Abstract

Sorghum is a type of cereal plant that easily adapts to stressful environmental conditions. Sorghum has many benefits as it can be used as food, feed, and energy sources, making it a promising crop for development on dry land in Indonesia. Drought impacts plants during their growth process, causing physiological and biological changes. This study aims to determine the effect of drought stress on the morphology and chlorophyll content of sorghum plants. Preliminary research was conducted by planting three varieties of sorghum (Numbu, Super 1, and Super 2). Subsequent planting involved selecting the variety most resistant to drought based on data analysis of several parameters. The basic design used in this experiment was a completely randomized design (CRD) with one factor: the level of drought stress, which had four levels -no drought stress as a day 0 (control) and drought stress for 3, 6 and 9 days by withholding water. The results showed a decrease in the total chlorophyll content of the leaves over time. Drought stress for 6 and 9 days without watering resulted in water content dropping below 25%, indicating severe drought stress and disrupting plant growth. Severe drought stress was characterized by low soil and leaf water content, yellowing and curling of the leaves.

Abstract

SCMV (Sugarcane Mosaik Virus) is sugarcane crop disease resulting in chlorosis in leaves with the formation of the colors yellow and green intermittents. Based on survey information obtained that the air observation of all varieties of sugarcane was already stricken with the virus SCMV. Even the most formidable attack is on PS881 varieties with the intensity of the attacks reached 80%, so that it is estimated will lose up to 40% of the harvest. The sugarcane is virus free can be obtained via organogenesis in tissue culture method directly on the apical meristem, somatic embryogenesis at, and also with the addition of khemoterapeutan (acyclovir).The workings of the khemoterapi materials are the chemotherapi will interfere with replication and synthesis of genetic material of the virus but also cause the same effect against the mechanism of synthesis of nuklet acid on plants hosts. This research aims to find the best concentration and exposure time the most good in eliminating viruses SCMV in the apical bud culture of sugarcane PS881, using the antiviral acyclovir in conditions of invitro, so that the resulting plant will be virus free. The methods used to detect the presence of the virus by using the two ways, the first is by serology test through the protein content of the virus / checking nucleic acid virus with ELISA and the second is by RT-PCR. The results of the analysis showed that the interaction between the concentration of chemotherapy and exposure time produces the best treatment in eliminating the virus was . treatment with acyclovirc oncentration of 20 ppm and exposure time of 5 weeks.

Abstract

Wee1 is a gene encoding for protein kinase that is located in the nucleus and it plays an essential role in determining the timing of mitosis. Overexpression of Wee1 in rice is resulting in increased plant size. However, the increasing plant size due to the increase of Wee1 protein expression is not elucidated. Immunodiagnostic using specific antibodies against Wee1 protein should be conducted to determine the role of the protein on cell division. This experiment was directed to construct Wee1 in an expression vector for synthesis of recombinant Wee1 protein using three strategies of construction. The three strategies were construction of full length (FL), deletion of putative binding site (BS), and deletion of N-terminal domain (ΔN) of Wee1. Restriction analysis with BamH1/Sal1 of FL-Wee1, BS-Wee1, and ΔN-Wee1 construct resulted in the different DNA fragments with molecular size at 1239, 1176, and 960bp, respectively. This result indicated that the Wee1 fragments have been successfully inserted in the expression plasmid which was further confirmed using DNA sequencing. Colony PCR analysis showed that Escherichia coli strain BL21 (DE3) has been transformed with the constructs. The protein analysis using SDS-PAGE revealed that the recombinant protein of Wee1 synthesized in the E.coli containing ΔN-Wee1 construct, but not in FL-Wee1 and BS-Wee1 constructs. The ΔN-Wee1 protein was synthesized in an insoluble fraction with a molecular size of 38.8 kDa which is the same as the size estimated using the software ExPASy. Interestingly, the level of synthesized ΔN-Wee1 protein was not induced by IPTG concentration. Collectively, the results indicated that the DNA construct of ΔN-Wee1 is suitable for recombinant protein production.

Abstract

Indica rice variety Ciherang is the most planted variety in Indonesia, but the micropropagation technique is restricted because it is known has low regeneration frequency and included as recalcitrant cultivar for tissue culture and tranformation activities. One of solution to resolve that problem is developing a technique of somatic embryogenesis in callus of ciherang rice cultivar. The aims of study were to determine medium composition for inducting embryogenic callus in Ciherang rice and to know the effectivity of rice regeneration using callus as explant. The methods were included induction of embryogenic callus in callus induction media (CIM) containing MS basal, Proline 600 mg/l, Casein Hidrolisat 300 mg/l, phytagel 2,5 g/l, BAP 0.25 mg/l, sukrosa 30 %, and 2,4-D in different concentration, from 2,4-D 0 mg/l as control (CIM 1), 2,4-D 2 mg/l (CIM 2), 2,4-D 3 mg/l (CIM 3), and 2,4-D 4 mg/l (CIM 4). It then be continued to regenerate the calli in RM 1 medium containing MS basal, NAA 0,2 mg/l, Kinetin 2 mg/l, Agarose 10 g/l, and sukrosa 30 %, pH 5,8 for six days in dark and RM2 medium containing MS basal , NAA 0,2 mg/l, Kinetin 2 mg/l, Agarose 8 g/l, sukrosa 30 %, pH 5,8 in light room. Results showed CIM 4 medium using 2,4-D 4 mg/l gave optimum result in calli induction with procentage 57,63% and CIM 3 using 2,4-D 3 mg/l gave optimum result in embryonic calli induction with procentage 53,63%. Result of embryonic calli gave regeneration frequency procentage is 9,6%. The total planlet obtained after five weeks old in regeneration medum were 11 planlets ready for aclimatization.
