Abstract

The aim of the study was to isolate lactic acid bacteria (LAB) from palm oil sap and to investigate the anti microbial activity of LAB against pathogen bacteria, Staphylococcus aureus and Escherichia coli using disc diffusion method. Palm oil sap were enriched using spesific medium MRS (Man Rogosa Sharpe) broth overnight, serial dilution were conducted and the culture were spread on the surface of MRS agar and then incubated for 48 hours. The result showed the total colonies of LAB were 1,4 x 107 CFU/mL. 6 colonies were randomly selected namely EHN1, EHN2, EHN3, EHN4, EHN5 and EHN6. The selected colonies are classified as Gram positive and bacilli in morphology. Antimicrobial activity against pathogenic bacteria was investigated using disc diffusion method. Antimicrobial activity of isolates against E.coli and S.aureus were 12,2 mm; 9mm; 11mm; 11mm; 10,5mm; 12 mm dan 13,5mm; 11mm; 12mm; 11,5mm; 12mm; 12,5mm respectively. from these result it can be concluded that LAB isolated from oil palm sap has potential as probiotic.

Abstract

Lactic acid bacteria (LAB) could be isolated from various fermented food products. One potential source of LAB is traditional fermented food. The aim of the study was to isolate and investigate antimicrobial activity of LAB isolated from traditional Batak food, “naniura”. The LAB isolates were characterized by Gram staining, fermentation type and catalase test. The investigation of antimicrobial activity of LAB against pathogenic bacteria were conducted using disc diffusion method. The results showed that 6 isolates of BAL were successfully isolated namely BN1, BN2, BN3, BN4, BN5 and BN6 had characteristics of Gram positive, rod shaped and catalase negative. All selected isolate have heterofermentation type. Four isolates (BN1, BN2, BN5 and BN6) were able to inhibit S. aureus, E. coli and S. typhi with inhibition zone diameters ranging from 6,9 to 12,3 cm. Based on the result, it was concluded that LAB isolated from naniura has potential as a source of probiotics.

Abstract

Kelapa sawit merupakan satu dari sekian banyak komoditas penyumbang terbesar bagi devisa negara. Pertumbuhan bibit kelapa sawit sangat ditentukan oleh ketersediaan unsur hara dengan pemanfaatan limbah organik seperti kulit pisang kepok dan urine domba. Hal tersebut akan mengurangi penggunaan pupuk kimia (anorganik) yang dapat menciptakan pertanian ramah lingkungan. Selain itu, berdasarkan refrensi yang penulis baca masih sangat jarang dijumpai penelitian mengenai pemanfaatan kulit pisang kepok menjadi pupuk organik cair terutama aplikasi pada pembibitan kelapa sawit. Penelitian ini memiliki tujuan untuk mengetahui respon pertumbuhan bibit kelapa sawit di tahap Pre-Nursery setelah diberikan kulit pisang kepok sebagai Pupuk Organik Cair (POC) dengan dosis yang berbeda. Penelitian ini menggunakan Rancangan Acak Kelompok (RAK) 1 faktorial. Faktornya adalah POC kulit pisang kepok yang terdiri dengan 4 taraf yaitu: Kontrol (A0), 50 ml (A1), 150 ml (A2), 300 ml (A3). Data dianalisis dengan menggunakan sidik ragam (analysis of variance) di lanjutkan Duncans Multiple Range Test dengan signifikan 5 %. Parameter yang diamati yaitu tinggi tanaman, diameter batang, jumlah daun, berat segar tajuk, berat kering tajuk, berat segar akar dan berat kering akar tanaman. Berdasarkan hasil dari penelitian ini bahwa pengaplikasian pupuk organik cair kulit pisang kepok tidak berpengaruh nyata terhadap respon pertumbuhan bibit kelapa sawit di tahap pre-nursery, namun bila berdasarkan data rataan, pengaplikasian 50 ml pupuk organik cair kulit pisang kepok mampu mendorong pertumbuhan bibit kelapa sawit pada tahap pre-nursery.

Abstract

Recently many researches have been carried out on Malacca fruit extract and its potential protective effect on in vivo organ toxicity. However, there is still no research on the protective effect of Malacca (Phyllanthus emblica L.) fruit extract on induced hepatotoxicity of anticancer drugs, especially doxorubicin in rats. The objective of this study is to determine the potential protective effect of ethanol extract of Malacca fruit (P. emblica L.) against doxorubicin-induced hepatotoxicity in rats. A preliminary phytochemical test was done followed by administration extract into doxorubicin-induced rats. Histological studies were carried out to observe the effect of the extract on liver tissue. Phytochemical test results show that the ethanol extract of Malacca fruit contains flavonoids, phenols, tannins, triterpenoids, terpenoids, and alkaloids. The increasing value of ALT and ALP of doxorubicin-induced liver tissue proved the toxicity in liver tissue caused by doxorubicin. The ethanol extract of P. emblica L. at dosage 200 mg/kg BW and dosage 400 mg/kg BW on doxorubicin-induced rats were successfully played as an antioxidant to decrease the ALT and ALPvalues and improve liver tissue doxorubicin-induced by normal hepatocytes more dominant than lysis and less mark of bleeding and congestion of blood vessels.

Abstract

Abstrak Banyak sekali jenis tanaman di Indonesia yang diduga memiliki potensi sebagai antifungi, tetapi belum dimanfaatkan secara optimal. Buah alpukat (Persea americana) adalah salah satu jenis tanaman yang dapat digunakan sebagai antijamur. Penelitian ini bertujuan untuk mengetahui efektivitas ekstrak antijamur kulit alpukat (Persea americana) terhadap pertumbuhan jamur Candida albicans dan Aspergillus niger.Ekstraksi dilakukan dengan maserasi menggunakan etanol 96%. Jamur uji yang digunakan adalah Candida albicans dan Aspergillus niger dengan konsentrasi 25%, 50%, 75%, dan 100% dengan pengulangan kerja 3 kali. Metode yang digunakan dalam penelitian ini adalah metode difusi cakram dengan pelarut DMSO.Hasil penelitian menunjukkan bahwa ekstrak kulit buah alpukat memiliki efektivitas antijamur pada jamur Candida albicans dan Aspergillus niger yang ditunjukkan oleh hasil zona hambat terbesar pada konsentrasi 100% dan zona hambat terkecil di 25%. Analisis data menggunakan uji hubungan antara perbedaan nilai rata-rata dilakukan oleh uji One Way ANOVA pada tingkat kepercayaan 95% (α = 0,05). Hubungan ini disebut bermakna jika p <0,05.

Abstract

Black soybean (Glycine max (L.) Merr.) is a plant that is widely planted and consumed in Indonesia. In addition, black soybean has unique content of isoflavones, such as daidzein, which is one of the active compounds that have the effect of fighting free radicals and can inhibit the aging process. The purpose of this study is to analyze the antioxidant potency possessed by black soybean extract (BSE) and daidzeinin inhibiting aging of the skin. The method used is a colorimetric test. The type of antioxidant test used is H2O2 scavenging and inhibiting the activity of the hyaluronidase enzyme for antiaging. BSE has better effectiveness of H2O2 scavenging (IC50: 286.24±11.16 (µg/mL)) than daidzein compound (IC50: 366.16±2.54 (µg/mL)). In the inhibition of hyaluronidase enzyme, the daidzein has more effective activities (IC50: 95.80±3.98 (µg/mL)) compared to BSE (IC50: 152.56±13.98 (µg/mL)). The antioxidant and anti-aging activities possessed by BSE make it possible to be used as a cosmetic ingredient for skin aging therapy.

Abstract

Tuberculosis (TB) is one of the leading causes of death in developing countries. One of the problems in controlling TB disease is that most anti-tuberculosis drugs are hepatotoxic. Citrus sinensis peel extract is the rich source of secondary metabolites with high potential effectiveness as an antioxidant. In the present study, we evaluate the hepatoprotective effect of Citrus sinensis peel ethanolic extract (CSPEE) on isoniazid and rifampicin-induced liver injury in Wistar rats. Twenty five adult male Wistar rats were divided into 5 groups of 5 each : control,  (INH+RIF) (50 mg/kg bw once a day for 14 days), (INH+RIF) + various dose of CSPEE (300, 450, 600 mg/kg bw).  CSPEE was given orally once a day for 14 days followed by administration of INH + RIF suspension. The measurement of serum ALT and AST were carried out on the 15th day. Histopathologic examination of the liver was also performed. The Serum ALT and AST of the rats that induced with INH + RIF were increased significantly (P<0.001) compare to those of control groups, and the histopathologic slides showed steatosis, vacuolation and necrosis of hepatic cells. The serum ALT and AST in groups treated with CSPEE were not significantly different (p>0.05) with those of control groups. The serum ALT and AST and histopathological examination of the liver of the group that administered 600 mg/kg CSPEE were closest to normal rats. Citrus sinensis peel extract exhibits hepatoprotective effect on liver injury induced with INH + RIF in Wistar rats.

Abstract

About 1.7%-58% of all cases of acute kidney failure are caused by gentamicin nephrotoxicity and consequently increasing urea and creatinine levels in the blood which are indications of damage to function. Oranges contain secondary metabolites such as flavonoids, alkaloids, coumarin, limonoid, keratinoid and essential oil that have pharmacological activities such as antioxidants, anti-inflammatory, anti-cancer, and also nephroprotector. The purpose of this study is to evaluate the nephroprotective activity of Citrus sinensis peel ethanol extract (EEKJS) on rats induced by gentamicin. Rats were induced using gentamicin at a dose of 80 mg/kgBW intraperitoneally on the 7th day after induced at a dose of 50 mg/kgBW, 100 mg/kgBW, and 200 mg/kgBW for 7 days. The results obtained from the ethanol extract of Citrus sinensis peel gave nephroprotective with the lowest serum creatinine and urea levels at a dose of 200 mg/kgBW which was 0.4±0.02 mg/dl and 43.33 ± 2.51 mg/dl and difference significantly (p<0.05) with a positive control group which was only induced by gentamicin and histopathological results showed significant cell damage in the positive control group that was only induced by gentamicin, and in the 50 mg/kgBW dose group, 100 mg/kgBW, 200 mg/kgBW had cell repair after gentamicin induction. Citrus sinensis are highly recommended to be a food supplement for kidney protection.

Abstract

Aging involves progressive physiological changes of the body, including skin dysfunction and death which can be accelerated by the presence of free radicals. Compounds with potential antioxidants and antiaging are contained in various fruits, including dragon fruit (Hyloceureus polyrhizus (F.A.C. Weber) Britton & Rose). This study aimed to compare the antioxidant and antiaging activities of dragon fruit rind extract (DFRE) and Kaempferol. The antioxidant and antiaging activities were measured by 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenger and inhibitory activity assay of tyrosinase, respectively and was conducted at the Biomolecular and Biomedicine Research Center, Aretha Medika Utama, from August to November 2018. Data acquired were then analyzed using ANOVA, followed by Post Hoc Test using Turkey HSD test with a confidence level of 95% (α=0.05). Phytochemical assay was also conducted to determine the content of active ingredients in thedragon fruit rind extract. Phytochemical test result showed that DFRE contained flavonoids, tannins, phenols, triterpenoids, and alkaloids compounds. Both DPPH scavenging and tyrosine inhibition assay indicated that Kaempferol compound also had antioxidant and antiaging activities. The IC50 values of DFRE and Kaempferol antioxidant activities were 164.98 μg/mL and 83.30 μg/mL, respectively, while the IC50 values of tyrosinase inhibition activity of DFRE and Kaemferol were 88.46 μg/mL and 59.34 μg/mL, respectively. DFRE had antioxidant activities and tyrosinase inhibition activities thus can be used as an alternative antiaging and antioxidant agents. Perbandingan Aktivitas Antioksidan dengan Antitirosinase Ekstrak Etanol Kulit Buah Naga (Hyloceureus polyrhizus (F.A.C. Weber) Britton & Rose)Aging atau penuaan adalah proses kompleks yang ditandai dengan penurunan progresif fungsi fisiologis tubuh, termasuk kulit yang diikuti oleh disfungsi dan kematian kulit. Faktor-faktor yang dapat mempercepat proses penuaan dini di antaranya radikal bebas dan gaya hidup yang tidak sehat. Buah-buahan banyak mengandung senyawa yang berpotensi sebagai antioksidan dan antiaging, salah satu di antaranya adalah buah naga. Penelitian ini bertujuan mengetahui potensi antioksidan dan antiaging ekstrak kulit buah naga (EKBN) yang dibandingkan dengan senyawa kaempferol (SK) sebagai pembanding. Penelitian yang dilakukan meliputi uji fitokimia untuk mengetahui kandungan bahan aktif pada kulit buah naga, uji pemerangkapan DPPH, dan uji penghambatan tirosinase dan dilakukan di Pusat Penelitian Biomolekul dan Biomedis, Aretha Medika Utama, dari Agustus hingga November 2018. Data lalu dianalisis menggunakan analisis varians (ANOVA) satu arah (one way) Hasil penelitian uji fitokimia menunjukkan EKBN mengandung golongan senyawa, flavonoid, tanin, fenol, triterpenoid, dan alkaloid serta tidak mengandung golongan senyawa saponin, steroid, dan terpenoid. Senyawa kaempferol memiliki aktivitas antioksidan melalui pemerangkapan DPPH dan aktivitas antiaging melalui penghambatan tirosinase yang lebih baik dibanding dengan EKBN. Aktivitas antioksidan dilihat dari nilai IC50 EKBN sebesar 164.98 μg/ml dan SK sebesar 83.30 μg/mL. Aktivitas penghambatan tirosinase memiliki nilai IC50 EKBN masing-masing sebesar 88.46 μg/mL dan IC50 SK sebesar 59.34 μg/mL. EKBN memiliki aktivitas antioksidan dan penghambatan tirosinase sehigga dapat dimanfaatkan sebagai alternatif bahan antiaging dan antioksidan.

Abstract

Asparagus is a vegetable that contains phenolic compounds with antioxidant properties that scavenges aging-triggering free radicals. This study aimed to investigate the components and anti-aging potentials of Ethanol Extract form Asparagus (EEA). The study was performed in February 2020 at the Pharmacy Laboratory, University of North Sumatera. The EEA was obtained through maceration using 96% ethanol. An antioxidant assay was performed and the total phenol and flavodoid content were determined using the spectroscopic method. Three gel formulas with different concentrations of EEA was prepared (F1: 1.5%, F2: 2.5%, and F3: 3.5%), and F0 was used as control. The parameters evaluated were moisture, oil content, texture, collagen, wrinkle, pigment, sensitivity, and pore. The result showed that asparagus had a moderate antioxidant activity (IC50: 118,992) with the total phenol and flavonoid contents of 15,9407 mg GAE/g and 3,2286 mg QE/g extract, respectively. The highest aging activities was seen in F3 (3.5%), followed by F2 (2.5%) and F1 (1.5%). The percentage of moisture, oil, texture, collagen, wrinkle, spot, sensitivity, and pore recovery were found to be 40.15%, 49.73%, 71.76%, 17.70%, 70.93%, 49.34%, 42.56% and 25.31%, respectively. Hence, it can be concluded that the EEA Gel at the highest concentration (3.5%) has a high content of phenol and flavonoid which can improve the skin moisture, oil content, texture, collagen, wrinkles, spots, sensitivity, and pores, which promotes anti-aging activities.
