Abstract

Ikan baronang (Siganus sp.) adalah salah satu jenis ikan yang potensial digunakan sebagai bahan baku surimi. Surimi kering adalah salah satu bentuk surimi yang telah mengalami proses pengeringan dan memiliki banyak keuntungan. Tujuan dari penelitian ini yaitu menentukan frekuensi pencucian dan suhu pengeringan terbaik dalam pembuatan surimi kering. Penelitian ini terdiri dari tiga tahap yaitu preparasi bahan baku, pembuatan surimi basah dan pembuatan surimi kering dengan ulangan sebanyak tiga kali. Pencucian dengan dua kali dipilih sebagai perlakuan terbaik dengan derajat putih 50,61%, protein larut garam 5,27%, pH 7,07 dan kekuatan gel 1.387,4 g.cm. Suhu pengeringan 60oC adalah suhu pengeringan terbaik dengan derajat putih 61,93%, daya ikat air 8,53 mL/g, protein larut garam 1,96%, densitas 0,43 mL/g, kapasitas rehidrasi 2,79 mL/g dan stabilitas emulsi 61,09%.

Abstract

Ikan Sidat merupakan komoditas hasil perikanan penting dengan nilai ekonomis tinggi. DNA barcoding telah cukup banyak digunakan sebagai metode berbasis penentuan urutan nukelotida untuk mendeteksi jenis spesies pada ikan. Proses pengolahan dengan suhu tinggi dapat menyulitkan proses identifikasi berbasis penentuan urutan nukelotida. Tujuan penelitian ini adalah untuk mengidentifikasi spesies berbagai produk olahan ikan sidat menggunakan marka molekuler mini-COI barcodes. Pengolahan suhu tinggi yang digunakan pada penelitian ini adalah: panggang, penggorengan dalam minyak sayur, panggang, pengasapan panas dan pengolahan suhu sterilisasi. Seluruh sampel segar dapat teridentifikasi sebagai spesies Anguilla bicolor bicolor (99-100%) dan Anguilla marmorata (100%). Sampel olahan sidat berhasil diamplifikasi dengan marka mini-COI barcodes, sehingga dapat digunakan untuk aplikasi pengawasan label produk seafood secara rutin.

Abstract

Fish gastric is a by-product of fishery waste that has the potential to be developed as a source of pepsin enzyme, such as tuna gastric. The purpose of this study was to extract pepsin from tuna gastric and characterize the enzyme pepsin after dialysis step. The extraction process of pepsin carried out by separating the gastric wall fluid by adding Tris-HCl buffer pH 7.5, then proceed by ammonium sulphate (NH4)2SO4 fractional precipitation from 20% to 80% followed by dialysis. The results showed that the crude extract of the pepsin enzyme had a specific activity of 0.251 mg/mL. Pepsin extract from precipitation with 30-40% fraction had 4,274 U/mg activity and after dialysis, pepsin had 5,137 U / mg activity. The pepsin obtained from gastric tuna had a working temperature in the range of 20-60°C and the working pH is in the pH range 2-3.5. The metal ion, namely FeCl3 and ZnCl2 increased the activity of the pepsin by 2.97 times and 1.92 times.

Abstract

Eel (Anguilla sp.) is a highly nutritious Indonesian marine commodity. However, information on the eel nutrient content in Indonesia is still limited. This study was aimed to determine chemical properties of Indonesian eels,Anguilla bicolor bicolor and A. marmorata. Proximate, amino acid, fatty acid, and vitamin A analysis were carried out on the fresh eel. The A. bicolor bicolor  ontained 65.51% of moisture, 1.34% of ash, 16.78% of protein, 13.26% of fat, and 3.1% of carbohydrate. Meanwhile, results on A. marmorata showed it contained 57.17% of moisture, 1.09% of ash, 17.30% of protein, 21.35% of fat, and 3.12% of carbohydrate. Leucine and glutamic acid represented the highest essential and non-essential amino acid of both A. bicolor bicolor and A. marmorata, respectively. The highest saturated and monounsaturated fatty acid were palmitate and oleic acid for A. bicolor bicolor as well as A. marmorata. The content of vitamins in A. marmorata was higher than in A. bicolor bicolor.

Abstract

Hairtail fishery is an important fishery commodity with high economic value as export commodity. Authentication based on DNA mini-barcodes can be applied to overcome seafood fraud and to support Indonesian traceability system. This study was aimed to design species-specific primers of DNA minibarcodes and authentication of various processed hairtail fish products such as smoked, baked, fried, canned, and boiled. Analytical method applied PCR-based of cytochrome c oxidase 1 (COI) full length andmini-barcodes. Mini-barcode primer specific for hairtail (Trichiurus lepturus) successfully amplified DNA of processed hairtail fish. The BLAST analysis showed the samples were identified as Trichiurus sp. and T. lepturus with homology level of 91% to 100%. Phylogenetic analysis showed the processed sample (LC dan LD) were in the same group with T. lepturus.

Abstract

Low temperatures could inhibit bacteria producing histamine during fish handling process. Longtail tuna (Thunnus tonggol) is kept in ice starting from catching process until delivering to retail market to maintain quality of fish. However, long period of this storage causes growth of bacteria and deformation of histidine into histamine. This study was aimed to determine the effect of different storage conditions to the quality of longtail tuna through sensory and histamine test. Samples of longtail tuna were kept for 18 days in room temperature storage (28±2°C), cold temperature (0±3°C) and freezing temperature (-10±4°C). The quality of tuna was evaluated using Quality Index Method (QIM). The formation of histamine was analysed on day 0, 1, 2, 3, 4, 8, 14, 18 using spectrofluorometry method. Sensory acceptability limit at room temperature was one day, while cold storage reached 10 days with QIM score 11. Longtail tuna was spoiled on day 2 at room temperature with QIM score >12 and concentration of histamin at 1713.88 mg/kg. The results of histamine in frozen and cold storage on the 18th day were 59.99 mg/kg and 77.98 mg/kg, these results exceeded the FDA limit (50 mg/kg) but still below the limit of FAO (100 mg/kg).The best storage condition is frozen storage, maintaining sensory acceptability and preventing histamine formation.

Abstract

Surimi-based processed products are prone to mislabeling using raw materials that are not in accordance with food safety requirements. There were cases reporting use of toxic fish tissue in commercial seafood products. This study was aimed to identify and determine the raw materials used in various processed surimi products using cytochrome oxidase I (COI) gene marker. The experiment consisted of DNA isolation, DNA amplification using several target primers namely full-length barcodes, mini-barcodes, as well as specific species primers against poisonous puffer fish Lagocephalus lunaris and genetic analysis. The results of bioinformatics analysis revealed that S1 samples were Coryphaena hippurus or mahi-mahi fish, S2 and S3 samples were Nemipterus bathybius or curated fish and CS samples were Evynnis cardinalis or kuro fish. Detection of samples with species specific primers of puffer fish Lagocephalus lunaris with annealing temperatures of 54°C, 57°C, and 60°C showed no DNA bands in the six samples analyzed.

Abstract

Amina biogenik merupakan komponen basa nitrogen yang terbentuk oleh dekarboksilasi asam amino. Amina biogenik yang sering terdeteksi pada produk perikanan di antaranya histamin, tiramin dan kadaverin. Gen pengkode dekarboksilasi asam amino tersebut yaitu histidine decarboxylase (hdc), tyrosine decarboxylase (tdc) serta lysine decarboxylase (ldc). Penelitian ini bertujuan untuk mendapatkan isolat DNA bakteri beserta konsentrasi dan kemurniannya, menyeleksi media pertumbuhan bakteri, mendeteksi gen hdc, tdc dan ldc menggunakan multiplex PCR serta identifikasi bakteri.. Sampel yang digunakan yaitu ikan tuna beku, tongkol beku, cakalang beku, tuna loin, pindang tongkol potong dan pindang tongkol bumbu kuning serta kontrol positif menggunakan ikan tuna yang disimpan pada suhu ruang selama 3 hari. Penelitian terdiri dari tahap pra-pengayaan bakteri pada media marine broth dan lactose broth, isolasi DNA sampel tanpa dan hasil pra-pengayaan, uji kemurnian dan konsentrasi isolat serta amplifikasi gen target hdc, tdc, ldc menggunakan multiplex PCR. Hasil yang diperoleh yaitu kualitas isolat DNA bakteri pada umumnya sudah murni, dengan konsentrasi isolat 15,75-157,84 ng/µL. Teknik multiplex PCR berhasil mendeteksi gen hdc, tdc dan ldc pada ikan scombridae. Kondisi optimum amplifikasi PCR yaitu annealing pada suhu 50°C selama 90 detik. Gen yang terdeteksi pada sampel tanpa pra-pengayaan yaitu tdc, sedangkan ldc dan hdc ditemukan pada sampel hasil pra-pengayaan media lactose broth dan marine broth. Bakteri pembentuk amina biogenik teridentifikasi sebagai Enterococcus fecalis, M. morganii, E. aerogenes, Citrobacter sp., Hafnia paralvei dan Obesumbacterium proteus. Abstract

Siput laut gonggong asal Bintan merupakan salah satu gastropoda laut yang belum dimanfaatkan secara optimal. Siput ini merupakan makanan laut khas Bintan dan harganya sangat mahal. Secara empiris, siput ini dipercaya dapat meningkatkan stamina dan vitalitas. Tujuan penelitian ini adalah untuk melakukan penapisan senyawa bioaktif pada siput laut gonggong asal Bintan yaitu aktivitas antioksidan dan aktivitas antimikroba. Aktivitas antioksidan dianalisis menggunakan metode DPPH dan aktivitas antimikroba dianalisis menggunakan metode difusi sumur. Aktivitas antioksidan ekstrak gonggong bercangkang tipis dengan IC50 1.433,08±0,01 ppm lebih tinggi daripada gonggong bercangkang tebal dengan IC50= 2.051,55±0,10 ppm. Aktivitas antimikroba pada ekstrak gonggong lebih baik pada bakteri Gram positif daripada bakteri Gram negatif. Ekstrak gonggong rebus bercangkang tebal memiliki aktivitas antimikroba yang paling tinggi dengan nilai rata-rata diameter daya hambat (DDH) yaitu 25,53±0,12 mm.

Abstract

Seafood product is an important commodity in the global trade market, however, seafood fraud has been increasingly reported. Therefore, a method to specifically identify species of seafood products is needed. DNA methods gain many interests for detection of seafood fraud due to its robustness. This study was aimed to determine the effectiveness of several methods for isolation of DNA from fresh and processed seafood. The DNA isolation methods used in this study were the CTAB DNAzol, TianGen, Qiagen, and Fasmac.. DNA concentration and purity were determinedusing nanodrop spectrofotometry and electrophoresis. DNA isolation showed that the DNA concentration of fresh seafood products was higher than the processed seafood products. DNA isolation using CTAB method have higher levels of DNA purity (1.60-2.00) than using commercial kits in all samples. The commercial kits offer a simple procedure and minimize the possibility of environmental contaminants.
