Abstract

One of the main public health issues with increasing morbidity and mortality rates in tropical countries like Indonesia is dengue hemorrhagic fever (DHF). Antiviral treatments are still unobtainable and the curative value of current vaccines is limited. Many bioactive compounds, including flavonoids and tannins, identified in the tropical fruit Psidium guajava, which are utilized extensively in traditional medicine, have shown possible antiviral, antibacterial, and antioxidant effects. The focus of this research is to determine Psidium guajava extract's antiviral activity against DENV-1, cytotoxic effects on Vero cells, and antibacterial activity towards Escherichia coli. The extract was tested on DENV-1 in Vero cells using the Viral ToxGloTM assay, and the selectivity index (SI) was determined by using the IC50. The  CellTiter 96Ò Aqueous One Solution Cell Proliferation Assay method was applied for cytotoxicity tests, while the disc diffusion method on Mueller-Hinton Agar was utilized for testing the antibacterial effect towards Escherichia coli. With an IC50 value of 67.74 µg/mL, the findings suggest that guava extract effectively inhibits DENV-1 replication. The level of cytotoxicity of this extract against Vero cells showed a good result, namely <200 µg/mL, which can be categorized as moderate cytotoxicity. Also, the Psidium guava extract indicated antibacterial effect against Escherichia coli, at a concentration of 100 mg/mL with an inhibition zone of 4.6 to 5.0 mm. Based on these findings, Psidium guajava extract has the potential to be a natural therapeutic substance with a good safety record towards antiviral, cytotoxicity and antibacterial effects.

Abstract

Troloyo Cemetery, located in Trowulan, Mojokerto, East Java, is a historical burial site that exemplifies the hybridity between Islam, Hindu-Buddhist traditions, and modernity. This study explores the cemetery as a cultural third space, a concept introduced by Homi K. Bhabha (1994), where different belief systems and historical legacies intersect, negotiate, and form new identities. The research applies a qualitative approach through semi-structured interviews and direct observation, focusing on the perceptions of local caretakers and community members. Findings reveal that Islamic elements are present in tombstone inscriptions, Arabic calligraphy, and pilgrimage practices associated with Syekh Jumadil Kubro, a key figure in Javanese Islamization. Simultaneously, Hindu-Buddhist influences persist through architectural structures like kijing and symbols such as Surya Majapahit, while modern features—such as tourism and infrastructure development—further shape the site's identity. Ritual practices like incense burning, flower offerings, and local cosmological concepts highlight syncretic expressions of spirituality rooted in Javanese culture. These complex layers of religiosity and symbolism challenge rigid categorizations of identity, instead revealing a dynamic cultural landscape formed through historical negotiation and continuity. The study concludes that Troloyo Cemetery functions not merely as a sacred Islamic site but as a hybrid space where cultural and religious forms co-exist and evolve, offering a deeper understanding of Indonesia's plural spiritual heritage.

Abstract

Local wisdom examines an essential aspect of cultural identity, social values, and community resilience, particularly within three heritage sites. This study aims to analyze the forms and functions of local wisdom embodied in three significant cultural sites in East Java: the tomb of Sunan Ampel, the tomb of Sunan Drajat, and Bajang Ratu Temple. Employing a qualitative descriptive method, data were collected through observation, documentation, and literature review to ensure systematic and valid interpretation. The findings reveal that each site reflects distinctive yet interconnected forms of local wisdom. The Sunan Ampel cemetery emphasizes moderation, spiritual devotion, and personal transformation, as seen in the figures of Sunan Ampel Lembu ,Peteng, Mbah Sholeh, and Mbah Bolong. Sunan Drajat site highlights social welfare, humanitarian values, and moral responsibility, particularly through the symbolic meanings of its stairways, which promote empathy, vigilance, perseverance, spiritual discipline, and charity. Meanwhile, Bajang Ratu Temple demonstrates a synthesis of Hindu-Buddhist philosophy and local wisdom through its architectural design, narrative reliefs, and symbolic ornaments, representing moral teachings, cosmic balance, spiritual protection, and transformation.

Abstract

Indonesia recorded the highest dengue mortality rate in Asia in 2022, with 45,387 cases and 432 deaths.  This study aimed to investigate the coexistence of arboviruses and insecticide resistance mechanisms in Aedes vectors mosquitos’ populations in Mulyorejo District, Surabaya, during the early monsoon season of 2021. A cross-sectional survey was conducted in January 2021, and Aedes larvae were collected and reared to adulthood under laboratory conditions. A total of 309 Aedes larvae (comprising 65% Aedes aegypti and 35% Aedes albopictus) were collected from 36 breeding sites, identified, and georeferenced using QGIS software. RNA extraction and RT-PCR assays were performed to detect Dengue virus (DENV), Japanese encephalitis virus (JEV), West Nile virus (WNV), and knockdown resistance (KDR) mutations. Results showed seven samples positive for DENV-2 and seven for JEV, with two samples (codes 9 and 12) exhibiting co-infection with both viruses, while no WNV was detected. The detection of JEV in Aedes larvae is a notable finding, suggesting a potential secondary vector role for these species in the study area, as Culex mosquitoes are the primary vectors of JEV. Mutations at codons 1016 (V1016G) and 1534 (F1534C), associated with pyrethroid and DDT resistance, were detected in eight and seven samples, respectively. These findings confirm the circulation of DENV-2 and JEV in local Aedes populations, highlighting the presence of KDR mutations that confer insecticide resistance. The results underscore the urgent need for integrated vector management in Surabaya, combining arbovirus surveillance, insecticide resistance monitoring, and community-based source reduction to mitigate arboviral disease transmission.

Abstract

Purpose of the study: This study examines the effect of integrating Inquiry-based learning with a pedagogical Deep Learning approach on students’ learning outcomes in a Learning Planning course. Methodology: Employing a one-group pretest–posttest quantitative design, the study involved N = 50 undergraduate students from two teacher education institutions in East Java, Indonesia, involving Economics Education students from Universitas Nusantara PGRI Kediri and Universitas PGRI Mpu Sindok Nganjuk. The instructional intervention emphasized Inquiry activities aligned with Deep Learning principles mindful, meaningful, and joyful learning. Main Findings: Data were collected using a validated learning outcomes test administered before and after the intervention and analyzed through paired-sample t-tests. The findings indicate a statistically significant improvement in students’ learning outcomes, with mean scores increasing from 73.40 (pretest) to 84.80 (posttest), yielding an average gain of 11.4 points (p < 0.05). Novelty/Originality of this study: These results suggest that the integration of Inquiry-based learning and Deep Learning principles holds potential for enhancing pedagogical competence in Learning Planning courses within teacher education programs.
