Abstract

The use of salicylic acid as resistance inducers agents in several plants species was well known. Salicylic acid has been believed to play an important role in inducing resistance against some pathogen. This research was aimed to study the role and effectiveness of salicylic acid as inducers for the resistance of some rice variety against bacterial leaf blight caused by Xanthomonas oryzae pv. oryzae. The experiment was arranged in completely randomized factorial design with three factors i.e. rice variety, salicylic acid, and pathotypes, each treatment was replicated three times. The results showed that salicylic acid could prolong the latent period of X. oryzae pv. oryzae pathotypes IV and VIII in paddy rice varieties Conde and Ciherang,  induce the resistance of rice varieties as well as reducing the rate of infection of the two mentioned pathotypes. Salicylic acid was also able to induce the resistance of Conde and Ciherang varieties in order to suppress area under disease progress curve (AUDPC)  of X. oryzae pv. oryzae pathotypes IV and VIII.

Abstract

ABSTRACTHybridization program was started in PKBT IPB Bogor in 2003, entangles of 12 parental cultivars, consisting of five type Smooth Cayenne cultivars and seven type Queen cultivars. The cross yielded 195 genotypes with various different character combinations. The result of cluster analysis based on morphological characters showed that there were 33 groups of hybrid at the degree of genetic similarity of 50%. The result of principal component analysis indicated that, between yield component characters and result most importantly, were fruit weight, fruit diameter and fruit length which were main supporting character of variance in hybrid result of the crosses. Descriptive results of fruit yield and quality characters showed three to five classes with the highest number of individuals around the mean value for each character. Fruit weight, crown weight, fruit length, fruit diameter, flesh thickness, core diameter, total soluble solid (TSS), total acid, vitamin C, pH, plant height, and peduncle length, were characters controlled by nuclear genes.Key words : hybridization, variability, Smooth Cayenne, Queen, genotype

Abstract

The F1 hybrid sterility in indica/japonica crosses is the major barrier in developing hybrid rice varieties between these two diverse germplasm. The sterility problem in japonica/indica hybrids can be overcome by using wide compatibility genes. The objective of this study was to identify wide compatibility varieties (WCVs) in some tropical japonica rice. Twenty five tropical japonica varieties as male parents were crossed with indica (IR64) and japonica (Akitakomachi) testers as female parents. The crosses were planted following a randomized complete block design with three replications. Varieties having average spikelet fertility of more than 70% with both the indica and japonica testers were rated as WCVs. Result from this study showed that six tropical japonica varieties were classified as WCVs, i.e., Cabacu, Grogol, Kencana Bali, Klemas, Lampung Lawer, and Napa. Hybrid sterility is caused by partial sterility of male and female gametes. The WCVs from the present study can be used in hybrid rice breeding program to solve hybrid sterility in indica/japonica hybrids.

Abstract

One of the important issues on rice breeding is to develop new rice lines suitable for nitrogen efficiency in the suboptimum environment. The study aimed to evaluate the tolerance and yield components of rice breeding lines selected under low and high nitrogen conditions. The F6 generation from two cross-combinations of Gampai/IR77674 and Progol/Asahan, were evaluated in the dry season of 2014 under N suboptimum and N optimum conditions. A total of 172 lines plus six check varieties derived from the low and high N environment selection were evaluated under low N rate (34.5 kg N ha-1) and high N rate (138 kg N ha-1), arranged in an augmented design. Six check varieties were relocated three times in each block. Plot size was 5.5 m2 and plant spacing 20 cm x 20 cm. Data were collected for grain yield and major yield components. Results showed that the different status of nitrogen fertilizer affected the number of productive fillers, number of filled grains, 100-grain weight, and grain yield. Different cross combinations exhibited different results in the progeny performance. Ten lines have a higher yield also tolerant to low N condition, i.e. B14250F-6-9, B14250F-1-4, B14250F-9-9, B14250F-6-4, B14250F-5-2, B14262F-15-6, B14250F-2-6, B14262F-12-4, B14250F-5-1, and B14250F-11-4. Thus, to obtain the N tolerant lines, selection at a low N environment was more effective compared with the optimum selection environment.

Abstract

Benih-benih kedelai dari varietas Argomulyo diiradiasi sinar gamma untuk meningkatkan keragaman genetik, memperbaiki morfologi tanaman dan nantinya untuk mendapatkan produksi hasil yang tinggi. Tujuan penelitian ini adalah untuk mengetahui radiosensitivitas tanaman kedelai [Glycine max (L) Merr ] varietas Argomulyo melalui irradiasi sinar gamma dan mengetahui respon pemberian tingkat irradiasi mikro sinar gamma terhadap pertumbuhan dan perkembangan dari benih kedelai turunan pertama (M ). Penelitian ini 1merupakan percobaan faktor tunggal dosis irradiasi yang terdiri dari delapan taraf, yaitu 0 Gy, 200 Gy, 400 Gy, 600 Gy, 800 Gy, dan 1000 Gy. Untuk mendapatkan nilai Lethal Dosis 50 (LD ), digunakan program Curve-fit 50Analysis. Untuk mengetahui respon pemberian tingkat irradiasi mikro sinar gamma, benih kedelai yang diuji adalah benih kedelai varietas Argomulyo yang diirradiasi dengan dosis rendah sinar gamma (micro mutation) dengan dosis sedikit dibawah LD yaitu 0 Gy, 50 Gy, 100 Gy, 150 Gy dan 200 Gy. Hasil penelitian menunjukkan 50 bahwa nilai Lethal Dosis 50 (LD ) terdapat pada dosis 457,178 Gy. Keragaman yang diperoleh dari tinggi 50 tanaman, jumlah cabang, jumlah polong, jumlah polong hampa dan jumlah biji pada generasi M mempengaruhi 1 pertumbuhan dan perkembangan tanaman baik secara kualitatif dan kuantitatif yang akhirnya akan mempengaruhi produksi tanaman.Kata kunci: irradiasi sinar gamma, radiosensitivitas, kedelai varietas Argomulyo

Abstract

The mutation could improve plant genetic variability. Some putative sugarcane mutants originating from the PS 881 variety have been produced through mutation induction using colchicine. The study aimed to determine the effect of colchicine induced on genome size and agronomical traits, and its correlation with sugarcane putative mutants production. The experiment was conducted at the UPBUP of IAARD, Bogor, July 2018-April 2019. The research was carried out in an augmented design in randomized complete block design, using 35 genotypes of the first generation G0 (30 putative mutants from colchicine 0.03 and 0.05%, five check varieties). The genome size, agronomical traits, i.e. stem number, length, diameter, and weight per meter; internode number and length; brix, and production, were evaluated. This result showed that colchicine increased 5.03-13.64% genome size of putative sugarcane mutants compared to the original variety PS 881. It is significantly different for almost all of agronomical traits. The genome size was significantly correlated very positively with brix and significantly positively with stem length and diameter, and production. Path analysis showed that stem (weight per meter and length) has a direct effect on production, in contrast, genome size, stem diameter, internode number, and brix have an indirect effect through stem weight per meter to production. Indirect selection to obtain high production can be done through stem (weight per meter and length), consider for genome size, stem diameter, internode number, and brix. This is  experimental preliminary information, validation on field is needed among direct and indirect of production components to production.Keywords: agronomical traits correlation, colchicine, path analysis, sugarcane production AbstrakPENGARUH KOLKISIN TERHADAP UKURAN GENOM DAN KARAKTER AGRONOMI SERTA HUBUNGANNYA DENGAN PRODUKSI MUTAN PUTATIF TEBUPeningkatan keragaman genetik dapat dilakukan melalui mutasi. Sejumlah mutan putatif tebu yang berasal dari varietas PS 881 telah dihasilkan melalui induksi mutasi menggunakan kolkisin. Penelitian bertujuan untuk mengetahui pengaruh kolkisin terhadap ukuran genom dan karakter agronomi, serta hubungannya dengan produksi mutan putatif tebu. Percobaan dilaksanakan di Unit Pengelola Benih Unggul Pertanian (UPBUP), Badan Litbang Pertanian, Bogor, Juli 2018-April 2019. Penelitian menggunakan rancangan augmented dalam rancangan acak kelompok lengkap, dengan 35 genotipe generasi awal G0 (30 mutan putatif tebu hasil perlakuan kolkisin 0,03 dan 0,05%, serta lima varietas pembanding). Karakter yang diamati adalah ukuran genom dan karakter agronomi (jumlah, panjang, diameter, dan bobot batang per meter; jumlah dan panjang ruas; brix; serta produksi). Hasil penelitian menunjukkan perlakuan kolkisin meningkatkan ukuran genom mutan putatif tebu 5,03-13,64% dibandingkan tetua PS 881 dan mengakibatkan perbedaan yang nyata pada hampir seluruh karakter agronomi. Ukuran genom berkorelasi positif sangat nyata dengan brix dan nyata dengan panjang dan diameter batang, serta produksi. Analisis lintas menunjukkan karakter bobot batang per meter dan panjang batang berpengaruh langsung pada produksi, sedangkan karakter ukuran genom, diameter batang, jumlah ruas, dan brix berpengaruh tak langsung pada produksi melalui bobot batang per meter. Seleksi tak langsung untuk mendapatkan produksi tinggi terutama melalui karakter bobot batang per meter dan panjang batang dengan mempertimbangkan karakter ukuran genom, diameter batang, jumlah ruas, dan brix. Hasil penelitian merupakan informasi awal yang memerlukan validasi hubungan pengaruh langsung dan tak langsung komponen produksi terhadap produksi di tingkat lapangan.Kata kunci : analisis lintas, kolkisin, korelasi karakter agronomi, produksi tebu

Abstract

ABSTRAKKlon kakao unggul berdaya hasil dan bermutu hasil tinggi sertaresisten terhadap penyakit utama perlu dikembangkan melalui pemuliaantanaman dan tersedianya informasi tentang parameter genetik diharapkandapat membantu memecahkan masalah tersebut. Pendugaan parametergenetik dapat dilakukan dengan analisis persilang dialel. Penelitianbertujuan untuk menduga parameter genetik ketahanan tanaman kakaoterhadap infeksi P. palmivora, menggunakan analisis persilangan setengahdialel. Bahan tanaman terdiri atas lima klon kakao (ICCRI 3, TSH 858,DR 1, ICS 13 dan Sca 6) yang tergolong rentan hingga tahan terhadapinfeksi P. palmivora yang digunakan sebagai tetua dan 10 galur hibrida F1hasil persilangan antar lima klon. Penelitian dilaksanakan di KebunPercobaan Kaliwining Pusat Penelitian Kopi dan Kakao Indonesia,Jember, Jawa Timur dari tahun 2008 hingga 2009. Untuk setiap kombinasipersilangan dievaluasi 20 bibit dan diulang tiga kali. Untuk mengetahuirespon bibit hibrida F1 terhadap infeksi P. palmivora, daunnya diinokulasidengan inokulum zoospora dan disungkup dengan plastik untuk menjagakelembapannya (>90%). Pengamatan luas bercak akibat infeksi P.palmivora dilakukan enam hari setelah inokulasi dan digunakan untukmenghitung intensitas penyakit. Hasil penelitian menunjukkan bahwakakao klon DR 1, ICS 13, dan ICCRI 3 mempunyai DGU yang palingtinggi dibandingkan dengan tetua lainnya. Selanjutnya, persilangan antarklon kakao DR 1 x ICS 13, dan TSH 858 x Sca 6 mempunyai DGKtertinggi sehingga kombinasi persilangan ini berpeluang untuk menjadipenghasil hibrida baru yang resisten terhadap P. palmivora. Kombinasipersilangan yang menunjukkan nilai heterosis tertinggi adalah DR1 x ICS13, DR1 x Sca 6, dan ICS 13 x Sca 6.Kata kunci: Heterosis, hibrida F1, DGU, DGK, intensitas penyakitABSTRACTEstimation of Heterosis and Combining Ability forResistance Against Black Pod Disease (Phytophthorapalmivora) in Cacao (Theobroma cacao L.)High yielding and disease resistance of cacao clone needs to bedeveloped through breeding program. Availability of genetic parametersfor various agronomic importance characters in cacao would be veryuseful and beneficial for cacao breeding activities. Estimation of variousgenetic parameters could be done by analyzing F1 arrays generated fromsemi-diallele crosses among a number of parents. The objectives of thisresearch were to estimate genetic parameters for resistance against P.palmivora infection in cacao using F1 arrays generated from semi-diallelecrosses among five cacao clones. Five cacao clones (DR 1, TSH 858, ICS13, ICCRI 3, and Sca 6), representing an arrays of clones with increasedresistance against P. palmivora infection, were used as parents to generate10 F1 hybrid arrays. This research was conducted at KaliwiningExperimental Station, Indonesian Coffee and Cacao Research Institute,Jember, Indonesia, during the period of 2008 to 2009. At most 20seedlings were evaluated for each F1 hybrid and the evaluation wasreplicated three times. To evaluate the response of the seedlings against P.palmivora infection, their leaves were inoculated with zoospore of P.palmivora. Relative humidity around inoculated leaves was maintained at>90% by wrapping them with plastic bag. The sizes of leaf necroseresponse due to P. palmivora infection were observed during six days afterinoculation and the disease intensity was calculated based on this recordedsymptoms. Results of the experiments indicated that cacao clones (DR 1,ICS 13, and ICCRI 3) were the highest in general combining ability(GCA) for resistance character than the other two clones. Moreover, F1hybrid originated from crosses between DR 1 x ICS 13, and TSH 858 xSca 6 were the highest in specific combining ability (SCA) for resistancecharacter. Therefore, this combination crosses might be used to developenew hybrid combinations resistance against P. palmivora infection.Combination crosses showing highest heterotic value for resistance againstP. palmivora infection were DR 1 x ICS 13, DR1 x Sca 6, and ICS 13 xSca 6.Key words : Heterotic effects, F1 hybrid array, GCA, SCA, diseaseintensity

Abstract

Percobaan untuk mengevaluasi efektivitas gen wide compatible dari enam genotipe padi telah dilaksanakan di kebun percobaan dan rumah kaca Muara, Bogor dari bulan April 2005 sampai Pebruari 2006. Percobaan ditata berdasarkan rancangan acak kelompok (RAK) yang diulang tiga kali. Untuk mengevaluasi efektivitas enam wide compatibility variety (WCV) dilakukan dengan menyilangkan antara enam WCV terdiri dari Cabacu, Napa, Kencana Bali,Lampung Lawer, Grogol dan Klemas disilangkan dengan 32 tester yang berasal dari subspesies dan ekotipe berbeda. Hasil pengujian menunjukkan bahwa genotipe Cabacu dan Grogol merupakan WCV yang efektif dalam meningkatkan pengisian gabah pada persilangan antar subspesies dan ekotipe berbeda.

Abstract

A study was conducted to study correlation between some characters of upland rice and path coefficient analysis, which will be helpful for further selection process to improve upland rice in acid soils. The results of evaluation of lines for Al-tolerant in acid soils revealed that number of tillers and filled grain per panicle were inherited with high heritability and directly affected grain weight and contributed indirectly to the correlation between grain weight and other characters. However, indirect selection through number of tillers and filled grain per panicle was less efficient than direct selection through grain weight.

Abstract

Teknik iradiasi sinar gamma dapat meningkatkan frekuensi dan memperluas spektrum mutasi makro namun belum banyak dikaji pengaruhnya terhadap mutasi mikro pada karakter kuantitatif dalam populasi termutasi. Penelitian ini bertujuan untuk mempelajari pengaruh perlakuan teknik iradiasi terhadap keragaman genetik dan heritabilitas karakter agronomi gandum pada generasi M2. Penelitian ini dilaksanakan dari bulan Juli sampai November 2014 di kebun percobaan Cibadak, Balai Besar Penelitian dan Pengembangan Bioteknologi dan Sumber Daya Genetik Pertanian, Kementerian Pertanian. Benih tiga galur gandum (F-44, K-95 dan WL-711) diberikan perlakuan tiga teknik iradiasi sinar gamma (akut, terbagi dan berulang). Generasi M1 ditanam secara bulk di lahan dan dipanen secara individu. Sebanyak 75 malai tiap kombinasi perlakuan ditanam satu baris per malai sebagai generasi M2 dan diamati karakter agronominya. Perlakuan teknik iradiasi sinar gamma menginduksi perluasan nilai kisaran semua karakter agronomi populasi M2. Teknik iradiasi terbagi mampu menginduksi nilai rataan yang lebih tinggi dengan kisaran yang lebih luas pada karakter panjang malai dan jumlah spikelet per malai dibanding teknik iradiasi yang lain. Teknik iradiasi terbagi dan berulang menghasilkan ragam yang lebih besar dibanding iradiasi akut pada karakter hasil biji per tanaman. Karakter agronomi jumlah anakan produktif, bobot malai, bobot biji per malai dan bobot biji per tanaman pada populasi M2 hasil induksi tiga teknik iradiasi memiliki heritabilitas yang tinggi. Tiga teknik iradiasi menginduksi perluasan keragaman genetik semua karakter agronomi populasi M2 yang diamati kecuali tinggi tanaman.
