Abstract

Kacang Tunggak (Vigna unguiculata L. Walp) merupakan tanaman Leguminosae yang bijinya dapat menjadi subsitusi bahan baku kedelai. Daunnya juga dapat dimanfaatkan sebagai sayuran. Penelitian ini bertujuan mempelajari pengaruh waktu pemetikan daun dan pemupukan NPK terhadap produksi biji kacang tunggak. Percobaan ini dilaksanakan di Kebun Percobaan Cikabayan IPB pada bulan Februari hingga Juni 2019 menggunakan kacang tunggak bertipe merambat. Rancangan percobaan yang digunakan adalah Rancangan Kelompok Lengkap Teracak (RKLT) dengan petak terpisah. Petak utama pada percobaan adalah faktor pemupukan NPK dengan dosis 100 kg ha-1 diaplikasikan pada awal tanam, 50 kg ha- 1 diaplikasikan pada awal tanam dan 50 kg ha-1 pada tanaman umur 4 MST, serta perlakuan tanpa pemupukan. Anak petak pada percobaan adalah waktu pemetikan daun dengan perlakuan pemetikan daun pada umur 5 MST, dua kali pemetikan daun pada umur 5 dan 7 MST, tiga kali pemetikan daun pada umur 5, 7, dan 9 MST, serta tanpa perlakuan pemetikan daun. Data dianalisis menggunakan Uji F dan diuji lanjut menggunakan Duncan Multiple Range Test (DMRT) pada taraf nyata α=5%. Pemupukan NPK 50-50 kg ha-1 menghasilkan hasil polong kering tertinggi sebesar 36.72 g per tanaman. Faktor waktu pemetikan daun tidak berpengaruh nyata terhadap semua peubah, kecuali pada jumlah tandan bunga tanaman umur 7 MST. Terdapat interaksi antara perlakuan pemupukan NPK dan waktu pemetikan daun terhadap total bobot polong kering dan indeks panen tanaman. Pemetikan daun dua kali pada 5 dan 7 MST dapat dilakukan tanpa mengurangi hasil polong dengan syarat adanya pemupukan NPK 50 kg ha-1 pada awal tanam dan NPK 50 kg ha-1 pada 4 MST dengan hasil sebesar 41.92 g bobot polong kering per tanaman. Abstract

Sorgum dapat dijadikan sebagai bahan pangan alternatif beras padi, namun beras dari biji sorgum kurang disukai masyarakat. Karena itu diperlukan adanya perbaikan terhadap kualitas biji sorgum untuk memperoleh biji sorgum yang disukai oleh masyarakat. Penelitian ini bertujuan memperoleh pengetahuan tentang keragaman genetik dan seleksi pada karakter agronomi dan tipe waxy pada populasi F3 sorgum persilangan Pulut 3 x PI-150-20A. Penelitian dilaksanakan pada bulan Juli 2022 hingga November 2022 di Kebun BSIP Biogen, Cimanggu, Kota Bogor. Hasil penelitian menunjukkan bahwa populasi F3 memiliki nilai tengah yang cenderung lebih kecil daripada kedua tetuanya. Aksi gen yang muncul yaitu epistasis komplementer dan epistasis duplikat dengan jumlah gen yang terlibat banyak pada mayoritas karakter. Heritabilitas arti luas dan koefisien keragaman genetik termasuk tinggi. Pengamatan tipe sorgum berdasarkan kandungan amilosa. Hasil dari pewarnaan biji menunjukkan bahwa populasi F3 sorgum persilangan Pulut 3 x PI-150-20A mayoritas bertipe waxy atau memiliki kandungan amilosa yang rendah. Abstract

Acidic soils with high aluminum (Al) toxicity are a major constraint to upland rice productivity in many tropical regions, including Kalimantan. Local rice cultivars often possess adaptive traits that can be utilized for breeding tolerant varieties. This study aimed to investigate the genetic diversity of Kalimantan local upland rice cultivars using SSR markers and their adaptation to aluminum toxicity stress. A total of 93 rice accessions obtained from the Agricultural Gene Bank of IAARD were subjected to both genetic diversity and Al stress adaptation analyses. The genetic diversity analysis used 11 SSR markers, while the adaptation study employed a hydroponic method with 45 ppm Al in Yoshida nutrient solution. Root and shoot growth were observed after 21 days of stress treatment. The SSR markers were highly polymorphic, grouping the accessions into four clusters at a 35% similarity level. Most accessions showed tolerance to Al stress based on relative root and shoot growth parameters. Accessions P. Pulut Timai, Padai Cina, Raya, Parai Paliping, and P. Pulut Ayang/Pang were the most adaptive. Root length, shoot length, and shoot fresh weight exhibited moderate heritability. These findings provide valuable insights for breeding Al-tolerant upland rice adapted to acidic soils. Keywords: adaptability; aluminum stress; molecular marker; SSR; upland rice

Abstract

Indonesian sorghum (Sorghum bicolor L.) germplasm with the highest genetic diversity can serve as a valuable resource for developing high-yielding cultivars. Therefore, the present research aimed to obtain information on genetic variability in the population and also determine the key traits associated with grain yield using correlation and path analysis in sorghum F2 population. The sorghum parental genotypes 'Demak 4' and 'Soraya 3', and a local high yielding IPB line, NS 19, NS 109, and NS 111, were used in the crosses. The F2 population was evaluated against parental cultivars and four check genotypes. The results showed that 64 individual plants were observed as transgressive segregants. Meanwhile, the number of leaves and plant height were controlled polygenically through additive genes. The traits of panicle diameter, dry panicle weight, and grain weight per panicle possessed the highest genotypic coefficient of variance. The traits dry panicle weight and diameter were significantly positively correlated with grain yield. The considerable positive correlation with dried panicle weight, panicle length, and diameter could be used as selection criteria. The path analysis reported that the observed agronomic traits explained 97.07% of the total variation in grain weight per panicle.

Radicle emergence as a rapid indicator of sorghum seed vigor to predict mean germination time, vigor index, and field emergence. This study aimed to develop a standardized radicle emergence test for sorghum (Sorghum bicolor) seeds by determining the optimal single-count timing and temperature for the simultaneous assessment of seed vigor and viability. This test was conducted by calculating the percentage of seeds whose radicles had emerged by at least 2 mm in length during a single observation period at the beginning of germination. Seed lots with low radicle emergence rates showed slow germination, reflecting an early physiological indicator of seed deterioration and a key factor contributing to reduced vigor. The experiment was arranged in a randomized complete block design with one factor: 12 sorghum genotypes, and four replications (blocks). Radicle length was measured at 2-h intervals from 40 to 48 h, with a minimum length of 2 mm. Results of radicle emergence (production of 2 mm radicle) at 25°C and alternating temperatures 20⇔30°C for 44 h (the optimal time), mean germination time, germination percentage, germination speed, vigor index, maximum growth potential, tetrazolium test, and field emergence were compared using 12 genotypes of sorghum seeds with standard germination above 83%. The single counts of radicle emergence performed at 25°C and 20⇔30°C were highly correlated with key vigor and viability indicators. Correlations with vigor-related traits, such as mean germination time (R²=0.87-0.81), vigor index (R²=0.83-0.89), germination speed (R²=0.73-0.71), and field emergence (R²=0.77-0.70), were strong, as were those with viability parameters, including standard germination (R²=0.66-0.59) and tetrazolium test results (R²=0.58-0.52). This study concludes that a single count of radicle emergence at 44 h, conducted at either 25°C or 20⇔30°C, is a method for simultaneous assessment of seed vigor (mean germination time, vigor index, germination speed, and field emergence) and viability in sorghum, offering a significant improvement over more time-consuming tests, such as germination test that requires 10 days.
