termasuk kriteria penghambatan sedang. Larutan yang jernih setelah inkubasi 24 jam adalah larutan dengan konsentrasi glutathion hasil isolasi dari S. cerevisiae 100%. Sedangkan ketika diinokulasikan ke media MHA, semua konsentrasi masih menghasilkan pertumbuhan P. aeruginosa. Hal tersebut berarti glutathion hasil isolasi dari S. cerevisiae bersifat bakteriostatik. Berdasarkan hasil penelitian tersebut, dapat disimpulkan bahwa glutathion hasil isolasi dari S. cerevisiae memiliki aktivitas antibakteri terhadap P. aeruginosa. Sehingga, glutathion hasil isolasi dari S.cerevisiae dapat digunakan sebagai alternatif pengobatan terhadap infeksi P. aeruginosa.

Abstract

Kelurahan Cepoko is one of 16 urban villages in Gunungpati sub-district. The source of income of Cepoko village residents is in the field of livestock, as much as 77 residents of Cepoko urban village who rely their life in the field of farms, both chicken farms, dairy cows and meat cows. Dairy cattle breeding activities do not provide sufficient income, because the selling price of milk purchased by cooperatives is too cheap, so it can not cover operational costs. Therefore, one of the citizens of SME RW 09 kelurahan Cepoko that is Ibu Ismiyati took the initiative to make processed foods made from milk. Mrs. Ismiyati as a pioneer of milk sticks production business that is personal with employ 5% of women PKK group. Dairy products that have been done just limited to milk stick. Though dairy products have the potential to be developed in order to increase farmers’ income. One of the obstacles to the development of dairy products is the lack of mastery of information on production and marketing techniques by mothers, the limited knowledge and skills of mothers in the processing of milk into products worth selling, causing dairy products are less varied and not devel- oped. The methods undertaken to improve knowledge on milk-based food production techniques are socialization, training and monitoring. PKK ladies are very enthusiastic to follow the milk process- ing into a variety of food products that are proclaimed with the presence of mothers despite the rain, they still spend time attending training activities. PKK ladies become aware of the diversification of products based , and skilled in processing milk into various foods as well as skilled in the produc- tion manejemn such as kenmas and marketing processed products kamanan Participants want more follow-up is more real in terms of assistance and continuous monitoring and marketing of dairy- based food products broadly and poured in MoU between FMIPA and Gunungpati District

Abstract

Results of the observation shows that students are still passive during the learning process in which influence their learning achievement. A learning model of Student Facilitator and Explaining (SFaE) is effective to make the students explain the materials to their friends also to train students in expressing their ideas bravely. This study combines SFaE with Roundhouse diagram that makes students easier to understand and explain the materials of learning. The research was conducted for the tenth grade students of SMA N 1 Jekulo. A purposive sampling was used as the sampling technique, and two classes were used as the sample. The T-test shows that the t-value is more than t-table for 2.71 > 1.67 with which means H0 is rejected. This condition means that the average results of the experimental class is higher than the control class. The results of analysis on students’activity in the experimantal class is around 91.76% students are in the category of active until very active, and 8.33% are in the category of medium. In the control class, around 80.56% students are in the category of active until very active, and 19.44% are in the category of medium. In conclusion, the use of learning model Student Facilitator and Explaining (SFaE) with a Roundhouse diagram is effective to improve students’activity and achievement in the learning material of Virus.

Abstract

PosPAUD is a form of early childhood education services (PAUD) whose implementation is integrated with the services of Bina Keluarga Balita (BKB) and Posyandu. PosPAUD "Melati" is one of the PAUD Posts in Gunungpati Subdistrict. Based on PosPAUD data "Melati" Getas Cepoko village, the number of children active in PosPAUD aged 0-6 years in 2017 reached 25 children, while in 2018 active PAUD students were 18 people, there was a decrease in the number of community participation in PAUD activities. Likewise in the Posyandu activities which are held routinely on Saturdays on the first week of each month, the longer the number of people who come there is less The main problems faced by this pospaud include the limited number of cadres, the low knowledge and skills of cadres about nutrition and health and nutritional supplementary feeding (PMT) based on local food ingredients, the limited facilities available, the lack of knowledge about administrative order and active-creative learning. Community service methods are applied through pospaud cadre training, and assistance in the implementation of pospaud. Pospaud cadre empowerment activities carried out for 4 months, contributed 1). Increased cadre knowledge in managing PosPAUD; 2) Posyandu cadre skills in providing counseling and counseling about nutrition and health to children participating in PosPAUD and mothers of children under five; 3) Increased knowledge of PosPAUD cadres about nutritional supplementary feeding (PMT) based on local food ingredients; 4) PosPAUD cadre skills in making PMT (Supplementary Feeding) increased, and cadres could innovate types of PMT based on local food ingredients; 5) PosPAUD room is more comfortable and attractive; 6) Establishment of Family Medicine Park (TOGA). The results of the posyandu cadre empowerment activities showed an average initial knowledge of posyandu cadres on various aspects of being cured by 40.45%; the average posyandu cadre final knowledge on various aspects of being educated and trained is 95.25%; thus increasing the final evaluation results by 54.80% (classified in the medium category). Abstract

The diversity of intestinal bacteria in geese correlates with environmental conditions, rearing methods, and consumed feeds. The intestinal bacteria composition is useful for the absorption of nutrition, improving the metabolism, and may be related to the immune system. This study was conducted to examine the intestinal bacteria composition and the diversity of maintained goose in aviaries and barns. This research was an observational exploratory. Five geese were taken purposively from local breeders in Gunungpati District, Semarang City. A total of 5 g of intestinal contents from each sample was used for microbial genome isolation. Then, the genome was amplified to collect 16S rRNA gene region V3-V4. The amplicons were then sequenced using the next generation sequencing (NGS) method (Illumina high-throughput sequencing; paired-end reads) and analyzed using QIIME2 to identify bacterial species. In addition, GC-MS was performed to identify and measure fatty acid contents in the intestinal. The results showed that both rearing and caged goose contained nine phyla of intestinal bacteria. The number of intestinal bacteria of barn geese (SU) reached 32,748 Operational Taxonomy Units (OTU); higher than aviary geese (SK), which was 11,646 OTU. The intestinal bacteria community in barn geese was approved by Phylum TM7 (Saccharibacteria candidate) (53.18%), followed by Firmicutes (32.51%) and Bacteriodetes (5.42%). Whereas on SK Firmicutes was compiled 49.3 4% of total OTU, TM7 (S. candidate) up to 21.17%, and Actinobacteria up to 15.99 %. The abundance of TM7 may contribute to high 9,12-octadecadienoic acid production, while Firmicutes was related to the high production of oleic acid. Based on these data, the reared geese had a more abundant diversity of bacteria than the caged one.

Abstract

Intestinal bacterial species play an essential role in the duck body metabolism and productivity. They are affected by various factors, such as heavy metal contamination. This research aims to study metal emission levels and their correlation with the composition of intestinal bacteria. A total of twenty-five duck samples (Anas platyrhynchos L.) were obtained from Semarang, Temanggung, Magelang, Pati, and Salatiga. Five g of intestinal substances were employed in the isolation 16S rRNA gene V3-V4 region. Data sequence in fastqc format was then analyzed using QIIME. Also, the heavy metals in water, feed, and meat samples were analyzed using inductively coupled plasm-optical emission spectroscopy (ICP-OES). The highest heavy metal contaminant was Hg, with a total concentration reaching 5.55±1.79 ppm of input and 4.42±0.80 ppm in meat. The bacterial composition comprises the four highest phyla, sequentially including Firmicutes, Actinobacteria, Proteobacteria, and Bacteriodetes. The relationship between heavy metals and intestinal bacteria shows a significant value with a coefficient of >0.90. Therefore, there was a correlation between heavy metals and intestinal bacteria composition, suggesting that bacteria are likely to protect ducks from the adverse effects of heavy metals in water and feed.

Abstract

Penelitian ini bertujuan untuk mengetahui  aktivitas enzim antioksidan superoksida dismutase (SOD) dan katalase dalam plasma tikus wistar yang diiunduksi Pb asetat dan suplementasi madu kelengkeng. Penelitian eksperimental laboratorik, dengan rancangan The Post Test-Only Group Designdilakukan pada bulan Maret 2015 menggunakan hewan coba tikus putih wistar umur 2-3 bulan dengan berat badan 150-250. Sebanyak 20 ekor tikus diadaptasi dan diberi pakan standar selama 2 minggu. Selanjutnya dibagi secara acak menjadi 4 kelompok, yaitu : kelompok kontrol hanya diberi pakan standar,  K(-): kelompok tikus yang diberi perlakuan Pb asetat 10 mg/kgBB, P1 : kelompok tikus yang disuplementasi madu kelengkeng 0,9 ml/200 grBB dan Pb asetat 10 mg/kgBB, P2: kelompok tikus yangdisuplementasi madu kelengkeng 1,8 ml/200 grBB dan Pb asetat 10 mg/kgBB dilakukan selama 14 hari. Setelah akhir perlakuan tikus diambil darahnya melalui plexus retroorbitalis untuk diukur kadar SOD dan CAT.  Hasil pemeriksaan dianalisis dengan uji Chi Square dengan batas signifikansi = 0,05.Hasil penelitian menunjukkan bahwa pada tikus kelompok kontrol kadar SOD 141,50μ/ml dan CAT 0,8μ/ml; K(-) kadar SOD 77,71μ/ml dan CAT 0,12μ/ml; P1 kadar SOD 289,82 μ/ml dan CAT 1,21. Sedangkan pada tikus P2,kadar SOD 391,59 dan kadar CAT 3,62. Secara statistik terdapat perbedaan signifikan antar kelompok. Kata kunci: antioksidan, katalase, madu kelengkeng, superoksid dismutase

Abstract

Pangan fungsional adalah pangan yang karena kandungan komponen aktifnya dapatmemberikan manfaat bagi kesehatan, di luar manfaat yang diberikan oleh zat-zat gizi yangterkandung di dalamnya. Tumbuh-tumbuhan merupakan salah satu bahan alami tertua yangdiketahui sebagai sumber pangan fungsional. Selain tumbuh-tumbuhan, beberapa bakteri jugamerupakan sumber pangan fungsional. Kajian tentang pangan fungsional dalam perspektifkesehatan telah banyak dilaporkan. Hasil penelitian tentang acemannan dan glucomanan lidahbuaya dan bakteri lactobacillus sebagai pangan fungsional akan dikaji dalam tulisan ini.Senyawa bioaktif acemannan dan glucomanan pada tanaman lidah buaya (Aloe vera) berperanpada keseimbangan sistem imun tubuh dan kadar lemak darah. Sedangkan bakteri lactobacillusberperan dalam menjaga keseimbangan mikroflora usus, membantu menurunkan kadar lipiddarah. Semoga kajian tentang peran senyawa bioaktif acemannan dan glucomannan serta bakterilactobacillus dapat menambah khasanah peran pangan fungsional dalam meningkatkan derajatkesehatan.

Abstract

BACKGROUND: One of the hormones that plays a role in glucose metabolism of Type 2 Diabetes Mellitus (T2DM) is melatonin. Its genetic variation is believed to play a significant role in the pathophysiology of obese and non-obese T2DM. The role of MTNR1B (melatonin receptor coding gene) and SLC2A1 (Glucose transporter 1/GLUT 1 transporter coding gene) on the risk of obese and non-obese T2DM patients is controversial. This study aims to analyze the association between the rs10830963 MTNR1B and rs841853 SLC2A1 polymorphism to the risk of Javanese obese T2DM.METHODS: This was a cross-sectional study that involved 107 Javanese T2DM patients from primary heath care in Semarang. Furthermore, obese T2DM was defined by a body mass index (BMI) more than 25 kg/m2. The genetic variations examined were rs10830963 MTNR1B and rs841853 SLC2A1 polymorphism by PCR-RFLP methods. Blood biochemistry parameters were also examined. Allele and genotype frequencies of rs10830963 MTNR1B and rs841853 SLC2A1 polymorphisms were analyzed using the x2 test with p≤0.05 and 95% CI.RESULTS: There was a significant association between rs10830963 MTNR1B polymorphisms in obese and nonobese T2DM (p=0.044) and the CG genotype increased the risk of obese T2DM. Furthermore, the allele and genotype frequency of rs841853 SLC2A1 polymorphism in both group had no significant difference, with a p=0.756 and p=0.802, respectively. There was also no significant difference in the biochemical parameters' in both groups of the two genetic variants studied.CONCLUSION: The rs10830963 MTNR1B polymorphism is associated with the risk of obesity in Javanese T2DM patients but not for the rs841853 SLC2A1 polymorphism.KEYWORDS: polymorphism, MTNR1B, SLC2A1, obese, diabetes mellitus

Abstract

Diabetes mellitus (DM) is a non-communicable disease and still a health problem in the world. Melatonin is responsible to metabolize carbohydrate and regulate blood sugar levels. Several studies proved Melatonin is useful in the diabetes therapy and its complication by performing antioxidant activity through various signaling pathways. The role of the melatonin hormone in the pathogenesis of complications of diabetes mellitus is still a lot of controversy. Animal studies are still incomplete to explain the mechanism of action of melatonin. Therefore, this review aims to discuss Melatonin role administration in DM and its complications. This review was started by searching articles using electronic databases such as Google Scholar, Science Direct, SpringerLink and Pubmed between 2009-2019. Melatonin increased respiration within mitochondria; acted as antioxidant that fight free radical production; and reduced oxidative stress. It diminished proinflammatory cytokines in diabetes and prevents various DM complications on eyes, cardiac and male reproductive organs through antioxidant effects. It inhibited proinflammatory within several signaling Pathways, included PI3K/Akt-Nrf2,cGMP-PKG, Nrf-2-HO-1 and MAPK, Syk, p38MAPK pathways. There are various melatonin roles in diabetes and DM complications as a promising new breakthrough of DM treatment. It is necessary to explore Melatonin role in other DM complications. Most of this research is on experimental animals, thus clinical trial studies should be carried out in humans to make melatonin a promising therapeutic or prevention for diabetes and its complications in the future
