Abstract

Snakehead fish with different geographical differences can have an impact on genetic diversity. DNA barcoding is a system designed to identify a species precisely and accurately by using short and standardized gene regions. This study aims to determine the diversity of snakehead fish populations in four areas in East Java. This study used the Cytochrome b (Cyt b) gene sequence in mitochondrial DNA to determine genetic variation in the wild population of Channa striata. The results of DNA barcodes with Cytochrome c oxidase subunit I (COI) showed that all samples were validly C. striata species (99.84-99.85%), while Cyt-b sequences had percentage identity with the same sequence species ranging from 99.74 to 100%. The phylogenetic tree reconstructed using MEGAX resulted in a separate clade in the sequences from East Java. This strengthens the similarity of the population of C. striata species in the Brantas watershed.

Abstract

Rasbora (Rasbora sp.) is one of the endemic freshwater fish species in East Java, which is currently being threatened due to environmental degradation, human activities and pollution. In East Java, it is known that there are several species of Rasbora (Rasbora sp.). Domestication is one method to preserve Rasbora in East Java, preceded by identifying potential species of Rasbora sp. from the natural catchment. This study aims to determine the diversity of Rasbora sp in East Java. Samples were taken at five different locations, specifically: Nganjuk (Brantas Watersheds), Ngawi (Bengawan Solo Watersheds, Silowo Springs Tuban, Umbulan Springs Pasuruan and the Setail Watersheds Banyuwangi. The main parameters observed were morphology and morphometry. The morphological analysis used identification guidelines, while morphometric data were obtained through 15 parameters measurement then analyzed using one-way Anova test on SPSS 25 software. Based on the morphological and morphometric analysis, it is known that the species of Rasbora in Nganjuk and Ngawi regencies are Rasbora argyrotaenia and the species of Rasbora in Tuban, Pasuruan and Banyuwangi regencies are Rasbora lateristriata. One-way Anova test showed Rasbora from the five locations were significantly different based on the result value of p<0.05.

Abstract

Budidaya rumput laut adalah salah satu komoditas ekspor utama di Indonesia. Jenis rumput laut yang memiliki nilai ekonomis salah satunya ialah Eucheuma cottonii. Karakteristik yang dimiliki oleh rumput laut yaitu dapat menyerap senyawa di perairan atau bersifat biofilter. Salah satu faktor yang dapat menurunkan mutu rumput laut adalah masuknya bahan pencemar seperti logam berat. Akibat dari masuknya logam berat di perairan ialah adanya akumulasi logam berat pada thallus rumput laut hingga terjadi bioakumulasi dan biomagnifikasi. Metode penelitian ini adalah survey atau observasi dimana data didapatkan dari lapang. Pengujian sampel secara ex situ diantaranya sampel air, sedimen dan rumput laut yang dilakukan menggunakan metode Inductively Coupled Plasma Mass Spectrometry (ICP-MS) untuk mengetahui kadar logam berat Pb, Cd, dan As. Hasil kemudian dibandingkan dengan standar baku.  Metode analisis data diukur menggunakan Contamination Factor (CF), Index Geo-Accumulation (Igeo), dan Bioconcentration Factor (BCF). Hasil pengujian menunjukkan akumulasi logam berat Pb, Cd dan As berada pada bawah ambang baku mutu dan berada dalam kondisi yang aman. Hasil logam berat Pb, Cd dan As berturut-turut pada rumput laut yaitu 0.0665 mg/kg, 0.516 mg/kg, dan 0.7253 mg/kg. Tingkat kontaminasi logam berat berdasarkan analisis data menggunakan Contamination Factor menunjukkan hasil kontaminasi rendah. Pada indeks Igeo menunjukkan tingkat pencemaran tidak tercemar. Sedangkan pada Bioconsentration Factor menunjukkan hasil akumulasi rendah.

Abstract

The enormous potential of marine resources possessed by Banda Aceh Province is expected to be utilised optimally. Accuracy in marine fish resource identification is a critical requirement to support their utilisation and preservation in Banda Aceh Province. In this study, a molecular identification approach was carried out in addition to conducting a morphological identification, commonly used by several scientists. The results were 47 COI sequences generated representing 33 genera, 19 families, and five orders. From the resulting COI partial sequences, there is one potential haplotype from the Scombridae family (Auxis thazard), two potential haplotypes from the Carangidae family (Elagatis bipinnulata and Decapterus macarellus), and two potential haplotypes from the Serranidae family (Variola albimarginata and Cephalopholis sonnerati). This study is essential for fisheries biological studies and other fisheries studies to support the sustainable utilisation of marine fisheries potential in Banda Aceh.

Abstract

Snapper is a type of demersal marine fish from the Lutjanidae family. The Lutjanidae family spread throughout the world and currently has 123 species in 21 genera, one of which is the Lutjanus genus (Miller and Thomas, 2007). To this day, the records of capture fisheries production data for snapper in Malang are still very limited to certain types. Morphological identification that has been carried out so far is still challenging to obtain accurate results because of the many similarities between the observed species or the loss of characteristics. Therefore, molecular identification is necessary to determine the types of snappers in this area and their conservation status. This study aims to determine the types of snappers using a molecular approach by Cytochrome Oxidase subunit I (COI) gene marker.  Phylogenetic tree reconstruction and genetic distance calculations were performed using Mega X software through the neighbour-joining (NJ) algorithm. The results of the identification snapper based on a molecular approach with DNA barcoding revealed that the four snapper samples were L. gibbus, L. rufolineatus, L. bengalensis, and L. erythropterus. Based on the results of the compilation of the phylogenetic tree, it can be seen that the L. bengalensis sample is closely related to L. rufolineatus while L. gibbus, and L. erythropterus each form a separate clade from the two previous Lutjanus species. Based on their conservation status at the IUCN, the four species of snapper found are in the Least Concern category.

Abstract

Antibiotics are compounds that are used to inhibit the growth of disease or are commonly referred to as drugs. Some examples of antibiotics most commonly used in shrimp culture are tetracycline, oxytetracycline, and chloramphenicol. In shrimp farming, antibiotics function as disease prevention or as a cure for the disease. But if the application is not under the dose, it will cause residues on the shrimp and harm human health if consumed. The purpose of this study was to determine the antibiotic content of tetracycline, oxytetracycline, and chloramphenicol in vannamei shrimp in intensive ponds in Kalipuro, Banyuwangi, East Java, and to determine the safety and feasibility of vannamei shrimp products consumed by the public based on the EDI (Estimated Daily Intake) and HQ (Hazzard) indexes. Quotients). The research was conducted using HPLC (High-Performance Liquid Chromatography) and ELISA (Enzyme-Linked Immunosorbent) methods to analyse the antibiotic content of vannamei shrimp then, the data obtained were processed using the EDI (Estimate Daily Intake) method to obtain daily estimates and HQ (Hazzard Quotient). to carry out an antibiotic risk assessment in the human body. The results obtained from this study were that vannamei shrimp in intensive ponds in Kalipuro, Banyuwangi, and East Java were negative for tetracycline, oxytetracycline, and chloramphenicol antibiotics. So the conclusion obtained from this study is that vannamei shrimp from intensive ponds in Kalipuro, Banyuwangi, and East Java is safe for consumption because it is negative for antibiotic residues.

Abstract

This study aims to determine the genetic character of cyprinid fish (Barbonymus sp.) using the mtDNA gene in the COI and Cyt-b gene regions. Samples were collected from the various areas in East Java Province, including Gresik, Mojokerto, Madiun, Malang, and Banyuwangi. The caudal peduncle of the cyprinid fish was used for molecular identification. Isolation and purification of its genomic DNA using the gSYNCTM DNA Extraction kit. We used a polymerase chain reaction machine for template DNA amplification with COI primers, including FishF1 and FishR1, while Cyt-b primers, including GluDG-L and CB2-H. The results showed that all cyprinid fish sequences were successfully amplified with the COI gene of 648 bp and the Cyt b gene of 408 bp. The results of the BLASTN analysis show that there are two species of cyprinid fish that have been found, namely Barbonymus gonionotus and Barbonymus schwanenfeldii, with percentage identities ranging from 99-100%. The species Barbonymus gonionotus was found in five regions (Gresik, Mojokerto, Madiun, Malang, and Banyuwangi), while Barbonymus schwanenfeldii was only found in Malang. The genetic distance value based on the COI gene ranged from 0.00000-0.10694, while the genetic distance value based on the Cyt-b gene ranged from 0.00000-1.15662. Conservation status based on IUCN Red List data, the species Barbonymus gonionotus and Barbonymus schwanenfeldii are included in the Least Concern (LC) category, and these species, according to CITES data, have not been evaluated or Not Evaluated.

Abstract

Polyethylene terephthalate (PET) adalah salah satu polimer termoplastik paling banyak tersedia di pasar. PET digunakan untuk produksi botol atau wadah untuk minuman, seperti air, minuman ringan berkarbonasi, minuman jus, dan industri makanan. Polyethylene terephthalate (PET) ditemukan melimpah pada sedimen ekosistem mangrove di Pasir Putih Wonorejo pada musim hujan sebanyak 59%. Studi terbaru telah menunjukkan bahwa bakteri indigenous sedimen mangrove mampu mendegradasi mikroplastik. Sehingga, tujuan penelitian ini adalah menemukan potensi agen biodegradasi mikroplastik dari bakteri indigenous sedimen ekosistem mangrove di Pasir Putih Wonorejo. Bakteri diisolasi pada April 2022 dari sedimen ekosistem mangrove. Metode yang digunakan diawali dengan isolasi bakteri dari sedimen ekosistem mangrove Pasir Putih Wonorejo, kemudian digunakan untuk uji degradasi mikroplastik polyethylene terephthalate dan mengetahui jenis bakteri yang memiliki kemampuan mendegradasi mikroplastik. Hasil isolat bakteri yang didapat sebanyak 16 bakteri. Potensi bakteri yang menjadi agen biodegradasi dengan penurunan berat mikroplastik isolat bakteri SPP.1.1.3. sebesar 8,73%. Identifikasi molekuler dengan cara ekstraksi DNA 16S rRNA menunjukkan jenis Pseudoaltoromonas caenipelagi dengan homologi 99% pada database National Center Biotechnology Information. Hasil ini membuktikan bahwa bakteri indigenous memiliki kemampuan sebagai agen biodegradasi mikroplastik. Polyethylene terephthalate (PET) is one of the most widely available thermoplastic polymers on the market. PET is used for the production of bottles or containers for beverages, such as water, carbonated soft drinks, juices, and the food industry. Polyethylene terephthalate (PET) is found abundantly in mangrove ecosystem sediments in Pasir Putih Wonorejo during the rainy season as much as 59%. Recent studies have shown that indigenous mangrove sediment bacteria are able to degrade microplastics. Thus, the purpose of this study was to find potential agents for biodegradation of microplastics from indigenous bacteria in mangrove ecosystem sediments Pasir Putih Wonorejo. Bacteria were isolated in April 2022 from mangrove ecosystem sediments. The method used begins with The isolation of bacteria from mangrove ecosystems sediments from Pasir Putih Wonorejo, then were used to assay biodegradation of polyethylene terephthalate microplastics and to determine the types of bacteria that has ability to degrade microplastics. The results of isolated bacterial obtained were 16 bacterias. The potential bacteria that becoming agents of biodegradation by decreasing the weight of microplastic isolate SPP.1.1.3. by 8,73%. Molecular identification by extraction of DNA 16S rRNA showed the type of Pseudoaltoromonas caenipelagi with 99% homology in the National Center Biotechnology Information database. These results prove that indigenous bacteria have the ability as microplastic biodegradation agents.

Abstract

A single specimen of saltwater crocodile Crocodylus porosus Schneider, 1801 (1.50 cm in length) was captured and photographed by a local fisherman by gill net at fishpond around mangrove ecosystem of east coast Surabaya on December 2020. The location where the estuarine crocodile caught is an important wetland area in Surabaya with an excellent mangrove cover. Wonorejo mangrove location is about 10 km from the centre of Surabaya, the second-largest city in Indonesia. This finding is considered as a first reported of C. porosus in Surabaya, and at the same time is proof of the importance of the mangrove area on the east coast of Surabaya as a habitat for wildlife that must be conserved appropriately. Routine monitoring of mangrove areas, including their wildlife, is essential so that they are not displaced by the development of the city which changes the land function to become a residential area.

Abstract

Mahseer (genus Tor dan Neolissochilus) adalah ikan dari famili Cyprinidae yang tersebar luas di Asia Selatan hingga Indonesia bagian barat. Saat ini keberadaan ikan tersebut di alam terancam akibat degradasi lingkungan, salah satunya di Jawa Timur, Indonesia. Berkurangnya jenis ikan tersebut mengakibatkan kepunahan spesies di masa yang akan datang dan terbatasnya peran spesies pada ekosistem karena kehilangan keragaman genetik. Keragaman genetik ini dapat diketahui dengan adanya karakteristik morfologi untuk mengetahui status genetik dari suatu ikan. Tujuan dari penelitian ini adalah untuk mengidentifikasi keanekaragaman jenis mahseer berdasarkan karakter morfologi di beberapa lokasi sungai di Jawa Timur. Sampel ikan mahseer diperoleh dari Kabupaten Pasuruan (DAS Banyubiru dan Umbulan), Kabupaten Tulungagung (DAS Song), dan Kabupaten Karanganyar (DAS Senatah). Identifikasi morfologi dilakukan berdasarkan panduan identifikasi. Karakteristik yang diamati adalah morfologi, meristik, dan fitur tambahan (matriks pola warna). Hasil kajian morfologi menunjukkan bahwa terdapat 2 jenis mahseer di Jawa Timur, yaitu Tor tambra dan Tor tambroides.
