Abstract

Aging is a natural process in human that can be characterized by the appearance of black spot on the skin due to hyperpigmentation. Aging may occur due to an excessive amount of free radicals in the body. Antioxidants possess ability to capture free radicals and inhibit tyrosinase which induces skin aging. Aloe vera has been used in traditional medicine because it contains several bioactive compounds that act as antioxidant and prevent aging process. This study aims to determine phytochemical content, antioxidant activity and tyrosinase inhibition activity of Aloe vera rind (AVRE) and gel (AVGE) extract. This research was carried out at the laboratorium of Aretha Medika Utama-Biomolecular and Biomedical Research Center in Bandung city in September–November 2018. Phytochemical assay was determined using modified Farnsworth method. Antioxidant assay was determined using 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity and antiaging assay was obtained using tyrosinase inhibition assay. AVRE contains flavonoid, phenol, steroid, and alkaloid. Meanwhile, AVGE contains steroid and alkaloid. IC50 DPPH scavenging activity of AVRE was 113.18 µg/mL followed by AVGE was 291.96 µg/mL. IC50 tyrosinase inhibition activity of AVRE was 65.04 µg/mL followed by AVGE was 111.89 µg/mL. AVRE had more active DPPH scavenging activity and tyrosinase inhibition activity than AVGE. AKTIVITAS ANTIOKSIDAN DAN ANTITIROSINASE KULIT DAN DAGING LIDAH BUAYA (ALOE VERA)Penuaan merupakan proses alamiah pada manusia. Penuaan dapat terjadi akibat kadar radikal bebas yang tinggi di dalam tubuh. Antioksidan memiliki kemampuan memerangkap radikal bebas dan menghambat kerja enzim yang berperan dalam proses penuaan. Lidah buaya (Aloe vera) telah digunakan dalam pengobatan tradisional karena diketahui mengandung senyawa bioaktif yang bermanfaat dalam menangkal radikal bebas dan menghambat penuaan. Penelitian ini bertujuan mengetahui kandungan fitokimia dalam ekstrak etanol kulit lidah buaya (EKLB) dan ekstrak etanol daging lidah buaya (EDLB), mengetahui aktivitas antioksidan, pemerangkapan 2,2-difenil-1-pikrilhidrazil (DPPH), serta aktivitas antitirosinase EKLB dan EDLB. Penelitian ini dilakukan pada September–November 2018 di laboratorium Aretha Medika Utama Biomolecular and Biomedical Research Center (BBRC). Analisis fitokimia dilakukan menggunakan metode Farnsworth yang dimodifikasi. Aktivitas antioksidan dianalisis menggunakan metode pemerangkapan DPPH, sementara uji antipenuaan dilakukan menggunakan metode uji aktivitas antitirosinase. Hasil uji fitokimia menunjukkan EKLB mengandung flavonoid, fenol, steroid, dan alkaloid; sedangkan EDLB mengandung senyawa steroid dan alkaloid. EKLB dan EDLB memiliki aktivitas pemerangkapan DPPH dengan nilai IC50 secara berurutan 113,18 µg/mL dan 291,96 µg/mL. Penghambatan tirosinase EKLB dan EDLB dengan nilai IC50 secara berurutan 65,04 µg/mL dan 111,89 µg/mL. EKLB memiliki aktivitas antioksidan dan penghambatan tirosinase yang lebih baik dibanding dengan EDLB.

Abstract

Diarrhea is caused by impaired intestinal absorption of food and beverages (such as carbohydrates, fats, and proteins) contaminated with bacteria such as Vibrio cholerae and Escherichia coli.  The purpose of this study was to determine the effect of giving young areca nut seeds and what percentage of the concentration had a very good effect on overcoming the growth of Vibrio cholerae and Escherichia coli bacteria. To determine the effect of giving young areca nut and what percentage of concentration has the best effect on overcoming bacterial growth of Vibrio cholerae and Escherichia coli bacteria. The sample used is 5kg. Type of research is True Experimental and the research design is Post-Test Only Control Design the data in this study will be analyzed using SPSS Version 27 software. The concentration of the extract was made into 4 concentrations, namely 20%, 40%, 60%, 80%, and positive control (Ciprofloxacin). Furthermore, the inhibition test was carried out on the two bacteria, namely Vibrio cholerae and Escherichia coli were conducted using NA (Nutrient Agar) media. Research shows that extracts from young areca nut (Arecha catechu L.) seeds can inhibit the growth of both bacteria at high concentrations 20%, 40%, 60%, 80%, and positive control (Ciprofloxacin) with an average diameter of the inhibitory zone test for Vibrio cholera were 13.3 mm, 11.5 mm, 13.4 mm, 13.8 mm and 21.3 mm. And for the power test, the inhibition zones of Escherichia coli were 12.8 mm, 11.5 mm, 11.9 mm, 11.9 mm, 19,4 mm. This study showed that the extract (Arecha Catechu L.) affected the growth effectiveness of Vibrio cholerae and Escherichia coli bacteria from 4 concentrations, namely 20%, 40%, 60%, and 80%.  The effect of young areca nut (Arecha Catechu L.) on bacteria was highest at a concentration of 60%.

Abstract

Skin aging due to the damage caused by ultraviolet radiation and toxic ingredients in cosmetics is still a problem. Tomato has antioxidant and skin protection activities. The study aimed to investigate the potential of tomato as an antioxidant and elastase inhibitor. A 170 g of tomato simplicial powder was extracted using ethanol 70% by the maceration method. Antioxidant activity was measured through 2,2’-Azino-bis 3-ethyl benzothiazoline-6-sulphonic acid (ABTS)-reducing activity. The antiaging activity was measured through anti-elastase activity. Tomato extract (SLE) showed strong ABTS-reducing activity (IC50 = 86.66 ± 10.58) and very strong anti-elastase activity (IC50 = 19.73 ± 0.44). In conclusion, there was a linear correlation between antioxidant activity and anti-elastase activity. However, the antioxidant activity and anti-elastase activity of tomatoes were still below lycopene (IC50 antioxidant = 49.23 ± 2.06 µg.ml-1 and IC50 anti-elastase = 10.39 ± 0.43 49.23 ± 2.06 µg.ml-1). However, it was worth to be developed as a natural product as an antioxidant and anti-elastase.  Further study is required to do fractionation to get the purer lycopene compound from tomato.

The banana species used in this study were bananas with seeds, and grew in abundance in several plantation areas. So that the potential is very disturbing to the main crop, namely oil palm, in other words it becomes a weed in plantations. In this study, bananas were used as a source of MOL. This study used an experimental method with a factorial randomized block design with 2 replications and 16 treatments. The first factor is the concentration of MOL banana weevil (Musa balbisiana Colla) (M) with 4 levels: M0 (0%), M1 (15%), M2 (30%), M3 (45%), the second factor is immersion time (P) with 4 levels: P0 (immersion), P1 (15 minutes), P2 (30 minutes), P3 (45 minutes). The data obtained were analyzed by means of analysis of variance with a significance of 5%. The results of data processing showed that anova did not significantly affect the interaction between the MOL concentration of banana weevil (Musa balbisiana Colla) and the immersion time on the growth of Mucuna bracteata. Various concentrations of banana weevil MOL (Musa balbisiana Colla) and soaking time did not significantly affect the growth of Mucuna bracteata plants. However, physically the growth of Mucuna bracteata is good, so that the presumption arises that this MOL actually acts as a nutrient/fertilizer for plants. Not as a growth regulator as the initial target of the research.

The research is aimed at obtaining the concentration of microorganism local bamboo shoots and long soaking the right to increase the growth of plants Mucuna bracteata. The study used an experimental method with a 2 factorial randomized block design (RBD). The first factor is the MOL concentration of the bamboo shoots which consist of 4 level that is: Control (R0), 100 ml/l (R1), 150 ml/l (R2), 200 ml/l (R3) and a factor of two is long immersion which consists of 4 level that is: Control (M0), 15 minutes (M1), 30 minutes (M2), 45 minutes (M3). The data were analyzed using (Analysis of Variance) with a significant 5% and followed by The DMRT (Duncan Multiple Range Test) with a significant 5% using SAS software version 9.3.1. Based on the results of data analysis, the results showed that the effect of local microorganism concentration (MOL) of bamboo shoots on Mucuna bracteata plants had a significant effect on tendril length at a concentration of 150 ml/l (R2) at the age of 4-5 MST and a concentration of 200 ml/l (R3) in age 6 MST, and root shoot ratio at a concentration of 150 ml/l (R2), but did not significantly affect the number of leaves, fresh and dry weight of shoots, fresh and dry weight of roots, number of root nodules, and root length. Soaking time and interaction had no significant effect on the number of leaves, tendrils length, weight of fresh and dry shoot, fresh and dry weight of root, root nodule number, root length, and the ratio of root header.

Golden snails (Pomacea canaliculata) contain minerals that can be used as nutrients for plants so that they can be used as MOL (local microorganisms). The purpose of this study was to determine the effect of giving MOL snails and the duration of immersion on the growth of Mucuna bracteata. This study used a factorial randomized block design experimental method with 2 replications. The first factor was the concentration of MOL snails (K) with 4 levels of treatment, namely K0 (0%), K1 (15%), K2 (30%), K3 (45%) while the second factor was immersion time (M) with 4 levels of treatment M0 (0 minutes), M1 (15 minutes), M2 (30 minutes), M3 (45 minutes). The results showed that the interaction between the concentration of MOL snails (Pomacea canaliculata) and the immersion time did not have a significant effect on all parameters. Various concentrations of MOL snail (Pomacea canaliculata) and immersion time did not significantly affect the growth of Mucuna bracteata.

Abstract

BACKGROUNDNon-alcoholic steatohepatitis (NASH) is a chronic liver disease associated with increased free fatty acids, trans fatty acids and free radical compounds due to the consumption of deep frying oil (DFO). Ginger has hypolipidemic and antioxidant properties, and acts as an insulin sensitizer. The present study was conducted to evaluate the hepatoprotective effect of an ethanol extract of Zingiber officinale Roxb var rubrum (EEZO) in DFO-fed male Wistar rats.METHODSA study of experimental design was conducted involving 24 non-alcoholic steatohepatitis (NASH) male Wistar rats that were randomly divided into 4 groups. Group K1 (control) was given DFO-3 (3 times heated DFO) at a dose of 10 ìl/g BW/day. The treatment groups (P1, P2, P3) were given DFO-3 and red ginger rhizome extract at doses of 100, 200, and 400 mg/kg BW orally for 30 days. The histopathology examination assessed fatty degeneration, and degree of fibrosis. The data were analyzed with the Kruskal-Wallis and Mann-whitney test.RESULTS    The Kruskal-Wallis test showed that fatty degeneration (steatosis, lobular inflammation, hepatocyte ballooning and NASH) were significantly different among the four groups (p<0.05). Mann-Whitney test proved that fatty degeneration was not significantly different between control and treatment groups (p>0.05).CONCLUSIONDeep frying oil increased fatty degeneration and NASH, and none of red ginger rhizome extract doses was able to inhibit fatty degeneration significantly. For the future research it is intended to prolong the induction time and the administration of red ginger rhizome extract.

Abstract

Amylase is the most studied enzyme in biotechnology and also accounts for 25% of the total enzyme market worldwide. In industry, amylase has potential applications in the bakery, textile, detergent, syrup, pharmaceutical, and medical industries. One of the potential sources of amylase is microbes, because it is easy to isolate, culture, and also genetically engineered. Liquid waste is one of the most potential and abundant sources of microbes. In this study, the authors used palm oil waste as a source of amylase-producing microbes. The purpose of this study was to isolate and screen bacteria that showed amylase activity. Bacteria were isolated from palm oil liquid waste samples using the serial dilution method and the number of CFUs was calculated. The isolates were then characterized morphologically including Gram staining, elevation, color, edge, cell shape and also catalase reaction. Characterization results obtained 7 gram positive isolates and 8 gram negative isolates, the shape of cocci (12 isolates) and bacilli (3 isolates) and convex elevation and white isolates. Amylase activity was detected using nutrient agar media supplemented with 1% starch and the amylolytic index was measured. Amylolytic activity was indicated by the presence of a clear zone around the isolate. Of the 15 isolates, only 1 isolate showed amylase activity, namely LCF 4 with an amylolytic index of 0.57. Further research is recommended to identify and determine enzyme activity.Keywords : Activity; amylase; bacteria; palm oil liquid waste

Abstract

Diabetes Melitus (DM) digolongkan sebagai penyakit metabolik yang salah satu tandanya diindikasikan dengan timbulnya hiperglikemia yang dipicu oleh terjadinya kelainan sekresi dan kerja insulin. Suplemen probiotik sebagai alternatif dalam pencegahan dan pengelolaan terapi diabetes. Tujuan dari diadakannya penelitian ini adalah untuk mengetahui aktivitas inhibisi ? glukosidase Lactobacillus casei. Isolat bakteri menggunakan media MRS (Man Rogosa Sharpe) Broth Karakterisasi isolate bakteri dilakukan secara morfologi meliputi pewarnaan gram, uji katalase, dan tipe fermentasi. Aktivitas antimikroba dengan metode disc diffusion agar. Toleransi pH rendah pada media MRS Broth dengan pH 3. Toleransi garam empedu yaitu dengan menumbuhkan isolat pada MRS Broth yang ditambahi dengan 0,3% garam empedu. Pengukuran absorban dijalankan dengan dibantu alat berupa spektrofotometer pada ? 400 nm yang ditujukan untuk mendapatkan aktivitas inhibisi ? glukosidase. Hasil yang terperoleh dari penelitian ini mengindikasikan jumlah koloni isolat bakteri asam laktat (BAL) Lactobacillus casei yang diisolasi dari minuman probiotik komersil sebesar 6,5x10-7 CFU/g. Karakterisasi termasuk jenis Lactobacillus casei, gram staining positif, basil, katalase negatif dan heterofermentatif. Aktivitas antimikroba BAL Lactobacillus casei terhadap S.aureus adalah 9,65 mm dan E.coli adalah 9,3 mm. Persentase viabilitas pertumbuhan BAL Lactobacillus casei terhadap asam yaitu 10,86%, dan garam empedu sebesar 27,5%. Persentase aktivitas inhibitor ? glukosidase BAL Lactobacillus casei sebesar 40% dan acarbose adalah 91%.

Abstract

Bakteri probiotik merupakan kelompok bakteri yang memberikan konstribusi terhadap keseimbangan mikroba usus. Setiap spesies bakteri asam laktat memiliki efek probiotik yang berbeda-beda sehingga diperlukan seleksi dan identifikasi untuk mendapatkan strain probiotik yang baik. Identifikasi bakteri asam laktat dalam penelitian ini menggunakan metode identifikasi secara konvensional. Didapatkan 3 isolat bakteri asam laktat (MF1, MF2, dan MF4) diidentifikasi memiliki karakteristik yaitu memiliki bentuk cirkuler, tepi entire dan elevansi flat selanjutnya Uji Biokima didapatkan katalase negative, tipe homofermentatif, pati negative, sitrat negative, motilitas positif dan gelatin negative serta pH 3,5-4,5. Hasil uji antibakteri didapatkan pada isolate MF1, MF2 dan MF3 yaitu 7,2 mm, 6,6 mm dan 7,1 mm. Hasil identifikasi konvensional pada bakteri probiotik didapatkan isolate bakteri yaitu Basillus sp.
