Abstract

Dampak suhu tinggi pada pertumbuhan dan produktivitas padi dapat dikurangi dengan menanam varietas toleran. Tujuan penelitian ini adalah memperoleh informasi tentang kendali genetik karakter hasil populasi bersegregasi padi pada dua tingkat stress suhu tinggi serta mengidentifikasi genotipe toleran. Penelitian dilakukan dari Agustus 2016 hingga Februari 2017. Saat fase vegetatif maksimum tetua dan populasi F3 diperlakukan di bawah dua kondisi suhu, yaitu di lantai jemur dan rumah kaca dengan suhu maksimum dan minimum rata-rata masing-masing 38.89/22,67 °C dan 41.62/23.32 °C. Karakter yang diamati adalah hasil dan komponen hasil. Hasil penelitian menunjukkan bahwa kondisi suhu tinggi mempengaruhi pertumbuhan dan hasil pada tetua dan populasi F3. Terdapat keragaman karakter agronomi padi pada kondisi cekaman suhu tinggi. Jumlah gabah isi per malai adalah satu-satunya karakter yang dikendalikan oleh aksi gen aditif dengan nilai heritabilitas tergolong tinggi pada cekaman suhu di lantai jemur dan dalam rumah kaca sehingga karakter ini dapat digunakan untuk mengidentifikasi individu toleran pada generasi awal. Terdapat 13 individu yang toleran terhadap cekaman suhu tinggi berdasarkan nilai SSI. Karakter bobot gabah isi dikendalikan oleh gen-gen non aditif sehingga dianjurkan seleksi untuk peningkatan bobot gabah isi dilakukan pada generasi lanjut. Metode SSD direkomendasikan sebagai metode untuk perbaikan toleransi terhadap suhu tinggi dikombinasikan dengan metode shuttle breeeding. Kata kunci: fertilitas, indeks, perubahan iklim, toleran

Abstract

Thefocus of wheat research in Indonesia is to obtained newpotential wheat lines that are adapted to low-mid elevationand heat tolerant. This study was aimed to obtaininformation on the effect of interaction of season x line xlocation and genetic variability of wheat lines in tropicalagroecosystem. This study was conducted at the Seameo-Biotrop (<400 masl) and Indonesian Ornamental CropsResearch Institute-Cipanas (>1,000 masl) experimental fieldfor two seasons. The results showed that there was an effectof interaction of season x line x location on plant height,days to flowering, number of spikelet and floret, seed/headweight, rate of grain filling, yield, flag leaf width, stomatadensity, chlorophyll b, and leaf greenness. Meanwhileseveral characters were only influenced by the interaction ofline x location, they were yield component characters, ie.empty floret percentage, number of seed/ head, 1,000 seedweight, number of head/m2 and seed/plant weight. Sevencharacters were not influenced by interaction of neitherseason x line x environment nor line x location, they werenumber of productive tillers, head length, number ofseed/head, chlorophyll a, ratio of chlorophyll a/b, totalchlorophyll, and leaf thickness. The characters that havehigh heritability and wide genetic variability for the twoanalysis models were the number of spikelet.

Abstract

As a tropical country, Indonesia certainly is not the most suitable place for producing wheat. Wheat flour consumption however, had increased tremendously during the last three decades, that import of wheat grain in 2012 reached 7.2 million ton. Producing wheat had been attempted since 1990’s, utilizing lands at high elevation with drier climate. However, crops competition, especially with the high land vegetables, had put wheat cropping become prohibitive. As early as in 1880, G Wallace experimented of growing wheat in the high land of Timor island. However the crop was never economically established. Selection for genotypes and varieties of wheat for the tropical area had been attempted since 1980’s, and adaptable varieties to the high land had been released, but farmers did not adopt the crop on their cropping system. Breeding research to develop tropical wheat varieties was revived in 2009. By using the modern breeding techniques, including biotechnology, cellular somatic mutation, as well as applying the conventional breeding techniques, varieties adaptable to the lowland tropics are expected to be identified.

Abstract

Morpho-physiological characterization of sixteen inbred lines of maize originated from CIMMYT and from Indonesian Cereals Research Institute Maros, was carried out in Tenjo, Bogor, from January to April 2013. The objectives of this research were to (1) estimate the genetic parameters of morphophysiological characters, (2) obtain morpho-physiological characters that can be used as selection characters and (3) obtain tolerance inbred lines on acid soil condition. The experiment was conducted using a Nested Design with three replications, nested in selection environment. The first factor was environments, the second factor was genotypes. The two environments were acid soil with eAl 0.2 me/100 g (represented the optimum soil) and acid soil with eAl 1.87 me/100 g (represented the medium acid soil). The results showed that environment had significant effect on inbred lines performances. Characters of anthesis silk interval, number of seeds/cob, number of rows/cob, cob dry weight, seed dry weight and cob weight/plot could be used as selection characters for tolerance to medium acid soil condition; the inbred lines tested indicated high genetic variabilities and showed high heritability under such condition. Inbred line CLA 84, CLA 46, CLA 95 and 1042-13 each was found tolerant genotype on medium acid soil condition

Abstract

The study was aimed to obtain the information on the genetic diversity on agronomic traits of wheat among the F3 and F4 generations, derived from crosses of Oasis x HP1744, planted at the highland and medium elevation. The second objective was to acquire the character for selection and to acquire the adaptable genotypes on the highland and on the plain medium elevation. Selection was carried out using Shuttle Breeding, where F3 generation was selected at the Experimental Station of Cipanas (1100 m asl) and F4 generation was selected at the plain medium elevation of Cisarua (600 m asl). The experiment used Augmented Designs. Genetic material consisted of 57 F3 pedigree families selected from the F2 generation of Oasis x HP1744 cross plus six check varieties. The F4 generation cinsisted 320 selected genotypes from the F3 generation plus six check varieties. Plant height, flag leaf area, percentage of empty florets per panicle, number of grains/panicle and grain weight/panicle of the F3 generation showed larger mid value than the two parents. In the F4 generation, flag leaf greenness, total number of tillers, number of productive tillers, main panicle seed weight, number of seeds/panicle, seed weight/panicle, number of seeds/plant and seed weight/plant indicated the mean value larger than that of the Oasis. Plant height, number of productive tillers, days to flowering, maturity, spikelet number, and the total number of florets of the F4 generation showed higher heritability than that of F3 generation. Character suitable for indirect selection in the F3 was plant height, which indicated the best indirect differential selection. Potential families for further selection included: O/HP 21, O/HP 82, O/HP 12, O/HP 100 and O/HP 28. Characters most suitable for indirect selection in the F4 generation were: main panicle seed weight which indicated the best indirect differential selection. Genotypes potential for further selection in the medium elevation were O/HP82-19; O/HP82-15, O/HP78-5, O/HP49-30 and O/HP78-2.

Abstract

The study was aimed to determine the productivity and yield stability of ratoon rice genotypes in three environments and to obtain rice genotypes suistable for ratoonning on specific environment of tidal land. The experiment was designed in a randomized complete block with three replicates. Seedling was planted at 21 days old with spacing of 20 x 20 cm, one seedling per hill. Plants were harvested 30 days after heading by cutting at a height of 10 cm from the soil surface. One day after harvest, the land was irrigated as high as 3 cm and fertilized with Urea 50 kg/ha, TSP 30 kg/ha, and KCl 25 kg/ha. Yield stability was analyzed according to Eberhart and Russel (1966). Ratoons were sensitive to the environmental changes. Genotype IPB97-F-13-1-1, IPB 4S, and IPB 3S produced the main crop + ratoon grain yield of 5.26 t/ha, 5.14 t/ha, and 5.64 t/ha dry milling grain (DMG), respectively. Based on yield of the main crop + ratoon, IPB97-F-13-1-1 and IPB 4S was each considered as adaptable to the suboptimum condition (bi<1). Ratoon crop yield of genotype IPB97-F-13-1-1, IPB 4S, and IPB 3S each was the highest in three locations. The ratoon crop contribution to the rice production was from 31.3% to 61.9% to the main crop.

Abstract

Fruit size is a quantitative character controlled by several genes and mutations in those genes could lead to phenotypic changes in fruits. Single nucleotide polymorphism (SNP) is a molecular marker that could be used to detect mutation in genes at the nucleotide level that could not be done by conventional markers. The aim of this study was to analyze nucleotide variations, primarily SNP, in mutant chili pepper genotypes using gene-specific primers related to fruit size characters to measure the genetic changes occured, identify the SNPs that were associated with fruit size character, and analyze the phylogenetic among the mutant genotypes. Sanger sequencing was conducted on six mutant[K21]  chili pepper genotypes from M2 generation and Lingga as origin variety using five pairs of newly designed gene-specific primers corresponding to fruit size characters. A total of 41 base changes were identified in the gamma ray-irradiated chili pepper mutants, consisting of 22 insertions/deletions (indels) and 19 SNPs. As many as 17 nonsynonymous SNPs which caused amino acid changes, in contrast to the other 2 synonymous SNPs, were found in these mutant genotypes. Association analysis between identified SNPs to fruit length and diameter showed nonsignificant association, therefore the functional marker related to targeted gene could not be developed. The genotype L269 showed a separation from Lingga in dendrogram reflecting a large number of genetic changes, showed its potential as a new germplasm in development of improved variety in the future.

Abstract

Heat stress is a major environmental factor limiting wheat productivity in tropical regions such as Indonesia. The objective of this  study was to investigate the adaptability of introduced wheat genotypes in tropical agro-ecosystems. Sixteen spring wheat genotypes were grown at two different altitudes i.e. low altitude (176 m asl) with an average temperature of 29.8 oC located at Leuwikopo Field Experimental Station, Bogor Agricultural University, Darmaga Bogor and high altitude  (1100 m asl) with an average temperature  of 20.6 oC at Cipanas Field Experimental Station (Ornamental Crop Research Station), Cianjur, West Java, Indonesia from July to November 2012. Plant height, number of tillers, flag leaf area, leaf angle, days to flowering, spike number per plant, empty spikelet number, grain weight per plant and 100 grain weight were observed following the standard methods. Heat susceptibility index was calculated based on grain weight per plant. The results showed that cultivation at a low altitude, hotter environment remarkably affected wheat growth and yield, as reflected in overall reduction of plant height, reduced number of tillers and leaf area, and ultimately reduced yield and yield components for most genotypes compared to the same measures taken at high altitude in lower temperatures. Plant growth  before  heading was similar in both locations, but the days to flowering  was longer in high altitude  than that in low altitude. High temperature stress in low altitude reduced the spike number/plant, grain weight/plant, 100 grain weight and increase number of empty spikelet/spike. Based on our results for heat susceptibility index, six genotypes, namely Sbr, Ymh, Astreb/Cbrd, Astreb/Ningma, H-20 and Nias, were characterized as heat tolerant genotypes.

Abstract

Plant breeding program consists of establishment of the population, selection, and evaluation. The study aimed to observe the variability of yield components, the heritability, and the distribution of the yield component characters in the F3 populations. The experiment was conducted at Muara Experimental Farm Bogor, from April to August 2012 on Latosol soil. The F3 populations derived from crosses of Bintang Ladang x US2, Gampai x IR77674, and Progol x Asahan and their parental were used as plant materials. Twenty one-day-old seedlings from each population were planted in plots of 2 x 12 m, with planting space of 20 x 20 cm and 3-5 seedlings per hole. Panicle length and weight were observed on 300 randomly selected plant samples from each population. The results showed that there was a variation of agronomic characters among genotypes. The heritability of characters, the panicle length and weight was low to high. Panicle length and weight were controlled by many genes with additive gene action in the Gampai x IR77674 derived population, while panicle weight was controlled by few genes with complementary epistatic additive gene action in both Bintang Ladang x US2 and Progol x Asahan derived populations. The more genes controlling a character, the more distribution classes formed and the greater variance among genotypes.

Abstract

Lemongrass (Cymbopogon nardus) as cover crop was suitable planted with agroforestry and monoculture system on post-coal mining revegetation land. The study investigated the influence of planting system, varieties, and plant spacing against the lemongrass growth on post-coal mining land under the agroforestry and monoculture system. Two lemongrass varieties were planted under on both planting systems. The growth variables of lemongrass measured were the tillers number per clump, leaf length, and canopy width. The results showed that the planting system and lemongrass varieties were able to increase the growth of lemongrass. Plant spacing treatment was able to increase the tillers number per clump. The agroforestry system was able to increase leaf length and canopy width of lemongrass. However, monoculture system was able to increase the tillers number per clump. The G2 variety had better growth on all measured variables either in agroforestry and monoculture system compared to Sitrona 2 Agribun variety. The plant spacing of 0.5 m × 0.5 m had greater tillers number per clump of lemongrass compared to plant spacing of 1 m × 1 m.
