Abstract

Abstract The purpose of research is to measure of physic-chemist aquatic parameter on Porong Estuary. Survey method used to collection of data. The average of chlorophyll-a index are 0-9.5 µg/l (3-68 µg/l rise tide and 1-30 µg/l low tide). Hight concentration of TSS on rise tide is 232-700 mg/l and low tide is 124-424 mg/l. Plankton analysis find out 73.3% diatom, 20% dinoflagelatta and 6.67% cyaniphyceae.

Highlight ResearchThe leopard coral grouper Plectropomus leopardus was identified and analysed based on molecular approach.Genetic diversity within two regions in Gorontalo, Sulawesi successfully performed using connectivity analysis.Three haplotypes of Plectropomus leopardus from two region in Gorontalo as one of economical important marine fish species. AbstractBar-cheek coral trout (P. leopardus) is the flagship of the grouper in the live fish market in Asia. Unfortunately, the potential of the grouper is still partly produced from natural catches. Even though hybridisation activities have also started to be carried out, there still have not been many studies on the genetic diversity of these fish. The application of molecular identification has been widely applied in marine aquatic animal species, which are very likely to occur due to errors in terms of shape and colour in the morphological character. DNA information has been beneficial in efforts to the breeding program and develop grouper aquaculture activities. DNA barcoding was used for the molecular identification and haplotype analysis of P. leopardus from two locations in Gorontalo, Sulawesi, Indonesia. A total of 14 fish samples were collected from two traditional fish markets around Kwandang and Sumalata Gulf in the northern part of Gorontalo Province, Sulawesi. This study identified and found three haplotypes from both regions. Molecular identification using Cytochrome c Oxidase subunit I (COI) gene region on mitochondrial DNA. Besides Mega7 for phylogenetic reconstruction, the data analysis using DnaSP6, Arlequin Ver.3.5.2.2, and Network 5.0.1.1. The first Haplotype is a mixed population between the Kwandang Gulf and the Sumalata Gulf, then the Kwandang Gulf haplotype and the Sumalata Gulf haplotype. The genetic distance between Kwandang Gulf haplotype and Sumalata Gulf haplotype is 0.003984, classified as a shallow genetic distance and needs more samples from another region to figure out leopard coral grouper around Indonesia.

Abstract

Artemia franciscana nauplii is a source of nutrients and pathogen vectors due to the non-selective nature of the filter feeder. Prevention of pathogens is done by adding iodine disinfectant to suppress the emergence of bacteria. One way to find out if a substance is toxic is by testing the toxicity and lethal concentration (LC50) on A. franciscana nauplii. The aim of this study was to evaluate the disinfectant effect of iodine and determine the lethal concentration of iodine in 50% of A. franciscana nauplii (LC50). Brine Shrimp Lethality Test (BSLT) method using Artemia as test animal. The results of the toxicity test with several doses of iodine showed that the toxic threshold value for 96 hours was 20.31 ppm. The results were supported by the presence of stunted growth at a dose of 100 ppm by measuring the length and weight. The conclusion of this study is that the administration of iodine at several concentrations is toxic to A. franciscana nauplii if it exceeds 20.31 ppm and affects the growth of A. franciscana nauplii.

Abstract

The recent adoption of environmental DNA (eDNA) represents an innovative method for assessing the presence of aquatic vertebrate species, providing a relatively straightforward approach with significant implications for conservation biology. In our investigation, we employed eDNA metabarcoding to explore the diversity of fish in the Makassar Strait. We collected eDNA from samples taken both at the surface and from the water column (15 m depth) at two specific locations within this region. The reliability of the MiFish-U primer set methodology in estimating fish diversity in the Makassar Strait was assessed. In a single survey, based on four water samples from the Makassar Strait, we successfully identified 11 marine fish taxa at the species level. These taxa belong to 8 families across 8 orders. The predominance of reef-dwelling species suggests that coral reefs play a dominant role as the primary ecosystem in this area. Among the surveyed sites, Barru Waters exhibited the highest species richness (7 species), while Pangkep Waters only revealed 4 species. Through the application of eDNA metabarcoding, this study provided a means to assess fish diversity, delivering crucial foundational information. Our findings highlight the cost-effectiveness of the eDNA metabarcoding method as a powerful scientific tool for the management and conservation of marine fish resources in the Makassar Strait.

(Eds.), Prosiding Seminar Nasional Lahan Suboptimal ke-10 Tahun 2022, Palembang 27 Oktober 2022. pp. 293-301. Palembang: Penerbit & Percetakan Universitas Sriwijaya (UNSRI).Plastic litter degrade very slowly in the natural environment. The different characteristics of plastic waste are one of the determining factors in the decomposition process. This study aimed to investigate the type, abundance and characteristics plastic waste in Segara Anakan Lagoon. Plastic waste was taken from Segara Anakan Lagoon on August, 2020 using survey method. Sample was collected from 1x1 meter transect. Analysis of the data used is descriptive analysis. The results showed that we found 7 types of plastics waste, i.e. PET, HDPE, PVC, LDPE, PP, PS, and OTHER. The highest plastic waste abundance was found in station 2 (82,67 items/m2), and the lowest was in station 9 (1,67 items/m2). PP is the dominant type of plastic with a total of 839 items (71.5%) followed by LDPE with a total of 189 items (16.1%), while PVC and PS were found in lowest number, with each of them 6 items (0.5%). The characteristics of plastic waste in Segara Anakan based on the type of plastic waste at station 2 have different types with other stations and the differences are clearly illustrated.

Abstract

Turmeric as a natural dye contains curcuminoids which are beneficial for the body and have the potential to be applied in food stuffs such as in amplang. Milkfish amplang (Chanos chanos) is a snack made from typical Indonesian fish which is quite popular in society. This study used experimental laboratory research methods which aimed to determine the effect of adding the amount of turmeric (Curcuma domestica) on the level of consumer acceptance of the chemical and organoleptic characteristics of milkfish amplang. The design used was a completely randomized design (CRD) with one factor, namely the addition of turmeric to the production of milkfish amplang consisting of four treatments and three repetitions. The results showed that there was a significant difference between the treatments (p<0.05), the highest nutritional value was found in milkfish amplang with the addition of 0.5 g of turmeric with an average value of water content (2.64%), ash content (1.75%), protein content (10.54%), fat content (7.51%), and carbohydrate content (77.53%). Organoleptic test results showed that the best treatment was the addition of 0.5 g turmeric with average values for color (8.60), smell (8.40), taste (8.53), and texture (8.80).

Abstract

Spirulina is known as a superfood because it is rich in nutrients, both macro and micro. Spirulina has high levels of protein, potassium and polyunsaturated fatty acids (PUFAs) which contain essential amino acids, phycocyanins, essential vitamins and antioxidants. The high nutritional content of Spirulina causes the higher utilization of Spirulina in food products. Taiyaki is a food product originating from Japan and also popular in Korea (bungeo-ppang). The research design consisted of four treatments and five replications. The treatment of adding Spirulina sp powder into the taiyaki filling as much as 1, 2, and 3% of the weight of the filling. The results of the Kruskal Wallis test showed that the addition of Spirulina sp powder had no effect on the parameters of appearance, aroma and texture, on the contrary it had a significant effect on the parameters of taste. The results of the Mann-Whitney follow-up test, the level of preference for taste parameters was not significantly different (p> 0.05) between P0 (without the addition of Spirulina sp powder) and the treatment with the addition of Spirulina sp powder 1% (P1), 2% (P2), and 3% (P3) of the filling weight. The addition of Spirulina sp. in taiyaki products can increase the nutritional value of taiyaki, especially the protein (3.61%) and fat (2.63%) components.

Abstract

Pedada fruit (Sonneratia caseolaris) contains phytochemicals such as steroids, tripenoids and flavonoids. Phytochemical compounds are antioxidants that neutralize free radicals that attack body cells. The application of making pedada fruit jam with the addition of carrageenan can function as a natural preservative. The purpose of this study was to determine the effect of adding carrageenan to the shelf life of pedada fruit jam. The research treatment consisted of 0 week of storage (P0), 1 week (P1), 2 weeks (P2), 3 weeks (P3), and 4 weeks (P4). Parameters observed in this study were the total plate count test, viscosity test, and antioxidant activity test with spektrofotometer Uv-Vis. The data obtained was tested with the ANOVA statistical test and continued with the DMRT test. The results of the research show that antioxidant activity and viscosity have decreasing values with each storage period, while microbiological activity is inversely proportional to antioxidant activity and viscosity, namely with increasing values with each storage period. The storage time for pedada fruit jam with the addition of carrageenan had a significant effect on antioxidant activity, viscosity and microbiological activity which had significantly different values (p < 0.05).

Abstract

Panceng water is a coastal area that has quite a lot of activities, and this can trigger an increase in waste pollution. This polluting waste can cause an increase in the bioaccumulation of heavy metals in waters. Therefore, this research aims to obtain information on environmental management monitoring activities, such as water quality conditions by determining the concentration of heavy metals Pb, Cu and Zn in the roots of the Avicennia marina mangrove, water and sediment in Panceng Waters, Gresik. The method used was observation, examination of the heavy metals Pb, Cu, and Zn using ICP-MS, and the data was analyzed using quantitative descriptive methods. The results of the analysis of the concentration of the heavy metal Pb in the roots of the mangrove averaged 0.3 mg/kg, Cu 1.205 mg/kg and Zn 0.14 mg/kg. The concentration of heavy metals Pb in sediment was 5.49 mg/kg, Cu 15.7 mg/kg, and Zn 30.6 mg/kg. Meanwhile, the concentration of heavy metals Pb, Cu and Zn in sea water was not detected (ND). Overall water and sediment are still below the threshold. However, the concentration of heavy metals in the roots of the A. marina mangrove is relatively high, above the quality standards of Minister of Environment Decree No. 22 of 2021. Calculation of the level of heavy metal pollution based on Enrichment Factor (EF), Geo-Accmulation Index (Igeo) and Contamination factor (CF) values shows that the heavy metals Pb, Cu and Zn indicate levels of pollution and contamination that are still low to moderate. The Bio Concentration Factor (BCF) of A. marina in Panceng Waters, Gresik is included in the excluder category (BCF<1). The results of this research indicate that the A. marina mangrove can be developed as a phytoremediation agent because it is able to absorb and transfer heavy metals from the environment to other body tissues.

Abstract

Aquatic mammals in Indonesia are officially protected. However, there is a lack of research on these mammals, particularly in relation to potential disease-causing bacteria. A study was conducted in a conservation pond to address this gap, where swab samples were taken from the blowholes of aquatic mammals. The focus was on identifying bacteria that could potentially cause infectious diseases in these animals. The results revealed V. parahaemolyticus bacterial isolates, which showed a 98% similarity to the registered V. parahaemolyticus in NCBI. These bacterial isolates exhibited hemolysin properties and demonstrated resistance to trimethoprim, streptomycin, cephalothin, and penicillin antibiotics.
