Abstract

Optimization conditions for thermostable xylanase production of Bacillus licheniformis TS10 by using substrate oil palm empty fruit bunches  has been conducted from November 2015 to January 2016 in the Laboratory of Genetics and Biotechnology, Department of Biology, Faculty of Mathematics and Natural Sciences, University of Sriwijaya, Indralaya. The aims of this study was to determine the potential of oil palm empty fruit bunches ( EFB ) as a substrate for the production of thermostable xylanase by Bacillus licheniformis TS10 and the optimum conditions of temperature, pH and substrate concentration in the fermentation process. The research method is to make the growth curve and the curve of Bacillus licheniformis TS10 enzyme production on EFB substrate. The number of bacterial cells was determined by using a UV-VIS spectrophotometer and total plate count (TPC) through a standard curve. Optimization performed at various pH (5, 6, 7, 8, 9), temperature (50 °C, 60 °C, 70 °C, 80 °C) and substrate (1%, 2%, 3%, 4%) , Test activity at each pH, temperature and substrate using methods DNS by measuring enzyme activity based on the reducing sugar released by the substrate by using dinitrosalicylic acid (DNS). Based on the results of this research showed that oil palm empty fruit bunches (EFB ) has potential as a substrate for the production of thermostable xylanase by Bacillus licheniformis TS10, thermostable xylanase production of Bacillus licheniformis TS10 on the substrate EFB has an optimum condition at pH 6, 80 °C and the substrate concentration of 4%.

Abstract

The research " The Effect of Applicated Ash Sawdust on Swamp Land (Indralaya) For Vegetative Growth of Red Pepper (Capsicum annum L.) var. Cemeti ’’ has done, from  September to November 2017. It was conducted at green house and Pest Diseases Plant Laboratory, Plant Protection Department, Agriculture Faculty, University of Sriwijaya. This research was aimed to know the effect of applicated ash sawdust on vegetative growth of red pepper. The Completely Randomized Design with treatments are : 10, 20, and 30 g/ kg marshland and without ash sawdust (control), with 6 replicates each treatments. The result showed that applicated ash sawdust on measure 10 g/ kg soil significantly on number of growth 53.28 cm and number of branch 9.2, but not differ on dry weight of red pepper.Keywords :  Swamp land, ash sawdust, branch, red pepper

Abstract

Culex spp are mosquito vectors that have a very wide distribution capability and are carriers of pathogens that can interfere with human and animal health. The wide distribution makes Culex spp a dangerous threat. DNA extraction is one of the important steps in obtaining genetic information and genetic analysis. Good quality DNA is used for activities such as the use of molecular markers, genome library creation, and sequencing. This study aims to determine the quality, concentration and purity of Culex spp mosquito DNA in Kemelak Bindung Langit Village, OKU Regency. It is hoped that the sample can be used for further research analysis on Mitochondria D-Loop Sequences in Culex spp mosquitoes. Quantitative measurement of DNA in the form of concentration and purity of DNA using Nanodrop Thermo cycle while qualitative DNA using electrophoresis technique. The results of the isolation of the mosquito genome DNA, obtained clear DNA bands without any degradation (smear) and the concentration results for the four samples ranged from 10-100 ng/µL and the DNA purity was good, ranging from 1.8 to 2.00.

Abstract

Ornamental plants are important horticultural commodities with considerable economic value, providing income for both growers and traders. Among them, Sansevieria trifasciata (commonly known as snake plant) is favored for its sword-shaped, rigid leaves. This study aimed to assess the incidence and intensity of disease in snake plants. The research was conducted from January to February 2025 using purposive sampling. Field observations revealed that the plants were affected by Colletotrichum sansevieria, causing necrotic black lesions. Disease incidence and intensity varied across locations, with the highest incidence (100%) observed in plants owned by Suwaji, Salikun, and Asep, and the lowest (10%) in those owned by Tina Purba and Sarkun. Disease intensity was greatest at Suwaji’s site (83.3%) and lowest at Tina Purba’s and Sarkun’s (3.3%). These differences were influenced by environmental factors such as temperature and humidity. Integrated disease management through physical, chemical, and biological methods is suggested as a strategy to suppress pathogen growth.
