Abstract

The purpose of this field verification was to determine the stability of yield of several drought-tolerant soybean genotypes selected from green house evaluation. The plants were planted at Muneng, Probolinggo in dry season (June-September 1995) with and without irrigation. From 5 tolerant genotypes, Mlg 2999 and Mlg 3474 gave a good stability of tolerance as evidence by less significance of growth and grain yield reduction. The tolerant genotypes of Mlg 2805 and Mlg 2984 suffered from tremendous leaves and pods abcission, leading to low yield.

Abstract

In this experiment. the changes on leaf osmotic potential and accumulation of proline and abscisic acid were identified from drought-tolerant and drought - sensitive soybean genotypes. Three drought - tolerant (Mlg 2805, Mlg 2984 and Mlg 2999) and two sensitive soybean genotypes (Mlg 2510 and Mlg 3541) were subjected to drought condition created by regulating water supply in greenhouse.  The results revealed that exposing plants to drought stress brought about a decrease of leaf osmotic potential. The decrease of which was greater in drought-tolerant genotypes (6.91 to 10.11 bars) than in sensitive genotypes (0.55 to 0.69 bars). The decreasing of leaf osmotic potential was followed with increasing praline accumulation, especialy for Mlg 2805. Only Mlg 2805 showed the significant ABA accumulation when the plants were subjected to drought stress. It is suggested that the drought tolerance was associated with the reduction of leaf osmotic potential (osmotic adjusment) in which proline might play an important role. The role of ABA could not be clarified since there had been a great variability in ABA content of all tolerant genotypes.

Abstract

Peanut and kidney bean gave similar response to salt stress showing a more severe growth reduction, as compared to spinach, leaf beet, barley cultivars and wild barley. Calcium showed the protective effect on salt injury in all plants, the effect of which was more pronounced in spinach, leaf beet and both types of barley plant than that in bean and peanut. High concentration of NaCl decreased the content of K, Ca, and Mg in both shoot and root of all plants. Elevated Ca in the nutrient solution, however. Considerably alleviated the inhibition of K uptake due to NaCl. The result suggested that the maintenance of K / Na selectivity by Ca in plant might result in the enhancement of salt tolerance.

Abstract

The purpose of this experiment was to investigate the effect of Si on the growth and ions uptake (K, Na, Ca, and Mg) in rice, bean and barley plants grown in saline conditions. Rice and bean plants were subjected to 0 and 30 mM NaCI in the nutrient solution with and without Si, whilst for barley plants 0 and 40 mM NaCI were used. In this investigation, 2.0 mM Si was used as silicilic acid. The results revealed that Si had the protective effect on salt injury only for rice plants. This alleviating effect of Si appeared to be associated with the interference of Si on the upward Na transport by reducing the content of Na in the shoots and retaining it in the roots. Generally, addition of Si did not affect the content of K in all plants tested. Except for rice, Si had no or less effect on Ca and Mg uptake in bean and barley plants.

Abstract

The aim of this study was to see the salt tolerance of turf grass Puccinellia distans in term of ion accumulation and ionic interactions under salt stress condition. Plants were grown hidroponically in culture solution with addition of NaCI (0, 25, 50, 100 and 200 mM). High concentrations of NaCI brought about growth reduction; the decrease of shoot dry matters was in the range of 30-84% and 30- 70% for root dry matters. Although the yield of plants decreased so much in the presence of 200 mM NaCI, but plants were able to remain alive without showing any visible symptoms of injury. Much more Na was accumulated in the roots than in the shoots. Exposing plants to salinity result in the decrease of K, Ca, and Mg contents.

Abstract

In this study, multi-compartment transport boxes were used to determine the mobility of ions in the execised roots. For measuring Na uptake, the concentrations of 1, 50 and 150 mM NaCI were used in the external medium, whilst Ca were 0 and 2. 0 mM. The uptake of K was measured from 1.0 mM KCI in the presence of Na (0.50 and 150 mM) and Ca (0.50 and 2.0 mM). In the present study, 22 Na and 86 Rb were used to label Na and K in the external medium, respectively. Addition of Ca inhibited Na uptake in the roots exposed to 1.0 mM and 50 mM NaCI, thought Ca was relatively less effective in 150 mM. High concentrations of NaCI brought about the reduction of K uptake. Calcium, however, did not show any importance in recovering the uptake of K from the adverse effect of Na when the roots were exposed 10 50 mM NaCI, thought stimulating effect of Ca was observed at 150 mM NaCI. The results revealed that the pattern of ionic interactions in Puccinellia distans differs much with that generally found in most of halophyte plants.

Abstract

Pot experiments and laboratory analysis were conducted to determine the effect of N fertilization and level of water table treatments of wet soybean culture on nodule formation and seed yield of soybean. The pot experiment was conducted in Cibungbulang and laboratorium analysis, was at three different laboratories: Soil, Agronomy, and Pasture, Bogor Agricultural University, started on August, 1993 up to February, 1 994. Three factors to be evaluated, namely varieties (Lokon and Lompobatang), N fertilizer applications of 0, 140, and 280 mg pot -1 (equivalent to 0, 25, and 50 kg N ha -1), and the level of water tables of (control, -15 cm, -10 cm, and -5 cm below the soil surface). They were arranged in randomized complete design. The highest value of nodule dry weight of Lokon variety (0.707 p. was resulted from the plant grown under -15 cm water table without N fertilizer (0 mg N pot- ). Lompobatang highest value (1. 727g) on -10 cm water table and fertilized with 140 mg N pot -1. The increases of nodule dry weight by the treatment were 0.300 g (73.7%) for Lokon and 1. 134 g (194.5%) for Lompobatang, respectively compared to control. Both varieties showed highest value of seed weight per plant if grown under -15 cm water table and fertilized with 140 mg N pot -1. Compared to control media, respective increased seed weight per plant 1.94 g (12.9%) for Lokon and 5. 40 g (25.4%) for Lompobatang.

water deficiency. Genetic engineering at the level of transcription factors (TF) is particulary a promising strategy in developing drought tolerant rice cultivar. HD-Zip genes are TF that function in plant adaptation to some environmental stresses including water deficit. The recombinant plasmid pC1301H Oshox6 which contained HD-Zip Oshox6 gene was placed under a drought inducible promoter called LEA promoter, gusA and hpt genes were driven with CaMV promoter. The aim of research was to obtain indica rice transgenic plants of Batutegi and Kasalath cultivars using pC1301H Oshox6 plasmid. Recombinant plasmid was transformed into immature rice embryos using Agrobacterium tumefaciens. Kasalath cultivar showed a better capacity to form embryogenic calli compared to Batutegi. Transformation efficiency of Batutegi is lower (1.5 - 0.3%) than Kasalath (2.2-28.3%). Regeneration efficiency is 25-83.3% and 7.7-100% for Batutegi and Kasalath, respectively. Number of putative transformant plantlets of Batutegi and Kasalath are 63 and 48 plantlets, respectively. Southern blot analysis (using hpt probe) on 12 independent lines of each Batutegi and Kasalath cultivars showed different gene copy number, ranging from one to four copies of gene.   Keywords: agrobacterium tumefaciens, LEA promoter, HD-Zip Oshox6, rice

Abstract

Melastoma malabathricum L. grows well in acid soil with high level of soluble aluminum. One of the important proteins in the detoxifying acid and aluminum stress is a plasma membrane H+  -ATPase protein encoded by PMA gene. The objective of this research was to isolate and clone the cDNA fragment of MmPMA encoding plasma membrane H+ -ATPase from M. malabathricum L. By reverse transcription, total cDNA had been synthesized from the total RNA as template. The fragment of MmPMA  cDNA  had been successfully isolated by PCR by using total cDNA  as  template and PMA primer designed from conserved region for corresponding gene. This fragment had been successfully inserted into pGEM-T Easy and the recombinant plasmid was successfully introduced into E. coli DH5". Nucleotide sequence analysis showed that the length of MmPMA fragment is 806 bp encoding 268 amino acids. Local alignment analysis based on nucleotide of mRNA showed that MmPMA fragment was 81% identical to part of PMA of Sesbania rostrata, Juglans regia, and Prunus persica. Based on deduced amino acid sequence, MmPMA was 94% identical to part of PMA of Juglans regia; 93% to PMA of S. rostrata, and Arabidopsis thaliana. MmPMA fragment has phosphorylation intermediate domain (DKTGT) and ATP binding domain (KGAP, DPPR, MITGD, and GDGVN).   Keywords: isolation, Melastoma malabathricum L., MmPMA fragment, sequencing

The study was conducted in the greenhouse of the University Farm, Bogor Agricultural University from August to October 2009. The experiment was carried out as a Factorial experiment in a Completely Randomized Design with three replications. The fi rst factor was sorghum genotypes consisted of Numbu (tolerant) and B-75 (sensitive), and the second factor was combination of lime and phosphorous fertilization consisted of: no lime-no P (R1), no lime-low P (R2), no lime-suffi cient P (R3), lime-no P (R4), lime-low P (R5) and lime-suffi cient P (R6). The results showed that all variables were signifi cantly affected by the interaction of media and genotype, except for length of shoot and total P uptake. Under low P and Al toxicity, sorghum variety Numbu showed a higher nutrient effi ciency than B-75 with ability of forming greater dry mass. The sensitive genotypes showed a higher nutrient uptake but a lower P nutrient use efficiency under Al toxicity and P defi ciency. Total P uptake and P nutrient use effi ciency were highly correlated with root growth and dry material accumulation.
