Abstract

Padina, one of the brown algae, grows in many tropical waters including in Lampung. Padina contains fucoidan, a sulfated polysaccharide, which has been shown to have a wide range of bioactivity. This study aims to determine the effectiveness of fucoidan Padina sp. from Lampung waters to enhance the non-specific immune response in Pacific white shrimp. Padina sp. was collected from Ketapang Beach, Pesawaran, Lampung then the Padina’s fucoidan was acidly extracted by previously method. Fucoidan then supplemented to 10 vananmei shrimp (size 10 g) each container (45 L) for 14 days. The study was conducted with a complete randomized design trial (CRD) consisting of 3 treatments and 4 repeats including fucoidan supplementation at a dose of 0 mg/kg (control); 750 mg/kg; and 1500 mg/kg of feed. Every day, shrimp were fed as much as 3% of biomass with a frequency of 4 times. Four nonspecific immune response of shrimp including total hemocyte count (THC), phagocytosis activity (PA), phagocytosis index (PI), and total plasma protein (TPP) were mesured on day 0; 7; and 14 after fucoidan supplementation. The results showed the administration of fucoidan extract Padina sp. gives significantly different results to THC and TPP enhancements. Supplementation treatment of 750 mg/kg fucoidan Padina is the best treatment to increase nonspecific immune response in Pacific white shrimp.

Abstract

Minyak bumi pada proses distilasi dipisahkan sesuai fraksi menurut titik didihnya, dengan adanya kondisi vakum maka akan menghasilkan parameter fisis yang dituju, sehingga memenuhi parameter produk yang diinginkan. Sistem produksi uap merupakan pengendalian cascade atau bertingkat, terdapat dua paramater pengendalian yang dikendalikan yaitu flow dan level. Analisa kinerja sistem produksi uap dengan prinsip two phase separator ini dilakukan dengan membuat model matematis sehingga mendekati keadaan aslinya menggunakan separator geometry dan perangkat lunak Matlab R2018b, maka diperlukan pengendali untuk menganalisanya, metode pengendali yang digunakan adalah metode Ziegler-Nichols Critical Gain dan Critical Period. dan tuning untuk menentukan gain parameter pengendalinya (Kp, Ki, Kd). Berdasarkan hasil pengujian, tipe pengendali dengan hasil terbaik adalah pengendali PID untuk pengendali flow dengan parameter Kp = 0.12, Ti = 0.28, Td = 0.4, dengan performansi Rise Time = 1.97, Settling Time = 7.17, Overshoot (%) = 9.01, dan Hasil terbaik untuk pengendali level adalah pengendali P dengan parameter Kp = 0.06, dengan performansi Rise Time = 3.16, Settling Time = 7.71, Overshoot (%) = 2.45.

Abstract

Kedensari Usaha Mikro, Kecil dan Menengah (UMKM) adalah UMKM yang bergerak di bidang produksi tas dan dompet. Namun, permasalahan yang terjadi pada UMKM Kedensari adalah sulitnya menetapkan harga pesanan yang mengakibatkan banyak konsumen yang membatalkan, hal ini dikarenakan tingginya harga yang diinginkan pelanggan. Sehingga perlu adanya evaluasi dan perbaikan dengan menggunakan metode full costing atas biaya yang di kenakan perusahaan yang kompetitif. Berdasarkan hasil penelitian didapatkan pada produk tas dengan total harga pokok pesanan sebesar Rp. 48.168.530,00 dan jumlah harga jual sebesar Rp. 72.000.000,00 diperoleh laba sebesar Rp. 23.831.470,00. Kemudian pada produk dompet jumlah harga pokok pesanan sebesar Rp. 23.286.030,00 dan jumlah harga jual sebesar Rp. 36.000.000,00 diperoleh laba sebesar Rp. 12.713.970,00. Sedangkan total laba yang diperoleh pada bulan November 2023 sebesar Rp. 36.545.440,00. Sehingga penggunaan metode full costing sangat cocok untuk meningkatkan efisiensi operasional dan meningkatkan daya saing di pasar.

Abstract

The object’s volume is a consideration in determining the quantity of products such as eggs, fruit, piles of rice, or sand. This research aims to obtain a system for determining the volume of a symmetrical object using the linear regression method in real-time, faster, more effective, and more enjoyable. This research uses segmentation methods and linear regression to determine the volume of a symmetrical object. The objects are a pile of rice and eggs which have symmetrical shapes. The shape of the symmetry in each object is a cone for a pile of rice and an oval for an egg. The results of this research are a system of symmetrical object volume determination using the linear regression method with an accuracy score of 96.48% for piles of rice and 97.84% for an egg. This system has limitations, there are the volume value must be in the data range that has been trained and the camera phone must be the same.

Optimum temperature, pH, and salinity can suppress the activity of the fucoidanase enzyme through the inhibition test. 4. Cytobacillus kochii GSD can produce fucoidanase enzyme with low molecular weight.     Abstract Extensive research has shown that low molecular weight fucoidan exhibits significantly greater biological activity than its high molecular weight. C. kochi GSD, a Sargassum symbiont bacterium, is proven to have the activity of hydrolyze fucoidan. This study proposes the growth optimization and fucoidanase enzymatic activity of C. kochii GSD bacteria under varying environmental conditions (temperature, pH, and salinity) cultured in basic liquid medium (BLM) for 48 hours. Based on Response Surface Methodology (RSM), the range of temperature, pH, and salinity for the growth optimization test of C. kochi GSD bacteria were 26.591, 30, 35, 40, and 43.49oC, the pH used starts from 3.636, 5, 7, 9, and 10.363, while the salinity to be used starts from 3.522, 5, 17.5, 30, and 38.522 ppt, respectively. The best conditions for growth of each environment were then continued with the test of fucoidanase enzyme activity in vitro. The results showed that C. kochii GSD bacteria grew optimally at temperature, pH, and salinity of 35oC, 7, and 30 ppt, respectively. The optimum enzyme activity of C. kochii GSD is at 72 hours with the forming of clear zones on media containing fucoidan and given Cetylpyridinium chloride (CPC) solution with clear zone diameters of 31.17 mm (temperature), 25.9 mm (pH), and 20.97 mm (salinity), respectively. The conclusion of this study is a high novelty finding to produce low molecular weight fucoidan enzymatically with C. kochii GSD bacteria to be used as an immunostimulant.

Abstract

Pada suatu perencanaan proyek konstruksi terdapat berbagai metode perhitungan yang dapat digunakan sesuai dengan pengaplikasian metode masing-masing untuk dapat memperoleh hasil yang paling efisien dalam berbagai hal dan memenuhi standar keamanaan suatu bangunan. Dalam perencanaan suatu bangunan terdapat berbagai metode perhitungan, dintaranya yaitu metode ultimit atau  Load and Resistance Factor Design (LRFD) dan metode tegangan ijin atau Allowable Stress Design (ASD). Dalam penelitian ini kedua metode perhitungan tersebut digunakan untuk menghitung demand/capacity ratio dan berat bangunan. Metode pada penelitian ini menggunakan metode kuantitatif. Penelitian ini bertujuan untuk mengetahui perbandingan nilai berat struktur baja dan demand/capacity ratio pada struktur baja menggunakan software ETABS 2019 dengan metode tegangan ijin dan metode ultimit. Berdasarkan hasil penelitian, diperoleh nilai rerata demand / capacity ratio balok pada seluruh story dengan menggunakan metode ultimit sebesar 0,6631 dan pada metode tegangan ijin sebesar 0,7520. Nilai rerata demand / capacity ratio kolom pada seluruh story menggunakan metode ultimit sebesar 0,4477 dan tegangan ijin 0,5783. Berat bangunan dengan metode ultimit 858684 kg dan berat bangunan dengan metode tegangan ijin sebesar 861326,5 kg. Berdasar hasil perbandingan demand/capacity ratio pada kolom, balok dan berat bangunan antar metode diperoleh kenaikan demand/capacity ratio sebesar 29% pada kolom, 13,4% pada balok dan kenaikan berat 0,3%  atau 2642,56 kg ketika menggunakan metode tegangan ijin

Abstract

Pembangkit Listrik Tenaga Surya (PLTS) adalah solusi alternatif untuk memenuhi kebutuhan energi listrik yang semakin meningkat. Penelitian ini berfokus pada analisis sistem PLTS On Grid yang terpasang di Gedung Mal QBIG BSD City. Tujuan dari penelitian ini adalah untuk menganalisis kinerja PLTS dalam memenuhi kebutuhan energi listrik gedung tersebut serta mengidentifikasi kapasitas dan kebutuhan beban yang diperlukan. Penelitian ini dimulai dengan pengumpulan data mengenai komponen utama dan konfigurasi sistem PLTS yang digunakan, diikuti dengan perhitungan kebutuhan beban listrik harian. Analisis dilakukan untuk menentukan kapasitas modul surya, inverter, dan komponen lainnya yang sesuai untuk mendukung operasional gedung. Hasil analisis menunjukkan bahwa sistem PLTS di Mall QBIG BSD City mampu menghasilkan energi listrik sebesar 2.2095 MWh per tahun, yang berkontribusi signifikan dalam memenuhi kebutuhan listrik Gedung. Manfaat dari penelitian ini adalah memberikan gambaran umum tentang proses dan kinerja sistem PLTS On Grid, serta dapat dijadikan referensi untuk pengembangan dan peningkatan sistem energi terbarukan di gedung-gedung komersial lainnya. Penelitian ini juga memberikan saran untuk optimalisasi dan perawatan sistem PLTS agar dapat beroperasi dengan efisien dan berkelanjutan.

Abstract

Seaweed Kaphapicus alvarezii is very popular in cultivation because it has high economic value and has quite large development potential in Ruguk Village, Ketapang District, South Lampung Regency. Water quality is a factor that influences the growth of seaweed, including the Kaphapicus alvarezii type, both physical and chemical water quality. This research aims to determine the condition of water quality in the seaweed cultivation area of Ruguk Village, Ketapang District, South Lampung Regency, meets the optimal living conditions for cultivating Kaphapicus alvarezii seaweed. This research is a type of descriptive research with a survey method carried out by taking water samples at seaweed cultivation locations and measuring the increase in seaweed growth every week for 4 weeks. The parameters observed in this research were the physical and chemical parameters of the water quality of the seaweed rearing media at the research location which consisted of temperature, brightness, current, pH, salinity, dissolved oxygen (DO), and phosphate and relative growth of the seaweed. Based on the results of observations during the research, it can be concluded that the water quality conditions, both physical and chemical water quality in the seaweed cultivation area in Ruguk Village, Ketapang District, South Lampung Regency, are in accordance with the standard quality requirements for the growth of Kaphapicus alvarezii seaweed, namely temperature 29.1-30.2⁰C, brightness 0,8-1.5 m, current strength 0.20-0.37m/sec, depth 0.8–1.5 m, pH 7.5-8.17, salinity 26-30 ppt, DO of 4.6-7.2 ppm, nitrate 0.2-0.92 mg/L and phosphate of 0.258-0.711 mg /L

Abstract

One of major problem of siamse catfish (Pangasianodon hypophthalmus) culture is Edwardsiliosis bacterial disease of Edwardsiella ictaluri, causes economic losses because it causes high mortality and reduces the quality of fishery products. Currently, the use of antibiotics as an effort to control the disease has been prohibited due to the impact of residual hazards, resistance and decreased food safety value. Therefore we need an alternative solution that is effective, efficient and applicable through the vaccines application. The aim of this research was to analyze the effectiveness of the Whole cell Edwardsiella ictaluri vaccine with different doses and different application methods to protect Siamese catfish (Pangasianodon hypophthalmus) from Edwardsiellosis disease. The research was conducted using a completely randomized design (CRD) method consisting of 3 methods, namely oral vaccine, immersion vaccine, and injection vaccine. Each method consisted of 4 different dose treatments with 3 replications. The results showed that the vaccine application using different methods resulted in different survival rates, relative survival rates, specific growth rates and mean time to death. The oral vaccine method using each different dose resulted in the best survival rate, relative survival percentage, and mean time to death. The immersion vaccine method with a dose of 10.9 resulted in the highest specific growth rate.

Abstract

Pada industri tekstil proses pemintalan benang dapat terjadi bercampurnya dua jenis benang dalam satu gulungan (cone). Umumnya memakai mika acrylic, namun mudah pecah, impor dan harga relatif mahal, maka diganti menggunakan triplek, tetapi cepat rusak (tidak ekonomis). Pemanfaatan fly ash sangat terbatas, maka perlu kajian potensi produk baru komposit.Tujuan penelitian mengetahui kekuatan mekanik komposit cerdas nano fly ash-epoksi sebagai bahan panel magazine mesin winding (penggulung benang). Bahan penelitian fly ash mesh 120, di-oven 110 ºC selama 45 menit (kadar air) 4%. Pencampuran fly ash-epoksi diaduk 6 menit ditambah katalis 1% diaduk lagi 2 menit dituang dicetakan. Hasil penelitian sifat mekanik tertinggi pada fraksi berat 30:70, ketangguhan impak 2,75 KJ/cm² dan kekuatan bending 48,03 MPa. Pengamatan foto SEM patahan impak, menunjukkan ikatan yang baik antara filler dan matrik dibanding variasi lainnya, adanya rongga/pori-pori ukuran kecil relatif sedikit, penyebaran matrik dan filler berlangsung lebih merata, berdampak peluang terjadinya inisiasi patah lebih kecil sehingga nilai ketangguhan impak tinggi. Hasil pengujian ketangguhan impak komposit fly ash-epoksi fraksi berat 30:70 lebih tinggi, dibanding bahan mika akrilik dan bahan triplek, sehingga disimpulkan komposit fly ash-epoksi sangat berpotensi dipakai sebagai panel/sekat magazine mesin winding, pengganti material sebelumnya.
